홍화유의 triglyceride 분자종에 관한 연구 (Molecular Species of Triglycerides in Safflower Oil)

  • Triglyceride 조성(組成)과 췌장리파제에 의한 PN별(別) 획분(劃分)의 ${\beta}$위치(位置)의 지방산조성(脂肪酸組成)의 결과로부터, 산정가능(算定可能)한 홍화유의 triglyceride 분자종은 23종류였다. 10% 이상을 차지하는 triglyceride분자종은 $C_{18:1}\;C_{18:2}\;C_{18:2}\;OLL;\;13.5%,\;C_{16:0}\;C_{18:2}\;C_{18:2}\;PLL;\;15.0%,\;C_{18:2}\;C_{18:2}\;C_{18:2}\;LLL;\;50.6%$로서 전체의 79.1%이었다.

Involvement of a Polyketide Synthetase ClPKS18 in the Regulation of Vegetative Growth, Melanin and Toxin Synthesis, and Virulence in Curvularia lunata

  • The clpks18 gene was first cloned and identified in Curvularia lunata. It contains 6571 base pairs (bp) and an 6276 bp open reading frame encoding 2091 amino acids. The ClPKS18 deletion mutant displayed an albino phenotype, and almost lost the ability to product 5-(hydroxymethyl) furan-2-carboxylate (M5HF2C) toxin, implying that clpks18 gene in C. lunata is not only involved in 1,8-dihydroxynaphthalene melanin synthesis, but also relatively associated with M5HF2C toxin biosynthesis of the pathogen. The pathogenicity assays revealed that ${\Delta}ClPKS18$ was impaired in colonizing the maize leaves, which corresponds to the finding that ClPKS18 controls the production of melanin and M5HF2C in C. lunata. Results indicate that ClPKS18 plays a vital role in regulating pathogenicity of in C. lunata.

  • ${\underline{O}},{\underline{O}}-Diethyl-{\underline{S}}-phenyl\;phosphorothiolate-^{18}O$와 관련화합물들을 합성하고 MCPBA로 산화시켰다. 각 반응을 $^{31}P$ NMR로 추적하였고 반응산물을 GC-MS로 분석하였다. ${\underline{O}},{\underline{O}}-Diethyl-{\underline{S}}-phenyl\;phosphorothiolate-^{18}O$은 메탄올에서 MCPBA에 의해 diethyl methyl phosphate로 전환되며 $^{18}O$를 포함하고 있어 이 산화반응은 Segall과 Casida가 제안하였던 반응기작을 따르고 있음이 증명되었다.

  • This study evaluated the effects of Dioscorea batatas and Gastrodia rhizoma on fatty acid composition of serum, liver and brain in rats which were fed control diet adding 5%, 10%, 15% Dioscorea batatas powder and 5%, 10%, 15% Gastrodia rhizoma powder for 3 weeks, respectively. In the other total fatty acid composition of serum lipids, the percentage of SFA was higher in the control group than in the other groups. The percentages 18:2 and 20:4 in serum lipids were significantly higher in the group 5, 6 and 7 than control group. In phospholipid fatty acid composition of serum lipids, the percentage of 18:2 was significantly higher in the group 5, 6 and 7 than in the other groups. 16:0, 18:0, 18:1, 18:2 and 20:4 were the main fatty acid in the total fatty acid and phospholipid fatty acid composition of liver lipids. 16:0, 18:0, 18:1, 18:2, 20:4 and 22:6 were the main fatty acid in the total fatty acid and phospholipid fatty acid composition of brain lipids.

  • Al2O3/(5~20vol%)LaAl11O18 composites in which the second phase was dispersed with a elongated grain shape were fabricated using Al2O3 and La2O3 composition by hot-pressing. In order investigate the in-fluence of LaAl11O18 on the toughening of LaAl11O18 on the toughening of Al2O3 matrix composites AE(acoustic emission) analysis was con-ducted together with an evaluation of fracture toughness using of SEPB technique. The degree of AE events occurred in composites were more than those in monolithic alumina. The occurrences of AE event increased with increasing the amount of LaAl11O18 phase in the Al2O3/LaAl11O18 composite is two times higher compared to monolithic alu-mina. The main toughening mechanism was attributed to the bridging of LaAl11O18 grains at tip of pro-pagating crack.

5·18 민주화운동의 지역성 극복을 위한 방안연구 -학술논문 저자와 학술지 편중분포를 중심으로 - (Plan Research to Overcome Regionality of 5·18 Democratization Movement: Focusing on biased distribution of academic paper writers and journals)

  • 5.18 광주 민중항쟁은 1993년 5월 김영삼 대통령에 의해 합법적인 민주화운동으로 인정받았고, 2011년 유네스코는 '5.18 광주민주항쟁'을 세계기록 유산으로 승인하였다. 이는 광주시민의 명예 회복에 전기를 마련하는 일이었다. 하지만 사건 발생 37년이 지난 지금도 이에 대한 평가는 전국적으로 통일되지 않았고, 어쩌면 광주만의 정신, 광주만의 자부심으로 기억되고 있는지 모르겠다. 이러한 상황이 지속되는 여러 요인들이 있겠지만, 본 연구자는 5.18의 진상규명과 시대정신에 맞는 재해석, 시민교육을 담당할 전문가들이 전라도와 서울에 집중적으로 분포되어 있다는 점이 가장 큰 요인으로 작용하고 있다고 생각한다. 게다가 5.18을 달리 평가하는 저널이 가해자 지역에 적을 두고 활동하고 있어 평가가 양분 된 채 통일되지 못하고 호남에 갇혀있게 된 것이라 생각한다. 본 연구는 5.18의 의미가 전국적으로 통일되지 못하고 호남에 갇혀있게 된 요인으로 5.18관련자들만의 "당사자주의"의 한계에 있다 판단하고 있다. 특히 이 사건을 객관적인 시각으로 분석하고 해명해야 할 연구자들이 연고지역에 편중분포(광주 전라도>서울>경남)되어 있어 5.18의 의미가 통일되지 못하고 있는 것이 아닌지 그 개연성에 주목하게 되었다. 이 연구에서는 학술연구논문과 저널 저자 및 출판 소재지가 양분되어 있고, 편중분포 되어 있음을 밝히고, 연구자들의 편중분포가 지역성 극복에 걸림돌이 되고 있다는 개연성을 밝히고자 한다.

둥굴레속 식물의 18S rDNA 염기서열의 특성 (Characterization of 18S rDNA in Polygonatum spp. Collections)

  • 둥굴레 유전자원의 유연관계를 위한 기초 자료를 얻고자 둥굴레속 식물 수집종 10종에서 18S ribosomal RNA를 암호화하는 18S rDNA 영역의 염기서열을 결정하고 그 특성을 분석한 결과를 요약하면 다음과 같다. 둥굴레속 10종의 18S rDNA 영역 전체의 길이는 $913{\sim}914bp$로 비슷하였으나, 총 8개 지점에서 염기의 치환 및 결실에 의한 변이가 발생하였다. 전위는 $T{\rightarrow}C$전위가 4개 지점에서 발생하였고, $A{\rightarrow}G$ 전위가 1개 지점에서 발생하였으며, 전좌는 $C{\rightarrow}A$ 전좌가 1개 지점에서 발생하여 전위가 전좌보다 5배 만큼 발생하였다. 결실은 2개 지점에서 발생하였다. 18S rDNA의 염기의 조성은 adenine $23.09{\sim}23.33%$, guanine $23.33{\sim}23.52%$, thymine $25.60{\sim}25.85%$, cytosine $27.38{\sim}27.79%$로 pyrimidine계가 purine계보다 많았다. 18S rDNA의 A + T 함량은 $48.80{\sim}49.18%$로 평균 48.99%였고, G+C 함량은 $50.82{\sim}51.20%$로 평균51.01%였다. 다중 정렬에 의해 염기서열을 비교한 결과 $99.7{\sim}100%$ 일치하여 종간에 차이가 적었다.

솔과 식물 종실유의 지방산 조성 (Fatty Acid Compositions of Some Seed Oils from the Pinaceae Family)

  • Levels of total lipids in the seeds of three species of the Pinaceae family were determined and their fatty acid compositions were also analyzed by a gas-chromatograph equipped with a capillary column coated with Carbowax 20M. The results are summarized as follows: Lipid contents of the seeds amounted to 56.9% in P. koraiensis, 29.9% in P. thunbergii, and 21.2% in P. rigida. In all lipids 19${\sim}$20 fatty acid were detected and, surprisingly, fatty acids having ${\Delta}^5$-non-methylene interrupted conjugate double bond such as ${\Delta}^{5, 9}-C_{18:2},{\Delta}^{5, 9, 12}-C_{18:3}\;and\;{\Delta}^{5, 11, 14}-C_{20:3}$ occurred in appreciable amounts. In the lipids of P. koraiensis, the main component was $C_{18:2}{\omega}_6(45.0%)$, followed by $C_18:1{\omega} _9(26.9%)$ and ${\Delta}^{5, 9, 12}-C_{18:3}(14.6%)$, and then ${\Delta}^{5, 9}-C_{18:2}(2.2%)$ and ${\Delta}^{5, 11, 14}-C_{20:3}$ were also present. Levels of saturated fatty acid such as $C_{16:0}\;and\;C_{18:0}$ were as low as 7.5%. The seed oil of P. thunbergii predominantly comprised $C_{18:2}{\omega}_6(45.2%)$, and was then occupied by equal amounts ${\Delta}^{5, 9, 12}-C_{18:3}(18.1%)$ and $C_{18:1}{\omega}_9(18.1%)$. Its ${\Delta}^5, 11, 14}-C_{20:3}(5.8%)$ level was the highest in the samples tested. ${\Delta}^{5, 9}-C_{18:2}(2.8%)$ was also detected with other minor components. In the oils from the seeds of P. rigida, $C_{18:2}{\omega}_6$ was present as a main component, accompanied by $C_{18:1}{\omega}_9(21.6%)$ and ${\Delta}^{5, 9, 12}-C_{18:3}(20.3%)$. The latter showed higher level than in any other samples. A minor component corresponding to ${\Delta}^{5, 9, 12, 15}-C_{18:4}$(not confirmed by GC-Mass) occurred in P. thunbergii and P. rigida.

Developmental Relationship of Unsaturated Fatty Acid Composition and Stearoyl-CoA Desaturase mRNA Level in Hanwoo Steers' Muscle

  • This study was conducted to investigate the developmental relationship between fatty acid composition in different lipid fractions and stearoyl-CoA desaturase (SCD) gene expression in steer muscles during growth. Twenty Hanwoo steers were used at 6, 12, 18, 24 and 30 months of age. Fatty acid composition and SCD mRNA level were analyzed. In the total lipid fraction, developmental profiles of C18:1, as the product of SCD enzyme, and SCD mRNA level were significantly increased between 6 months and 12 months of age. During this period, the percentage of C18:1 increased from 31.9% to 49.5% in the total lipid. The increased C18:1 level was maintained until 30 months of age within the range of 44.8- 49.9%. In contrast, the C18:0 composition decreased with age and this decrease was compensated by the increase of the C18:1. However, the sum of C18:0 and C18:1 was changed before and after 12-month old by a 20% increase. Unlike the C18 fatty acids, the C16 fatty acids such as C16:0 and C16:1 did not show a consistent change with age in steers' muscle. On the other hand, C18:2 proportion as a major polyunsaturated fatty acid in muscle was significantly reduced from 21.1% at 6 months of age to 4.4% at 12-months old and then this reduced level was maintained until 30 months within the range of 7.4-11.4%. As in the C18:1 composition during early stages, a 2-fold significant increase was observed in the $\Delta^9$-desaturase index of C18 fatty acid as a measure of SCD activity, but not in that of C16 fatty acid. Also, the steady-state level of SCD mRNA reached a peak at 12 months of age. Thus, the positive relationship between the C18:1 composition and the $\Delta^9$-desaturase (SCD enzyme) index of C18 fatty acid or SCD mRNA level was demonstrated during growth, but the negative relationship between the C18:2 composition and the above three indices was demonstrated at the same time, indicating that the sharp induction of SCD mRNA may be closely related to the dramatic reduction of C18:2, which is known as a suppressor of SCD gene expression during growth.

유지의 가열 및 저장에 따른 Trans지방산 생성에 관한 연구 -제2보 ; Trans지방산 함량 및 조성 변화를 중심으로- (The Formation of Trans Fatty Acids with Heat Treatment and Storage of Fats and Oils (II) -The contents and the kinds of Trans fatty acids-)

  • In this study, the cis to trans isomerization of unsaturated fatty acids were investigated about Soybean oil (SBO), Corn germ oil (CGO), Cottonseed oil (CSO), Margarine (MG), Shortening (ST) incubated at $40{\pm}2^{\circ}C$ for 35 days and heated at $185{\pm}2^{\circ}C$ for 24 hours. The contents and kinds of trans fatty acids in each sample were determined by GLC after seperating by HPLC. The results were obtained as follows; 1. When samples were incubated at $40{\pm}2^{\circ}C$ for 35 days, the contents of total trans fatty acids were increased from $0.5{\sim}12.3%$ to $0.8{\sim}20.5%$. The kinds of trans fatty acids found were $t-C_{18:1},\;t,c-C_{18:2},\;t,t-C_{18:2},\;t,c,c-C_{18:3}$ in $SBO,\;t-C_{18:1},\;t,c-C_{18:2}$ in $CGO,\;t-C_{16:1},\;t-C_{18:1},\;t,c-C_{18:2},\;t,t-C_{18:2}$ in CSO. Processed oil such as MG and ST showed more complicated composition of trans fatty acids than SBO, CGO and CSO. $t-C_{18:1},\;t,c-C_{18:2},\;t,t-C_{18:2},\;t,c,t-C_{18:3},\;t-C_{20:1}$ were detected in ST. 2. In the case of heating at $185{\pm}2^{\circ}C$ for 24 hours, the contents of total trans fatty acid were $1.6{\sim}37%$. 2% in all samples. Heating made more remarkable isomerization than incubation $(40{\pm}2^{\circ}C)$. Specially, $c,c,t-C_{18:3}$ in $SBO,\;t,t,t-C_{18:3}$ in $MG,\;t,c,t-C_{18:3},\;t,t,t-C_{18:3}$ in ST were detected. 3. The total contents of trans fatty acids of processed oil were higher than vegetable oils. During incubation, trans fatty acids increased in the order of MG>ST>CGO>SBO>CSO, and during heat treatment, MG>ST>SBO>CGO>CSO.
