Cryopreservation of Mouse Embryos by Vitrification (Mouse 배의 Glass화 보존)

  • Eight-cell mouse embryos equilibrated with vitrification solution(VS3), consisting of glycerol and polyethlene glycol, were plunged directly into liquid nitrogen. The embryos were cryopreserved by vitrification without intra- and extracellular ice formation. After the vitrified embryos were warmed at 4$^{\circ}C$, sucrose dilution procedures were examined and the survival embryos were transferred to recipients after 48h incubation in vitro. The results were obtained as follows. 1. Mouse embryos equilibrated with VS3 were diluted with 1.5m sucrose-HB 1 solution, 0.5M sucrose-HB1 solution for 5min at room temperature, respectively. The proportion of vitrified-warmed embryos developed to blastocyst(85.7%) was as high as that of the embryos diluted with 1.04M sucrose-HB1 solution at 4$^{\circ}C$(85.5%). 2. Normal live youngs were obtained in 53.9%(55/102) of the vitrified-warmed embryos after tranfer to pseudopregnant recipients.

Optimization of Culture Conditions for the [+]-Eudesmin Production in Magnolia Sieboldii Cells (함박꽃나무의 현탁배양세포로부터 [+]-Eudesmin의 생산을 위한 최적화)

  • In order to product the furofuranoid lignans, (+)-eudesmin which is one of the secondary products from Magnolia sieboldii. through cell suspension cultures; various culture media, initial sucrose concentration, elicitations, shaking speeds, and inoculum sizes. Among the culture media tested, MS medium had a pronounced effect on suspension cell growth and (+)-eudesmin contents. The maximum dry cell weight (DCW) of 3.71 g per flask was obtained at inoculum size of 0.5 g and in MS medium supplemented with $3\%$ sucrose plus 0.5 mg/L 2,4-D after 8 weeks. (+)-Eudesmin biosynthesis was stimulated with high initial sucrose concentration ,and the maximum (+)-eudesmin production of $3.2{\mu}g/g$ DCW was achieved at 200mg/L chitosan and $5\%$ initial medium sucrose. The optimal shaking speeds for dry biomass accumulation and (+)-eudesmin contents was 130 rpm. This work is considered to be helpful for large-scale bioprocessing of Magnolia sieboldii suspension cell cultures in bioreactor.

Determination of Soluble Carbohydrates in Soybean Seeds

  • This study was conducted to identify the soluble carbohydrates in soybean seeds using on-line HPLC-RID-ES/MS and HPLC behavior, and to deter­mine their contents for high quality soybean breeding. The monosaccharide (glucose) and three oligosaccharides (sucrose, raffinose, and stachyose) were identified in Korean soybeans by their chromatographic behavior and results of on-line HPLC-RID-MS with Electro­spray Ionization mode. On the basis of HPLC with a RID detector, the 32 Korean major soybeans contain $0.37{\pm}0.26\%$ glucose, $4.55{\pm}0.91\%$ sucrose, $1.19{\pm}0.19\%$ raffinose, and $2.72{\pm}0.37\%$ stachyose on a dry basis. In 468 soybean germplasms, the ranges of glucose, sucrose, raffinose, and stachyose were $0.03 - 0.98\%$, $2.33 - 6.96\%$, $0.08 -1.87\%$ and $0.75 - 3.18\%$, respectively. Among 500 soybean samples, oligosaccharide contents of 32 Korean major cultivated soybeans and 468 soybean germplasms were varied $5.83 - 10.06\%$ and $3.66 - 10.32\%$, respectively. The composition of glucose, sucrose, raffinose, and stachyose in soluble carbo­hydrates of 500 soybean samples were $2.07 {\pm} 1.75\%$, $58.01{\pm}5.82\%$, $10.13{\pm}2.28\%$ and $29.80{\pm}4.54\%$, respectively. Sucrose appeared to be most prevalent in soy­bean soluble carbohydrates.

  • Effects of the glycerol addition and removal medium containg 10% sucrose on the mouse embryo survival after freezing in a liquid introgen container were determined using the FDA test. The summarized results are the following. 1. The FDA score was higher (P<0.05) when embryos were frozen in the glycerol addition medium with sucrose that the one without it(3.4 vs 3.0). 2. No difference in the score was found between the glycerol removal medium containing 10% sucrose(3.2) and PBS+10% sucrose(3.3). 3. The score was higher(P<0.01) at morular stage than blastocyst stage of embryos.

  • This study were carried out to investigate the effective concentration of cryoprotective agents and sucrose by one-step straw method, and to determine the optimum thawing temperature and equilibration time of frozen porcine embryos. The porcine embryos foflowing dehydration by cryoprotective agents and a various concentration of sucrose were directly plunged into liquid nitrogen and thawed in 3$0^{\circ}C$ water. Survival rate was defined by FDA test. The results are sunnnarized as follows : 1. The survival rates of porcine embryos after ultrarapid frozen4hawing in the freezing medium with a various concentration of glycerol, DMSO and propanediol added 0.25M sucrose were higher survival rate than those of sucrose concentration of 0.50M. 2. The survival rates of porcine embryos after ultrarapid ftozen4hawing in the freezing medium added 0.25M and 0.SOM sucrose were higher survival rate than those of sucrose concentration of 0.75M and 1.00M. 3. The temperature thawed at 2$0^{\circ}C$ and 3$0^{\circ}C$ resulted in a significantly higher embryos survival rate after 72 hrs in culture than did at 35$^{\circ}C$. 4. The equilibration time on the survival rate of porcine embryos was attained after short period of time(2.5~5 min.) in the freezing medium higher than long period of time(10~20 min.).

Effect of different levels of xylooligosaccharide in sugar on glycemic index and blood glucose response in healthy adults (자일로올리고당 함유비율이 다른 설탕이 건강한 성인의 혈당지수와 혈당반응에 미치는 영향)

  • Purpose: In the present study, we aimed to evaluate the effect of sucrose containing 2 different levels of xylooligosaccharide on the glycemic index (GI) and blood glucose response in healthy adults. Methods: Healthy adults (4 male participants and 6 female participants, n = 10) were randomized to receive glucose, sucrose, sucrose containing 7% xylooligosaccharide active elements (Xylo 7), or sucrose containing 10% xylooligosaccharide active elements (Xylo 10). Each participant was administrated one of these materials once a week for 8 weeks and an oral glucose tolerance test was performed. Results: We found a reduction in the glycemic response to sucrose that included xylooligosaccharide active elements (Xylo 7 and Xylo 10). The glycemic indices of sucrose, Xylo 7 and Xylo 10 were 68.9, 54.7, and 52.5, respectively. The GI values of Xylo 7 and Xylo 10 were similar to that of foods with low GI. The percentage reduction of GI value caused by sucrose containing xylooligosaccharide active elements was significantly different and dose-dependent as compared to that caused by sucrose alone (p < 0.05). The reduction in the glycemic response to Xylo 7 and Xylo 10 was 21% and 24%, respectively, as compared to the glycemic response to sucrose. The attenuation of the glycemic response to Xylo 10 tended to be higher than that for Xylo 7 when the percentage of body fat was increased. Conclusion: These results demonstrated that xylooligosaccharide active elements may be effective in protecting humans against overconsumption of sucrose.

  • Sucrose is added to feed materials to alter the taste and texture of extruded products. Emulsifier can affect extrudate properties by forming complexes with amylose during extrusion-cooking. These ingredients may improve the cell structure and texture of cornmeal extrudates obtained by using $CO_{2}$ as a bubble forming agent. The objective of this study was to evaluate effects of sucrose (5% and 10%) and glyceryl monostearate (GMS) (0.75% and 1%) on properties of cornmeal extrudates produced with $CO_{2}$ at injection pressures from 1.04 to 2.07 MPa. Dough temperature increased and die pressure decreased when $CO_{2}$ was injected into barrel. The addition of sucrose to cornmeal resulted in decreasing dough temperature, specific mechanical energy (SME) input, and die pressure. SME input was not significantly influenced by GMS addition but die pressure was decreased when GMS was added. Extrudate density was decreased over observed $CO_{2}$ injection compared to GMS. WSI was significantly decreased with the addition of GMS. Paste viscosity was also decreased with addition of sucrose or GMS, but significant differences of paste viscosity among $CO_{2}$ injection pressures were not found. Stucture forming and texture of cornmeal extrudates by $CO_{2}$ injection was improved by adding GMS.

  • This study was carried out to effects of ethylene glycol concentration, sucrose and culture day of in vitro production embryo on slow-down freezing in Hanwoo. 6, 7, 8 and 9 day embryos produced in vitro were frozen using 1.8M EG+0.1M sucrose, 1.8M EG+0.5% BSA and 1.5M EG+0.1M sucrose media. Survivability was confirmed after frozen-thawed 24 and 48h and ICM, TE cell number were counted by Hoechst 33342 and PI staining after frozen-thawed 24h. As a result, 1.8M EG+0.1M sucrose group was most significantly (p<0.05) higher compared with the other treatment groups on survivability, TE and total cell number after frozen-thawed 24h ($94.2{\pm}2.6%$, $94.67{\pm}3.4$ and $129.67{\pm}5.5$). ICM number did not found significant (p<0.05) differences between the three treatment groups. in 6, 7, 8 and 9 day of embryos using three types of freezing media, frozen-thawed, 1.8M EG+0.1M sucrose groups with embryos cultured 8 day was significantly (p<0.05) highest survivability to $98.3{\pm}1.7%$ after frozen-thawed 24h. 1.5M EG+0.1 sucrose group with embryos cultured 9 day was significantly higher survivability than group of embryos cultured 8 day after frozen-thawed 24 and 48h. In conclusion, 1.8M EG+0.1M sucrose media is considered to be effective to cryopreservation of embryos cultured 8 and 9 day.

  • We cryopreserved mouse morulae by a simple ultra-rapid method of freezing embryos directly in $LN_2$ after holding 2min in a $LN_2$ vapor, and thawed them in $37^{\circ}C$ water bath. The time requirements for permeation and dehydration by 2.0 M glycerol and 0.2 M sucrose before freezing were studied. When the embryos were equilibrated for 10 min, the optimun post-thaw survival was obtained. Embryos those developed normally to blastocyst after in vitro culture for over 24hrs were regarded as survival ones. Two experiments to assess post-thaw survival following predehydration in various mixtures of glycerol and sucrose were also accomplished. When sucrose was held constant (0.2 M) and glycerol concentration varied (1.5-3.5 M), post-thaw survival was best (78.0%) in 3.0 M glycerol. When glycerol was held constant (3.0M) and sucrose concentration varied (0.0-1.0M), optimun post-thaw survival (78.0%) was found in 0.2 M sucrose.

  • Separation performance was measured for the disc plate and frame type reverse osmosis module using NaCl and sucrose solutions. An analysis of membrane performance was done following the equations proposed by Kimura-Sourirajan[8]. The membrane permeability was $2.17 \times 10^{-6}$(gmol/$cm^2$-sec-bar) and independent of operating pressure. The effect of concentration polarization for sucrose solution was higher than that of NaCl. Permeation flux for sucrose solution above 40 bar was decreased as operating pressure was increased. Solute rejection for NaCl solution was decreased, but that of sucrose was increased as operating pressure was increased.
