Multiple Shoot Induction from Ex Vitro and In Vitro Derived Stein Node Culture of Populus alba L.$\times$P.grandidentata Michx. (줄기 절간조직 배양에 의한 교잡종 사시나무의 대량증식)

  • Physiologically modified stem nodes derived from ex vitro and in vivo explants of hybrid aspen (Populus alba L.X P.grandidentata Michx. 'Crandon') were tested for their multiple shoot regeneration capacity using a broad spectrum dosage of cytokinins. Ex vitro derived stem nodes with excised axillary buds at the time of culture produced 11 to 13 multiple shoots on 20 to 30 $\mu$M zeatin containing Woody Plant Medium (WPM) after 6 weeks. Excision of axillary bud sprouts after 2 weeks of culture and culture of the remaining stem nodes on WPM with 1.0 to 2.0 $\mu$M BA or 10 to 30 $\mu$M zeatin produced 13 to 15 and 7 to 8 shoots per explant, respectively, Multiple tiny shoots were produced when in vivo derived stem nodes (on which all leaves were removed) were cultured on WPM with 30 to 50 $\mu$M 2iP or 20 to 50 $\mu$M zeatin. The greatest number of multiple tiny shoot proliferation (32 to 50 shoots per explant) were obtained when the explants were cultured on media containing 20 $\mu$M zeatin. Successful transplanting of these multiple shoots into the greenhouse and/or nursery was achieved.

Growth characteristics and distribution pattern of a brackish water clam, Corbicula japonica along an estuarine salinity gradient in Seomjin River (섬진강 하구역에서 염분구배에 따른 일본재첩의 분포와 성장특성)

  • The purpose of this study was to determine the growth characteristics and distribution pattern of a brackish water clam Corbicula japonica in Seomjin River. Field samples were taken from 14 stations with salinity gradients during spring. Salinity at the bottom layer ranged from 1.0 psu to 32.9 psu, with low salinities in the upper area of the river. In particular, salinity at St.11 was decreased drastically to be ca. 15.0 psu, indicating an intermediate salinity zone. The distribution pattern of C. japonica was related to the salinity gradient, with the highest densities of $2,102ind.m^{-2}$ at Station 13, followed by $1,507ind.m^{-2}$ at Station 11. Here, we focused on the growth characteristics of collected C. japonica collected at two stations with different salinity values. The relationship between shell length and total weight was highly correlated ($R^2=0.91$, P<0.001) at Station 13 compared to that at Station 11 ($R^2=0.72$, P<0.001). On the other hands, the degree of correlation between shell length and shell height (SH) or shell width (SW) at Station 11 (SH: $R^2=0.91$, P<0.001; SW: $R^2=0.69$, P<0.001) was higher than that at Station 13 (SH: $R^2=0.64$, P<0.001; SW: $R^2=0.48$, P<0.001). In addition, fatness index of C. japonica at Station 13 was significantly (P < 0.001) higher than that at St. 11 (t-test value=-22.8, p<0.001). This implies that C. japonica at Station 13 might have enhanced their somatic growth, whereas C. japonica at Station 11 might have this kind of defense mechanism their internal organization against the salinity stress. Ecologically, this kind of defense mechanism of C. japonica against salinity flucuation may play an important role in their survival strategy.

  • This study not only carried out to grasp about the sununarized characteristics of the relationship between international timber market and production trend of wood products, but also focused on the analysis of korean wood demand and the long-term forecast with econometric analysis. The result of regression analysis for wood demand in Korea is that coniferous roundwood demand(CIWD) is explained by coniferous foreign roundwood price(CWRI), Gross domestic product(GDP), a dummy variable. Non-coniferous roundwood demand(NCIWD)is explained by non-coniferous roundwood price(NCWRI), coniferous roundwood price(CWRI), a dummy variable. As the result of long-term forecast by base case, the total roundwood demand was forecasted $11,107,000m^3$ in the year 2000, $11,781,000m^3$ in 2005, $12,565,000m^3$ in 2010. As the result of scenario 1, total roundwood demand was forecasted $11,027,000m^3$ in 2000, $11,435,000m^3$ in 2005, $11,952,000m^3$ in 2010. And as the result by scenario 2, total roundwood demand was forecasted $11,341,000m^3$ in 2000, $12,208,000m^3$ in 2005 $13,257,000m^3$ in 2010.

  • The citrus leafminer (CLM), Phyllocnistis citrella Stainton, is an oligophagous pest of Rutaceae family, especially Citrus spp. occurring in most worldwide citrus-growing areas. This study was conducted to evaluate a sex pheromone chemical of CLM, (Z,Z)-7,11-hexadecadienal (7Z,11Z-16:Al) in monitoring CLM by trap types, the diel activity and the influence of some weather factors on trap catch. CLM was well attracted on a trap baited 7Z,11Z-16:Al 1mg. Sticky wing trap was more effective than bucket trap. Most caught CLM were attracted at 2$\sim$6 a.m. regardless of season, and activity time of CLM was affected by sunrise time as well as sunset time. The trap catch of CLM was more influenced by wind velocity than temperature for activity time of CLM. The number of caught CLM was fallen at below 13$^{\circ}C$, but there was little effect for trap catch at over that temperature. The average wind velocity at over 2.0 m/sec made the number of caught CLM drop down. The precipitation did not affect the number of caught CLM when the average wind velocity was lower than at 2.0 m/sec.

  • mST3GaIV synthesizes ganglioside GM3, the precursor for simple and complex a- and b- series gangliosides, and the expression and regulation of mST3GaIV (CMP-NeuAc: lactosylceramide $\alpha$2,3-sialyltransferase) activity is central to the production of almost all gangliosides, a class of glycosphingolipids implicated in variety of cellular processes such as transmembrane signaling, synaptic transmission, specialized membrane domain formation and cell-cell interactions. To understand the developmental expression of mST3GaIV in mice, we investigated the spatial and temporal expression of mST3GaIV mRNA during the mouse embryogenesis [embryonic (E) days; 19, E11, E13, E15] by in situ hybridization with digoxigenin-labeled RNA probes. All tissues from 19 and E11 were positive for mST3GaIV mRNA. On E13, mST3GaIV mRNA was expressed in various neural and non-neural tissues. In contrast to these, on E15, the telencephalon and liver produced a strong expression of mST3GaIV which was a quite similar to that of E13. In this stage, mST3GaIV mRNA was also expressed in some non-neural tissues. These data indicate that mST3GaIV is differently expressed at developmental stages of embryo, and this may be importantly related with regulation of organogenesis in mice.

  • Following a preceding study if Shin et al.(2004), wave fields for a month if September if 2003 are simulated based on the modified WAM cycle 4 model that enables the precise wave hindcasting with fine spatial meshes, and characteristics of extreme waves at the south roast if Koreo are analyzed The accuracy if applied wave model is verified by comparing computed wave parameters and corresponding ones measured at Ieodo ocean research station. The wave hindasting if typhoon 'Maemi' with an hour time interval reveals the extreme wave characteristics at 4 primary locations if south coast of Korea as follows: 1) At the front sea of Chaguido in the south of Jeju-do, the maximum significant wave height, moon wave period and mean wave direction appear to be 7.41m, 13.65s and $6.4^{\circ}$, respectively at 16:00 KST if Sep. 12, 2003. 2) At the entrance of Masan Bay, 12.50m, 13.65s and $1.2^{\circ}$ at 21:00 KST if Sep. 12. 3) At the front sea of Suyoung Bay, 13.85m, 13.81s and $0.2^{\circ}$ at 22;00 KST if Sep. 12. 4) At the front sea of Ulsan port, 11.00m, 13.25s and $28^{\circ}$ at 23:00 KST if Sep.

Reproductive Maturity Onset and Tree Size in a Garcinia kola (Heckel) Coastal Humid Tropical Climate Plantation

  • Little is known of the life history of Garcinia kola; the objective of this study, therefore, was to assess the fruiting age and tree size of the species in a coastal humid tropical climate plantation condition. A total 103 trees were used in the study viz; 80 ten-year-old trees at reproductive maturity onset and 13 thirty-year-old trees with several cycles of reproduction that constitute two independent variables. Data collected were age of onset of flowering and size at reproductive maturity onset. Relative size at reproductive maturity onset (RSOM) was estimated as size at reproductive maturity onset (SOM) divided by asymptotic maximal size (AMS). Data analysis was conducted using pairwise t-test and principal component analysis (PCA). Reproductive maturity onset (flowering) was recorded in the ten-year-old stand eight (8) years after planting. Mean size at reproductive maturity onset (SOM) was height 5.32±1.7 m, dbh 0.11±0.03 m, total number of branches was 29.6±7.3, crown depth 5.24±1.05 m, crown diameter was 4.78±0.7 m, branch diameter 0.098±0.01 m, leaf length 0.13±0.02 m, leaf breadth 0.37±0.01 m, twig length 0.35±0.11 m and leaf per twig 6±0.84 and asymptotic maximal size (AMS) was height 19.85±0.76 m, dbh 0.95±0.09 m, total number of branches 62±5, crown depth 18.83±0.7 m, crown diameter 12.5±1.64 m, branch diameter 0.5±1.6 m, leaf length 0.16±0.023 m, leaf breadth 0.45±0.12 m, twig length 0.37±0.11 m and leaf per twig 19±7.5. Pairwise t-test analysis showed there was significant differences between SOM and AMS in all growth factors except leaf length, leaf breadth, and twig length. Highest relative size at reproductive maturity onset (RSOM) was recorded in leaf length 0.82, twig length 0.82, and leaf breadth 0.80, while, the lowest was branch diameter 0.11. Four components out of the total of eleven were extracted to explain the relationship in RSOM: Principal component one (PC1) explained 37.23%; PC2 26.4%, PC3 22.73%, and PC4 13.64%.

A STUDY ON THE CALCIFICATION OF SECOND MOLARS IN SKELETAL CLASS II MALOCCLUSION (골격형 II급 부정교합자의 제2대구치 석회화 과정에 관한 연구)

  • To study the calcification of second molars in skeletal class II malocclusion, the author examined cephalograms, orthopantomograms, periapical films of 391 children from 7y 0m to 15y 11 m years old who had skeletal class II malocclusion, and observed the difference in the calcification stage between upper and lower second molars. The result s are as follows. 1. The mean ages of crown completion of upper and lower second molars are $8.7{\pm}1.75$, $8.8{\pm}1.13$ in boys, and $8.4{\pm}0.81$, $8.5{\pm}0.91$ in girls. 2. The mean ages of root completion of upper and lower second molars are $14.0{\pm}1.09,\;14.5{\pm}0.52$ in boys, and $13.7{\pm}1.15,\;13.8{\pm}1.18$ in girls. 3. The calcification stages of upper second molars are more advanced than those of lower second molars at $5\%$ level in both sexes.

SEARCH FOR $H_2COH+\;AND\;H_2^{13}CO$ IN DENSE INTERSTELLAR MOLECULAR CLOUDS

  • We have searched for the 2 mm transitions of $H_2COH^+(2_{02}-1_{01})$ and $H_2\;^{13}CO(2_{02} - 1_{01},\; 2_{12}-1_{11},\;and\;2_{11}-1_{10})$ toward the dense interstellar molecular clouds Orion A, TMC-1 and L134N using the FCRAO 14 m telescope. None of the transitions have been detected except the $H_2\;^{13}CO$ transitions toward Orion-KL. We set upper limits for the abundances of the protonated formaldehyde ion $(H_2COH^+)$, which are close to the abundances expected from ion-molecule chemistry.

Purification and Characterization of Aryl Acylamidase from Pseudomonas sp. (Pseudomonas sp. Aryl Acylamidase의 정제 및 성질)

  • Aryl acylamidase [EC 3.5.1.13] present in an acetaminophen-assimilating Pseudomonas sp. has been purified to a homogeneity using series of ammonium sulfate fractionation, DEAE-Sephacel anion exchange, Phenyl-Sepharose CL-4B hydrophobic, and Sephadex G-100 gel-permeation chromatography. The molecular weight, which was estimated by gel-permeation filtration and sodium dodecyl sulfate polyacylamide gel electrophoresis, was about 57 kDa and 56 kDa, respectively, indicating that this enzyme is a monomeric protein. The optimum pH was 10.5 and the optimum temperature was 40$^{\circ}C$. After incubation of the enzyme at 50$^{\circ}C$ for 30 min, residual activity of the enzyme was 34% compared to its original activity. The Km values for acetaminophen and 4'-nitroactanilide were 0.10 mM and 0.11 mM, respectively.
