Single-Strand Conformation Polymorphism Analysis by Microchip Electrophoresis for the Rapid Detection of Point Mutation in Human Obesity Gene

  • We describe an effective method of microchip electrophoresis (ME) based on single strand conformation poly-morphism (SSCP) analysis to rapidly detect the point mutation, Leu72Met, in a human obesity gene. The 207-bp dsDNA in the Leu72Met region, an estimate of the child obesity DNA mutant, was amplified by polymerase chain reaction (PCR) and submitted to a conventional glass microchip analysis with a sieving matrix of 1.75% poly(vinylpyrrolidone) (Mr 1 300 000), 1.0% poly(ethyleneoxide) (Mr 600 000) and 5.0% w/w glycerol. When combined with base stacking (BS) with hydroxide ions, the SSCP-ME provided rapid analysis as well as sensitive detection. The detection sensitivity was effectively enhanced in the OH- concentration range of 0.01-0.025 M NaOH. The sensitivity and speed of this ME-based SSCP methodology for the rapid detection of Leu72Met point mutations makes this an attractive method for diagnosing childhood obesity in a clinical diagnostic laboratory.

A Comparative QSPR Study of Alkanes with the Help of Computational Chemistry

  • The development of a variety of methods like AM1, PM3, PM5 and DFT now allows the calculation of atomic and molecular properties with high precision as well as the treatment of large molecules with predictive power. In this paper, these methods have been used to calculate a number of quantum chemical descriptors (like Klopman atomic softness in terms of $E_n^{\ddag}\;and\;E_m^{\ddag}$, chemical hardness, global softness, electronegativity, chemical potential, electrophilicity index, heat of formation, total energy etc.) for 75 alkanes to predict their boiling point values. The 3D modeling, geometry optimization and semiempirical & DFT calculations of all the alkanes have been made with the help of CAChe software. The calculated quantum chemical descriptors have been correlated with observed boiling point by using multiple linear regression (MLR) analysis. The predicted values of boiling point are very close to the observed values. The values of correlation coefficient ($r^2$) and cross validation coefficient ($r_{cv}^2$) also indicates the generated QSPR models are valuable and the comparison of all the methods indicate that the DFT method is most reliable while the addition of Klopman atomic softness $E_n^{\ddag}$ in DFT method improves the result and provides best correlation.

  • This study outputs calculation of regression of each items for production of torso basic pattern according to 6 body types as the result of another study and intends to present drawing method of torso model by short measure method modified and supplied and supplied by experiments of wearing clothing. SAS(Statistical Analysis System) is used for figures management and methods for analysis used are Frequency Analysis, Means Analysis, Regression Analysis, Correlation Analysis, etc. Results are as follows. 1. Correlation analysis is used to output the size necessary for torso prototype drawing by sort measure method and waist front length, back length, crotch length, shoulder point-cerricale-shoulder point, bust circumference, waist circumference, weight, etc, are set up as representative items calculation of regression of each type is suggested. 2. In the result of experiment of the first wearing clothing intended for 5 in each type and the whole 30, to develop torso prototype drawing method by short measure method, as we find some problems of the shape and propriety of neck root circumference line, the position of shoulder point, pulling or hold armpit parts, waist circumference line, the degree of dissatisfaction is high, so the second experiment of wearing clothing is propriety of each part is improved, all items except the length and quantity of shoulder dart, waist in back bodice, clearance quantity of hip circumference, and the place of shoulder line in side bodice. So, it was modifed and supplied and then the third torso prototyped drawing method by shout measure method was suggested. The third prototype drawing method was suggested, by modifying and supplying.

고해상도 상용카메라를 사용하는 영상변환을 이용한 탄착점 좌표획득 (Bomb Impact Point Location Acquisition by Image Transformation using High-Resolution Commercial Camera)

  • In the bomb impact test, to acquire the bomb impact point location the high-priced embedded equipments such as the Bomb Scoring System or the EOTS are needed. Recently, a high-resolution image processing could be possible since the resolution of the commercial camera is growing rapidly. In this paper we first propose an image transformation method for acquiring the real bomb impact image using a high-resolution commercial camera, and then present the process calculating the real bomb impact point location coordinate from the transformed image. Based on the experimental results we found the possibilities that the real bomb impact point information could be effectively earned just using the commercial camera.

지리공간 웹 서비스 기반의 기준점 자동추출 기법 연구 (Automatic Extraction Method of Control Point Based on Geospatial Web Service)

  • 본 논문에서는 지리공간 웹 서비스 기반의 기준점 자동 추출 기법을 제안한다. 제안하는 기법은 3단계로 구성된다. 1) 첫 번째 단계에서는 지리공간 웹 서비스를 통해 대상영상의 촬영지역에 해당하는 기준자료를 자동으로 획득하고, 2) 두 번째 단계에서는 획득된 기준자료와 대상영상에 SURF 알고리즘을 적용하여 후보 기준점을 찾는다. 3) 마지막 단계에서는 RANSAC 알고리즘을 이용하여 추출된 후보 기준점 중 정 정합점을 최종 기준점으로 산출한다. 제안하는 기법은 기준자료를 획득하기 위해 지리공간 웹 서비스를 활용하였다. 이를 통하여 제안하는 기법은 기준영상과 고도자료의 관리 및 획득 시 사용자 편의성을 증대 시켰고, 표준을 따르기 때문에 높은 확장성을 가진다. 본 논문에서는 제안하는 기법을 SPOT-1, SPOT-5, IKONOS 위성영상에 적용하여 실험을 수행하였다. 실험지역에 적용한 결과, 제안하는 기법은 대상영상의 촬영센서, 촬영일자, 해상도 변화에도 RMSE 5화소 미만의 일관된 정확도를 산출하였고, 대상영상의 해상도가 좋아짐에 따라 정확도의 지속적인 향상 가능성을 확인하였다. 또한 기준영상과 고도자료로 군 표준 자료를 사용함으로써 제안하는 기법의 군사적 활용가능성을 확인하였다.

Excel의 추세선을 이용한 표준곡선 검증 (Standard Curve Validation using Trendlines in Excel)

  • Insulin 83건을 검사 대상으로, 장비 WIZARD(PerkinElmer, USA)와 DREAM- G-10(Shinjin, KOREA)의 Graph Algorithm 중에 표준곡선과 신뢰성이 가장 높은 Excel의 추세선은 다항식 추세선이다. 다음으로 다항식 추세선식을 이용하여 표준농도 1개씩을 제외하여 표준농도의 회귀식에 미치는 영향을 비교한 결과 최고농도($315{\mu}IU/m{\ell}$)의 평균값이 표준물질 6개로 실시한 표준 회귀식을 이용한 평균값과 비교하여 49%나 평균값이 저하되었다. 단 낮은 농도에서는 영향이 미비하였다. 마지막으로 WIZARD의 Point to Point형식과 DREAM G-10의 Point to Point형식이 적합성이 높고, DREAM G-10(Point to Point)와 Excel 다항식 추세선이 적합성이 높으며, Excel 다항식 추세선과 DREAM G-10(2'nd order Polynomial)의 적합성이 높다.

A Study on the Goal Setting Method for Increasing the Holed Probability in Slope Putting Stroke on an Artificial Putting Surface

  • Objective: The purpose of this study was to develop a goal setting method for increasing the probability of a holed in a side inclined putting stroke. Method: Three-dimensional video data was recorded at a frequency of 120 hz per second after synchronizing 19 infrared motion capture systems (Qualisys, Gothenburg, Sweden). Putting green used a polycarbonate plate ($1.2{\times}2.4{\times}0.01meter$) with coefficient of friction (${\mu}=0.062$) and a real curve of the actual hole. Results: The velocity ratio between the club and the ball was 1:1.6 under various ball speed conditions in this study. The overall position of the break is 1 m to 1.4 m from the point where the ball leaves. If there is a slope, the ball follows the target line by the straightening force, and when it reaches 1 m position, the straightening force decreases by 30~50% and reaches to the deviation (break) point which is severely influenced by the slope. From here, the ball is aimed in a direction other than the target, and the size is affected by the slope. Conclusion: If there is a side slope, the ball moves away from the straight line, and the larger the slope, the closer the break point is to the starting point of the ball. Therefore, it is necessary to calculate the degree of departure according to the slope carefully, and it is preferable that the slower the speed is, the more the influence of the slope becomes. It is preferable to use the center of the hole as a reference when calculating the departure.

DNN과 HoG Feature를 이용한 도로 소실점 검출 방법 (Method for Road Vanishing Point Detection Using DNN and Hog Feature)

  • 소실점이란 실제 공간의 평행한 선들이 영상 내에 투영되면서 한곳에 모이는 점으로, 도로 공간에서의 소실점은 매우 중요한 공간정보이다. 도로 공간에서의 소실점을 이용해 추출된 차선의 위치를 개선하거나, 깊이지도 영상을 생성할 수 있다. 본 논문에서는 자동차의 시점을 기준으로 도로를 촬영한 영상을 Deep Neural Network(DNN)과 Histogram of Oriented Gradient(HoG) Feature를 이용한 소실점 검출 방법을 제안한다. 제안하는 알고리즘에서는 영상을 블록별로 나눠서 주요 에지 방향을 추출하는 HoG Feature 추출 단계와 DNN 학습 단계, 그리고 Test 단계로 나뉜다. 학습단계에서는 자동차 시점으로 기준으로 도로 영상 2300장으로 학습을 진행한다. 그리고 Test 단계에서는 Normalized Euclidean Distance(NormDist) 방법을 사용하여 제안하는 알고리즘의 효율성을 측정한다.

선삭에서 공작물 지지조건이 채터진동발생에 미치는 영향 (Influence of Chucking Conditions on the Chatter Vibration Commencing Point in Turning)

  • With increasing demands on automatic and high-capability manufacturing, the dynamic performance of machine tools becomes more and more important. In this paper, the correlation between dynamic compliance of the cutting system and the commencing point of chatter vibration in turning is checked by impulse excitation method and cutting tests for some cutting system. The correlation between chucking conditions of workpiece and the commencing point of chatter vibration is clarified, and it is proven that there is a mutual relations between them. Chatter vibration commenced at certain level of dynamic compliance of the cutting system regardless of the kind of the system. It shows the possibility of dynamic performance test of a lathe by means of impulse excitation method.

게임요소의 규모가중치 기준 연구 (The Weight of Game Factor of Game Contents Costing)

  • Function Point(FP) is well spread and advanced methodology in software costing. But FP is not common in game contents costing. It is one reason that Game Contents is a new area in software costing. And, ISO/IEC FDIS 19761(final draft) recommends not to use FP method in game contents costing. So we suggested new methodology in that area which is called Game Factor Point Method(GFPM). In this paper, we studied more about the weight of the game factor. We hope this will clarify them. We made definition of game factor, and studied more detail about Mission which was a little ambiguous in prior study. We also studied difficulties of game factor which will be a very important factor in game contents costing.
