  • Robinia pseudoacacia var. umbraculifera, commonly called umbrella black locust were regenerated after co-cultivation of internode segments with Agrobacterium tumefaciens which included yeast cadmium factor 1 (YCF 1) gene. The tolerance to cadmium and lead for plants can be increased by the YCF1 gene expression. Moreover, the recent studies have shown that YCF1 gene transgenic plants increase the accumulation of cadmium and lead into plant vacuoles. The effect of plant growth regulator such as 2,4-dichlorophenoxyacetic acid (2,4-D), ${\alpha}$-naphthaleneacetic acid (NAA), 6-benzyladenine (BA), and thidiazuron (TDZ) were studied to evaluate the propagation of plants through internode explants. The efficient induction of multiple adventitious shoots and callus were observed on a medium supplemented with 0.1 mg/L TDZ + 0.2 mg/L BA. To induce shoot elongation and rooting, regenerated shoots were transferred into basal MS medium without any plant growth regulator. Successful Agrobacterium tumefaciens mediated transformation was obtained by 20 min vacuum-infiltration with $50{\mu}M$ acetosyringone on the optimal multiple shoot induction medium with 30 mg/L hygromycin and 300 mg/L cefotaxime. To confirm the integration and expression of transgene, Polymerase Chain Reaction (PCR) and Reverse Transcriptase PCR (RT-PCR) were performed with specific primers. The frequency of transformation was approximately 18.94%. This study can be used to genetic engineering of phytoremediator.

  • In this study, three cold-rolled TRIP steels containing different Al content (0.04wt%, 1.0 wt.% and 2.00wt%) were fabricated to understand the complex effects of Al in TRIP steel. The influences of Al on microstructural evolution of cold-rolled TRIP steels have been analyzed by using advanced analysis techniques, such as transmission electron microscope (TEM) and three dimensional atom probe tomography (3D-APT). TEM results revealed that second phases such as bainte and retained austenite decrease with increase of Al content. In addition, 3D-APT was used to characterize atomic-scale distribution of alloying elements at the constituent phases. Through these analysis techniques, the advanced characteristics of constituent microstructure in TRIP steels were identified depending on Al contents in TRIP steels.

  • The purpose of this paper is to present the optimal investment conditions of SOC facilities for maximizing regional social welfare in implementing the urban development project in the theoretical fashion. Particularily, SOC facilities are divided into both supply-side($P_s$) and demand-side SOC ($P_d$) in the paper. General equilibrium analysis from the intra-regional viewpoint by utilizing Pareto's Optimal Conditions and by revising Samuleson's Conditions for public goods($P_s$ and $P_d$) results in the optimum pattern of SOC investment. The following are important implications from the analysis. First, rather than the pursue social equity, SOC investment is to resolve the issue of efficiency to activate the regional economy. Second, the marginal rate of transformation (MRT) between $P_s$ and $P_d$ in the region is to play a significant role in structuring SOC investment plant of local government for social welfare maximization. Third, the optimal SOC investment policy based on this regional economy but also to generate the enhancement of soical amenities of the residents.

  • In the present paper, a method of nearly orthogonal grid generation in an arbitrary simply-connected 3D domain will be presented. The method is a new direct and non-iterative scheme based on the concept of the decomposition of the global orthogonal transformation into consecutive mapping of a conformal mapping and an auxiliary orthogonal mapping, which was suggested by King and Leal [4]. In our numerical scheme. Kang and Leal's method is extended from 2D problems to 3D problems while the advantage of the non-iterative algorithm is maintained. The essence of the present mapping method is that an iterative scheme can be avoided by introducing a preliminary step. This preliminary step corresponds to a conformal map and is based on the boundary element method(BEM). This scheme is applied to generate several nearly-orthogonal grid systems which are orthogonal at boundaries.

  • This study was carried out to develop an efficient transformation protocol via particle bombardment with PLBs (protocorm-like bodies) in Phalaenopsis. To achieve this aim, osmoticum treatment and an increasing shooting chances in particle bombardment process were applied for this study. In addition, pCAMBIA3301: ORE7 vector which contains a herbicide-resistance bar gene as a selectable marker and ORE7 gene as a gene of interests were employed. With regard to the increasing chances of shooting in particle bombardment, double shooting was the best results with 1.5 ~ 2.5 times higher than those of a single or triple shooting treatment in the productioon of PPT (D-L-phosphinothricin)-resistant PLBs. However, regeneration rate of shoots in double shooting was not high as a single shooting. Further, double shooting showed 35 ~ 40% higher than that of a single shooting in the frequency of browning. Regarding effects of different osmotic treatments, combination of 0.2 M sorbitol with 0.2 M mannitol showed the best results in transformation efficiency, regeneration of transformants and reduction of browning. Putative transgenic Phalaenopsis plants were analyzed by PCR analysis and confirmed the presence of bar and ORE 7 gene. Also, real-time PCR was conducted by using 21 transgenic plants and showed only 4 plants had one copy of transgene; whereas, the other 17 plants had more than 2 copies of transgene. Transgenic phalaenopsis plants produced in this study were transferred to pots and flowered normally without morphological variations in flower and leaf.

  • In order to improve forage characteristics of Italian ryegrass by genetic transformation, an efficient callus induction from mature seed and optimal plant regeneration system were established using a domestic cultivar 'Kospeed'. Addition of 5 mg/L of 2,4-D showed highest frequency of embryogenic callus induction from mature seeds. N6 medium showed higher frequency of both callus induction and plant regeneration as compared with MS and SH medium. The highest plant regeneration frequency 67% was obtained when embryogenic calli were transferred to N6 medium containing 1 mg/L 2,4-D and 5 mg/L BA. Supplementation of regeneration medium with sucrose at 30 g/L level maximized regeneration frequency as compared to the other concentrations. These data would be very helpful for molecular breeding of domestic Italian ryegrass cultivar through genetic transformation.

  • In an effort to increase the data capacity of one-dimensional symbology, 2D barcodes have been proposed a decade ago. In this paper, we present an effective 2D barcode detection algorithm from gray-level images, especially for the handheld 2D barcode recognition system. To locate the symbol inside the image, a criteria based on the block contrast is adopted, and a gray-scale projection with sub-pixel operation is utilized to segment the symbol precisely from the region of interest(ROI). Finally, the segmented ROI is normalized using the inverse perspective transformation for the following decoding processes. We also introduce the post-processing steps for decoding the QR-code. The proposed method ensures high performances under various lighting/printing conditions and strong perspective deformations. Experiments shows that our method is very robust and efficient in detecting the code area for the various types of 2D barcodes in real time.

  • The first and second harmonics have been measured in the iron whisker with the axial current that produces a circular field. The observed harmonics profiles have been discussed in terms of theoretical analysis based on the nonlinear, asymmetric magnetization that are related to the nucleation, transformation and annihilation of domains. The change of second harmonics profile is more sensitive to the domain variation than that of first harmonics.

  • The materials to develop a computer-based method for the wave resistance of a ship within the frame of Guilloton's wedge concept are presented in this paper. A systematic reliable procedure to retrieve the linearized hull corresponding to a given real hull form(the so-called inverse transformation) has been devised. The algorithm based on the present materials produces evidently accurate values of the H-functions, and the wave profiles and the wave resistance coefficients in good agreement with the experimental measurements.
