Temperature Effect on the Growth and Odorous Material (2-MIB) Production of Pseudanabaena redekei (온도가 남조류 Pseudanabaena redekei의 성장과 냄새물질(2-MIB) 생산에 미치는 영향)

  • Cyanobacteria Pseudanabaena strains are known to produce 2-MIB(odorous material) in freshwater systems, thereby causing problems in water use. However, their physiological responses to environmental factors in relation with 2-MIB production is not well explored. This study was conducted to evaluate the effect of temperature on the growth and 2-MIB production of Pseudanabaena redekei. The experimental cyanobacteria strains were separated from the Uiam Reservoir (North Han River) and cultured in the BG-11 medium. Temperature was set to 10, 15, 20, 25, and 30℃ for the experiment, in the reflection of the seasonal water temperature variation in situ. For each temperature treatment, cyanobacterial biomass(Chl-a) and 2-MIB concentration (intra-cellular and extra-cellular fractions) were measured every 2 days for 18 days. Both maximal growth and total 2-MIB production of P. redekei appeared at 30℃. While intra-cellular 2-MIB contents were similar (26~29 ng L-1) regardless of treated temperatures, extra-cellular 2-MIB concentration was higher only in high temperature conditions (25~30℃), indicating that the extents of 2-MIB biosynthesis and release by P. redekei vary with temperature. The 2-MIB productivity of P. redekei was much higher in low-temperature conditions (10~15℃) than high temperature conditions (25~30℃). This study demonstrated that temperature was a critical factor contributing to 2-MIB biosynthesis and its release in cell growth (r=0.605, p<0.01). These results are important to understand the dynamics of 2-MIB in the field and thereby provide basic information for managing odorous material in drinking water resources.

Morphological characteristics of the mesiobuccal root in the presence of a second mesiobuccal canal: a micro-CT study

  • Objectives: This study investigated the internal morphology of mesiobuccal (MB) roots of maxillary molars with a second mesiobuccal (MB2) canal. Materials and Methods: Forty-seven maxillary first or second molars from Brazilians were scanned using micro-computed tomography. The following measurements were obtained from the MB roots: root thickness, root width, and dentin thickness of the buccal aspect of the first mesiobuccal (MB1) canal, between the MB1 and MB2 canals, and the palatal aspect of the MB2 and MB1 canals at 3 mm from the root apex and in the furcation region. For statistical analysis, the Student's t-test and analysis of variance with the post-hoc Tukey test were used (α = 0.05). Results: In maxillary molars with an MB2 canal, MB roots were significantly thicker (p = 0.0014) and narrower (p = 0.0016) than in maxillary molars without an MB2 canal. The dentin thickness of the palatal aspect of the MB1 canal was also significantly greater than that of MB roots without an MB2 canal at 3 mm from the root apex (p = 0.0007) and in the furcation region (p < 0.0001). In the furcation region of maxillary molars with an MB2 canal, the dentin thickness between the MB1 and MB2 canals was significantly smaller than that in the buccal and palatal aspects (p < 0.0001). Conclusions: The internal morphology of MB roots of maxillary molars with an MB2 canal revealed differences in dentin thickness, root diameter, and distance between the canals when compared with maxillary molars without an MB2 canal.

Analysis of Geosmin and 2-MIB in Water by Stir Bar Sorptive Extraction (SBSE) and GC/MS (교반막대추출법(SBSE)과 GC/MS를 이용한 수중의 Geosmin과 2-MIB의 분석)

  • Musty and earth odors caused by geosmin and 2-MIB are the major complaints from the drinking water consumers. Therefore, early detection of geosmin and 2-MIB is the key to prevent off-flavor occurrence. In this study, a rapid method using stir bar sorptive extraction (SBSE) in combination with the GC/MS was investigated to analyze geosmin and 2-MIB in water. The SBSE method, a solventless extraction technique, was optimized and then applied to the analysis of geosmin and 2-MIB in water. The SBSE technique was found to be a simple and fast procedure that allows many samples to be extracted simultaneously using very small volume (10~20 mL). In addition, the SBSE method offered high recovery and good linear regression coefficient for the geosmin and 2-MIB. The good repeatability of this method can be deduced from the low RSD (7.2~7.6%) at 10 ng/L for geosmin and 2-MIB. The limit of detection was determined 1~2 ng/L and the limit of quantitation was 3~6 ng/L. Above all, the SBSE method proved to be a very practical technique for the analysis of geosmin and 2-MIB in water.

Apical periodontitis of endodontically treated maxillary first molar: Cone-Beam Computed Tomography study (근관치료된 상악 제1대구치의 치근단 병소: CBCT 연구)

  • Objective: The purpose of this study was to analyze the Cone-beam computed tomograghy(CBCT) scan of endodontically treated maxillary first molars and investigate how second mesiobuccal (MB2) canal is treated, how the prognosis of mesiobuccal (MB) root is different compared to other roots and the prognosis factor on apical periodontitis. Methods: Subjects were endodontically treated maxillary first molars whose were collected from CBCT scans taken from January 2018 until December 2019. A total of 525 maxillary first molars were analyzed by an endodontist to determine the presence of the MB2 canal, the quality of the root canal filling, and the presence of apical periodontitis. The chi square test and Fisher's exact test was used to examine the relationship between each variable. Results: MB2 canals were found in 46.3%, of which 76.5% were not treated. The more main canal of mesiobuccal root (MB1 canal) was well filled, the more significantly MB2 canal was well filled (p<0.001). The apical periodontitis of MB root was not related to the filling quality of MB1 canal (p=0.370) and was related to the filling quality of MB2 canal (p=0.004). The apical periodontitis of MB root was related to the quality of canal filling of MB2 canal and the apical periodontitis of DB and P root. Conclusions: It was found that the majority of MB2 canals were not treated. The apical periodontitis of MB root was analyzed to be related to the quality of canal filling of MB2 canal. The apical lesion of the MB root was not correlated with the treatment of the MB1 canal, but it was significantly related to the quality of filling of MB2 canal.

  • The present study is synthesis of derivatives of N'-(4-amino-5-sulfanyl-4H-1,2,4-triazole-3-yl)-2-(1H-benzimidazole-2-ylsulfanyl) acetohydrazide (IV). Antibacterial activity tested against the E. coli and A. Substilis. Biological activities conducted by disc diffusion method. Compound $2MB_1$, $2MB_3$, $2MB_5$ inhibit the appreciable microbial growth while rest of the compound possess the moderate activities. Anti-inflammatory activity tested by reduces local edema induced in the rat paw by injection of phlogestic agent. Compound $2MB_1$, $2MB_8$, $2MB_5$, $2MB_3$ and $2MB_6$ exhibit satisfying anti-inflammatory activity while analgesic activity conducted by acetic acid induced writhing effect in mice while compound $2MB_1$, $2MB_4$ and $2MB_7$ having the good analgesic activity. The chemical structures of all newly synthesized compounds were confirmed by their IR, $^1H$ NMR and mass spectral data.

  • This paper describes the design of a 155 Mb/s multiplexer-demultiplexer chip. This device for a 2.5 Gb/s SDH based transmission system is to interleave the parallel data of 51 Mb/s into 155 Mb/s serial data output, and is to deinterleave a serial input bit stream of 155 Mb/s into the parallel output of 51 Mb/s The input and output of the device are TTL compatible at the low-speed end, but 100K ECL compatible at the high-speed end The device has been fabricated with a 0.7${\mu}m$ BiCMOS gate array The fabricated chip shows the typical phase margin of 180 degrees and output data skew less than 470 ps at the high-speed end. And power dissipation is evaluated under 2.0W.

A cone-beam computed tomography study of the prevalence and location of the second mesiobuccal root canal in maxillary molars

  • Objectives: This study aimed to investigate the incidence and location of the second mesiobuccal root (MB2) canal in maxillary molars with the aid of various measuring points and lines using cone-beam computed tomography (CT). Materials and Methods: A total of 205 images of patients who underwent cone-beam CT examinations between 2011 and 2015 as part of their dental diagnosis and treatment were included. There were 76 images of the maxillary first molar and 135 images of the maxillary second molar. Canal orifices were detected at -1 mm from the top of the pulpal floor on cone-beam CT images. Image assessment was performed by 2 observers in reformatted image planes using software. Assessments included measurement of the distance between the MB1 and MB2 canals, and the angles between the lines connecting the MB1-MB2 and distobuccal (DB)-palatal (P) canals. The data were analyzed using the student's t-test. Results: The prevalence of the MB2 canal was 86.8% in the first molar and 28.9% in the second molar. The angle between the lines connecting the MB1-MB2 and DB-P canals was 2.3° ± 5.7° in the first molar and -3.95° ± 7.73° in the second molar. The distance between the MB1 and MB2 canals was 2.1 ± 0.44 mm in the first molar and 1.98 ± 0.42 mm in the second molar. Conclusions: The angles between the lines connecting the MB1-MB2 and DB-P canals was almost parallel. These findings may aid in the prediction of the location of the MB2 canal orifice.

Rejection Behavior of 2-MIB and Geosmin Using Polyethersulfone Nanofiltration Membrane and Surface Hydrophilicity Effect Using $TiO_2$ Particles (2-MIB와 Geosmin의 Polyethersulfone 나노 분리막 배제율 거동 및 $TiO_2$ 적용 표면 친수화 효과 관찰)

  • In this study, rejection behavior of 2-MIB (methylisoborneal) and geosmin which are known as taste-and-odor (T & O) causing micropollutants in drinking water source was investigated using hydrophobic polyethersulfone (PES) nanofiltration "loose" membrane (MWCO : 400 Da). It was found that the rejection of the geosmin was higher than that of the 2-MIB in all experimental conditions tested. This study also showed that the rejections of 2-MIB and geosmin were increased by increasing solution pH due to enhancing electrostatic repulsions between micropollutants and membrane surface. The presence of natural organic matter led to increase the rejection of the hydrophobic 2-MIB and geosmin and the effectiveness was more pronounced at higher solution pH. Increasing hydrophilicity of the hydrophobic membrane surface by coating with $TiO_2$ particles resulted in the significant increase in the rejection of 2-MIB and geosmin. In addition to the charge repulsion, this result suggests that hydrophobic-hydrophobic interaction should be one of main rejection mechanisms of T & O compounds by NF membrane.

The Effect of Mibaeksan(MB) on Melanin Synthesis and Gene Expression (미백산(美白散)이 멜라닌 생성 및 유전자 발현에 미치는 영향)

  • Purpose: This study was performed to elucidate the inhibitory effect of Mibaeksan (MB) on melanin synthesis in B16F10 mouse melanoma cell. Methods: To demonstrate the inhibitory effects of MB on melanin synthesis, we measured the amount of released and produced melanin in B16F10 melanoma cell. Also, we evaluated tyrosinase-activity in vitro as well as in B16F10 melanoma cell. And to investigate the action mechanism, we assessed the gene expression of tyrosinase, TRP-1, TRP-2, MMP-2, PKA, $PKC{\beta}$, ERK-1 ERK-2, AKT-1 and MITF in B16F10 melanoma cells. Results: 1. MB decreased the release and production of melanin in B16F10 melanoma cells. 2. MB decreased tyrosinase activity in vitro and in B16F10 melanoma cells. 3. MB decreased the expression of tyrosinase, TRP-1, TRP-2, PKA, $PKC{\beta}$ and MMP-2 in B16F10 melanoma cells. 4. MB increased the expression of ERK-1, ERK-2 and AKT-1 in B16F10 melanoma cells. 5. MB decreased the expression of MITF in B16F10 melanoma cells. Conclusion: From these results, it may be concluded that MB has the antimelanogenetic effects.

Adsorptive Removal of 2-Methylisoborneol and Geosmin in Raw Water Using Activated Carbon and Zeolite (활성탄과 제올라이트를 이용한 상수원수 중 이취미물질(2-MIB, Geosmin)의 흡착제거)

  • This study was performed to investigate the recovery efficacy of 2-methylisobomeol (2-MIB) and Geosmin, odor contaminants produced by algae, by pretreatment techniques, and also to investigate both adsorption characteristics and removal efficiency to get some information for the effective removal of 2-MIB and Geosmin by batch experiments. In pretreatment experiments, the best recovery efficiency of both odorants at 0.2 and $2\;{\mu}g/L$ in raw water was 30 mL of sampling size, 9 g NaCl for salting out headspace of sampling phase and 40 minutes of adsorption. At the best condition, the recovery efficiency of 2-MIB was 85% at $0.2\;{\mu}g/L$ and 95% at $2\;{\mu}g/L$, whereas the efficiency of Geosmin was lower than that of 2-MIB : 61% at $0.2\;{\mu}g/L$ and 81% at $2\;{\mu}g/L$. In batch experiments, the removal efficiency of the Geosmin and 2-MIB by adsorbents using distilled water were increased in comparison with raw water, the efficiency in raw water was little different by their concentrations. When these results were applied to the Freundrich adsorption isotherm, the K value of 2-MIB for zeolite, coal activated carbon, and coconut activated carbon was 0.671, 1.811, and 1.340, respectively, and the value of Geosmin was 0.6125, 1.771, and 1.5191, respectively. Thus the adsorption efficiency of 2-MIB and Geosmin was in the order of zeolite, coconut activated carbon, coal activated carbon.
