하수슬러지와 토양 중 다핵방향족 탄화수소의 정량 (Analysis for 16 Polynuclear Aromatic Hydrocarbons (PAH) in Sewage Sludge and Soil)

  • 하수슬러지와 토양시료 중 16가지의 다핵방향족 탄화수소(PAH)를 KOH와 $CHE_3OH$에 녹인 용액으로 Soxhlet 추출후 내부표준물질로 2-ethylantracene을 이용하여 GC/MS-SIM 방법으로 분석하였다. 12종의 PAH가 하수슬러지에서 검출되었고 공업지역 하수슬러지 중 가장 특징적인 화합물은 naphthalene($0.78{\mu}g/g$), 도시 하수슬러지에서는 pyrene($0.26{\mu}g/g$)이었으며, 총 PAH는 각각 $1.74{\mu}g/g$$1.19{\mu}g/g$이었다. 논 토양과 밭 토양에서의 PAH는 phenanthrene을 포함한 8종이 검출되었으며 그 농도범위는 $0.01-0.04{\mu}g/g$이었고, 총 PAH의 양은 각각 $0.19{\mu}g/g$$0.22{\mu}g/g$이었다. 논 토양 중의 총 PAH함량에 비하여 공업지역과 도시지역 하수슬러지 중의 총 PAH양은 각각 9.2배와 6.3배가 높게 나타났다.

  • The results of individual PAH source profiles that can be applied to receptor model are as follows. The sum of 16 PAH concentrations was 391.41 ng/S㎥ in a tunnel. Phenanthrene was the most abundant compound among 16 PAH, and then pyrene, fluoranthene, anthracene, and naphthalene can be seen in elevated contents. 11,056.61 ng/S㎥ of 16 PAH concentrations in BC oil boiler was two times higher than 6,582.57 ng/S㎥) of those in LNG boiler. Naphthalene was the most abundant compound in both facilities. Phenanthrene, anthracene, and acenaphthylene were the second dominant compound group in order from both facilities. BC oil boiler had relatively high concentration of pyrene compared to LNG boiler that had high concentration of fluorene and did not detect pyrene. The sum of 16 PAH concentrations emitted from MSW incinerators after APCD (air pollution control device) was three times higher than those from MSW incinerators before APCD. However, the concentrations of more than 4-ring PAH compounds (e.g., benzo (a)anthracene) before APCD were higher than those after APCD. This fact implies that PAHs generated by combustion process are eliminated in APCD and they are continuously produced in stack or atmosphere by PAHs precursors.

고양이의 신장에서 Ethacrynic Acid가 PAH 분비에 미치는 영향 (Effect of Ethacrynic Acid on Renal Tubular Secretion of PAH in Anesthetized Cat)

  • The effect of ethacrynic acid (EA) on the renal secretion of PAH was examined in cat kidney. $C_{PAH}$ and $T_{PAH}$ were measured before and after infusion of EA $(0.5{\sim}50mg/kg)$ through the femoral vein. The following results were obtained: 1) In the dosage range of 0.5 to 25 mg/kg, EA increased the urine flow, and sodium and potassium excretion in dose-dependent manner, but the glomelular filtration rate was decreased as the dosage of EA was increased. 2) $C_{PAH}$ and $T_{PAH}$ were decreased by EA in the dosage range of 3 to 25 mg/kg and 1 to 50 mg/kg, respectively, in dose·dependent manner with the dosage to cause 50% inhibition of about 5 mg/kg. 3) With dosage of 0.5mg/kg, EA appeared to exert a great effect on diuretic response without the influence on $T_{PAH}$. At 10min after infusion of EA, a potent diuretic effect appeared, while $T_{PAH}$ did not show a significant change. These results suggest that the action mechanism of EA on tubular secretion of PAH may be different from that on natriuresis. 4) With dosage of 5 mg/kg, EA did not inhibit the Na-K-ATPase activity in microsomal fractions from both cortex and medulla. 5) The double reciprocal plot ($l/T_{PAH}$ versus $l/P_{PAH}$) suggested that EA inhibited the P AH secretion by a competitive pattern. However, probenecid, a prototypic inhibitor of the organic acid pump, had no influence on both the inhibitory effect of $T_{PAH}$ and the natriuretic effect by EA. These results suggest that in vivo EA altered tubular secretion of P AH through interactions with receptors that are not identical with the Na-K-ATPase.

Assessing the Formation of Polycyclic Aromatic Hydrocarbons in Grilled Beef Steak and Beef Patty with Different Charcoals by the Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) Method with Gas Chromatography-Mass Spectrometry

  • This study investigated the effects of different charcoals on the occurrence of 16 polycyclic aromatic hydrocarbons (PAHs) in grilled beef steaks and beef patties. Seven different charcoals were used as follows: from oak wood (C1), from orange wood (C2), from Valonia oak wood (C3), from Marabu wood (C4), extruded charcoal from beech wood (C5), from coconut shells (C6), and from hazelnut shells (C7). The grilling times for each charcoal type were 6 min for the beef patties and 7 min for the beef steaks, until the internal temperature reached at least 74℃. The total concentration of 16 PAHs (PAH16) in beef steaks grilled with C1 (35.75 ㎍/kg) and C7 (36.39 ㎍/kg) was higher than that of C3 (23.80 ㎍/kg) and C6 (24.48 ㎍/kg; p<0.05). The highest amounts of PAH16 (216.40 ㎍/kg) were determined in the beef patty samples grilled using C5 (p<0.05). The summation of benzo[a]anthracene, chrysene, benzo[b]fluoranthene and benzo[a]pyrene, referred to as PAH4, was not detected in any of the beef steaks, whereas it was determined in the beef patties grilled with C2 (7.72 ㎍/kg) and C5 (22.95 ㎍/kg; p<0.05). The PAH16 concentrations of the beef patty samples in each charcoal group were significantly higher compared to the beef steaks (p<0.05). To avoid the formation of high PAH levels, the use of extruded charcoal and hazelnut shell charcoal should therefore be avoided when charcoal grilling beef steaks and beef patties, and low-fat meat products should be preferred.

The Presence of Polycyclic Aromatic Hydrocarbons (PAHs) in Grilled Beef, Chicken and Fish by Considering Dietary Exposure and Risk Assessment

  • Polycyclic aromatic hydrocarbons (PAHs) are dangerous chemical compounds that can be formed by cooking foods at high temperatures. The aim of this study is to determine the level of contamination of PAH compounds with high performance liquid chromatography (HPLC) on heat treated meat samples and the consumption of PAH compounds in meat samples, as well as the dietary exposure status and possible health risk estimation. In five different heat treated meat samples (meat doner, chicken doner, meatballs, grilled chicken, and fish), the total PAH (Σ16PAH) contamination level was 6.08, 4.42, 4.45, 4.91, and 7.26 ㎍/kg, respectively. Benzo[a]pyrene (BaP) in meatballs and grilled fish samples had a level 0.70 and 0.73 ㎍/kg. All of the samples analyzed were found to be below the EU permitted limit (5 ㎍/kg) in terms of BaP. Estimates of daily intake (EDI) for a total of 16PAH in heat treated meat doner, chicken doner, meatballs, grilled chicken and fish samples were 3.41, 3.71, 2.49, 4.12, and 1.77 ng/kg bw/day, respectively. In this study, the average margin of exposure (MOE) value calculated was found in the range of 179.487 and 425.000 for BaP and PAH4. This study is the first study to provide important information in terms of evaluating the possible health risk that PAH compounds can create in people's diets due to heat treatment of meat and meat products in Sivas, Turkey.

Atmospheric Concentrations of PAHs in the Vapor and Particulate Phases in Chongju

  • Four intensive seasonal sampling campaigns between October 1998 and October 1999 were undertaken at an urban site of Chongju, in which polyurethane foam (PUF) sampler was used to collect particulate- and vapor-phase polycyclic aromatic hydrocarbons (PAHs). The contribution to total (particulate+vapor) PAH concentration by the vapor phase component exceeded the particulate phase contribution by factor of ${\sim}2.6$. Summed concentrations of phenanthrene (30.9%), pyrene (16.6%), naphthalene (11.3%) and fluoranthene (11.0%) account for significant amounts of the vapor-phase, while chrysene (12.5%), benzo[b]fluoranthene (11.6%), indeno[123-cd]pyrene (9.9%), benzo[ghi]perylene (9.5%), benzo[k]fluoranthene (9.4%), pyrene (8.9%), and benzo[a]pyrene (8.3%) are found to be the most common PAH compounds in the particulate phase. The results from application of principal component analysis to particulate-phase PAH data demonstrate that a combination of PAH and $PM_{2.5}$ inorganic data is a more powerful tracer of emission sources than PAH species data alone. Particulate-phase PAH species were found to be associated predominantly with emissions from diesel engine vehicles and incineration.

환경대기중 다환방향족탄화수소의 포집과 분석방법에 관한 연구 (Evaluation of Sampling and Analytical Methods for the Determination of Polycyclic Aromatic Hydrocarbons in the Ambient Atmosphere)

  • 본 연구에서는 대기중 저농도 수준의 다환방향족탄화수소(PAH) 농도를 측정하기 위하여 이들의 포집과 분석방법을 평가하였다. PAH의 포집매체로서 기체상은 Tenax 흡착제를, 입자상은 PTEE 및 유리섬유필터를 이용하였으며 양자 모두 저유량샘플러에 의해 포집되었다. 필터 및 흡착제에 함유된 유기성분은 Soxhlet 장치로 추출하였으며 신속하고도 간편한 Silica 미니칼럼을 이용하여 PAH 함유성분을 분리하였다. PAH 성분은 역상 HPLC로 분리되어 형광 및 UV 검출기로 검지하였다. 분석은 환경보건학적 중요성을 갖는 18개의 PAH에 대하여 행하여졌다. 분석방법의 타당성을 검토하기 위하여 추출능(시료의 회수율), 시료의 전처리단계, HPLC 분리능과 재현성 및 검출한계치 등이 평가되었으며 이울러 서로 다른 포집매체의 이용에 따른 회수율의 영향도 비교 검토되었다. 본 연구에서 채택된 포집 및 분석방법을 실제 환경시료에 적용하여 가스상 및 입자상 PAH 분석을 위한 실례를 제시하였다.

상용 휘발유로부터 분리한 다환 방향족 탄화수소(PAH) 분해 세균의 특성 (Characterization of PAH (Polycyclic Aromatic Hydrocarbon)-Degrading Bacteria Isolated from Commercial Gasoline)

  • BACKGROUND: Recent studies have described the importance of bacteria that can degrade polycyclic aromatic hydrocarbons (PAHs). Here we screened bacterial isolates from commercial gasoline for PAH degraders and characterized their ability to degrade PAHs, lipids and proteins as well as their enantioselective epoxide hydrolase activity, salt tolerance, and seawater survival. METHODS AND RESULTS: One hundred two bacteria isolates from commercial gasoline were screened for PAH degraders by adding selected PAHs on to the surface of agar plates by the sublimation method. A clear zone was found only around the colonies of PAH degraders, which accounted for 13 isolates. These were identified as belonging to Bacillus sp., Brevibacterium sp., Micrococcus sp., Corynebacterium sp., Arthrobacter sp., and Gordonia sp. based on 16S rRNA sequences. Six isolates belonging to Corynebacterium sp., 3 of Micrococcus sp., Arthrobacter sp. S49, and Gordonia sp. H37 were lipid degraders. Arthrobacter sp. S49 was the only isolate showing high proteolytic activity. Among the PAH-degrading bacteria, Arthrobacter sp. S49, Brevibacterium sp. S47, Corynebacterium sp. SK20, and Gordonia sp. H37 showed enantioselective epoxide hydrolase activity with biocatalytic resolution of racemic styrene oxide. Among these, highest enantioselective hydrolysis activity was seen in Gordonia sp. H37. An intrinsic resistance to kanamycin was observed in most of the isolates and Corynebacterium sp. SK20 showed resistance to additional antibiotics such as tetracycline, ampicillin, and penicillin. CONCLUSION: Of the 13 PAH-degraders isolated from commercial gasoline, Arthrobacter sp. S49 showed the highest lipid and protein degrading activity along with high active epoxide hydrolase activity, which was the highest in Gordonia sp. H37. Our results suggest that bacteria from commercial gasoline may have the potential to degrade PAHs, lipids, and proteins, and may possess enantioselective epoxide hydrolase activity, high salt tolerance, and growth potential in seawater.

코크스오븐 작업자들의 코크스오븐배출물 및 다핵방향족탄화수소 노출에 관한 연구 (A Study on exposure assessment of Coke-Oven Workers to Coke Oven Emissions and Polynuclear Aromatic Hydrocarbons)

  • Coke-oven workers are regularly exposed to coke oven emissions (COE), which consist primarily of polynuclear aromatic hydrocarbons (PAH) and volatile organic components. This study assessed the exposure of coke-oven workers to COE and PAH. The results of this study are as follows. 1. Among the 136 samples on coke-oven workers 33.1% exceeded the occupational exposure limit to the Coal Tar Pitch Volatiles ($0.2mg/m^3$) established by the Korean Ministry of Labor and American Conference of Governmental Industrial Hygienists(ACGIH). 2. The coke-oven workers were classified into twelve job categories and there was statistically significant difference among the airborne concentrations of COE and PAH in twelve job categories(p<0.001). 3. There was no significant difference of the airborne concentrations of COE between NIOSH method and OSHA method applied. 4. There was a significant correlation between the airborne concentrations of total PAH and COE(r=0.733, p<0.01). 5. The percentage of naphthalene in total PAH was 55.9% and the highest among those of 16 PAHs. There was a significant correlation between the airborne concentrations of naphthalene and total PAH(r=0.90, p<0.01) and this study recommends naphthalene as the index component of PAH.

순천만 갈대근권 토양으로부터 얻은 PAH 분해세균의 특성 분석 (Characterization of PAH-Degrading Bacteria from Soils of Reed Rhizosphere in Sunchon Bay Using PAH Consortia)

  • 본 연구는 농업과 어업, 그리고 생태체험과 같은 인간들의 활동으로 인하여 상당히 영향을 받는 갯벌환경 중의 하나인 순천만을 모델장소로 갈대의 환경정화 기능에 있어 근권에 분포하는 미생물의 역할에 대한 기초 자료를 얻고자 수행하였다. 우선, 순천만의 갈대근권 토양을 시료로하고 anthracene, naphthalene, phenanthrene, pyrene 등이 첨가된 다환성 방향족 화합물(polycyclic aromatic hydrocarbons; PAH)을 탄소원 및 에너지원으로 하는 농화 배양을 통하여 두 개의 consortium을 획득하였다. 두 consortium으로부터 순수 분리된 우수한 PAH분해능을 갖는 4개의 균주(SCB1, SCB2, SCB6,그리고 SCB7)를 형태 및 생리학적 특성과 16S rRNA유전자서열을 기초로 분석한 결과 각 균주는 $99{\%}$ 이상의 신뢰도로 Burkholderia sp., Aicaligenes sp., Achromobacter sp., and Pseudomonas sp.로 동정되었다. 주목할 만한 점은 Burkholderia sp. SCB1과 Alcaligenes sp. SCB2는 naphthalene이나 phenanthrene보다 훨씬 안정되어 있는 구조의 anthracene이나 pyrene에서 더 빠른 성장률과 기질 분해율을 나타내는 것으로 밝혀졌다. 반면,Achromobacter sp. SCB6와 Pseudomonas sp. SCB7은 pyrene을 제외한 다른 시험기질에 대하여 유사한 성장 및 분해패턴을 나타내었다. 이러한 결과는 주요한 염습지 식물중의 하나인 갈대의 근권에서 살아가는 이들 PAH 분해 균주들이 PAH와 같은 물질로 오염된 근권 환경의 정화작용에 중요한 역할을 할 수 있음을 제시해 주었다.
