Effect of an extract of Bauhinia variegata leaves on chronic arsenic intoxication in mice (Mus musculus): A preliminary study

  • Ethanolic leaf extract of Bauhinia variegata has been tested for its possible antioxidant potentials against sodium arsenite induced toxicity in mice. Mice were randomized into two groups of five and fifty mice. Group I consisting of 5 mice without any treatment with food and water ad libitum which served as normal control. Group II mice were fed with sodium arsenite in drinking water at 100 ppm concentration for two monthsthen they were segregated into five groups which were treated differently. Group II a mice received only arsenic as sodium arsenite with drinking water, Group II b were fed chronically 1 : 20 alcohol to distilled water (vehicle), Group II c, d, e mice were orally fed 50 mg/kg, 150 mg/kg and 250 mg/kg of B. variegata leaf extract of once daily for 15 and 30 days respectively along with arsenic. Several toxicity marker enzymes such as gamma glutamyl transferase, lactate dehydrogenase, aspartate and alanine aminotransferase, acid and alkaline phosphatase, catalase and superoxide dismutase along with haematological variables such as glucose 6-phosphate dehydrogenase, creatinine, bilirubin, haemoglobin and sugar in different groups of treated and control mice were studied. Results obtained from the in vivo experiment revealed that administration of sodium arsenite caused a significant increase in some enzymes while decrease in some. A similar trend was also observed with haematological variables. In contrast B. variegata treatment at 150 mg/kg favourably modulated these alterations and maintained the antioxidant status than other two doses i.e. 50 mg/kg and 250 mg/kg thereby making it a good candidate to be used as supportive palliating measures in arsenic induced toxicity.

벼의 약 부유배양에서 캘러스 유도 및 식물체 분화에 미치는 생장조절제와 항산화제의 영향 (Effect of Growth Regulators and Antioxidant Mixture on the Anther Floating Cultures of Rice)

  • 벼 소포자 유래의 반수성 세포집단을 기내 돌연변이 유기 연구에 이용하기 위하여, 액체배지를 이용한 약 부유배양에서 캘러스 유도에 미치는 생장조절물질의 영향과 유도된 캘러스의 갈변화에 따른 활력저하를 막기 위하여 배지내 항산화제의 첨가효과를 조사하였다. 약 부유배양을 위한 액체배지는 N6배지에 1 mg/L 2,4-D와 1 mg/L kinetin을 첨가한 배지에 서 캘러스 형성율 48.5%, 녹색체 분화율 6.8%, 총 식물체 분화율 9.0%로 가장 높았다. 약 부유배양에서 항산화제 (antioxidant mixture, Sigma Chemical Co.)의 첨가는 캘러스 형성율에 영향을 미치지 않았으며, 캘러스 갈변방지와 총 식물체 분화율 향상에 유의한 효과를 보였다. 항산화제의 적정농도는 250mg/L 였으며, 캘러스 유도배지와 식물체 분화배지에 모두 첨가하는 것이 가장 좋았다.

고지방식이 마우스에서 산마늘의 간과 신장 보호효과 (Hepatoprotective and Nephroprotective Effects of Allium victorialis Leaf Extracts on the High Fat Diet Supplied Mice)

  • 고지방식이로 유발된 간과 신장의 손상에서 산마늘 메탄올 추출물의 효과를 알아보고자 하였다. 12주 동안 산마늘 메탄올 추출물 62.5, 125, 250 mg/kg/day를 투여한 결과 고지방식이로 유발된 혈청중의 AST, ALT, BUN, creatinine의 증가 억제와 간의 조직병리학적 변화를 감소시켜 지방간염과 신장의 손상이 현저하게 감소하였다. 또한, 항산화 효소인 superoxide dismutase, glutathione, catalase의 증가와 malondialdehyde의 감소로 항산화방어체계(antioxidant defense system)를 강화시켰다. 산마늘 메탄올 추출물 125 mg/kg 투여군은 대조군인 silymarin 100 mg/kg 투여군과 유사한 효과를 나타내었다. 결론적으로 산마늘 메탄올 추출물은 고지방식이로 유발되는 간과 신장의 손상을 완화하는 유용한 작용을 하는 것으로 나타났다.

Anti-obese related pharmacological effects of standard potato protein extracts on the 45%Kcal high fat diet supplied mice

  • Objectives : In present study, therefore, possible beneficial pharmacological activities of standard potato protein extracts (SPE) were observed on the mild diabetic obese mice. Methods : After end of 12 weeks of continuous oral administrations of three different dosages of SPE 400, 200 and 100 mg/kg, or metformin 250 mg/kg, analyzed the hepatoprotective, hypolipidemic, hypoglycemic, nephroprotective and anti-obesity effects, separately. In addition, liver antioxidant defense systems were additionally measured with lipid metabolism-related genes expressions and hepatic glucose-regulating enzyme activities for action mechanism. Results : All of diabetes and related complications including obesity were significantly inhibited by treatment of SPE 400, 200 and 100 mg/kg, dose-dependently, and they also dramatically normalized the hepatic lipid peroxidation and depletion of liver endogenous antioxidant defense system, the changes of the hepatic glucose-regulating enzyme activities, also changes of the lipid metabolism-related genes expressions including hepatic $AMPK{\alpha}1$ and $AMPK{\alpha}2$ mRNA expressions, dose-dependently. Especially, SPE 200 mg/kg constantly showed favorable inhibitory activities against type II diabetes and related complications as comparable to those of metformin 250 mg/kg in HFD mice, respectively. Conclusions : The present work demonstrated that SPE 400, 200 and 100 mg/kg showed favorable anti-diabetic and related complications including obesity refinement activities in HFD mice, through AMPK upregulation mediated hepatic glucose enzyme activity and lipid metabolism-related genes expression, antioxidant defense system and pancreatic lipid digestion enzyme modulatory activities.

Effect of Triton X-100 on Compactin Production from Penicillium citrinum

  • Glucose alone was found to be the most effective carbon source for producing compactin. An initial glucose concentration of 40 g/L gave the highest compactin concentration of 250mg/L. Among the various nitrogen sources, when 5g/L of pharmamedia and soybean meal as the sole nitrogen source were used, respectively, the compactin concentration was higher than 250mg/L. Especially, in the case of the mixture of 6 g/L of pharmamedia and 8 g/L of soybean meal, the compactin concentration was 400mg/L. To select the best surfactant for effective compactin production, various surfactants were investigated. When Triton X-100 was used, the maximum compactin concentration was 445mg/L. With the initial concentration ranging from 1.5 to 2.0 g/L, the compactin concentration was the highest at 465-450mg/L. The cell concentration was similar to that of the control without the addition of Triton X-100. On the other hand, when the above 4.0 g/L of Triton X-100 were used, the cell concentration decreased. Using the based results, the continuous fed-batch cultures by adding the Triton X-100 were carried out for 10 days in an air-lift bioreactor. When 1.5 g/L of Triton X-100 was added to the culture broth at 0, 4, and 8 days of culture, respectively, the compactin production was increased with the increase of culture time. The maximum compactin concentration after 10days of culture was 1,200mg/L, which was about 2.0-fold higher than that of the control without the addition of Triton X-100.

Hepatoprotective effect of cordycepin-enriched Cordyceps militaris extract powder on high fat diet-induced hepatic steatosis in obese (ob/ob) mice

  • Herbal medicinal mushroom Cordyceps militaris has been traditionally used as tonic medicine for metabolic syndrome. Cordycepin, main extract of C. militaris, has been reported with immunomodulatory, anticancer, and hepatoprotective effects. This study was conducted to evaluate the potential hepatoprotective effect of cordycepin-enriched Cordyceps militaris, against high fat diet (HFD)-induced hepatic steatosis (HS) in male obese (ob/ob) mice. HFD was provided to ob/ob mice ad libitum (except negative control). Cordycepin-enriched C. militaris extract powder (CM) was orally administered once daily at dose levels of 0, 125, 250, and 500 mg·kg-1 for 4 weeks. During the study, body weight gain was statistically increased in all HFD fed groups compared to negative control, but body weight gain in CM 500 mg·kg-1 treated group shows a low tendency compared to HS model group. In organ weights, absolute and relative weights (to body weight) in liver and perirenal adipose tissue were increased in all HFD treated groups except CM 500 mg·kg-1 treated group compared to the negative control. In clinical chemistry, serum glucose and total cholesterol levels in CM 250 and/or 500 mg·kg-1 treated groups were lower than HS model group. In microscopical examination, hepatocyte vacuolation with macrovesicles in HS model group was increased compared to negative control, but this finding was decreased in CM 500 mg·kg-1 treated group compared to HS model group. In this study, CM exhibited hepatoprotective effects against hepatic steatosis at mg·kg-1 in ob/ob mice.

Antihyperlipidemic effects of alcoholic extract of Pongamia pinnata Linn. leaves on high fat diet fed rats

  • The objective of the study was to investigate the antihyperlipidemic activity of alcoholic extract of Pongamia pinnata Linn. (PPAE) leaves in rats fed with high fat diet (HFD). PPAE was administered orally in the divided doses of 250 and 500 mg/kg/day for 30 days in HFD fed rats. Body weights were observed and the analysis of serum lipid profile was carried out on day 30. Marked decrease in the body weight, total cholesterol (TC), triglycerides (TG), low density lipoprotein (LDL) and very low density lipoprotein (VLDL) whereas significant increase in the levels of high density lipoprotein (HDL) were observed after treatment with PPAE. However, PPAE in a dose of 250 mg/kg did not show significant (P < 0.05) increase in HDL levels. The PPAE also lowered TC: HDL-c and LDL: HDL-c ratios significantly suggesting it's antihyperlipidemic and cardioprotective potential. The present work reveals that PPAE at the dose of 500 mg/kg/day exhibited significant (P < 0.01) antihyperlipidemic effects.

Lindera erythrocarpa Makino extract reduces obesity induced by high-fat diet in rats

  • Lindera erythrocarpa Makino (LE) is widely distributed on Jeju Island, where it has been used for various traditional therapies. Effects of a crude extract of LE were examined in rats with obesity induced by a high-fat diet (HFD). Anti-obesity effects were followed in rats receiving orally administered vehicle, 100mg/kg extract, or 250 mg/kg LE extract, for 56 days. LE extract (250 mg/kg) suppressed increases in body weight and epididymal fat, with amelioration of fatty changes in the liver. Additionally, serum levels of alanine aminotransferase, aspartate aminotransferase, and total cholesterol were significantly decreased compared with those of vehicle-treated groups (p < 0.05). These results suggest that oral administration of LE extract reduced rat obesity induced by HFD, possibly through the reduction of fat accumulation.

지유 에탄올 추출물의 식품부패균에 대한 항균효과 (Antimicrobial Effects Against Food-borne Pathogens of Sanguisorbae Officinalis L. Ethanol Extract)

  • This study was performed to investigate the antimicrobial effect against food-borne pathogens of Sanguisorbae officinalis L. ethanol extract. The antimicrobial activity of the ethanol extract was determined using a paper disc-diffusion method and the diameter of the clear zone was measured. The diameters of the clear zone in the presence of 10 mg of the ethanol extract were the maximum against Staphylococcus aureus among the tested 4 gram-positive bacteria and Pseudomonas aeruginosa among the tested 7 gram-negative bacteria. Analysis of the minimum inhibition concentrations (MIC) showed that the ethanol extract exhibited a similar efficacy as sorbic acid, well-known chemical preservatives. The growth inhibitory effects of the ethanol extract in the concentrations of 250, 500, 1,000 and 2,000 mg/L on food-borne pathogens were determined against Staphylococcus aureus, Bacillus subtilis, Salmonella Typhimurium and Pseudomonas aeruginosa. The growth of the microorganisms was significantly (p<0.05) inhibited by the ethanol extract in the concentrations higher than 250 mg/L. Thus, the results of the present study demonstrate that the ethanol extract exhibits antimicrobial effects against food-borne pathogens, suggesting that Sanguisorbae officinalis L. could be used as natural antibacterial agent in food.

Central nervous system depressant activity of Leucas aspera root

  • The ethanolic extract of Leucas aspera root was studied for its effect on the central nervous system (CNS) using pentobarbitone induced sleeping time test, the open field test and the hole cross test in Swiss albino mice. The present investigation revealed that the extract at the doses of 250 and 500 mg/kg, significantly prolonged the pentobarbitone induced sleeping time in mice though the onset of sleep was delayed as compared to control. In open field test the depressing effect was prominent from the second observation period (30 min) and persisted throughout the entire experimental period (240 min). In the hole cross test, the depressing effect was observed significant from the third observation period (60 min) and persisted up to the seventh observation period (240 min) except at fourth observation (90 min) for 250 mg dose group and depressing effect was significant from second observation (30 min) up to seventh observation period (240 min) for 500 mg dose group. These results support the finding that Leucas aspera root may contain biologically active constituent(s) having CNS activity.
