쇄양(鎖陽)의 Diphenyl-picryl-hydrazyl (DPPH) 소거 활성 및 HepG2 세포에 대한 항산화 효과 (Antioxidant Effect of Cynomorii Herba on HepG2 Cells and Diphenyl-picryl-hydrazyl (DPPH) Radical Scavenging Activity)

  • The purpose of this study was to investigate the anti-oxidant effect of Cynomorium songaricum. The extract of Cynomorii Herba was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, HepG2 cell viability and $H_2O_2$-induced cytotoxicity by a modified MTT assay. DPPH radical scavenging activity was measured after 30 minutes. The extract was tested by 1. 5, 10, 50, 100 and 500 ${\mu}g/ml$ concentrations. HepG2 cell viability by a modified MTT assay was measured in the concentrations of 10, 50, 100, 250, 500 ug/ml for 24 h. The results showed that the extract scavenged DPPH radical up to 52.2% with 50 ug/ml concentration. The extract did not reduced the cell viability and $H_2O_2-induced$ cytotoxicity (69.4%) was blocked by the extract in the concentrations of 50, 100, 250 and 500 ${\mu}g/ml$. In conclusion, the extract of Cynomorii Herba has potent antioxidant activity.

남성생식세포 Sertoli cell에 미치는 복분자(覆盆子)의 항산화 효과 (Antioxidant Effect of Rubi Fructus on TM4 Sertoli Cells)

  • Objectives : The purpose of this study was to examine the antioxidant effects of the extract of Rubi Fructus on TM4 Sertoli cells. Methods : The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity and cell viability assays on Sertoli cells. In addition, hydrogen peroxide-induced oxidative stress on Sertoli cells were examined by MTT assay. The antioxidant enzyme of Cu/Zn SOD, Mn SOD, catalase protein expression on Sertoli cells were also measured. Results : The results showed that the extract scavenged DPPH radical dose-dependent manner. The extract showed no cytotoxicity at concentration of 1, 5, 10, 50, $100{\mu}g/ml$. The hydrogen peroxide-induced cytotoxicity of Sertoli cells was protected to 88.3% by the extract at concentration of $100{\mu}g/ml$. Cu/Zn SOD and Mn SOD protein expression were significantly increased on Sertoli cells, but catalase protein expression was not significantly changed. Conclusions : In conclusion, the extract of Rubi Fructus has antioxidant effects on Sertoli cells and protect male reproductive system against oxidative stress.

  • 온도, pH, 배양시간을 세가지 인자로 하고 반응표면 분석을 통하여 Bacillus subtilis DC-2균에 의한 색소 생성 및 DPPH($\alpha$,$\alpha$-diphenyl-$\beta$-picryl-hydrazyl)에 의한 전자공여성의 최적 조건을 조사하였다. 색소생성능에는 배양온도가 10% 유의수준에서 영향을 미치는 것으로 나타났으며, DPPH에 의한 전자공여성의 경우 배양온도가 5% 유의수준, pH가 10% 유의수준에서 영향을 미치며, 배양시간은 10% 유의수준에서 영향을 미치지 않는 것으로 나타났다. Bacillus subtilis DC-2의 색소생성 최적 조건은 배양온도 39.$25^{\circ}C$, pH 8.83 및 84.41 시간으로 나타났으며 DPPH에 의한 전자공여성은 배양 온도 39.19$^{\circ}C$, pH 8.84 및 82.21시간에서 가장 높은 것으로 나타났다.

Effects of Salviae Miltiorrhizae Radix Hot Aqueous Extract on Nitric Oxide and Prostaglandin E2 Production and on 1,1-diphenyl-2-picryl hydrazyl Radical Scavenging in Macrophages

  • Objectives: The objective of this study is to investigate the effects of Salviae Miltiorrhizae Radix hot aqueous extract on nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) production and on 1,1-diphenyl-2-picryl hydrazyl (DPPH) free-radical scavenging in macrophages. Methods: Salviae Miltiorrhizae Radix (300 g) was heated at $100^{\circ}C$ with distilled water (2 L) for 4 hours. The extract was filtered and concentrated to 100 mL by using a rotary evaporator, was frozen at $-80^{\circ}C$, and was then freeze-dried by using a freezing-drying system. The RAW 264.7 macrophage was subcultured by using $10-{\mu}g/mL$ lipopolysaccharide (LPS). In order to evaluate cytotoxicity, we performed 3-(4,5-dimrthylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays and measured the cell viability. The NO production was measured by using Griess assays, and the $PGE_2$ production was measured by using enzyme immunoassays. The antioxidant activity, the 1,1-diphenyl-2-picryl hydrazyl (DPPH) free-radical scavenging capability, was measured by using the DPPH method. Results: Cell viability with the 1-, 5-, 25-, 125- and $625-{\mu}g/mL$ Salviae Miltiorrhizae Radix hot aqueous extract was not significantly decreased compared to the cell viability without the extract. When 125 and $625{\mu}g/mL$ of Salviae Miltiorrhizae Radix hot aqueous extract were used, nitric oxide (NO) production in LPS-stimulated RAW 264.7 macrophages was significantly inhibited compared to that in the control group. When 25, 125, and $625{\mu}g/mL$ of Salviae Miltiorrhizae Radix hot aqueous extract were used, $PGE_2$ production in LPS-stimulated RAW 264.7 macrophages was significantly inhibited compared to that in the control group. The 125- and $625-{\mu}g/mL$ Salviae Miltiorrhizae Radix hot aqueous extracts had high DPPH free-radical scavenging capabilities in RAW 264.7 macrophages. Conclusion: This study indicates that Salviae Miltiorrhizae Radix hot aqueous extract suppresses NO and $PGE_2$ production and improves DPPH free-radical scavenging capability. Thus, it seems that Salviae Miltiorrhizae Radix hot aqueous extract may have an anti-inflammation effect and antioxidant activity.

산수유(山茱萸)가 남성 생식세포 GC-1의 항산화에 미치는 영향 (Antioxidant Effects of Corni Fructus in GC-1 Cells)

  • The purpose of this study is to examine the antioxidant activity in the germ cells of the extract of Corm fructus. The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, GC-1 cell viability by a modified MTT assay, the effects on $H_2O_2$-induced cytotoxicity by MTT assay and lipid perixidation by malondialdehyde (MDA) formation, respectively. The results showed that the extract scavenged DPPH radical with the IC50 being $200{\mu}g/mL$. The extract at concentrations of $10-500{\mu}g/ml$ showed dose-dependent in growth of GC-1 cell. $H_2O_2$-induced cytotoxicity (63.0%) was blocked by the extract (10, 50, 100, 250 and $500{\mu}g/ml$) concentration-dependently. Furthermore, the extract (50, 100 and $250{\mu}g/ml$) also displayed a dose-dependent reduction of MDA formation on $H_2O_2$-induced lipid peroxidation. In conclusion, the extract of Corm fructus has potent antioxidant activity.

백자인(柏子仁)이 수컷 생쥐의 생식세포에 미치는 항산화 효과 연구 (Study of Antioxidant Effects of Platycladi Semen on Male Reproductive cells)

  • Objectives : The purpose of this study is to examine the antioxidant effects on male mouse reproductive cells of the extract of Platycladi Semen. Methods : The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, cell viability by a modified MTT assay, the effects on $H_2O_2$-induced cytotoxicity by MTT assay, lipid peroxidation by malondialdehyde (MDA) formation and super oxide dismutase (SOD), respectively. Results : The results showed that the extract scavenged DPPH radical in a dose-dependent manner by up to 74.87%. The cell viability of the extract was within 72~96% on Leydig cells and GC-2 cells at concentrations of 1, 5, 10, 50 and 100 ug/ml. The hydrogen peroxide-induced cytotoxicity of Leydig cells was protected to 72.09% by the extract at concentration of 100 ${\mu}g/ml$. The hydrogen peroxide-induced lipid peroxidation of MDA formation was decreased to 1.80 and 1.65 nmoles/mg protein by the extract at concentrations of 50 and 100 ${\mu}g/ml$. The extract at all concentrations, SOD activity was not significantly changed. Conclusions : In conclusion, the extract of Platycladi Semen has antioxidant effects on Leydig cells and protect male reproductive system against oxidative stress.

An Antioxidant Capacity Assay Using a Polyvinyl Alcohol-Based DPPH Pellet

  • To assay for antioxidant capacity of natural products considered important in producing human health benefits, a practical and economical method using pellet techniques was developed. A standard visualizing reagent, 1,1diphenyl-2-picryl-hydrazyl (DPPH), was mixed with a water-miscible polyvinyl alcohol (PVA), serving as a solid phase support for the DPPH reagent. A DPPH pellet was prepared by dropping a small volume of the DPPH solution onto PET film, and drying in an oven. The PVA-based DPPH pellet was dissolved into water, in which the water-miscible PVA plays as a non-ionic surfactant to help the DPPH reagent to be dissolved into the solvent. Using the DPPH assay, the antioxidant capacity of water-soluble extracts of black soybean, barley, green tea, and green gram was examined. Among the natural products tested, green tea showed the highest antioxidant capacity. This PVA-based DPPH antioxidant assay can be further applied in the natural food, raw plant material, and health product inspection field.

참죽나무 잎 추출물의 생리활성 (Physiological Activities of Extracts of Cedrela sinensis leaves)

  • 본 연구를 통하여 요즘 기능성 측면에서 관심을 끌고 있는 참죽나무 잎의 열수추출물과 유기용매 추출물을 이용하여 항산화능과 면연증강능을 확인하였다. 열수 추출물의 총 폴리페놀 함량은 46.5-59.6 mg/100 g으로 유기용맹 추출물보다 그 수치가 더 높았다. 열수 추출물과 유기용맹 추출물의 항산화 활성은 2,2-diphenyl-picryl-hydraryl (DPPH) radical 소거활성으로 6-33%로 기준물질보다 낮은 값을 나타내었다. 참죽나무 잎 추출물의 면역증강능을 확인하기 위하여 박테리아 감염에 대항하는 숙주방어체계에 필요한 NO 생성능을 확인하였다. 열수 추출물은 마크로파지 세포주인 RWA 264.7의 NO 생성을 증가시켰다. 그러나 유기용매추출물은 별다른 변화를 가져오기 않았다. 항염효과를 증명하기 위하여 인지질다당류 (LPS) 유발 NO생성에 미치는 영향을 조사하여, NO 생성을 억제하는 것을 증명하였다. 즉, 면역증강능의 확인 실험을 통하여 참죽나무 잎의 열수추출 분획에 대식세포의 활성을 증가시키는 성분이 포함되어 있고, 유기용매 추출 분획에서는 대식세포의 활성을 감소시키는 성분이 포함되어 있는 것으로 판단된다. 이상과 같은 결과를 미루어 보았을 때, 참죽이 한 가지 식물인데도 불구하고 유효 성분에 대한 추출 방법에 따라 상이한 면역 증강 및 항균, 항염 작용을 보이는 것으로 나타났다.

수삼, 연질 홍삼, 수액 홍삼의 항산화 효과 (Antioxidant Effects of Raw Ginseng, Soft Red Ginseng, and Red Ginseng Sap)

  • 고려 인삼(Panax ginseng)은 일반적으로 안전성이 우수하고 ginsenosides or panaxosides 같은 많은 생활성적인 물질을 함유하고 있다. 특히 고려 홍삼은 교감 신경계를 안정시키고 인지력을 향상시키고 개인의 의학적 효과를 높이는 데 도움을 줄 수 있다. 새로운 가공 기술로 부드러운 홍삼을 생산하였다. 본 연구는 새로운 가공 기술에 따라 생산된 연질 홍삼이 항산화 효과를 감소시키거나 개선했는지 여부를 조사하는 데 초점을 맞췄다. 연질 홍삼과 시판 홍삼액은 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) 소거활성능에 있어 유의한 차이를 나타내지 않았다(p<0.05). 연질 홍삼은 수삼과 시판 홍삼액에 비해 DPPH (22-Diphenyl-1-pichrylic)와 OH(hydroxyl radical) 소거활성능이 높게 나타났다. OH 소거활성능은 세 그룹(수삼, 연질 홍삼과 시판 홍삼액) 간 유의한 차이를 나타내었다(p<0.05). NO (nitric oxide) 소거활성능 역시 수삼, 연질 홍삼, 홍삼 수액생산품 사이에 유의한 차이가 있었다(p<0.05). 연질 홍삼에서 많은 칼슘 결정체가 전자현미경상에서 관찰되었다. 마그네슘과 칼륨은 연질 홍상과 경질 홍삼 사이에 유의한 차이가 없었다. 연한 홍삼의 추출물은 DPPH와 OH의 존재로 인해 서로 다른 활성산소를 효율적으로 소거할 수 있어 자유 라디칼로 유도된 질환에 도움이 될 수 있다.

Antioxidant Activity in Rice Cultivar, Wild Rice, and Barley

  • The antioxidant activities of methanol extracts of sixteen samples were tested using 1.1-diphenyl-2-picryl hydrazyl(DPPH) reactivity and TBARS substances assay in vitro. The methanol extracts of the rice brans from three wild rice -O. minuta, O. rufipogon, and O. barthii-were found to be the most effective in DPPH radical scavenging activity. The next effective ones were the rice brans of Heugjinjubyeo and leaves of Tapgolbori. When tested on lipid peroxidation using a lipid peroxidation generation system mediated by $H_2O$$_2$/Fe$^{2+}$ in rat liver homogenates, the brans and hull of wild rice (O. minuta, O. rufipogon, and O. barthii) and rice bran of Heugjinjubyeo exhibited protective activities against lipid peroxidation in the order of effectiveness.s.
