담배 인산수송자 유전자를 이용한 벼의 형질전환 (Transformation of Rice (Oryza sativa L.) with Phosphate Transporter cDNA from Tobacco (Nicotiana tabacum L.))

  • 저인산 조건하에서 인산 흡수 효율이 높은 벼 품종을 개발하고자 담배의 인산수송자 유전자를 벼에 형질전환하였다. 동진벼로부터 유도된 callus를 담배의 인산수송자 유전자가 도입된 A. tumefaciens LBA 4404와 공동배 양한 후, 선발배지에서 증식된 callus를 250 mg/L carbenicillin, 2 mg/L kinetin, 0.1 mg/L NAA가 첨가된 MS배지에 옮겨 약 2주 후부터 소식물체를 얻었다. Carbenicillin 250 mg/L 첨가된 MS 기본배지에서 소식물체의 발근을 유도하여 재분화 식물체를 얻었다. 선발된 callus는 약 52%의 식물체 재력화율을 보였다. 선발된 재분화 식물체에 대한 PCR 분석을 수행하여 형질전환 벼 식물체에 담배의 인산수송자 유전자가 삽입되었음을 확인하였다. 형질전환체의 genonlic DNA로부터 PCR에 의해 증폭된 DNA에 대한 Southern blot분석 결과, 대조 식물체에서는 band가 검출되지 않았으나 형질전환 식물체에서는 인산수송자 유전자와 동일한 1.7kb의 band가 검출되어 외래유전자인 담배 인산수송자 유전자가 안전적으로 도입되었음이 확인되었다.

  • 동화산물의 분배 효율 개선을 통한 생산성 향상 가능성을 조사하고자 감자의 sucrose 수송자 유전자를 벼에 형질전환 하였다. 동진벼로부터 유도된 callus를 감자의 sucrose 수송자 유전자가 도입된 A. tumefaciens LBA 4404와 공동배양한 후, 선발배지에서 증식된 callus를 250 mg/L carbenicillin, 2 mg/L kinetin, 0.1 mg/L NAA가 첨가된 MS 배지에 옮겨 약2주 후부터 소식물체를 얻었다. Carbenicillin 250 mg/L 첨가된 MS 기본 배지에서 소식물체의 발근을 유도하여 재분화 식물체를 얻었다. 선발된 callus는 약 150%의 높은 식물체 재분화율을 보였다. 재분화 식물체에 대한 PCR 분석을 수행하여 감자의 sucrose수송자 유전자가 삽입된 형질전환 벼 식물체를 선발하였다. 선발된 형질전환 식물체에 대한 Southern blot 분석을 통하여 외래유전자인 감질 sucrose 수송자 유전자가 벼 genome 내에 안정적으로 도입되었음을 확인하였다.

  • High temperature deformation behavior of AZ31 Mg alloy was investigated in this study on the basis of a processing map $(\varepsilon\approx0.6)$. To construct a processing map, compression tests were carried out at wide range of temperatures and strain rates $(T=250\~500^{\circ}C,\;\varepsilon=10^{-4}\~100/s)$. Two regions of high deformation efficiency $(\eta)$ were identified as: (1) a dynamic recrystalization (DRX) domain at $250^{\circ}C$ and 1/s and (2) a superplasticity domain at $450^{\circ}C$ and $10^{-4}/s$. Possible deformation mechanisms operating at high temperature were discussed in relation to the activation energy. A two-stage deformation method was found to be effective in enhancing the superplasticity of AZ31 Mg alloy. From the two-stage deformation method, tensile elongation of $1200\%$ was obtained at the superplastic domain.

  • In this study, the effects of initial texture on the twinning formation of AZ31 Mg rolled sheet was investigated. Uniaxial compression tests were carried out on samples cut along the normal direction(ND) and roiling direction(RD), respectively, of rolled AZ31 Mg alloy sheet at various temperatures (RT, 200, 250, 300, 350, $400^{\circ}C$) with the fixed strain rate($10^{-2}/s$). The results showed that deformation twining occurred actively only in the RD specimens, which promoted homogeneous deformation as compared to the ND specimens. The effect of temperature on the formation of deformation twins was also investigated, and the slip/twin transition temperature was found to be approximately $250^{\circ}C$.

  • Magnesium(Mg) alloys have been evaluated as replacements for steel or aluminum parts in the automobile industry because of the fuel economy they can provide through reducing weight. The application of Mg alloys has been limited due to its low formability at room temperature, which results from a small number of active slip systems. In the current study, an extruded TAZ711(Mg-7Sn-1Al-1Zn) alloy was warm forged into an automotive control arm to evaluate its formability at various forging temperature. Warm forging was conducted at temperatures of 200, 250, 350 and 450℃. Static strength evaluation was performed on the as-forged specimen at 250℃. The results showed good static strength.

금은화(金銀花)가 Cationic Bovine Serum Albumin 투여로 유발된 Membranous Nephropathy Mouse Model에 미치는 영향 (Effects of the Lonicerae Flos Extract on the Membranous Nephropathy Induced by Cationic Bovine Serum Albumin in Mice)

  • Membranous nephropathy(MN) is the most common cause of adult nephrotic syndrome worldwide. But treatment of MN is not defined. This study was to evaluate the effects of Lonicerae Flos Extract(LFE) on the MN induced by cBSA in mice. Mice were divided into 4 groups. The first group named for 'Normal' was injected with a saline solution. The second group named 'Control' treated with cBSA(10 mg/kg i.p) only. The third group named 'LFE-250', treated with cBSA(10 mg/kg i.p) and LFE(250 mg/kg, p.o). The fourth group named 'LFE-500'treated with cBSA(10 mg/kg i.p) and LFE(500 mg/kg, p.o). After cBSA and LFE treatment for 4 weeks, we measured change of body weight, 24hrs proteinuria, serum albumin, total cholesterol, triglyceride, BUN, creatinine, TNF-$\alpha$, IL-6, IL-$1{\beta}$, IL-10, IFN-$\gamma$, IgA, IgM and IgG levels. The morphologic changes of renal glomeruli were also observed with a light microscope. The levels of 24 hrs proteinuria, total cholesterol, IgG , IgM, IgA, IL-6 were significantly decreased in both LFE groups. The level of triglyceride, IL-$1{\beta}$ was significantly decreased in LFE-500 group. The level of Albumin was significantly increased in LFE-250 group. The level of TNF-$\alpha$, IFN-$\gamma$ were significantly decreased in LFE-250 group. The mRNA expression of IL-$1{\beta}$ in splenocytes was consideraly decreased in LFE-500 group. In histological findings of kidney tissue, thickening of GBM decreased in both LFE groups. This study shows that the LFE might be effective for treatment of MN. More clinical data and studies are to be done for efficient application.

마그네슘 합금에 대한 DLC 코팅의 온도에 따른 마찰기구 해석 (Friction Behavior of DLC Coating Slid Against AZ31 Magnesium Alloy at Various Temperatures)

  • Sheet-forming of Mg alloys is conducted at elevated temperatures (250℃) due to the low formability at room temperature. The high-temperature process often gives rise to surface damage on the alloy (i.e. galling.) In the current study, the frictional characteristics of DLC coating slid against an AZ31 Mg alloy at various temperatures were investigated. The coating has been used widely for low-friction processes. Dry-sliding friction and galling characteristics of an AZ31 Mg alloy (disk), which slid against uncoated and a DLC-coated STD-61 steel (pin), were investigated using a reciprocating-sliding tribometer at room temperature and 250℃. To represent the real sliding phenomena during a sheet metal forming process, single-stroke tests were used (10mm stroke length) rather than a reciprocating long sliding-distance test. The DLC coating suppressed adhesion between the alloy and the tool steel at room temperature, and exhibited a low friction coefficient. However, during sliding at 250℃, severe adhesion occurred between the two surfaces, which resulted in a high friction coefficient and galling.

팔미지황환의 Propylthiouracil로 유발된 Rat 갑상샘기능저하성 웅성 생식기 손상 제어 효과 (Favorable Control Effects of Palmijihwang-Whan on the Propylthiouracil Induced Hypothyroidism Related Rat Male Reproductive Organ Damages)

  • Objectives: This study was to evaluate the effect of Palmijihwang-whan (PMJHW) aqueous extract in the regulation of hypothyroidism related reproductive organ damages in propylthiouracil (PTU)-induced rat model. Methods: PMJHW aqueous extract (yield=17.90%) were administered, once a day for 42 days from 2 weeks before start of PTU treatment as oral doses of 500, 250, and 125 mg/kg (body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28 days. Results: PTU-induced hypothyroidism and related male reproductive organ damages-atrophic changes of testis, epididymis and prostate, were favorably and dose-dependently inhibited by treatment of PMJHW 500, 250, and 125 mg/kg. They also effectively regulated the PTU-induced abnormal antioxidant defense system changes in the testis. Although levothyroxine also favorably inhibited PTU-induced hypothyroidism, it deteriorated the hypothyroidism related male reproductive organ damages through testicular oxidative damages. The results suggest that oral administration of 125, 250, and 500 mg/kg of PMJHW has favorable effects on the hypothyroidism and related reproductive organ damages through augmentation of antioxidant defense system in the testis. Conclusions: This study suggest that PMJHW may be help to ameliorate the hypothyroidism and related organ damages in clinics.

도시 내 중금속 오염지의 관상식물로서 자생 맥문동(Liriope platyphylla)의 적용성 평가 (Application of Liriope platyphylla, Ornamental Korean Native Plants, for Contaminated Soils in Urban Areas)

  • 본 연구는 토양 내 중금속(카드뮴, 납, 아연) 처리 농도에 따른 자생 맥문동의 외형적인 생장 변화를 살펴봄으로써, 도시 내 중금속 오염지의 관상효과 증진을 위한 수종탐색 자료로 활용하고자 수행하였다. 중금속은 카드뮴, 납, 아연등 3종류이고, 처리 농도는 Control, 100, 250, $500mg{\cdot}kg^{-1}$ 등 4가지로서, 총 12가지 처리구로 구성하였다. 2009년 3월에 각각의 실험구에 10개씩 3반복으로 각 처리구에 정식하여, 같은 해 9월까지 약 7개월간 온실에서 실험을 실시하였다. 생장이 가장 활발한 시기라 볼 수 있는 5월부터 8월까지 약 3개월 동안 엽장, 엽폭, 총엽수, 고사엽수, 신엽수, 엽록소함량, 관상가치 등을 중심으로 생장 변화를 모니터링하였다. 카드뮴(Cd) 처리구의 경우, 엽장과 엽폭의 상대생장률이 식재 후 4개월이 경과된 시점에서 급속히 감소하였다. 총엽수, 신엽수, 엽록소함량, 관상가치 등의 형태적 항목에서도 Control> $Cd_{100}$ > $Cd_{250}$ > $Cd_{500}$ 순으로 감소하는 경향을 보였다. 납(Pb) 처리구에서 엽장의 상대생장률은 $Pb_{250}$$Pb_{500}$ 처리구에서만 변화율이 감소한 반면, 엽폭은 납 처리 농도가 높을수록 성장률의 감소양상이 뚜렷하였다. 총엽수, 신엽수, 고사엽수등은 대조구와 $Pb_{100}$ 처리구에서 비교적 유사하였으나, 엽록소함량, 관상가치 등은 납 처리 농도가 증가할수록 감소양상이 매우 뚜렷했다. 아연(Zn) 처리구는 카드뮴와 납 처리구와는 달리 식재 후 4개월이 경과된 시점에서도 변화율의 차이가 있을 뿐, 엽장은 꾸준히 증가하는 경향을 보였다. 엽폭 상대성장율 또한 $Zn_{500}$ 처리구를 제외하고, 비교적 완만한 성장세를 보였다. 총엽수, 신엽수, 고사엽수, 관상가치는 $Zn_{500}$ 처리구에서 가장 낮은 값을 나타낸 반면, Control, $Zn_{100}$, $Zn_{250}$ 처리구간에 차이가 뚜렷하지 않았다. 본 연구를 통해, 맥문동의 관상가치적 관점에서 생장 변화를 파악함으로써 향후 현장적용의 기반 연구로서 큰 의미를 갖는다고 하겠다. 하지만, 중금속에 대한 식물의 내성은 중금속 자체의 특이성과 환경조건에 따라 변화할 수 있다는 개연성을 가지므로, 실내연구와 현장적용성 연구가 병행되어야 할 것이다.

흰점박이꽃무지(Protaetia brevitarsis) 추출물의 항혈전 효능 (Antithrombotic Efficacy of Protaetia brevitarsis Extract)

  • We aimed to evaluate antithrombotic efficacy of Protaetia brevitarsis extract during 21 days. Rats (SPF rat, weight 240~260 g) were divided into 16 groups (5 rats per group), they were: control group and Protaetia brevitarsis extract groups with dose of 0.1, 0.5, 1, 2.5, 5, 10, 25, 50, 75, 100, 200, 250, 500, 750, 1,000 mg/kg kg of body weight. Thromboplastin time (PT) and activated partial thromboplastin time (aPPT) as antithrombotic efficacy were tested in this animal experiment (at 7, 14 and 21 days). Overall, the admistration dose of Protaetia brevitarsis extract over 50 mg/kg at 7, 14 and 21 days for PT and over 25 mg/kg at 7, 14 and 21 days for aPPT tented to be longer than that of other groups. In addition, the optimal admistration doses of Protaetia brevitarsis extract to improves antithrombotic efficacy were 75, 100, 200 and 250 mg/kg at 7, 14 and 21 days for PT (p<0.05) and 50 and 100 mg/kg at 7 days, 75 mg/kg at 14 days, or 50, 100, 200 and 250 mg/kg at 21 days for aPPT (p<0.05). It can be concluded that Protaetia brevitarsis extract at optimal levels have antithrombotic efficacy.
