Estimation of Nitrate Sources in Cheju Island Groundwater using $\delta$$^{15}$ N ($\delta$$^{15}$ N을 이용한 제주도 지하수 중의 질산염 오염원 조사)

  • 18 boreholes with nitrate contaminated were selected. Samples were collected 4 times between both 1995 and 1996. Stable nitrogen isotope ratio for them all was measured and the contribution to contamination from several sources like fertilizer, sewage, cropland. and landfill was analysed. Nitrogen source for 11 sampling sites of T-3, L-1. O-1∼O-4, F-2∼F-5, and G-2 considered to come from chemical fertilizer and its contribution was around 60% or more. T-4. T-5 were located downward the downtown, which were influenced bydomestic sewage and its contribution were 70.7% and 54.7%. Nitrate concentration of G-2 was 17.7 mg/L, among which 60.7% was estimated to come from landfill leachate. T-1 and T-2 were located in the small village, in which 42.2 and 43.4% of nitrogen was to come from domestic sewage but 52.8% and 56.0% were from fertilizer sprayed in the cropland. L-2 was near livestock by which it was estimated to be influenced, in which 59.9% of nitrogen was from cropland. F-1 was in the cropland, by which 50.0% was influenced and 49.5% was estimated from organic matter of animals.

Effects of Charcoal Application on Ammonia Emission and Nitrogen Use Efficiency of Pig Slurry in the Vegetative Growth of Maize (Zea Mays L.)

  • The objective of this study was to prove the effect of pig slurry application with charcoal on nitrogen use efficiency (NUE), feed value and ammonia (NH3) emission from maize forage. The four treatments were applied: 1) non-pig slurry (only water as a control), 2) only pig slurry application (PS), 3) pig slurry application with large particle charcoal (LC), 4) pig slurry application with small particle charcoal (SC). The pig slurry was applied at a rate of 150 kg N ha-1, and the charcoal was applied at a rate of 300 kg ha-1 regardless of the size. To determine the feed value of maize, crude protein, dry matter intake, digestible dry matter, total digestible nutrient, and relative feed value were investigated. All feed value was increased by charcoal treatment compared to water and PS treatment. Also, the NUE for plant N was significantly higher in charcoal treatments (LC and SC) compared to PS treatment. On the other hand, there is no significant difference for feed value and NUE between LC and SC. The NH3 emission was significantly reduced 15.2% and 27.9% by LC and SC, respectively, compared to PS. Especially, SC significantly decreased NH3 emission by 15% compared to LC. The present study clearly showed that charcoal application exhibited positive potential in nitrogen use efficiency, feed value and reducing N losses through NH3 emission.

  • In this study, the impact of Nitrogen implantation process on deep-level defects and lifetime in 4H-SiC Epi surfaces was comparatively analyzed. Deep Level Transient Spectroscopy (DLTS) and Time Resolved Photoluminescence (TR-PL) were employed to measure deep-level defects and carrier lifetime. As-grown Schottky Barrier Diodes (SBDs) exhibited energy levels at 0.16 eV, 0.67 eV, and 1.54 eV, while for implantation SBD, defects at 0.15 eV were observed. This indicates a reduction in defects associated with energy levels Z1/2 and EH6/7, known as lifetime killers, as impurities from nitrogen implantation replace titanium and carbon vacancies.

  • Seasonal changes of symbiotic nitrogen-fixation activity and growth characteristics of four different natural populations of kummerowia striata were quantitatively analyzed during the growing period. the nitrogen-fixation activity of root nodules attained the maximum rates of 148, 132, 102 and 100$\muM\;C_2H_4\;\cdot\;g\;fw\;nodule^{-1}\;\cdot\;hr^{-1}$, respectively for sunny, multibranched, shade and unibranched populations at the optimum growth conditions. and the seasonal changes showed fluctuations by environmental conditions such as light, temperature, nutrient contents, water stress and plant ages, etc. The multibranched plant showed the greater amount of leaf and root nodule biomass, and the higher nitrogenase activity than the unibranched plant. the optimum conditions of leaf chloropht11 and water content of each organ indicated the active growth and the maximum fresh biomass of 4 different populations were 1.92, 1.85, 0.97 and 0.56 g $fw\cdotplant^{-1}$ for shade multibranched, sunny and unibranched populations, respectively.

  • The effects of various nitrogen soruces on the expression of nif gene were investigated using nif-lac fusants of Klebsiella pneumoniae. K. pneumoniae UK 2979 was infected with Mudl lysate prepared by heat induction of K. pneumoniae UK 4482. About 80 nif-lac fusants were greatly repressed. Amino acids, such as serine, glutamine and asparagine, were found to support the growth of K. pneumoniae M5al quite well, and showed a repressive effect on .betha.-galactosidase activities of nif-lac fusants LX-9 and LX-22 in NFHM. Glutamic acid, histidine and arginine rendered poor growth but high activities of .betha.-galactosidase. Good cell growth and high enzyme activity were observed when complex nitrogen sources, such as casitone, proteose pepone, were employed. .betha.-Galactosidase activities of LX-9 and LX-22 in nitrogen free minimal medium increased sharply within first 4 hours.

  • The tribological behaviour of WS$_2$ solid lubricant was investigated using a ball-on-disk type tester. The WS$_2$ powder was spray-coated at room temperature and the effects of specimen configuration, atmosphere, applied load and rotating spccd on the friction coefficient was examined. WS$_2$ coated ball and disk showed the lowest friction coefficient of 0.05 in the nitrogen atmosphere under 0.3 kgf and 100 rpm, whereas relatively high coefficient of 0.1 - 0.15 was observed in air atmosphere. This confirmed that the spray-coated WS$_2$ solid lubricant was effective in reducing the friction coefficient and improving wear life in nitrogen atmosphere.

  • The effect of feeding freshly-cut common grasses on growth performance of Black Bengal goats during the pre-monsoon and pre-dry season was evaluated. Four castrated goats of 5-6 months age with a mean initial body weight were 11.17 and 10.23 kg for pre-monsoon and pre-dry season respectively, were used as experimental animal. Nutrient during the pre-monsoon season contained higher nitrogen(15.6g/kg) and less DM(196.8g/kg) than the grass harvested in pre-dry season where nitrogen was 12.8 g/kg and DM was 454.9 g/kg. Intake of DM, OM, N and NDF were 73.12 g, 67.12 g, 964.68 mg and 50.14 g/kg $W^{0.75}$ respectively in pre-monsoon were higher than those in pre-dry season at p<0.01 level of significance. Higher(p<0.01) growth(35.71 g/day) rate was found in pre-monsoon season. It may concluded that grasses grown during the pre-monsoon season was higher nitrogen value and appeared to be more palatable.

  • Improvement of L-asparate ${\beta}-decarboxylase$ production from Pseudomonas dacunhae ATCC 21192 was attempted by optimizing fermentation conditions. Optimum carbon and nitrogen sources for cell growth and enzyme production were determined. L-Glutamate (2%) was the most suitable carbon source, and D-glucose, D-glycerol and fumarate repressed enzyme production. Yeast extract (2%) was the most effective as nitrogen source. A slight change of pH to 6.5 from medium pH resulted in a meaningful increase in the production of enzyme. The production of the enzyme was highly improved by using 2% yeast extract and 2% L-glutamate in culture media. Maximum L-asparate ${\beta}-decarboxylase$ activity reached up to over 24 U/mL-broth by 15 h flask fermentation.
