Aldh2 knockout 마우스에서 8주간 에탄올 노출에 따른 뇌조직의 thiobarbituric acid reactive substances 농도 (Thiobarbituric Acid Reactive Substances Levels in Brain Tissue of Aldh2 Knockout Mice Following Ethanol Exposure for 8 Weeks)

  • 과다한 음주는 알츠하이머 및 파킨슨 질병과 같은 각종 만성 퇴행성 뇌질환의 대표적인 원인 중 하나로 알려져 있다. 체내에 유입된 에탄올은 알코올 탈수소효소(alcohol dehydrogenase, ADH)에 의해 아세트알데히드로 대사된 후 다시 알데히드탈수소효소 2(aldehyde dehydrogenase 2, ALDH2)에 의해 아세트산으로 대사되어 배출된다. 에탄올의 대사과정 중에는 다량의 free radical이 생성되어 체내에서 산화적 스트레스를 유발하는 것으로 알려져 있고, 아세트알데히드는 활성산소를 생산하는 독성물질로 잘 알려져 있다. 본 연구에서는 8주간 에탄올에 노출된 Aldh2 knockout 마우스를 사용하여 ALDH2 효소 활성이 뇌 조직과 소변의 지질과산화에 미치는 영향에 대하여 살펴보았으며, 지질과산화 정도를 측정하기 위해 HPLC를 통한 TBARS 정도를 측정하였다. 연구결과, 마우스에서 만성 에탄올 섭취는 뇌 조직 TBARS 생성에 영향을 주지 않는 것으로 나타났으나, 소변 TBARS는 Aldh2 (-/-) 마우스에서 에탄올을 투여함에 따라 유의한 증가를 보였다(p<0.05). 본 연구 결과로부터 8주간 에탄올을 경구 투여한 마우스에서 ALDH2의 활성은 체내의 전반적인 활성산소 생성에는 중요하게 관여하는 것으로 보이지만 뇌조직에서의 활성산소 생성에는 영향을 주지 않는 것으로 보이며, 이는 에탄올 노출과 이에 따른 활성산소가 다양한 만성 뇌질환을 유발한다는 기존의 가설에서 ALDH2의 활성이 중요하게 관여하지 않을 가능성을 시사한다.

Antioxidative Activity of the Korean Wild Leafy Vegetables: Aster scarer mild Ligularia fischeri

  • The purpose of this study was to evaluate the antioxidative potential of Korean wild leafy vegetables in vivo as well as in vitro. Antioxidative activities of Aster scaber and Ligularia fischeri were evaluated against a reference of Spinacia oleracea Forty rats were fed either control diet or respective vegetable diets for four weeks. The level of thiobarbituric acid reactive substances (TBARS) and the activities of catalase and superoxide dismutase (SOD) in liver were compared. The plasma concentrations of total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C) and triglyceride (TG) were also compared. Korean wild leafy vegetables were assayed for $\beta$-carotene, vitamin C and vitamin E and total phenolic compound including flavonoid and thiobarbituric acid (TBA) value using the linoleic acid model system. SOD activity in rats fed Aster scaber was significantly higher(as much as 289%) than those fed Spinacia oleracea. Compared to control animals, the Aster scaber fed animals had significantly lower TC and lower atherogenic index. Compared to Spinacia oleracea, Aster scaber and Ligularia fischeri had vitamin C concentration of 150% and 400%, flavonoid concentrations of 470% and 310%, and phenolic compound concentrations of 326% and 203%, respectively, but tended to have lower $\beta$-carotene and significantly lower vitamin E concentrations. TBA values were only 18% of the control value in Aster scarier and Ligularia fischeri and 41% of the control value in Spinacia oleracea. These results suggest that Aster scaber could have potent antioxidative activity in vivo as well as in vitro and potential vague as a functional food to improve the plasma lipid profiles. Furthermore, phenolic and flavonoid compounds may be a major contributing factor to the antioxidative potential of Korean wild leafy vegetables.

LDL 산화에 대한 달맞이꽃의 플라보노이드 추출물의 항산화 활성 (Antioxidative activity of flavonoid rich extract of Oenothers odorate Jacquin on oxidation of low density lipoprotein)

  • 달맞이꽃 추출물로써 항산화활성을 알아보기 위하여 사람 low densty lipoprotein(LDL)에 대하여 실험하였다. 사람 LDL에 CuSO$_4$를 첨가한 후 배양하면서 달맞이꽃 추출을 일정 농도씩 증가시키면서 첨가하여 배양하였다. 달맞이꽃 추출물로서 LDL의 산화에 대한 개시와 연쇄 반응에서 thiobarbituric acid reactivy substances (TBARS)와 전기 영동 및 공액 2중 결합으로 측정하였다. 달맞이꽃 추출물을 LDL에 첨가한 후 macrophage J774로써 배양한 결과 TBARS값은 용량 의존형의 항산화 활성을 나타내었으며 전기 영동 이동상에서도 대조구에 비하여 각 시험구에서 LDL의 산화를 억제하였다. 또 공액 2중 결합에 대한 실험에서 용량 의존형산화 억제 효과를 나타내었다. 달맞이꽃 추출물의 항산화 활성은 40 $\mu\textrm{g}$/ml의 농도에서도 항산화 효과가 있었으나 80$\mu\textrm{g}$/mL의 농도에서는 거의 완전한 억제효과가 있었다. 그리고 비타민 C 및 $\alpha$-dl-tocopherol과 비교한 결과 달맞이들 추출물이 약간 높은 항산화 활성을 나타내었다.

율무의 지질에 관한 연구 (A research on the characteristics of Job's tears oil)

  • The contents of proximate components of the polished adn unpolished Job's tears, Coix agrestis, were determined. And also the phisical and the chemical properties of fatty acids composition and the oxidative rancidity of the crude oil extracted from Job's tears were examined. RESULTS : 1.The contnets of moisture, total ash, crude protein, crude fat, and carbohydrate in Job's tears were shown to be about 9.2%, 2.1~5.0%, 19.5~20.8%, 3.7~7.2%, 60.7~67.0%. 2. The average values of specific gravity, refractive index iodine value, saponification value, acid value of the crude oil extracted from Job's tears were 0.917~0.920, 1.47574, 107~111, 198~199, 1.3~2.0, respectively. 3. The fatty acids composition in the crude Job's tears oil showed oleic acid 48.97~49.97%, linoleic acid 37.33~36.16%, palmitic acid 13.70~13.87%. 4. Generally, the peroxide values and thiobarbituric acid values of the Job's tears oils and control during 40 storage days at 40±1℃ showed very low increase. After 32 days, the peroxide values and TBA values of soybean oil showed higher than those of the Job's tears oils. Conclusively, the Job's tears oils were shown to be more stable than the soybean oil.

  • Several new spiro compounds were synthesized via one-pot ternary condensation of isatin, malononitrile and each of thiobarbituric acid, barbituric, 3-methyl-pyrazolin-5-one, 1-phenyl-3-methyl-pyrazolin-5-one, acetylacetone, benzoylacetone, ethyl acetoacetate, phenacyl cyanide or ethyl-cyanoacetate dimer. Structures and reaction mechanisms were reported and supported via a second synthetic route.

Antioxidant Effect of Monascus koji in Sausage Mixture

  • The antioxidative capacity of Monascus koji was studied in raw bologna-type sausage by measuring thiobarbituric acid reactive substances (TBARS) and peroxide values (POV) during storage at $25^{\circ}C$ for 7 days. The addition of 2% Monascus koji effectively inhibited lipid oxidation in the mixture without vitamin C and NaNO$_2$, evidenced by its prevention of an increase in TBARS and a significantly reduction in increases in POV. Monascus koji in raw minced processed meat significantly improved the shelf life of the food.

  • Acanthopanax species (Araliaceae) traditionally has been used as analgesics, stimulant of immune system, and replenishment of body functions. Acanthopanax divaricatus var. albeofructus is indigenous plant to Korea. The antioxidant activities of compounds from A divaricatus var. albeofructus were determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and thiobarbituric acid reactive substance (TBARS) assay on human plasma low-density lipoprotein (LDL). The triterpenoid and lignan constituents from this plant showed antioxidant activities and the lignan, l-sesamin exhibited the most potent antioxidant activity in Cu$^{2+}$ -induced LDL oxidation.n.

Structural and Physiological Characteristics of Rhamnogalacturonan II from Fruit Wines

  • To characterize the polysaccharides which exist as soluble forms in fruit wines, crude polysaccharides were isolated from red, white, raspberry, wild grape, and pear wine, respectively. Among them, the crude polysaccharide (RW-0) in red wine showed the highest yield and considerable amounts of thiobarbituric acid (TBA)-positive materials. The pectic polysaccharide RW-2 was purified to homogeneity from RW-0 by subsequent size-exclusion chromatography using Sephadex G-75 and its structure was characterized. RW-2 consisted of 14 different monosaccharides which included rarely observed sugars in general polysaccharides, such as 2-O-methyl-fucose, 2-O-methyl-xylose, apiose (Api), 3-C-carboxy-5-deoxy-L-xylose (aceric acid, AceA), 3-deoxy-D-manno-2-octulosonic acid (Kdo), and 3-deoxy-D-lyxo-2-heptulosaric acid (Dha). Methylation analysis indicated that RW-2 comprised at least 20 different glycosyl linkages such as 3,4-linked fucose, 2,3,4-linked rhamnose, 3'-linked apiose, and 2,3,3'-linked apiose, being characteristic in rhamnogalacturonan II (RG-II). High performance size-exclusion chromatography indicated that RW-2 mainly comprised RG-II of higher molecular weight (12,000), and that the changes of molecular weight to apparent 7,000 under less than pH 2.0 were observed. These analyses indicated that the higher molecular weight polysaccharide in RW-2 was mainly present as a RG-II dimer.

바질의 항산화 물질 측정에 관한 연구 (Measurement of Antioxidation Substances in Basil)

  • Basil is known to contain six types of polyphenols that engage in physiological activation; protocateuic acid, caffeic acid, chlorogenic acid, courmaric acid, rosmarinic acid and quercetin. In this study, the antioxidants in eight types of basil were evaluated. Specifically, the antioxidative activation of basil was evaluated based on the relationship between active oxygen scavenging (DPPH radical-scavenging), which was used as an index showing the content and functionality of the polyphenol compounds in basil, and $Fe^{2+}$/ascorbate (FTC) and 2-thiobarbituric acid (TBA). The total polyphenol content of the different types basil occurred in the following order: Dark-opal> Lettuce> Bush> Greek> Lemon> Sweet> Geno> Holy. The highest content Dark-opal was 173.3 mg, which was about three times greater than the lowest content Holy, which was 49.85 mg. In addition, DPPH radical-scavenging by Dark-opal 51%, which was the highest scavenging activity occurred in the folling order: Dark-opal> Lettuce> Bush> Greek> Lemon> Sweet> Geno> Holy. The antioxidative activation values measured using the FTC and TBA were the same as the value obtained using the DPPH method. Finally, the level of antioxidative activation measured using FTC, TBA and DPPH methods showed that the higher the content of polyphenol substance was, the stronger the antioxidative activation became.
