Optimal culture conditions for mass production of rock polypody (Polypodium vulgare L.)

  • This study aimed to develop a suitable method for inducing the proliferation of prothallus and producing sporophytes of rock polypody (Polypodium vulgare L.). The prothalli used in all experiments were obtained from spore germination and sub-cultured for 8-week intervals. The most appropriate media for prothallus propagation were investigated by culturing 300 mg of prothallus in MS ($1/4{\times}$, $1/2{\times}$, $1{\times}$, and $2{\times}$ strength) medium and in Knop medium for 8 weeks. Cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $30{\pm}1.0{\mu}mol-m-2{\cdot}s-1$, and a photoperiod of 16/8 h (light/dark). Fresh weight of prothalli was 4.8 g on $1{\times}$ MS, 4.5 g on $1/2{\times}$ MS and 4.3 g on 1/4 MS medium. To select a suitable soil combination for sporophyte formation, 1.0 g of prothallus was ground with distilled water, spread in five combinations onto different soil substrates (decomposed granite, horticultural substrates, peat moss, and perlite), and then cultivated for 13 weeks. The sporophyte cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $43{\pm}2.0{\mu}mol-m-2{\cdot}s-1$, humidity of $84{\pm}1.4%$, and a photoperiod of 16/8 h (light/dark). The results showed that a mixture containing a 2:1 (v:v) ratio of horticultural substrate and perlite, increased sporophyte formation to 462.5 sporophytes per pot (7.5 cm2). The other soil substrates produced from 314.5 to 405.3 sporophytes per pot. Therefore, our results will provide conditions suitable for mass production of Polypodium vulgare L.

In Vitro Propagation of Persimmon(Diospyros kaki) by Embryo Culture (감나무의 배 배양법에 의한 기내 증식)

  • The embryos(6-8mm) isolated from seeds of Diospyros kaki were cultured on Murashlge-Skoog(MS), Woody Plant Medium(WPM), Campbell Durzen(CD), Lictvay's Medium(LM), Kao-Michaluk(KM), Nitsch, White, Heller, Wolter-Skoog(WS) media. The results showed that MS and WPM media were most suitable to the development of embryos into plantlets with length of $5.4{\pm}1.2 cm$ and 5-6 leaves. However, when LM and KM media were used, the addition of 1 to $2{\mu} moles/\ell GA_3$ was required for the germination of the embryos. Superoxide dismutase (SOD) activities, one of the changing factors in leaves according to physiological status displayed to be exceptionally significant in the leaves of plantlets germinated from seeds in potting sand soil contrary to those of cultured embryos specially around germination period.

Efficient transformation of Actinidia arguta by reducing the strength of basal salts in the medium to alleviate callus browning

  • An efficient transformation system for high-throughput functional genomic studies of kiwifruit has been developed to overcome the problem of necrosis in Actinidia arguta explants. The system uses Agrobacterium tumefaciens strain EHA105 harbouring the binary vector pART27-10 to inoculate leaf strips. The vector contains neomycin phosphotransferase (nptII) and ${\beta}$-glucuronidase (GUS) (uidA) genes. A range of light intensities and different strengths of Murashige and Skoog (MS) basal salt media was used to overcome the problem of browning and/or necrosis of explants and calli. Callus browning was significantly reduced, resulting in regenerated adventitious shoots when the MS basal salt concentration in the culture medium was reduced to half-strength at low light intensity ($3.4\;{\mu}mol\;m^{-2}\;s^{-1}$) conditions. Inoculated leaf strips produced putative transformed shoots of Actinidia arguta on half-MS basal salt medium supplemented with 3.0 $mg\;l^{-1}$ zeatin, 0.5 $mg\;l^{-1}$ 6-benzyladenine, 0.05 $mg\;l^{-1}$ naphthalene acetic acid, 150 $mg\;l^{-1}$ kanamycin and 300 $mg\;l^{-1}$ $Timentin^{(R)}$. All regenerated plantlets were deemed putativ transgenic by histochemical GUS assay and polymerase chain-reaction analysis.

  • Silene seulensis Nakai was used as traditional medicines in Korea, we collected plant from demilitarized zone (DMZ). S. seulensis was extracted with 30, 50, and 70% ethanol and partitioned successively with n-hexane, EtOAc, dichloromethane and BuOH. These extracts (30, 50 and 70% ethanol) were evaluated the cytotoxicity on B16F10 and Hacat cell lines. The LC-MS/MS data of each fractions (n-hexane, EtOAc, dichloromethane, and BuOH) were compared with MS library, combined with ultraviolet/visual (UV/Vis) and MS data for faster determine structure by database search results. This led to the identification of four compounds (1-4) from S. seulensis. These compounds was isolated first time from S. seulensis. Their chemical structures are elucidated by combinations of NMR and mass spectrometry techniques.

  • Ocean surface wind speed was estimated using TRMM (Tropical Rainfall Measurement Mission) TMI (TRMM Microwave/Imager) data. It is used the TRMM TMI brightness temperature and National Data Buoy Center's buoy winds speed dataset near North-America to estimate by the algorithm of the ocean surface wind speed retrieval over North America. Comparing with the buoy data by D-matrix equation, the result that RMSE, BIAS, and correlation coefficient are 2.19 $ms^{-1}$, 1.10 $ms^{-1}$, and 0.81, respectively. Therefore the estimated oceanic surface wind speed by TRMM TMI brightness temperature data show that available to ocean research over upper ocean.

Strength and Durability Properties of Concrete with Starch Admixture

  • This paper examines some properties of concrete, such as strength, oxygen permeability and sorptivity using starch [cassava (CA) and maize (MS)] as admixtures. Concrete cubes containing different percentages of the CA and MS by weight of cement (0, 0.5, 1.0, 1.5 and 2.0 %) were cast. Compressive strength tests were carried out after 3, 7, 14, 21, 28, 56, 90, 180, 270 and 365 days of curing. Oxygen permeability and sorptivity tests were carried out on another set of concrete specimens with the same percentages of starch at 7, 28, 90, 180, 270 and 365 days. Oxygen permeability and sorptivity tests data obtained were subjected to Kruskal-Wallis one-way analysis of variance by ranks. The strength increase after 1 year over the control for CA 0.5 and CA 1.0 are 2.7 and 3.8 % respectively, while MS 0.5 and MS 1.0 gave 1.5 % increase over control. These results showed a decrease in oxygen permeability and rates of sorptivity, with concretes containing starch as admixtures giving better performance than the control concretes.

Callus induction and high-efficiency plant regeneration via somatic embryogenesis in Papaver nudicaule L., an ornamental medicinal plant

  • We describe culture conditions for a high-efficiency in vitro regeneration system of Papaver nudicaule through somatic embryogenesis and secondary somatic embryogenesis. The embryogenic callus induction rate was highest when petiole explants were cultured on Murashige and Skoog (MS) medium containing 1.0 mg $1^{-1}$ ${\alpha}$-naphthaleneacetic acid (NAA) and 0.1 mg $1^{-1}$ 6-benzyladenine (BA) (36.7%). When transferred to plant growth regulator (PGR)-free medium, 430 somatic embryos formed asynchronously from 90 mg of embryogenic callus in each 100-ml flask. Early-stage somatic embryos were transferred to MS medium containing 1.0 mg $1^{-1}$ BA and 1.0 mg $1^{-1}$ NAA to germinate at high frequency (97.6%). One-third-strength MS medium with 1.0% sucrose and 1.0 mg $1^{-1}$ $GA_3$ had the highest frequency of plantlet conversion from somatic embryos (91.2%). Over 90% of regenerated plantlets were successfully acclimated in the greenhouse. Secondary somatic embryos were frequently induced directly when the excised hypocotyls of the primary somatic embryos were cultured on MS medium without PGRs. Sucrose concentration significantly affected the induction of secondary embryos. The highest induction rate (89.5) and number of secondary somatic embryos per explant (9.3) were obtained by 1% sucrose. Most secondary embryos (87.2-94.3%) developed into the cotyledonary stage on induction medium. All cotyledonary secondary embryos were converted into plantlets both in liquid and on semisolid 1/3-strength MS medium with 1.0% sucrose.

The Study on the Composition of Rehmanniae Radix Extracts by Supercritical Carbon Dioxide Extraction and by Hydrodistillation Extraction (생지황(生地黃) 증류 추출 약침액과 초임계 유체 추출물의 성분 연구(硏究))

  • Objectives : The purpose of this study was to investigate the composition of Rehmannia glutinosa's essential oils with Rehmanniae Radix herbal acupuncture Methods : I obtained the essential oils of Rehmannia Radix by hydrodistillation extraction method and supercritical fluid extraction(SFE) method, and then I analyzed those by GC/MS(gas chromatography/mass spectrum). Results : 1. With GC(gas chromatography) and GC/MS(gas chromatography/mass spectrum) analysis. I identified 9 compounds in the Rehmanniae Radix's essential oil obtained through the SFE method. The main compounds were as follows : Hexachloroethane(2.24%), N-Butyl-benzenesulfonamide(2.05%), hexadecanoic acid(1.93%), hexadecanoic acid, ethyl ester(3.49%), 9,12-Octadecadienoic acid(z,z)(2.70%), (9E)-9-Octadecenoic acid(6.14%), ethyl linoleate(4.43%), ethyl oleate(5.80%). 2. I failed to get Rehmanniae Radix's essential oil obtained through the hydrodistillation method. 3. With GC(gas chromatography) and GC/MS(gas chromatography/mass spectrum) analysis. I identified 4 compounds in the Rehmanniae Radix's essential oil obtained through the hydrodistillation method. The main compounds were as follows : Ethylbis(trimethylsilyl)amine(1.04%), 2-(Trimethylsiloxy)benzoic methyl ester(2.65%), Hexadecanoic acid trimethylsilyl ester(12.61%), octadecanoic acid, trimethylsilyl ester(6.28%). Conclusions : The substances by hydrodistillation method may not perfectly match with the substances by supercritical fluid extraction(SFE) method in essential oils extracted form Rehmanniae Radix. But, the main substances was assumed Hexadecanoic acid and octadecanoic acid.

Microbiological Elimination of 2,4-Dinitrotoluene and 276-Dinitro-toluene by an TNT-degrading Bacterium in Stirred Tank Reactors (교반탱크 반응조에서 TNT 분해세균에 의한 2,4-Dinitrotoluene/2,6-Dinitrotoluene의 미생물학적 제거)

  • An aerobic microbiological process was tested in 1.5 L stirred tank reactors for the treatment of dinitrotoluenes (DNTs)[e.g., 2.4-dinitrotoluene (2,4-DNT), 2,6-dinitrotoluene (2,6-DNT)] in the test culture of Stenotrophomonas maltophilia, which had previously characterized. Both 2,4-DNT and 2,6-DNT were completely degraded within 10 days and 14 days of incubation, respectively. Addition of the secondary carbon was essential to degrade DNTs, and little degradation was achieved in the absence of the secondary carbons. The effect of additional nitrogens on the degradation of DNTs was evaluated. Complete degradation of DNTs was observed in the absence of any additional nitrogens, whereas DNTs were partially degraded in the growth media with additional nitrogens. Both HPLC and GC-MS were used to detect and verify the residual DNTs and their intermediates. As the results, the HPLC and GC-MS chromatograms demonstrated that the both parent compounds, 2, 4-DNT and 2,6-DNT, and respective intermediates, 2-amino-4-nitrotoluene and 2-amino-6-nitrotoluene, could be successfully identified under the analytical conditions.

Design of a 6-bit 500MS/s CMOS A/D Converter with Comparator-Based Input Voltage Range Detection Circuit (비교기 기반 입력 전압범위 감지 회로를 이용한 6비트 500MS/s CMOS A/D 변환기 설계)

  • A low power 6-bit flash ADC that uses an input voltage range detection algorithm is described. An input voltage level detector circuit has been designed to overcome the disadvantages of the flash ADC which consume most of the dynamic power dissipation due to comparators array. In this work, four digital input voltage range detectors are employed and each input voltage range detector generates the specific clock signal only if the input voltage falls between two adjacent reference voltages applied to the detector. The specific clock signal generated by the detector is applied to turn the corresponding latched comparators on and the rest of the comparators off. This ADC consumes 68.82mW with a single power supply of 1.2V and achieves 4.9 effective number of bits for input frequency up to 1MHz at 500 MS/s. Therefore it results in 4.75pJ/step of Figure of Merit (FoM). The chip is fabricated in 0.13-um CMOS process.
