The Inhibitory Effect of Gooseberry on DNCB-induced Atopic Dermatitis in vivo and in vitro

  • Generally, berry fruits have various pharmacological activities such as anti-inflammation, anti-oxidation and anti-cancer effects. The effects of gooseberry, a berry fruits, on atopic dermatitis (AD) have not been widely examined. The aim of this present study is to investigate whether gooseberry modulates AD. We examined the pharmacological effects of gooseberry on 2, 4-dinitrochlorobenzene (DNCB)-induced AD symptoms in mice. To determine the anti-atopic mechanism of gooseberry, we investigated its effects on the production of inflammatory cytokines and activation of nuclear factor-${\kappa}B$ in PMA + ionophore -stimulated human mast cells (HMC-1). The results demonstrated that gooseberry attenuated AD clinical symptoms such as erythema, edema and dryness as well as histamine and IgE serum levels in DNCB-induced AD model mice. Additionally, gooseberry suppressed the expression of inflammatory cytokines and activation of nuclear factor-${\kappa}B$ in stimulated HMC-1. These findings demonstrate that gooseberry is potential agent for treating AD and allergic inflammation.

감맥대조탕이 DNCB로 유발된 생쥐의 아토피피부염에 미치는 영향 (Effects of Gammakdaejo-tang(GMD) on DNCB induced Atopic Dermatitis in Mice)

  • Objectives : Atopic dermatitis is a chronic inflammatory skin disease with frequent relapses. This study was to investigate the effects of Gammakdaejo-tang(GMD) in DNCB induced atopic dermatitis mice. Methods : The study was divided into five comparion groups. 2,4-dinitrochlorobenzene(DNCB) solution was applied to Nc/Nga mice to induce atopic dermatitis, followed by normal group, negative control group with distilled water, positive control group with Dexamethasione and GMD 200mg/kg or 400mg/kg. The control group was orally administered 200㎕ once daily for 4 weeks. Visual skin condition, Immunoglobulin E, Histamine, Cytokine, Immune cells, Tissue biomarkers were observed. Results : As a result of the dermatitis score evaluation, it was confirmed that the GMD-administered group improved symptoms compared to the negative control group. As a result of measuring IgE, the GMD-administered group significantly decreased compared to the negative control group. As a result of measuring Histamine, GMD group except 200mg/kg of GMD significantly decreased compared to negative control group. As a result of measuring cytokine, GMD 200mg/kg significantly reduced IL-1β, IL-6 and TNF-α compared to the negative control. 400mg/kg significantly reduced IL-1β, IL-4, IL-5, IL-6, IL-10, TNF-α and significantly increased IL-2, IFNγ. As a result of confirming the immune cells, all experimental groups showed no difference in basophil, GMD group significantly reduced monocyte and eosinophil compared to negative control group, and GMD 400mg/kg group significantly reduced white blood cell and neutrophil. And significantly increased lymphocytes. As a result of measuring the gene expression level, all GMD group significantly increased TGF-β1 compared with the negative control group, and filaggrin, VEGF and EGF were significantly increased in GMD 400mg/kg group. Epidermis, dermis thickness, and eosinophil infiltration were found to be decreased in all GMD groups compared with the negative control group. Conclusions : GMD is effective in atopic dermatitis by reducing imbalance of immune response of T cells (Th1 / Th2) and reducing skin tissue damage and inflammatory response.

한국에서의 피혁방미에 관한 연구 2 (Studies on the Antifungal Action of Leather in Korea (Part. 2))

  • It seems like that the characteristics and drug-resistances of fungi are respectively different in various circumstances. Scores of chemicals were applicated to the leather-fungi in this study. M-dinitrobenzene, 2, 4-dinitrochlorobenzene and phenyl mercuric acetate inhibited the growth of Aspergilli which were isolated from Korean-leather. The minimum fungicidal limits of p-nitrophenol, 8-hydroxyquinoline and sodium-pentachlorophenolate against the Korean-originated strains are different from that of other country. In the mass-screening of fungicides, artificial "Leather-extracts media" have been designed and used, and the media contributed to screening-tests. Fat and oils which are the materials of fat-liquoring in leather manufacture affects the drugresistance of the leather-fungi. It is found that the accelerating-method on malt-agar plate is effective to determinate the fungicidal action of chemicals in short time.hort time.

대청룡탕(大靑龍湯)이 DNCB로 유발된 생쥐의 아토피피부염에 미치는 영향 (Effects of Daecheonglyong-tang on Atopic Dermatitis Induced by DNCB in Mice)

  • Objectives : This study was designed to examine the effects of Daecheonglyong-tang(DCL) on atopic dermatitis induced by DNCB in mice Methods : The Nc/Nga mice were divided into 5 groups, and four groups excluding the normal group were applied by 2,4-dinitrochlorobenzene(DNCB), to cause AD and were orally administered with distilled water(negative control), dexamethasone(positive control), and DCL 200 or 400mg/kg once a day for 4 weeks respectively. The visual changes on skin, changes in skin tissue thickness and eosinophil infiltration were observed. IgE, Histamine, Cytokines, immune cells and the amount of gene expression of filaggrin, VEGF, $TGF-{\beta}1$, EGF were measured. Results : Dermatitis score showed a gross improvement on all DCL groups, similar to or better than positive control. All DCL groups showed no significant change in the basophils, while neutrophils and eosinophils decreased. In only DCL 400 mg/kg groups, white blood cells and mononuclear cells were decreased and lymphocytes were increased. In particular, neutrophils had similar or better effects than the positive control. In all DCL groups, IgE, Histamine, $IL-1{\beta}$, IL-4, IL-5, IL-6 and $TNF-{\alpha}$ were decreased and IL-2 was increased. In only DCL 400 mg/kg groups, IL-10 decreased and $IFN-{\gamma}$ increased. In particular, $IL-1{\beta}$ and $IFN-{\gamma}$ showed a similar rate of increase and decrease comparing positive control in DCL 400 mg/kg. $TGF-{\beta}$1 was increased in all DCL groups, filaggrin and VEGF were increased in only DCL 400 mg/kg groups. EGF did not make any changes. Epidermis, dermis thickness and eosinophil infiltration were also decreased in all DCL groups. Conclusions : By increasing Th1 cytokine and decreasing Th2 cytokine, DCL extracts appear to be effective in controlling immune response imbalances, anti-inflammatory and skin regeneration and are likely to be available as a treatment for AD.

Mycobacterium bovis 감염(感染)이 화학적(化學的)으로 유발(誘發)된 지연형(遲延型) 피부과민증(皮膚過敏症)에 미치는 영향(影響) (Chemically-Induced Delayed Cutaneous Hypersensitivity in Guinea Pigs and Rabbits Experimentally Infected with Mycobacterium bovis)

  • In order to evaluate the effects of the cell-mediated immunity of the animal in the chronic diseases the guinea pigs and rabbits were inoculated with Mycobacterium bovis. After 6 weeks these animals were sensitized and challenged with 2,4-dinitrochlorobenzene. The cutaneous reactions observed in these animal species were similar each other. Macroscopic and microscopic responses in the animal experimentally infected with M. bovis were markedly reduced compared to those in the control animals. The results indicated that the cell-mediated immunity of the animals was depressed by infection of M. bovis.

Anti-atopic dermatitis properties of Cordyceps militaris on TNFα/IFNγ-stimulated HaCaT cells and experimentally induced atopic dermatitis in mice

  • [Purpose] This study evaluated the anti-atopic dermatitis (AD) properties of Cordyceps militaris (CM) aqueous extract in keratinocytes in vitro and in vivo. We investigated the nutraceutical composition of the CM extract, including its protein, carbohydrate, and selected phytochemical content. [Methods] The expression of pathogenic cytokines in keratinocytes was assayed using an in vitro model. The CM extract downregulated extracellular signalregulated kinase 1/2 (ERK1/2) and p38 kinase expression in TNFα/IFNγ-stimulated HaCaT cells. We also established an in vivo AD model by repeatedly exposing the ears of mice to local Dermatophagoides farinae extract (DFE; house dust mite extract) and 2,4-dinitrochlorobenzene (DNCB). The epidermal and dermal ear thickness, mast cell infiltration, and serum immunoglobulin levels were measured following a 4-week oral administration of the CM extract. [Results] Histopathological examination showed reduced epidermal/dermal thickness and mast cell infiltration in mouse ears. The CM extract also suppressed serum immunoglobulin levels and gene expression of T helper (Th)1/Th2 cytokines in mouse ear tissue. [Conclusion] These results suggest that the CM extract may be useful for the treatment of AD-like skin lesions.

오가피열수추출물이 DNCB로 유도된 알레르기성 접촉피부염 흰쥐의 지질대사, 항산화능 및 표피회복능에 미치는 영향 (Effect of Acanthopanacis cortex Water Extract on Antioxidative Activity, Lipid Profile and Epidermal Thickness in DNCB-induced Allergic Contact Dermatitis Animal Model)

  • 본 실험은 오가피열수추출물이 알레르기성 접촉 피부염을 유발한 흰쥐의 체내 지질 조성, 항산화활성, serum Ig E 수준과 표피회복능에 미치는 영향을 조사하기 위해 행해진 것으로 1-chloro-2,4-dinitrochlorobenzene(DNCB: 2.5% DNCB 1 mL로 감작, 4일 경과 후 1% ONCB 1 mL로 유발) 처리로 알레르기성 접촉 피부염이 유발된 Sprague-Dawley 수컷 흰쥐에 오가피열수추출물 (100 mg/체중 kg/day)을 경구투여 한 다음 혈장과 간의 지질조성과 항산화활성, 혈중 Ig E 농도, 표피두께변화를 관찰하여 다음과 같은 결과를 얻었다. 혈장의 total cholesterol과 LDL-cholesterol은 정상군에 비해 ACFG군에서 유의적으로 감소하여(p<0.05) 오가피 열수추출물이 지질개선에 효과적인 것으로 나타났다. 혈장과 간의 지질과산화물 함량은 정상군에 비해 CG군과 ACFG군에서 지질과산화물 함량이 유의하게 높았으며(p<0.05), ACFG군이 CG군에 비해 과산화물 함량이 감소하는 경향을 보였다. 간의 SOD효소활성은 CG군에서 정상군에 비해 유의하게 증가하였으며(p<0.05), ACFG군에서 정상군의 수준으로 유의하게 감소하였다(p<0.05). Serum 중 Ig E 수준은 정상군에 비해 CG군에서 유의하게 증가하였으며(p<0.05), ACFG군에서 CG군에 비해 정상군의 수준으로 유의하게 감소하였다(p<0.05). 표피두께는 정상군에 비해 CG군에서 표피두께가 유의적으로 증가하였으며(p< 0.05), ACFG 군에서 CG군에 비해 유의하게 감소하였다(p<0.05). 혈장과 간의 지질과산화물 함량과 표피두께의 상관계수가 각각 r=0.644(p<0.01), r=0.516(p<0.05) 로 비교적 높은 정의 상관관계를 보였다. 이상의 결과로 미루어 볼 때 정상 흰쥐에 비하여 DNCB에 의해 유도된 접촉성 피부염을 가진 흰쥐의 혈장과 간의 지질과산화물 생성이 증가되며 이로 인해 표피의 두께가 증가하고, SOD 활성이 증가하는 것으로 나타났다. 오가피열수추출물의 섭취로 알레르기성 접촉피부염으로 유발된 흰쥐의 체내 지질과산화물 생성 억제, Ig E 농도감소, 표피두께 감소되는 효과가 나타났다.

DNCB로 유도된 NC/Nga 아토피피부염마우스에서 부신피질자극호르몬방출인자 활성에 따른 모발세포의 초기세포사멸 연구 (Acceleration of DNCB-induced Early-apoptosis via Activation of Corticotropin Releasing Factor in the Hair Root of NC/Nga Mice)

  • 피부에 가해지는 스트레스는 헤어조절 및 사이클에 직 간접적으로 중요한 영향을 미친다고 알려져 있다. 특히, 모근세포는 스트레스에 의한 부신피질관련호르몬과 세포손상 및 사멸과 밀접한 관련이 있다고 보고되고 있지만, 현재까지 실험적으로 입증된 사실은 매우 제한되어 있다. 보고에 의하면, 부신피질자극호르몬방출인자가 증가되면 모근세포의 마이토콘드리아 활성을 저해하여 초기단계의 세포사멸을 가져올 수 있다고 임상학적으로 보고된바가 있다. 특히 아토피 피부염으로 인한 스트레스는 부신피질자극호르몬방출인자와 부신피질관련 호르몬의 양을 증가시키며, 이는 모발의 outer epithelial sheath에 영향을 준다고 알려져 있으며, 이러한 스트레스의 변화는 마이토콘드리아 손상을 초래하여 초기단계세포손상을 준다고 한다. 따라서 본 연구는 아토피피부염스트레스가 피부의 모근세포에 주는 영향에 대하여 연구를 하였는데, 이에 대한 연구는 현재까지 전무한 실정이다. 우리는 NC/Nga 마우스에 2,4-dinitrochlorobenzene (DNCB)로 아토피피부염을 유발 후, 피부 스트레스 생성에 의한 초기단계 세포손상을 스트레스관련 인자, 부신피질자극호르몬방출인자 및 그 관련 인자, annexin V 및 마이토콘드리아 반응을 이용하여 연구하였다. 그 결과, 아토피피부염에 의한 스트레스는 체내의 부신피질 자극호르몬방출인자 및 관련인자의 활성을 증가시킬 뿐 아니라, 모근세포에 영향을 주어 초기단계세포사멸을 초래하는 것으로 나타났다. 이는 아토피피부염관련 헤어손상을 일으킨다는 중요한 연구결과를 보고하는 바이며, 부신피질자극호르몬 조절관련 의약품 및 화장품 등과 같은 보조적 요법이 필요함을 제안한다.

볏집 도초(稻草)이 DNCB로 유발된 생쥐의 알레르기성 접촉성 피부염에 미치는 영향 (Effects of Rice Straw (RS) on Allergic Contact Dermatitis (ACD) Induced by DNCB in Mice)

  • Objectives In the theory of Korean medicine, rice straw (RS) has been used effectively as treatments for dyspepsia, diarrhea, enteritis, inflammatory epigastric diseases and various dermatitis. However, the theory has not been studied intensively yet about anti-inflammatory effects for human. This study was to investigate effects of RS for a treatment of allergic contact dermatitis (ACD) induced by 2,4-dinitrochlorobezene (DNCB) in mice. Methods In this experiment, effects of RS were investigated on changes in body weights, dorsum skin thickness, clinical aspects on the dorsum skin, spleen and body weight among these four groups; normal group (NOR), control group (CON), RS spread group (RSS) and RS spread and administer group (RSS+Adm). In addition, the effects on proliferations of splenocytes were also investigated in vitro and in vivo study. Results RSS group and RSS+Adm group showed increasing in body weights, diminished dorsum skin thickness and treated dermatitis on dorsum skin. In RSS+Adm group, spleen weights were lowered significantly compared to CON group. Conclusions In conclusion, these data suggest that RS can decrease symptoms of ACD significantly, and it shows the anti-inflammatory and immunosuppressant effect as well. Therefore, RS can be useful to treat patients with ACD.

The Beneficial Effect of Avocado on Skin Inflammation in a Mouse Model of AD-like Skin Lesions

  • Avocado, superfood, contains a variety of essential nutrients and phytochemicals. The purpose of this study was to explore whether avocado could modulate skin inflammation in vivo. We elucidated the pharmacological effects of avocado on compound 48/80- or histamine-induced scratching behaviors and 2, 4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis (AD)-like skin lesions in mice. Additionally, we investigated the anti-inflammatory activity of avocado and its underlying mechanism including its effect on the expression levels of inflammatory-related genes and nuclear factor-κB (NF-κB) in DNCB-induced AD-like skin lesions. The findings of this study demonstrate that avocado attenuated AD-clinical symptoms including itching, eczematous, erythema and dryness and histamine levels in mice. Moreover, avocado suppressed both inflammatory cytokines expression as well as NF-κB and caspase-1 activation in AD-like skin lesions in mice. Taken together, these results demonstrate that avocado may be a potential candidate for treating skin inflammatory diseases like AD.
