Preparation of Defatted Grape Seed Meal Protein Composite Films (포도씨박 단백질을 이용한 가식성 필름의 제조)

  • Defatted grape seed meal protein (DGP) was extracted, and DGP films containing various plasticizers were prepared. To improve the mechanical properties of DGP film, nanoclay or gelatin was incorporated in the film-forming solution to manufacture DGP composite films. Among the plasticizers of fructose (FRU), sucrose (SUC), and polypropylene glycol (PPG), DGP film containing a SUC : PPG ratio of 2.5:0.5 exhibited the best film-forming ability. Addition of Cloisite $Na^+$ improved the mechanical properties of DGP film. Tensile strength (TS) and elongation at break (E) of the film containing 5% Cloisite $Na^+$ were 1.45 MPa and 71.97%, respectively. Regarding the DGP/gelatin composite films, TS and water vapor permeability (WVP) increased with increasing gelatin amount, whereas E decreased. In particular, the DGP/gelatin (2:2) composite film exhibited 20.95 MPa TS and 12.25% E. These results suggest that the DGP film prepared from defatted grape seed meal can be used as a food packaging material.

Nutritional Components Content of Oriental Melon Fruits Cultivated under Different Greenhouse Covering Films (시설하우스 외피복재 종류에 따른 참외과실의 성분 함량)

  • This study was conducted to investigate the effect of functional cover film on oriental melon fruit quality under unheated plastic greenhouse cultivation in cold period. The 6 kind of films having different characteristics were covered at plastic greenhouses and oriental melon seedlings were cultivated at 2 regions of Seongju, respectively. The air-temperatures in plastic greenhouses of J-1 and J-2, having high infrared absorption rate, were about $2{\sim}5^{\circ}C$ higher than K-3. The contents of ${\beta}-carotene$ and sugar of fruit showed significant difference between functional and normal films. The sucrose, determining a sweetness during maturation, and soluble solids content of fruits cultivated in J-1, J-2, J-3, and K-1 were higher than those of K-2 and K-3. Ascorbic acid of fruits was highest in K-2 having low light transmission rate and thermo-keeping capacity. There was no significant difference in mineral content among all of cover films. These results indicated that the use of functional greenhouse covering films could improve fruit quality such as ${\beta}-carotene$ and sugar content of oriental melon.

  • This research aims the production of anti-tumor substances through in vitro culture of hairy roots transformed by Agrobacterium rhizogenes in Artemisia sylvatica MAX and the effect of culture conditions on optimum growth of hairy roots. We investigated the optimum medium, pH, carbon source, sucrose, light, Fe and polyamine conditions of various lines of hairy roots (NK3, NK4, YX. NK3-10) induced from Artemisia sylvatica to increase the optimum growth of hairy roots. MS medium was the best for optimum growth of hairy root clone, NK3-S10. The optimum culture period was 4 weeks for NK3-S10. The optimum sucrose concentration was 3.5%. The optimum concentration of FeSO$_4$, spermine and spermidine was 0.1 mM, 10 mM and 100 mM, respectively.

In Vitro Fertilization and Development of Frozen-thawed Bovine Follicular Oocytes (동결융해 소 난포란의 체외발생에 관한 연구)

  • Immature nocytes and in VitrO matured Oocytes collected from the slaughtered Korean cattle were frozen slowly with 10% ethylene glycol+5% polyvinyl pyrolidine+0.05M trehalose (l0EPT), 10% ethylene glycol+5% ficoll+0.05M sucrose (1OEFS), or 10% ethylene glycol+5% ficoll+0.05M trehalose (l0EFT) by cell freezer (experiment 1). And also,They were ultra-rapidly frozen with 30% ethylene glycol+10% polyvinyl pyrolidine+0.5M trehalose (3OEPT) or 30% ethylene glycol+18% ficoll+0.5M sucrose (3OEFS) using electron microscope grid (experiment 2). In experiment 1, the cleavage rate was 23.0% when immature oocytes were frozen slowly using various cryoprotectants descrihed above, and 5.1% of cleaved oocytes developed to over morula stage after in Vitro fertilization (IVF). There were no significant differences among these groups. When matured oocytes were frozen slowly, the total cleavage rate was 19.7%, and over morula stage was 3.2%. lOEPT (4.8%) and EFS (4.4%) were slightly more effective than l0EFT (0.0%) for development in vitro. Only in l0EFT treated group, immature oocytes have higher developmental capacity than matured ones, when they were frozen slowly and IVF after thawing. In experiment 2, oocytes were ultra-rapidly frozen using the electron microscope grid with two kind of cryoprotectants described above. In immature oocyte group, the cleavage rate was 13.9% and 5.8% of cleaved oocytes developed to over morula stage after IVF, and in matured group, 25.7 and 7.6%, respectively. There were no significant differences between two kind of cryoprotectants, but in ultra-rapid freezing using electron microscope grid, the efficiency is slightly higher in matured oocyte group.

  • The strain of Nuruk yeat No. 15 (NY-15) which ferments lactose in milk was Isolated from Nuruk and identified as Saccharomyces marxianus according to the API 20C profile index. The lactose hydrolysing ability of NY-15 was similar to that Saccharomyces fragilis ATCC 8583 which has ${\beta}-galactosidase$ activity. Its optimum growth temperature, pH and time for the production of maximum enzyme activity showed $28^{\circ}C$, 4.5 and 28hr, respectively. Galactose as well as sucrose as carbon sources, and urea as nitrogen source Increased the production of enzyme. In order to test the production of alcohol, NY-15 was inoculated in whey medium and whey medium added with sugar. In the former, NY-15 produced 2% alcohol and in the latter, it showed 12% alcohol production. The optimal medium pH for lactose hydrolysis of NY-15 is 4.5, whereas that of Saccharomyces fragilis ATCC 8583 is 3.5

  • As a study on the acid protease production by Rhizopus japonicus S-62, the culture conditions for the enzyme production and characteristics of the crude enzyme were investigated. The results are summarized as follows: 1. The optimum conditions for solid culture on wheat bran were 48 hrs of culture period and $100{\sim}120%$ addition of tap water. 2. Of the several media, wheat bran medium was the most excellent in the enzyme production. 3. The addition of sucrose, fructose, $NH_4Cl$ and $NaH_2PO_4$ on wheat bran, respectively, increased largely the enzyme production. 4. The optimum pH for the enzyme action was pH $2.4{\sim}2.6$, the optimum temperature was about $40^{\circ}C$, and the stable pH range was $pH\;2.5{\sim}5.0$, the enzyme was stable below $40^{\circ}C$ and was inactivated abruptly above $40^{\circ}C$.

  • Fifty strains of mold which isolated from the various sources were screened for the production of Naringinase which hydrolyse naringin, the 7-rhamnoe-glucoside of 4'.5.7. - trihydroxyflavanonin, the main bitter principle of citrus fruits and grape fruits. Of the 4 strains yielded naringinase with significant activity, S-1 strain was selected on the criterion of industrial application, and some properties of crude naringinase of this S-1 was investigated. The results obtained were as follows. 1. Naringenase obtained from S-1 strain has optimum pH range from 3.0 to 5.0 for its activity. 2. Production of naringinase was increased on the addition of naringin to the medium. 3. Hydrolysis of naringin with approporiate concentration of naringinase was carried out linerly up to 80% on the 0.1% substrate solution. 4. The optimum temperature for its activity was $50^{\circ}C$, and this enzyme was inactivated 80% of its total activity at $70^{\circ}C$ for 10 minutes, 40% at $60^{\circ}C$ for 30 minutes. But signifiant decrease of activity were not occurred by heat treatment at $50^{\circ}C$ for 2 hours. 5. Crude enzyme of the naringinase obtained from S-1 strain was competitively inhibited by addition of glucose on the substrate, and inhibitor constant of the glucose on the this enzyme was 1.5 Mol, and inhibition rate were linearly increased according to the increase of sucrose concentration and 56% of its total activity was inhibited at 1 Mol sucrose solution.

  • Fructans are the major vegetative storage carbohydrate in wheat(Triticum aestivum L.). The depolymerization of fructans occurs by the sequential removal of terminal fructosyl residues by a specific fructan exohydrolase(FEH). The objective of this study was to isolate and characterize this enzyme in wheat. From stems and sheaths of field-grown wheat(cv. Clark), FEH was purified 356-fold using salt precipitation and a series of chromatographic procedures including size exclusion, anion exchange, and affinity chromatography. FEH had a molecular weight of 63.7 kD and an optima at pH 5.5 and 3$0^{\circ}C$. The $K_{m}$s for $\beta$(2 longrightarrow1) linked oligofructans varied, from 10 to 37mM, with the lowest $K_{m}$ for tetrasaccharide. The $V_{max}$ increased as degree of polymerization (DP) increased. Wheat FEH hydrolyzed only, $\beta$(2 longrightarrow1) linked fructans but not, $\beta$(2 longrightarrow6) linked timothy fructan or sucrose. The role of this FEH in fructan metabolism in wheat is discussed.sed.

Cultural Condition of Enterobacter agglomerans U-1 for Polysaccharide Production (Enterobacter agglomerans U-1의 다당류 생산을 위한 배양조건)

  • The cultural condition of Enterobacter agglomerans U-1, a polysaccharide producing soil bacterium, was examined. The optimal medium composition was that contains the following components per liter of distilled water; $sucrose(23.75\;g/L),\;peptone(2.06\;g/L),\;yeast\;extract(0.5\;g/L),\;KH_2PO_4(1.0\;g/L)$ and $MgSO_4{\cdot}7H_2O(1.0 g/L)$. The optimum temperature for polysaccharide production was $30^{\circ}C$, where 8.5 g/L of polysaccharide was produced. The apparent viscosity of fermentation broth after 3 days was 240 mPa.s. at $70sec^{-1}$. The product yield and specific productivity were 36% and 142.07 mg/g/h.

  • Chemical components relevant to the characteristic antimicrobial activities of the Korean chestnut (Castanea crenata S. et Z.) leaves and flowers were analyzed. The composition of free sugar were sucrose, maltose in the chestnut flowers and sucrose, glucose, fructose in the chestnut leaves. The contains of tannin were 0.16% in the chestnut flower and 1.98% in the chestnut leaves. In fatty acids case, the linoleic contents were significantly high in the chestnut leaves and flowers. The organic acids showed high composition to succnic and citric acid in the chestnut leaves. The ammo acid compositions showed high contents to aspatic acid, proline, glutamic acid, glycine and methionine in the chestnut leaves and flowers. The total amino acid showed significantly higher in the chestnut leaves than flowers. The major minerals contained in the chestnut leaves and flowers were K, Ca, Ng, Fe, Mn and Al. Ascorbic acids were detected highly in the chestnut leaves and flowers.
