Effects of dietary Antrodia cinnamomea fermented product supplementation on antioxidation, anti-inflammation, and lipid metabolism in broiler chickens

  • Objective: This study was investigated the effects of dietary supplementation of Antrodia cinnamomea fermented product on modulation of antioxidation, anti-inflammation, and lipid metabolism in broilers. Methods: Functional compounds and in vitro antioxidant capacity were detected in wheat bran (WB) solid-state fermented by Antrodia cinnamomea for 16 days (FAC). In animal experiment, 400 d-old broiler chickens were allotted into 5 groups fed control diet, and control diet replaced with 5% WB, 10% WB, 5% FAC, and 10% FAC respectively. Growth performance, intestinal microflora, serum antioxidant enzymes and fatty acid profiles in pectoral superficial muscle were measured. Results: Pretreatment with hot water extracted fermented product significantly reduced chicken peripheral blood mononuclear cells death induced by lipopolysaccharide and 2,2'-Azobis(2-amidinopropane) dihydrochloride. Birds received 5% and 10% FAC had higher weight gain than WB groups. Cecal coliform and lactic acid bacteria were diminished and increased respectively while diet replaced with FAC. For FAC supplemented groups, superoxide dismutase (SOD) activity increased at 35 days only, with catalase elevated at 21 and 35 day. Regarding serum lipid parameters, 10% FAC replacement significantly reduced triglyceride and low-density lipoprotein level in chickens. For fatty acid composition in pectoral superficial muscle of 35-d-old chickens, 5% and 10% FAC inclusion had birds with significantly lower saturated fatty acids as compared with 10% WB group. Birds on the 5% FAC diet had a higher degree of unsaturation, followed by 10% FAC, control, 5% WB, and 10% WB. Conclusion: In conclusion, desirable intestinal microflora in chickens obtaining FAC may be attributed to the functional metabolites detected in final fermented product. Moreover, antioxidant effects observed in FAC were plausibly exerted in terms of improved antioxidant enzymes activities, increased unsaturated degree of fatty acids in chicken muscle and better weight gain in FAC inclusion groups, indicating that FAC possesses promising favorable mechanisms worthy to be developed.

Somatic Embryogenesis and Plant Regeneration from Embryogenic cell Suspension Cultures of Schisandra chinensis Baill

  • An efficient somatic embryogenesis and plant regeneration protocol was developed for Schisandra chinensis Baill, using embryogenic cell suspensions and optimized media conditions. Friable embryogenic callus was induced from cotyledonary leaf and hypocotyl explants of 7 days old seedlings on MS agar medium supplemented with 1.0 to $4.0\;mg\;l^{-1}$ of 2,4-dichlorophenoxyacetic acid (2,4-D). Fast growing and well dispersed embryogenic cell suspensions were developed within two months when embryogenic calli were transferred to MS liquid medium containing $1.0\;mg\;l^{-1}\;2,4-D$. One third strength of MS medium was the best for both overall growth and development of somatic embryos in liquid culture. Over 3400 viable somatic embryos were produced from each 150 ml flask with an initial cell density of 30 mg in 30 ml medium. Germinated somatic embryos developed in liquid medium converted into plantlets after transferred to half-strength MS semi-solid medium. Approximately 90% of the converted plantlets were successfully transplanted to soil and grew into fertile plants.

Utilization of Fungal Treated Wheat Straw in the Diet of Late Lactating Cow

  • Eight primiparous Holstein cows, in late lactation (255$\pm$10 days in milk) and yielding 10.3$\pm$1.3 kg/d of 4% fat corrected milk (FCM) were allocated into two groups randomly. Two diets containing 30% wheat straw either untreated (UWS) or treated with Pleurotus ostreatus (FTWS) were offered as total mixed ration (TMR). In vivo digestibility of the diets was determined, using acid insoluble ash as a marker. Daily milk production was recorded and milk samples were collected and analysed. Diet FTWS resulted in significantly (p<0.05) higher dry matter intake (DMI) (12.2$\pm$0.86 vs. 10.6$\pm$1.3), DM digestibility (58.8 vs. 52.3) and milk yield (9 vs. 7.5 kg). Milk fat contents were 34.2 and 35.6 g/liter that did not differ between cows fed treated or untreated straw. However, the concentrations of lactose, solid non fat, total solids and milk protein for diets UWS and FTWS were 57.3 and 54.9, 98.9 and 93.2, 134.5 and 127.4, 35.7 and 32.3 g/l, respectively, which differed significantly (p<0.05). The average body weights gain (BWG) for UWS and FTWS were 272 and 743 g/d, respectively (p<0.05). The FCM yield per kg of DMI was similar (0.68 and 0.67 liter) for the two groups, but BWG/kgDMI was higher in the FTWS diet.

Altered Protein Expression in Peach (Prunus persica) Following Fruit Bagging

  • Fruit bagging has been widely practiced in peach cultivation to produce high quality and unblemished fruit. Moreover, fruit bagging has been utilized to study the effect of shading on the quality of fruit. We conducted a proteomic analysis on peach fruit to elucidate the biochemical and physiological events that characterize the effect of bagging treatment. Comparative analysis of 2D electrophoresis (2-DE) gels showed that relative protein levels differed significantly at 125 DAFB (days after full bloom), as well as at 133 DAFB in fruit that had been bagged until 125 DAFB, followed by exposure to sunlight. Most of the proteins with altered expression were identified by MALDI TOF/TOF. Twenty-one proteins with differential expression among the groups were identified at 125 DAFB, while thirty proteins with differential expression among the groups were identified at 133 DAFB. The analysis revealed that expression of proteins involved in photosynthesis, stress responses, and biochemical processes influencing metabolism were altered during bagging treatment, suggesting that regulation of the synthesis of carbohydrates, amino acids, and proteins influenced fruit size, solid/acid ratio, and peel color. This work provides the first characterization of proteomic changes in peach in response to fruit bagging treatment. Identifying and tracking protein changes may allow us to better understand the mechanisms underlying the effects of bagging treatment.

Feasibility Test of Biohydrogen Production from Food Waste (음식물쓰레기의 수소발효 타당성 평가)

  • Although extensive studies were conduced on hydrogen fermentation of organic wastewaters, little is known about biohydrogen production from organic solid wastes. The leaching-bed reactor treating food waste by heat-shocked anaerobic sludge was, therefore, operated at D of 2.1, 3.6, 4.5 and $5.5d^{-1}$ to find optimal D for hydrogen production. Successful operation of a reactor can be accomplished when it is operated at proper dilution rate (D). Operation at high D leads to the washout of biomass in the reactor while operation at low D leads to product inhibition due to the accumulation of excess VFA. These appear to limit the production of hydrogen to reach a higher level. All the reactors showed that, on day 1-3, hydrogen production was dominant and VFA concentration was higher than ethanol. Butyrate and acetate were major components of VFAs over the whole operation, though lactate was very high on day 1-2. Compared with other D values, D of $4.5d^{-1}$, resulted in higher butyrate/acetae (B/A) ratios during the fermentation. The trend of B/A ratios was similar to the hydrogen production, suggesting that butyrate formation favored hydrogen production. Ethanol increased significantly from day 4 when hydrogen Production stopped. It indicated that heat-shocked sludge was able to induce a metabolic flow from hydrogen-and acid-producing pathway to solvent-producing pathway. Operation at D of $4.5d^{-1}$ led to higher fermentation efficiency (58%) than those (51.5, 55.3 and 53.7%) at 2.1, 3.6 and $5.5d^{-1}$. The COD removed was convened to hydrogen (10.1%), VFA (30.9%), and ethanol (17.0%).

Evaluation of Antimicrobial, Antioxidant, and Antithrombin Activities of the Rhizome of Various Dioscorea Species (마 품종별 메탄올 추출물의 항균, 항산화 및 항혈전 활성 평가)

  • Yams (Dioscorea spp.) have been used as medicinal and edible resources worldwide. In the present study, methanol extracts were prepared from various kinds of yam, including D. batatas (Gyeongbuk No. 1 and No. 4), D. alata L. (Gyeongbuk No. 5 and No. 6), and D. bulbifera and D. nipponica, and the antioxidant, antithrombin, and antimicrobial activities of these extracts were evaluated. The water content of the various yams was 54.7~83.2% (D. batatas> D. alata L.>D. nipponica>D. bulbifera) and the methanol extracts contained 0.59-1.59% (w/v) solid matter (D. alata L.>D. batatas >D. nipponica>D. bulbifera). The mean concentrations of total polyphenols, total flavonoids, and total sugars in the different types of yam were 49.82 mg/g, 9.79 mg/g, and 173.81 mg/g, respectively. Analysis of DPPH radical-scavenging activity showed that D. alata L. Gyeongbuk No. 6 had a strong antioxidant capacity (IC50=$142.30{\mu}g/mL$), whereas the other yams showed reduced levels of activity (IC50=371-$486{\mu}g/mL$). D. alata L. Gyeongbuk No. 6 showed a strong antithrombin activity against human thrombin. Upon treatment with a D. alata methanol extract (1.5 mg/mL), thrombin time was increased 7.35-fold compared with that when methanol alone was used. In an antimicrobial activity assay, the D. nipponica extract showed moderate antibacterial activity against Bacillus subtilis, Staphylococcus aureus, Proteus vulgaris, and Salmonella typhimurium. Our results indicate that different varieties of Dioscorea spp., including D. batatas, have useful biochemical attributes, including antioxidant, antithrombin, and antibacterial activities.

A Comparative Analysis between 3D Geological Modeling and Magnetic Data of Fe-Mn Ore in Ugii Nuur, Mongolia (몽골 우기누르 철-망간 부존 지역의 3차원 지질모델과 자력탐사 결과의 비교분석)

  • This study constructed a 3D geological model for Uggi Nuur Fe-Mn mineralization zone in Mongolia, and the 3D geological distribution is cross-analyzed with magnetic anomaly distribution to figure out relationship between ore zone and subsurface geology. As a result of 4 step 3D modeling procedures including geological cross section, surface modeling, foliation modeling and solid modeling, the geology of the both study area is bordered by faults in NW direction with Munguntessj formation being located in the west side of the fault while Yashill formation is located on the other side of the fault. Moreover, the strike direction of foliation in the both formation shows same directional pattern with the NW faults. The magnetic anomaly distribution reveals that higher anomaly values are concentrated to near the ground surface. The analyses of 3 dimensional distribution between subsurface geology and magnetic anomaly indicates that higher anomaly is mainly distributed over the Munguntessj formation as a elongated lens bodies whereas the magnetic anomaly is evenly found in the both of Munguntessj formation and Yashill formation in the study area 2. It infers that volcanic activities associated mineralization occurred during silurian period, and the mineralized zone is thought to be realigned along the geological structures caused by later stage tectonic activities.

Photoluminescence and Concentration Quenching Properties of BaMoO4:Tb3+ Phosphors (BaMoO4:Tb3+ 형광체의 발광과 농도 소광 특성)

  • $BaMoO_4:Tb^{3+}$ phosphor powders were synthesized with different concentrations of $Tb^{3+}$ ions using the solid-state reaction method. XRD patterns showed that all the phosphors, irrespective of the concentration of $Tb^{3+}$ ions, had tetragonal systems with two main (112) and (004) diffraction peaks. The excitation spectra of the $Tb^{3+}$-doped $BaMoO_4$ phosphors consisted of an intense broad band centered at 290 nm in the range of 230-330 nm and two weak bands. The former broad band corresponded to the $4f^8{\rightarrow}4f^75d^1$ transition of $Tb^{3+}$ ions; the latter two weak bands were ascribed to the $^7F_2{\rightarrow}^5D_3$ (471 nm) and $^7F_6{\rightarrow}^5D_4$ (492 nm) transitions of $Tb^{3+}$. The main emission band, when excited at 290 nm, showed a strong green band at 550 nm arising from the $^5D_4{\rightarrow}^7F_5$ transition of $Tb^{3+}$ ions. As the concentration of $Tb^{3+}$ increased from 1 to 10 mol%, the intensities of all the emission lines gradually increased, approached maxima at 10 mol% of $Tb^{3+}$ ions, and then showed a decreasing tendency with further increase in the $Tb^{3+}$ ions due to the concentration quenching effect. The critical distance between neighboring $Tb^{3+}$ ions for concentration quenching was calculated and found to be $12.3{\AA}$, which indicates that dipole-dipole interaction was the main mechanism for the concentration quenching of the $^5D_4{\rightarrow}^7F_5$ transition of $Tb^{3+}$ in the $BaMoO_4:Tb^{3+}$ phosphors.

Anaerobic digestion for food wastewater using HADS Pilot Plant and analysis of microbial community in the digester (HADS Pilot Plant를 이용한 음폐수의 혐기성 소화 및 미생물 군집 변화 분석)

  • We(Hansol EME Co. Ltd.) proceeded anaerobic digestion test for domestic food wastewater applying to two operating method for increasing of OLR. The methods are as follows. One was the rapidity operating method which was increasing the OLR continuously and rapidly and the other was the terraced operating method which was increasing the OLR having adaptation period for each step. As a result of this tests, the ratio of VFA/Alkalinity of the process was very unstable under the rapidity operating method then the volume of produced biogas was dramatically decreased. However the process was shown stable performance under the terraced operating method maintaining the ratio of VFA/Alkalinity less than 0.4. Also, the process was performing the biogas recovery of $0.8Nm^3/kgVS_{rem}/d$ and the VS removal ratio of 85%. T-RFLP analysis about the community of bacteria and methanogen is also conducted to check the change of the microbial community according to the methods of OLR increasing operation. The microbial community was changed by the methods of OLR increasing operation according to the result of T-RFLP analysis. Although the anaerobic digestion test was executed by same pilot plant, the reactivity and the tolerance of microbial community for surrounding environment could be considerably changed by the operating method for the process.

Topology Optimization of Offshore Wind-Power Turbine Substructure Using 3D Solid-Element Model (3 차원 고체요소모델을 활용한 해상풍력터빈 하부구조의 위상최적화)

  • The structural layout of mechanical and civil structures is commonly obtained using conventional methods. For example, the shape of structures such as electric transmission towers and offshore substructures can be generated systematically. However, with rapid advancements in computer graphic technology, advanced structural analyses and optimum design technologies have been implemented. In this study, the structural shape of a jacket substructure for an offshore wind turbine is investigated using a topology optimization technique. The structure is subjected to multiple loads that are intended to simulate the loading conditions during actual operation. The optimization objective function is defined as one that ensures compliance of the structure under the given boundary conditions. Optimization is carried out with constraints on the natural frequency in addition to the volume constraint. The result of a first step model provides quick insights into the optimum layout for the second step structure. Subsequently, a 3D model in the form of the frustum of a quadrilateral pyramid is developed through topology optimization.
