Effects of Daminozide Spray and Floral Preservatives on Longevity and Ethylene Production of Cut Lily (Lilium longiflorum Thunb.) (수확전(收穫前) Daminozide의 살포(撒布)와 수확후(收穫後) 절화보존제(切花保存劑)의 처리(處理)가 절화백합(切花百合)의 수명(壽命)과 Ethylene발생(發生)에 미치는 영향(影響))

  • This study was conducted to clarify the effects of daminozide (butanedioic acid mono-(2,2 - dimethylhydrazide)) sprayed 2 days be fore harvest and floral preservative solutions on longevity, quality and ethylene production of 'Georgia' cut lily for prolonging vase life and improving quality of cut lily flowers. And also the relation between ethylene production and flower senescence of cut lily was studied. 1. Lilies held in silver nitrate ($AgNO_3$) at 25 and 50 ppm were increased in solution uptake, fresh weight, and flower longevity. The solution of 2.5% sucrose+50 ppm $AgNO_3$+200 ppm aluminum sulfate+10 ppm 6-benzylamino purine prolonged vase life and improved quality of cut lily flowers. 2. Lilies sprayed with 500 ppm daminozide 2 days be fore harvest and then held in the preservative solution (5% sucrose+50 ppm $AgNO_3$+150ppm 8-hydroxyquinoline) after harvest were significantly increased in fresh weight and vase life as compared with non-sprayed (control) flowers. 3. Ethylene production from cut lily was increased by ethephon ((2-chloroethyl) phosphonic acid) treatment. The flowers producing a lot of ethylene, however, were senesced slowly instead of rapid wilting. 4. The preservative solution markedly reduced ethylene production of cut lily but prolonged vase life for only a few days. 5. These results suggest that ethylene be one of the most important factors promoting flower senescence of cut lily, but it be very difficult to prolong vase life remarkably by only inhibition of ethylene production in cut lily.

CHANCES IN THE SOME INGREDIENTS OF LOW GRADE TOBACCO LEAVES BY HEAT TREATMENT (하급 잎담배의 열처리에 의한 성분변화 연구)

  • low grade leaves of flue-cured (N.C. 2326) and air-cured (Br 21) have been heated at $140^{\circ}C$ for 10 and 20 minutes, respectively. As heating duration increased, each contents of nicotine, total nitrogen, total sugar and free amino acid decreased in both leaves. The result obtained from this experiment are as following 1. Nicotine content decreased in both leaves. Heating for 10 and 20 minutes showed respectively nicotine decrease of 13.2% and 21.6% for Br 21 and 8.2% and 13.2% for N.C.2326. Decrease of total nitrogen and total sugar was inversely proportional to the heating duration. N.C. 2326 in the decrease out numbered Br.21 by the ratio 2: 1 for 10min.-heating, and 5:4 for 20 min.-heating. 3. Total free amino acid was decrease by 17.1% and 13.85 for N.C. 2326 and Br 21, respectively when heated for 10 min., and 25.6% and 26.5% respectively when heated for 20 min. When sucrose was added to Br 21, th decrease ratio of total free amino acid of Br.21 was increased. It is suggested that the addition of sucrose could promote the browning reaction in Br 21 leaves.

Growth and Pigment Formation of Genus Monascus on Medium Compositions (배지에 따른 Genus Monascus의 증식도와 색소함량)

  • Growth and pigment formation in Genus Monascus(M. pilosus and M purpureus) related 15 kinds of culture media(Lin medium, SP medium, YM medium, YE medium, GMIN medium, SMO medium, MY medium, GY medium, Nishikawa medium, sucrose medium, stock culture, Mizutani medium, modified Lin medium, Toya medium and rice medium) were investigated. Mizutani medium and Lin medium among 15 kinds of the culture media showed good growth for M. pilosus, M purpureus, fresh mycelium weight cultivated for 10 days at 30$^{\circ}C$ was each 24.5∼26.9 or 15.9∼17.2 g/100 mL. The culture media which showed higher content of pigment production in two fungi were Lin medium(OD: 1.2 ∼ 1.6 and Mizutani medium(OD: 0.8 ∼ 1.0) that showed higher in M. pilosus.

Promotion of in vitro growth and rooting of micropropagated shoots in Spathiphyllum floribundum by the addition of liquid medium (액체배지 첨가에 의한 Spathiphyllum floribundum 신초의 기내생육 및 발근 촉진)

  • This experiments were carrid out to examine the effects of liquid medium addition in same vessels on shoot elongation and rooting, and soil survival of plantlets after the shoot cluster sections of Spathiphyllum floribundum 'Cupid' were pre-cultured. The shoot clusters with 3 to 4 small shoots were proliferated on LS medium supplemented with 2.0 mg/L BA for 8 weeks, and then 15 mL of various kinds of liquid medium was added in the same vessels. The addition of 15 mL liquid medium containing l/2 MS macro elements, 50 g/L sucrose and 5.0∼10.0 g/L activated charcoal was significantly stimulated the elongation and rooting of proliferated shoots. The medium addition was resulted in the enhanced soil survival of plantlets.

  • This experiment was carried out to investigate proper conditions for protoplast isolation and reversion from Ganoderma lucidum and Gctnoderma sp.. In G. lucidum, 10 mg. $ml^{-1}$ Novozyme 234 with 0.6 M sucrose was proper for protoplast isolation. The optimal reaction time of mycelium with lytic enzyme was five hrs. Protoplast isolation from four-day-old mycelium was the most effective. Protoplast isolation from four-day-old mycelium in G. sp. was optimum in the combination of N ovozyme 234 and ${\beta}-glucuronidase$ with 0.6 M sucrose. MCM was suitable for reversion in G. lucidum while SCM was good for G. sp.. The most effective osmoticum stabilizer for protoplast reversion in G. lucidum and G. sp. was 0.6 M sucrose.

Isolation and Characterization of Hydrogen Producing Bacterium (수소생산균 Enterobacter cloacae YJ -1의 분리 및 특성)

  • The hydrogen-producing bacterium was isolated from fresh water and identified as Enterobacter cloacae. The isolated was named Enterobacter cloacae YJ-1. In batch culture, The optimum cultivation temperature and pH of strain YJ-1 was 35℃ and 7.5, respectively. All of the added glucose was consumed completely during fermentation even though pH was not controlled. Amount of hydrogen produced on each condition of 2% glucose, 4% sucrose and 5% fructose was 950, 1000 and 948 mL/L, respectively and resulted in increasing hydrogen production approximately 2.5-times more than controlled condition. The maximum hydrogen production was obtained when 50 mM phosphate was added. In repeated-batch culture, hydrogen gas of 1920 mL/L was totally produced for 48. The maximum hydrogen was produced on the condition of 0.5% yeast extract, but the production amount was not changed on the condition of over 0.5%. Most of the organic acids produced during the fermentation were formic and acetic acid, and propionic acid was moiety also generated.

  • The young cells of Clostridiunm saccaroperbutylacetonicum were rapidly autolysed by exposing them to the hypertonic solution of sucrose(0.3-0.6M) without any other supplement to decompose the rigid cell wall. The cells were converted into the spherical cells by lysis. The spherical cells had following properties: (1) they were absent in the cell wall and osmotically fragile. (2) they were stabilized in the existence of 0.4M sucrose and 5mM $MgSO_4$ (3) they were resistant against adsorption of phage particles. (4) they allowed infection of the isolated phage DNA and produced progeny phage particles. (5) they were able to biosynthesize their macromolecules for a few hours according to a balanced manner of biosynthesis. (6) they were able to produce the bacteriocin particles by mitomycin C treatment. (7) they were unable to multiply. These results were all in the level of typical properties of bacterial protoplasts. It was apparent that the spherical cells formed by lysis occcurring by treatment with hypertonic sucrose were protoplasts.

Anthraquinone Productivity by the Cultures of Adventitious Roots and Hairy Roots from Curled Dock (Rumex crispus) (소리쟁이 (Rumex crispus)의 부정근과 모상근 배양에 의한 Anthraquinone 생산성)

  • In order to survey anthraquinone productivity from in vitro root culture, transformed hairy roots of Rumex crispus were induced from leaf segments by infection with Agrobacterium rhizogenes strain $A_4$ and compared with adventitious roots. The optimum condition of adventitious root formation from leaf segments was 5 $\mu$M NAA added to MS medium. Mannopine was detected in the extract of hairy roots by paper electrophoresis, but not in adventitious roots. Secondary root tips of both adventitious roots and hairy roots elongated without lateral root branching in hormone free MS medium, but primary root tips showed more rapid growth with extensive lateral root branching. MS basal medium was the best for growth of the adventious roots and hairy roots for anthraquinone content. Adventitious root tips and hairy root tips cultured in liquid MS medium supplemented with 0.05 $\mu$M NAA and 0.1 $\mu$M kinetin (contained 5% sucrose) showed the maximal growth and anthraquinone content. Anthraquinone content of hairy roots was increased by the culture periods, but was reduced after 25 days of culture.

Statistical Approach to Development of Culture Medium for Ansamitocin P-3 Production with Actinosynnema pretiosum ATCC 31565

  • The Plackett-Burman design and the response surface method (RSM) with a central composite design (CCD) were employed to develop a culture medium for ansamitocin P-3 production using Actinosynnema pretiosum ATCC 31565. Among the 11 nutrients tested using the Plackett-Burman design, two carbon sources, sucrose and dextrin, and two nitrogen sources, polypeptone and yeast extract, were selected. Optimization of the concentrations of the selected nutrients was then performed using RSM with CCD. After two rounds of RSM, the optimum concentrations ($\%w/v$) of sucrose, dextrin, polypeptone, and yeast extract were identified as 4.5, 4.5, 0.16, and 0.89, respectively. The maximum ansamitocin P-3 titer was 45.2 mg/l with the optimized medium, which was about 6 times higher than that (7.315 mg/l) obtained with an $R_{2}YE$ medium before optimization.

The Effect of Addition of Egg Yolk and Sucrose on the pH and Specific Gravity for Heated Egg Albumen (난백의 가열처리에 있어서 난황과 자당 첨가가 pH 및 비중의 변화에 미치는 영향)

  • This study was divided into experiment I(129g egg-albumen plus 150g sucrose) and experiment II(161g egg-albumen plus 150g sucrose) which were subdivided into groups treated with 0, 8.71, 17.43 and 26.149 egg yolk. These experiments were incubated in a shaking water bath(5$0^{\circ}C$) with a speed of 92(turnaround) per minute for a period of times. The pH and specific gravity were measured after 2, 4, 6, 8, 10 and 12 hour incubations. The results obtained were as follows: 1. The pH and specific gravity were steadily increased by shaking time, but variably was influenced by the amount of egg yolk and egg-albumen. 2. In the groups treated with egg-albumen, egg yolk, and sucrose of experiments I and II, pH was increased by the amount of egg-albumen in conjunction with shaking time, but specific gravity was decreased. 3. Ten hours after shaking maximum pH 8.60 was shown in the groups treated with 161g egg-albumen : however, in :he groups treated with 129g egg-albumen the maximum pH 8.39 was shown 12 hours after shaking. The different time exhibited maximum pH resulted from the amount of egg-albumen used. 4. The pH specific gravity were higher in the ,groups treated without egg yolk than in the groups with egg yolk. 5. In the experiment I which was added four levels of egg yolk to 1509 of sucrose and 129g of egg albumen, specific gravity(Table 2) had a high (r=0.9692$^{* *}$) correlation with pH (Table 1) and the regression equation between specific gravity and pH (X) was Y=0.050+0.145x. 6. In the experiment II which was added four levels of egg yolk to 150g of sucrose and 161g of egg albumen, specific gravity(Table 4) had a high (r=0.8963$^{* *}$) correlation with pH (Table 3) and the regression equation between specific gravity and pH (X) was Y=0.294+0.110X.10X.
