Temperature Effect on the Growth and Odorous Material (2-MIB) Production of Pseudanabaena redekei (온도가 남조류 Pseudanabaena redekei의 성장과 냄새물질(2-MIB) 생산에 미치는 영향)

  • Cyanobacteria Pseudanabaena strains are known to produce 2-MIB(odorous material) in freshwater systems, thereby causing problems in water use. However, their physiological responses to environmental factors in relation with 2-MIB production is not well explored. This study was conducted to evaluate the effect of temperature on the growth and 2-MIB production of Pseudanabaena redekei. The experimental cyanobacteria strains were separated from the Uiam Reservoir (North Han River) and cultured in the BG-11 medium. Temperature was set to 10, 15, 20, 25, and 30℃ for the experiment, in the reflection of the seasonal water temperature variation in situ. For each temperature treatment, cyanobacterial biomass(Chl-a) and 2-MIB concentration (intra-cellular and extra-cellular fractions) were measured every 2 days for 18 days. Both maximal growth and total 2-MIB production of P. redekei appeared at 30℃. While intra-cellular 2-MIB contents were similar (26~29 ng L-1) regardless of treated temperatures, extra-cellular 2-MIB concentration was higher only in high temperature conditions (25~30℃), indicating that the extents of 2-MIB biosynthesis and release by P. redekei vary with temperature. The 2-MIB productivity of P. redekei was much higher in low-temperature conditions (10~15℃) than high temperature conditions (25~30℃). This study demonstrated that temperature was a critical factor contributing to 2-MIB biosynthesis and its release in cell growth (r=0.605, p<0.01). These results are important to understand the dynamics of 2-MIB in the field and thereby provide basic information for managing odorous material in drinking water resources.

Effects of Environmental Factors on Cyanobacterial Production of Odorous Compounds: Geosmin and 2-Methylisoborneol

  • Geosmin and 2-methylisoborneol (2-MIB), responsible for earthy or musty smell, are a major concern for safe drinking water supplies. This study investigated the effects of environmental factors on odorous compound production and cell growth in cyanobacterial strains. Anabaena sp. FACHB-1384, a 2-MIB producer, was sensitive to low temperature (<$20^{\circ}C$). However, geosmin producers, Anabaena sp. Chusori and Anabaena sp. NIER, were sensitive to high light intensity (>$100{\mu}mol/m^2/sec$), but not to low temperature. Geosmin concentrations increased under higher nitrate concentrations, being linearly proportional to cell density. A P-limited chemostat showed that P-stress decreased the geosmin productivity and extracellular geosmin amount per cell in Anabaena sp. NIER. However, only 2-MIB productivity was reduced in Planktothrix sp. FACHB-1374 under P-limitation. The extracellular 2-MIB amount per cell remained constant at all dilution rates. In conclusion, high light intensity and P-stress can contribute to the lower incidence of geosmin, whereas 2-MIB reduction could be attainable at a lower temperature.

  • Two strains S-P and S-P-2, both Streptomyces sp., have been isolated and were found to have relatively high specific enzyme activity compared to other organisms reported. The specific activity of the enzyme produced from these two strains were 0.25 and 0.2 international units respectively. The productivity of the enzyme achieved was about 50 IU/l/hr. Glucose isomerase form these strains was found to be stable under the temperature of heat treatment (at $65^{\circ}C$) for fixation of enzyme inside the dell. This organism has an advantage in that it did not require toxic metalic ion for enzyme activity and could utilize xylan in leu of xylose as an inducer. The optimal temperature and pH of enzymatic reaction purpose of using these data for the optimal operation and designing of enzyme reactor system. The reaction mechanism was found to follow the single substrate reversible reaction kinetics. The kinetic constants determined experimentally are : $K_{mf}=0.33M,\;K_{mb}=1.0M,\;V_{mf}=0.88{\mu}mole\;per\;min.,\;V_{mb}= 2.96{\mu}mole\;per\;min.\;and\;K_{eq}=0.74.

Studies on the Composition, Productivity, Regrowth and Soil Properties of Grasslands in Kimhwa and Chuiwon Areas (금화 철원지역 자연초지의 군락구조, 생산력, 재생력 및 토양특성에 관한 연구)

  • The investigation was made on the relationship among vegetation, net productivity, regrowth and soils of Moonheri-Wasoori, Soodowon-Sungilkyo and Mt. Kumhak grasslands in Chulwon area. The following conclusions have been made as the results of this study. The vascular flora of the grassland in Moonheri-Wasoori was composed of 102 species, the most of which were Medicago denticulata, Miscanthus sinensis, Arundinella hirta, Festuca ovina and Cassia nomame. The flora of Soodowon-Sungilkyn grassland was composed of 74 species, the most of which were Arundinella hirta, Miscanthus sinensis, Carex nanella, Festuca ovina and Cassia nomame. It of Mt. Kumfiak grassland was composed of 78 species, the most of which were Arundinella hirta, Carex nanella, Festuca ovina, Miscanthus sinensis and Aster scaber. These five species contributed greatly to the standing crop of live material which was in excess of 60% of it. The peak standing crop of each 413.7, 307.6 and $348.7g /m^2$ in Moonheri-Wasoori, Soo- dowon-Sungilkyn and Mt. Kuruhak grasslands wars reached in August under flooded conditions as a result of the growth of dominant species, Medicago denticulata and Arundinella hirta. The net prodution of organic matter was slight in some intervals but occurred throughout the growing season After cutting of grasses, the regrowth of new leaves mainly depended on growing stage and edaphic factors. Characteristics of the natural grassland soils, especially, concerning water, total nitrogen, avail-able phosphorus and exchangeable calcium were clarified and might be depend upon the quantity of plant production. Nitrogen, phosphorus and calcium concentration of leaves of Medicago denticulata, Arundinella hirta and Miscanthus sinensis were higher than of stems. Even if diem we diffurmem in the a-mb of nutrients among Medicago denticulata hirta and Miacanthus sinensis for single species and stands. they were covered by variations within species in the present survey. Simulation of Local Climate and Crop Productivity in Andong after Multi-Purpose Dam Construction (임하 다목적댐 건설 후 주변지역 기후 및 작물생산력 변화)

  • A simulation study was carried out to delineate potential effects of the lake-induced climate change on crop productivity around Lake Imha which was formed after a multi-purpose dam construction in Andong, Korea. Twenty seven cropping zones were identified within the 30 km by 25 km study area. Five automated weather stations were installed within the study area and operated for five years after the lake formation. A geostatistical method was used to calculate the monthly climatological normals of daily maximum and minimum temperature, solar radiation and precipitation for each cropping zone before and after the dam construction. Daily weather data sets for 30 years were generated for each cropping zone from the monthly normals data representing "No lake" and "After lake" climatic scenarios, respectively. They were fed into crop models (ORYZA1 for rice, SOYGRO for soybean, CERES-maize for corn) to simulate the yield potential of each cropping zone. Calculated daily maximum temperature was higher after the dam construction for the period of October through March and lower for the remaining months except June and July. Decrease in daily minimum temperature was predicted for the period of April through August. Monthly total radiation was predicted to decrease after the lake formation in all the months except February, June, and September and the largest drop was found in winter. But there was no consistent pattern in precipitation change. According to the model calculation, the number of cropping zones which showed a decreased yield potential was 2 for soybean and 6 for corn out of 27 zones with a 10 to 17% yield drop. Little change in yield potential was found at most cropping zones in the case of paddy rice, but interannual variation was predicted to increase after the lake formation. the lake formation.
