The effects of heat treatment of soy protein and of various nutrients added to soy protein isolate (SPI)-based yogurt on the growth of Lactobacillus acidophilus were investigated. The sensory evaluation of the yogurt beverage prepared from SPI was also performed. Soy milk was prepared from SPI (4.2% as protein) and various nutrients, such as glucose, lactose, sucrose, yeast extract, peptone and tryptone. Mild $(60^{\circ}C)$ or medium $(95^{\circ}C)$ heat treatment of soy protein did not inhibit the growth of L. acidophilus and the optimum degree of heat treatment was approximately $95^{\circ}C$ for 20 min. Glucose and lactose enhanced the growth and acid production by L. acidophilus. The optimum concentration of these two sugars in the medium was approximately 3% each. Yeast extract of approximately 0.5% stimulated the acid production by L acidophilus. Concentration of soy protein did not affect the growth of L. acidophilus, whereas it affected the viscosity of SPI-based yogurt markedly.
As a part of the survey on the chemical compositions of Korean Sweet potatoes, the changes of chemical compositions of Suwon No. 118 and Suwon No. 147, which are representative varieties of sweet potatoes in Korea, were studied during storage. The result of the study showed the followings. 1. The moisture content of the both varieties were fairy uniform with higher value of Suwon No. 147 than another. 2. The starch content and crude starch yield in the stored root tuber were shown to be considerable uniform except decrease at the earlier part of storage and in January. 3. Reducing sugar content of them showed increase at earlier part, then almost uniform except gradual decrease in later part of the storage. 4. The content of total soluble sugar were gradually increased at the earlier part, then became almost uniform at the later part of the storage. 5. The sucrose content showed gradual increase except at the earlier part of the storage in which both varieties showed different feature in decrease of the content. 6. The content of crude protein and soluble protein were almost uniform throughout the storage.
This study was carried out to examine growth characteristics and seed qualities of 1,296 Korean soybean landraces. The range of days to flowering, and days to maturity was 38 to 83 days and 47 to 102 days, respectively. The range of growth days were 105 to 160 days, and 38% was belonged to maturity group III. The 100 seed weight was 19.5g, showing the range of 2.4 g to 40.4 g, and 19.5 g in mean. 35.3% was in the range from 13.1 g to 20.0 g, and 29.4% in the range from 20.1 g to 25.0 g. Mean contents of crude protein was 41.8%, showing the range from 32.7% to 49.2%. Mean contents of crude oil was 18.0%, showing the range from 11.8% to 22.2%. The composition of unsaturated fatty acids were from 81.8% to 94.2%, and 85.4% in mean. Sucrose contents were in the range from 1.24% to 7.91% with the mean 5.21%, and oligo-saccharide contents from 2.45% to 11.13% with the mean 8.01%. Total isoflavone contents were in the range from $278.4\;{\mu}g/g$ to $2,736.9\;{\mu}g/g$ with the mean $1,066.8\;{\mu}g/g$. Among isoflavone contents, daidzein, glycitein, and genistein contents were in the range from $48.8\;{\mu}g/g$ to $1,709.6\;{\mu}g/g$ with the mean $483.2\;{\mu}g/g$, from $0.98\;{\mu}g/g$ to $892.3\;{\mu}g/g$ with the mean $111.6\;{\mu}g/g$, and from $79.8\;{\mu}g/g$ to $1242.3\;{\mu}g/g$ with the mean $472.0\;{\mu}g/g$, respectively.

The study was aimed to investigate the mixing ratio of kiwi and persimmon juices for the production of good quality wine by Saccharomyces cerevisiae Y28. Firstly, the optimum condition of rapidase treatment for the kiwi and persimmon juices was established, thereafter various mixing ratio (10:0, 9:1, 8:2, 7:3, 6:4, 5:5) of kiwi and persimmon was investigated regarding physiochemical properties and flavor compounds of wine. As the result, the optimum conditions were obtained as 0.3% rapidase for 1 h in kiwi and 0.3% rapidase for 3 h in persimmon. According to higher ration of persimmon, the pH of wines increased from 3.69 to 3.77, while the acidity of wines decreased from 2.07% to 1.51% at 14 days fermentation. The ranges of brix and reducing sugar in wines were decreased which ranges around 9.6 to 8.8 and 6.07 to 6.90 g/L, respectively, after fermentation. Major organic acid in wines were identified as tartaric acid, malic acid, and citric acid. A small amount of free sugar such as sucrose and glucose were detected in wines, but fructose was completely absent. The soluble phenolic contents were decreased that ranges around 1.00 to 1.25 g/L, in contrast, browning degree were increased ranges around 0.212 to 0.412 after fermentation. The major flavor components were identified as ethyl acetate and hydrazine, and 1,1-dimethyl. Importantly, phenylethyl alcohol was detected from the all wines that have a typical rose like flavor. But sensory test results and preference of kiwi-persimmon (7:3) mixing wine was better than the other wines.

The chemical components in different parts of Artemisia argyi was investigated to provide industrial possibilities as functional foods The analysis result of proximate composition in leaves, stems and roots of Artenisia argyi was substantially as follows. The crude protein contents were 19.87, 6.14 and 5.68%, the crude lipid contents were 4.56, 1.30 and 1.20%, the crude fiber contents were 16.80, 29.70 and 29.45%, respectively. The major mineral components in Artemisia argyi were potassium, calcium and magnesium. Contents of potassium and calcium in leaves were 4,270.24 and 617.64 mg/100 g, respectively, they were more than double the contents of root. Sucrose and glucose as main free sugars were detected in the leaves and roots. However, glucose and fructose were identified in the stem. Total amino acids showed 17 amino acids. Contents of total amino acid in the leaves was the highest as 4,864.11mg/100g, and the stems and roots showed 1,953.99 and 1,601.73mg/100g, respectively. The major amino acids in the leaves and stems were proline(963.91 and 407.52mg/100g) and aspartic acid(577.38 and 299.17mg/100g), respectively. Glutamic acid(206.34mg/100g) and arginine(193.23mg/100g) were main amino acids in the roots. The major fatty acids in all parts were linoleic acid($C_{18:2}$), behenic acid($C_{22:0}$), and palmitic acid($C_{16:0}$). Eupatilin(35.0mg/100g) and jaceosidin (107.63mg/100g) as physiological compounds contents were higher in leaves than other parts.

This study was carried out to investigate the quality characteristics of ginseng JungKwa produced with hot-water extract from Maegmundong (MGJ) instead of sugar. Chemical characteristics of MGJ and control ginseng JungKwa produced by sucrose (CGJ) were measured by proximate constituents, free sugars, ginsenosides, total phenolic compound, flavonoid content, and antioxidative activity was measured by electron donating abilities and hydrogen peroxide scavenging activity. Carbohydrate, crude protein, crude ash contents of MGJ were 76.40, 1.46 and 3.49%, where as those of CGJ were 80.01, 0.59 and 0.96%. Free sugar compositions of MGJ were fructose (32.42%), oligosaccharide (16.91%), disaccharide (13.91%) and glucose (13.16%). Eleven ginsenosides types were detected on MGJ and 9 kinds on CGJ; their contents were 740.1 and 675.6 mg%, respectively. Total phenolic compound and flavonoid content were 5.46 and 0.016% on MGJ, 5.02 and 0.014% on CGJ, respectively. In physiological characteristics, the $IC_{50}$ of electron donating ability by DPPH on MGJ was about 34.5 mg/mL and ability was slightly higher than CGJ, although there's not much difference in ability between MGJ and CGJ. Hydrogen peroxide scavenging activities were 92% on MGJ and 96% on CJG when treated at $50{\mu}g$/mL.
The culture condition and medium composition for the enhanced mycelial growth of Inonotus mikadoi IMSNU 32058 were investigated. The optimal temperature and pH for the mycelial growth were $27^{\circ}C$ and pH 4.5, respectively. Among the complex media tested, the malt extract medium and Phellinus igniarius medium were very good for mycelial growth of I. mikadoi. When Czapek-Dox medium was used as a minimal medium for cultivation of mycelia, xylose, raffinose and carboxymethyl cellulose were very excellent as a carbon and energy source. With respect to carbohydrate, sucrose and glucose were very good carbon sources. In general, organic complex nitrogen sources were better than other inorganic ones. As the organic complex nitrogen sources tested, yeast extract, soytone, proteose peptone and bacto peptone were the best as a source of organic nitrogen. When ammonium sulfate as an inorganic source of nitrogen was used, the enhanced mycelial growth was shown. p-Aminobenzoic acid was proved to be most appropriate source of vitamin. After the mycelia of I. mikadoi was cultivated at $27^{\circ}C$ for 5 days in MEM broth (pH 4.5), the activities of both exomycelial and endo-mycelial enzymes were determined. Among endomycelial enzymes assayed, the specific activity of laccase was much higher than those of other enzymes. When the fungus was grown in MEM broth, exomycelial specific enzyme activity of laccase was comparatively high. However, little or no enzyme activities of protease, chitinase and lipase were found.

Radish (Raphanus sativus L.) is a species of the Brassicaceae family and an important root vegetable crop, produced worldwide. A total of 82 radish accessions with various morphological and physiological characteristics analyzed for total sugars content. These accessions includes five subspecies and classified as wild, wild-relative, traditional and improved cultivar. The four sugars, glucose, fructose, sucrose and maltose, showed various contents in 82 accessions. Total sugar content ranged from 5.64 to 46.68 mg/g and showed average 25.33 mg/g. Total sugar content was not statistically significant among the five subspecies, but individual sugar ratio varied. The wild, wild-relatives and traditional cultivars were not significantly among average total sugars content compared with improved cultivars. On the other hand, the wild and traditional cultivars were showed high ratio of individual sugars. These results could be valuable information for the development of new radish cultivars and regulation of sugars biosynthesis in radish.
This study was conducted to examine the effects of electric stimulation conditions on in vitro developmental ability of caprine embryos after somatic cell nuclear transfer. Recipient oocytes were surgically collected after superovulation by using CIDR and FSH, PMSG, hCG and estrous synchronization in Korean native goats. The caprine ear cells were cultured in vitro in serum-starvation condition (TCM-l99 + 0.5% FBS) for 3 to 5 days of cell confluence. The zona pellucida of in vivo and in vitro matured oocytes were partially drilled using laser system. Single somatic cell was individually transferred into the enucleated oocyte. The reconstructed oocytes were electrically fused with 0.3M mannitol. After the electofusion, embryos were activated by electric stimulation or Ionomycin + 6-DMAP. Nuclear transfer embryos were cultured in mSOF medium supplemented with 0.8% BSA 6∼7 days at 39 , 5% $CO_2$, 5% $O_2$, 90% $N_2$. The fusion rate of donor cells was 60.4% and 40.3 % in ear cell and fetal fibroblast, and cleavage rate were 40.6% and 48.2%, respectively. No significant difference was found in the fusion and cleavage rate in different donor cells. Nuclear transferred oocytes were fused by electric pulses of 1.30∼1.40, 2.30∼2.39 and 2.40∼2.46 ㎸/cm. There was no significant difference among different electric pulses in fusion rates (26.7, 34.8 and 43.8%). The cleavage rate was higher (p<0.05) in 1.30∼1.40 ㎸/cm (82.9%) than 2.30∼2.39 ㎸/cm (43.8%) and 2.40∼2.46 ㎸/cm. (51.8%). The fusion rates of recipient oocyte source were 1st (43.5% and 23.6%), 2nd (55.7% and 39.2%) and 3rd (66.1% and 52.8%) in in vivo and in vitro oocytes. However, fusion ratee were significantly higher (p<0.05) in in vivo than in vitro oocyte. The cleavage rate of fused oocytes from in vivo and in vitro sources were 52.6% and 54.4%, respectively. No significant difference was found in the cleavage rate according to the recipient oocyte source. These results suggest that factors such as field pulse of electric stimulation and oocyte source could affect in vitro developmental ability of nuclear transplanted caprine oocytes.

Bacterium capable of using biphenyl as a sole source of carbon and energy were isolated from soil, and based on the results of 16S rDNA sequence, strain BK10 identified as a Sphingobium yanoiktiyae. The optimum cultural conditions were as follows; $NH_4NO_3$ 1g, $K_2HPO_4$ 1g, $MgSO_4{\cdot}7H_2O$ 0.5g, $CaCO_3$ 0.2 g per 1 liter of distilled water. The Sphingobium yanoikuyae BK10 strain was completely utilized biphenyl in mineral salt media containing biphenyl at concentration 500 $\mu$g/ml of biphenyl as a sole carbon and energy source within 48 hours. Optimumal pH and temperature for biphenyl degradation and cell growth of strains were 6.0$\sim$8.0 and 20$\sim$50$^{\circ}C$, respectively. Especially, at 30$^{\circ}C$, cell-growth were higher than other temperature. Cell grown on biphenyl has been shown to have a higher removal rate for biphenyl than grown on sucrose. This study shows that Sphingobium yanoikuyae BK10 strain had a high biodegradation capability of biphenyl and can be simulate a candidate compounds the bioremediation of PCBs (Polychlorinated biphenyl) contaminant soil and water.
