A Novel Polyacetylene from Cirsium spp. (Cirsium속 식물로부터 새로운 polyacetylene의 분리)

  • A novel polyacetylene was isolated from Cirsium spp., as well as five known ones, and its chemical structure was determined as heptadeca-1-en-11.13-diyne-8R, 9S, 10R-triol(1) on the basis of spectral data and chemical reactions. $^{1}$H-and $^{13}$C-NMR data of these polyacetylenes were completely assigned by the appfication of 2D-NMR techniques.

Quality Control of Adenophorae Radix (사삼의 품질 관리에 관한 연구)

  • This study is to establish the quality control method of Adenophorae Radix. (6R,7R)-E,E-tetradeca-4,12-diene-8,10-diyne-1,6,7-triol (1) isolated from this plant was adequate as an analytical marker. Content of 1 in Adenophorae Radix was $0.006\;{\pm}\;0.003%$ (n = 43). In addition, total ash content was $6.5\;{\pm}\;4.0%$, and loss on drying was $12.1\;{\pm}\;2.1%$.

  • Phytochemical investigation of the 80% MeOH extract from the leaves of Hosta longipes resulted in the isolation of sixteen compounds (1 - 16). The structures of the compounds were elucidated by spectroscopic methods to be methyl 10,10-dimethoxydecanoate (1), methyl 10-hydroxy-8E,12Z-octadecadienoate (2), methyl coriolate (3), trans-phytol (4), phytene-1,2-diol (5), phyton (6), (3S,5R,6S,7E,9R)-7-megastigmene-3,6,9-triol (7), (3S,5R,6S,9R)-3,6,9-trihydroxymegastigman-7-ene (8), shikimic acid (9), p-coumaramide (10), trans-N-p-coumaroyltyramine (11), cis-N-coumaroyltyramine (12), tryptophan (13), thymidine (14), adenosine (15), and deoxyadenosine (16). Compound 1 was synthesized, but not yet isolated from natural source, and compounds 2-16 were isolated for the first time from this plant source.

Effect of Red Ginseng Triol Saponin Fractions on the Spatial 1 Memory Function Studied with 12-Arm Radial Maze (12-Arm Radial Maze를 이용한 쥐의 공간 인지능력에 미치는 홍삼 PT계 사포닌의 효과)

  • Male rats aged 15 months, which had been over-trained with 12 arm radial maze up to the criterion of 1 or 2 errors an average per trial, were divided into two groups and 7 months after the over-training and were studied the effect of red ginseng trial saponin fraction (PT) on the spatial memory function. The rats which could be improved the performance of 9.75$\pm$1.26 (within 185$\pm$8.2 1 sec) were classified into "normal group" (n=4) and the ones, which showed R-maze performance of 7.0$\pm$2.87 within 300 sec, "deteriorated memory group" (n=5). PT was dissolved in distilled water and injected into the deteriorated memory group intraperitoneally at 30 min before R-maze for 3 consecutive days. The injected amount of PT on the 1st day was 10mg/rat, 1mg/rat both on the 2nd the 3rd day. As results, the performance was restored to 9.4$\pm$2.0 after stopping ginseng administration, although the lower performance values (1.4$\pm$0.89, 2.8$\pm$0.83, 3.8$\pm$0.84, respectively) were exerted during the three days of PT administration than before PT administration (7.0$\pm$2.87) since 1 day after administratitan of PT. The restored performance values were continuously maintained up to the level of the "normal group". However, any lower performance values were not observed when PT was administered via intraperitoneal route to the normal group in about of 2 mg/rat except 10mg1ra1. The number of errors when the foods in arms were put at only 6 places was clearly reduced during the 9 days of consecutive administration of PT as compared with untreated control. These results indicated that PT administration may enhance their cognitive function after a long lapse of time not only in the memory deteriorated rats but also in normal ones. The reason exerted lower performance values during PT administration in this paper was discussed as compared with the results of T-maze behavior of another 3-month-aged rats. of another 3-month-aged rats.

  • From the polar fractions of a 70% EtOH extract of the flower buds of Lonicera japonica (Caprifoliaceae), ten constituents were isolated and identified as iridoid glycosides 7-dehydrologanin (7-ketologanin, 2), secologanin dimethyl acetal (3), (E)-aldosecologanin (centauroside, 5), dimethyl secologanoside (6), secoxyloganin (7) and epivogeloside (8). Other identified constituents were 1-O-$\beta$-D-glucopyranosyl-(2S,3S,4R,8E/Z)-2-[(2R)-2-hydroxy(docosanoyl, tricosanoyl, tetracosanoyl, pentacosanoyl)amino]-8-octadecene-1,3,4-triol (1), uracil (4), D-mannitol (9), and sucrose (10). Among them, 1, 2, 4, and 10 were isolated for the first time from this plant.

  • A new ceramide alomacrorrhiza A was isolated from the ethanolic extract of the plant Alocasia macrorrhiza (L.) Schott. Its chemical structure was elucidated as (2S,3S,4R)-2N-[(2'R)-2'- hydroxy-hexacosanoyl]-tetradecane-1,3,4-triol based on extensive 1D, 2D NMR, EI-MS, FABMS, HR-FAB-MS spectroscopic data and chemical degradation studies.

  • Adenophorae Radix, the roots of Adenophora triphylla var. japonica Hara (Campanulaceae) is known to be an anti-inflammatory and antitussive drug used for the treatment of lung disease. However quality control method is not established yet. This study is to establish the quality control method of Adenophorae Radix.From the roots of this plant, we isolated (6R,7R)-E,E-tetradeca-4,12-diene-8,10-diyne-1,6,7-triol (1) as a marker compound for the quality control and determined its content by HPLC-UV detector.   • To study on the changes in saponins from callus mass by tissue culture, the callus was derived from the petiole of Korean Ginseng (Panax Ginseng C.A. Meyer) and cultivated on Murashige and Skoog's agar medium supplemented with 2.4-dichlorophenoxyacetic acid and kinetin for 8 months. Then, well-grown callus was analyzed for its components estimation. The results obtained are as follows: (1) When saponins isolated from callus mass were chromatographed on a silca gel plate, and determined by the thinchrograph TFG-10, the ratio of Rb, c to Rg(f) in saponins was 2.16 to 1 and Rb, c, d to Re, g (f) was 1 to 1.63, while in the case of saponins from the root of Panax Ginseng grown by soil culture, the ratio of Rb, c to Rg(f) was 1.03 to 1 and the ratio of Rb, c,d to Re, g(f) was 1 to 1.17. (2) Sapogenins were obtained from the hydrolysates of saponins, and determined by thinchrograph TFG-10. The ratio of panaxadiol to panaxatriol in sapogenins from callus saponins was 2.66 to 1, while the ratio of panaxadiol to panaxatriol in sapogenins from ginseng root saponins was 1.86 to 1. From the results above mentioned, we concluded that the relative contents of sapogenins in saponins from callus mass by tissue culture were different from those in saponins from ginseng root by soil culture.

Neuroprotective Compounds Isolated from Lysimachia christinae

  • We previously reported that dried Lysimachia christinae whole plant extract exerted significant neuroprotective activity. In this study, we tried to isolate neuroprotective compounds of L. christinae. We evaluated the neuroprotective activity of the four fractions (hexane, chloroform, ethyl acetate, and n-butanol fractions) of methanol extract. Among them, ethyl acetate and n-butanol fractions showed most potent neuroprotective activity against glutamate excitotoxicity. Nine compounds were isolated from ethyl acetate and n-butanol fractions of L. christinae extract and identified as cynaroside (1), (3,4,5,6-tetrahydroxytetrahydro-2H-pyran-2-yl)methyl-3-hydroxy-2-octyldopentaconta-23,33-dienoate (2), androst-16-ene-3,6-diol (3), 2-hydroxy-24-propoxytetracos-4-enoic acid (4), 2-hydroxy-24-methoxytetracos-4-enoic acid (5), 12-(stearoyloxy)octadec-9-enoic acid (6), β-sitosterol (7), (E)-4-(3,4-dimethoxyphenyl)but-3-en-1-yl palmitate (8) and (1S,2S,3R,4R)-4-(((2S,3R,4R,5R,6S)-2-(((2R,3R,4S,5R,6R)-2-(3,4-dimethoxyphenethoxy)-3,5-dihydroxy-6-(hydroxymethyl) tetrahydro-2H-pyran-4-yl)oxy)-4,5-dihydroxy-6-methyltetrahydro-2H-pyran-3-yl)oxy)cyclohexane-1,2,3-triol (9) by spectroscopic data such as UV, IR, NMR, Mass spectroscopy. Their neuroprotective activity was evaluated by MTT assay. Cynaroside (1) and androst-16-ene-3,6-diol (3) had significant neuroprotective activity against glutamate-injured HT22 cells. The neuroprotective efficacy of cynaroside (1) and androst-16-ene-3,6-diol (3) was related to their anti-oxidative activity.

Studies on the Optimum Light Intensity for Growth of Panax Ginseng II. Effect of Light Intensity on the Contents of Saponin and Free Sugar in the Ginseng Leaf (인삼생육의 최적광량 구명에 관한 연구 제2보 광도가 인삼엽내 Saponin 및 유리당함량에 미치는 영향)

  • This study was conducted to define the effects of light intensity on the amount of saponin and free sugar and the ratio of triol group saponin and diol group saponin (PT/PD) in the leaf of Panax ginseng C.A. Meyer. 4-Year-old ginseng plants were grown under the shadings of different light transmittance rate(LTR) of 5%, 10%, 20% and 30% for 5 months and the leafiets were sampled from 2nd low at late August to determine the amount of saponin and free sugar.-Rd was main ginseuoside in the diol group saponin but in triol group saponin, ginsenoside-Re showed highest value and next was ginsenoside-$Rg_1$ and $Rg_2$ respectively. Up to 20% of light transmittance rate (LTR), the ginseng leaves grown under high light intensity showed an increase in the amount of total saponin and the ratio of PT/PD but the amount of total saponin and the ratio of PT/PD but the amount of total saponin and the ratio of PT/PD was decreased at the ginseng leaves grown under the shading of 30% LTR. The ginseng leaves grown under the shading of 20% LTR showed a significant increase in the amount of glucose and fructose but a significant decrease of sucrose content. A significant positive correlation ($r=0.992$^{**}$) was recognized between the of amount of total saponin and glucose.
