Improvement of the Strains Degrading Recalcitrant Aromatic Compounds by Cell Fusion Between Arthrobacter spp. and Pseudomonas putida (Arthrobacter spp. 와 Pseudomonas putida 의 세포융합에 의한 난분해성 방향족 화합물 분해세균의 균주개량)

  • To develope the new strains of microorganisms having the degradative ability for various aromatic hydrocarbons. spheroplast cell fusions were performed with Arthrobacter spp. degrading phthalate ester and Pseudomonas putida degrading alkylbenzen sulfonate(ABS) and the characteristics of the fusants were investigated. The spheroplasts of P. putia KUD15 and Arthrobacter sp. were formed effectively by lysozyme-EDTA treatment and by Ampicillin-lysozyme-EDTA treatment. respectively. The Spheroplast formation frequency and the regeneration frequency of the strains were 98-99% and 5-8%, respectively. For cell fusion. 40% PEG6000 was used as a fusogenic agent and the formation frequencies of fusion product were $1.8{\times}10^{4}-$2.9{\times}10^{4}$ Most of the fusants, which were selected in complemented antibiotics media showed the degradative ability in minimal selective medium added phthalate ester or ABS as sole carbon source. ABS degradation by fusant strain was increased about 20% with compared with the parental strain, while the degradative ability of phthalate ester was simillilar to that of parental strain.

The development of CAl Courseware for Basic Life Support - Centered on the Foreign-Body Airway Obstruction in Adult- (기본 인명구조술 교육을 위한 CAI 코스웨어 개발 - 성인의 이물질에 의한 기도폐쇄를 중심으로 -)

  • With the rapid development of information and communication technology, a lot of multi-media learning programs are being developed and reported in the field of Emergency medicine both home and abroad. In this connection, this study was aimed at developing a foreign-body airway obstruction courseware in adults for EMT. The development period of CAI courseware lasted from May 2003 through November 2003. Among CAI courseware patterns, private instruction and repeat practice and simulation patterns were used as an instruction-learning strategy. The learning contents of the CAI courseware consisted of five chapters concerning (1) A relief of partial FBAO in the responsible victim, (2) A relief of complete FBAO in the responsible victim, (3) In case of unconsciousness in the responsible victim without removing all foreign body, (4) In case of consciousness in all victims after getting removed all foreign body and (5) A complete airway obstruction in victims without consciousness on the basis of assess responsiveness and the degree of airway obstruction. The way to use this courseware, with just a click on one specific chapter, was developed to proceed a course with progressive algorithm, a method of solving problems by choosing one between two situations. A characteristic of this CAI courseware is the enhanced efficiency of an instruction-learning method by providing an opportunity of choice based on situations in its effort to encourage learners to use a self-initiated learning method, not one-way method and to enhance problem solving skills among situations. Moreover, this courseware went through the diverse phases such as development, application, feedback in connection with learning process by practicing teachers, so that the courseware could be used frequently in the future. The contents of this courseware were written with the web, so that, if necessary, the contents could be continuously modified and complemented and handed out in the form of CD-ROM. This study indicates that the development of a variety of CAI courseware requires institutional and financial assistance and initiatives reflecting a reality in terms of learning process, technical assistance and resources.

A Study on the Image Changes of an Eye Shape according to Eyelash Design (속눈썹디자인에 따른 눈 형태의 이미지변화 연구)

  • The purpose of this study was to examine the need of the eyelash design area and the changes of an eye image according to eyelash design. Methodology included both a theoretical research and an empirical research. The theoretical research reviewed previous researches and technical books. The empirical research divided eye shapes, complemented the defect of eye shapes, and examined resulting image changes. A mechanical way was performed for treatment because it was more efficient than a manual way. A J-curl type was used. Eye length was divided into five and the point was determined. The length and density of curl were changed in each point to maximize a treatment effect. As a result, eye images were changed as follows. First, round eyes became stable. Slit eyes became vivid and soft. Wide-set eyes became intelligent and narrow-set eyes became smooth. Large eyes were naturally gradated and small eyes became larger. Double-eyelid eyes became stable and elegant and single-eyelid eyes became gentle and soft. Bulging eyes became easy and soft and sunken eyes became smooth. Peaked eyes became gentle and soft and sleepy eyes became strong and vivid. Based on the findings, eyelash design can play a role in changing an image according to eye shapes. In particular, a speed eyelash extension device to extend eyelash can reduce working hours and gradate only by angle control. It can complement the defect of eyes according to the design of each shape and change into a better image.

Comparative Analysis of the Prerequisite Items Applicable to the HACCP in Livestock Processing Plants (축산물가공장 HACCP 선행요건 평가항목 개선을 위한 비교분석)

  • We analyze the HACCP Prerequisites of National Veterinary Research and Quarantine Service (NVRQS), Korea Food and Drug Administration(KFDA), and US Food Safety and Inspection Service (FSIS) and recommend contents and ranges to be complemented and used for the preparation of guidelines. We used the HACCP Notice of the Processing Livestock Act, the HACCP Application Manual for Livestock Processing Plants, the HACCP Notice of the Food Sanitation Act, US GPO 9 CFR and 21 CFR, US FSIS Directive 11,000.1, and HACCP-based Inspection reference guide of Consumer Services, North Carolina Department of Agriculture. The Prerequisites of NVRQS and KFDA are composed of 66 and 84 items respectively, without detailed guidelines for their application. This may decisively affect the application in the field and the evaluation activities. Water supplies, washing and disinfection monitoring tools, and examining and correcting plan are required to be improved. If the standards of compliance of each Prerequisite item as well as performance guidelines are given, the application and evaluation will be performed more effectively. The evaluation items should be associated with Prerequisite operations such as non-compliance complement, self-evaluation, and record keeping. Hazards found during official inspection should be promptly controlled not to contaminate the work places and processing items. As the HACCP is expected to spread from farms to tables, the standardized Prerequisite program among the official governments should be prepared.

  • This study is to help creative idea generation based on the theory of the 'reconstruction of character shape image elements', and aims to extrusion of creative and diverse shapes with combination of image elements upon computing creative image generation. In order to suggest the design generation methodology for the maximization of idea generation ability and to overcome restriction of thinking out of existing idea generation methodology, it has suggested the CGS(Character Generation System) that is a creative idea generation methodology identified and complemented the problem of the existing computerized idea generation(PDS with Proportion) method out of the preceded studies on the creative idea generation methodologies. this study is expected to have effectives as one method for idea generation or creative image generation assistance during the 3D character development process, and to serve as an assistance to overcome the restriction of the character shape image generation through diverse idea generations.

Delamination and concrete quality assessment of concrete bridge decks using a fully autonomous RABIT platform

  • One of the main causes of a limited use of nondestructive evaluation (NDE) technologies in bridge deck assessment is the speed of data collection and analysis. The paper describes development and implementation of the RABIT (Robotics Assisted Bridge Inspection Tool) for data collection using multiple NDE technologies. The system is designed to characterize three most common deterioration types in concrete bridge decks: rebar corrosion, delamination, and concrete degradation. It implements four NDE technologies: electrical resistivity (ER), impact echo (IE), ground-penetrating radar (GPR), and ultrasonic surface waves (USW) method. The technologies are used in a complementary way to enhance the interpretation. In addition, the system utilizes advanced vision to complement traditional visual inspection. Finally, the RABIT collects data at a significantly higher speed than it is done using traditional NDE equipment. The robotic system is complemented by an advanced data interpretation. The associated platform for the enhanced interpretation of condition assessment in concrete bridge decks utilizes data integration, fusion, and deterioration and defect visualization. This paper concentrates on the validation and field implementation of two NDE technologies. The first one is IE used in the delamination detection and characterization, while the second one is the USW method used in the assessment of concrete quality. The validation of performance of the two methods was conducted on a 9 m long and 3.6 m wide fabricated bridge structure with numerous artificial defects embedded in the deck.

  • A combination of CFT column and RC flat plate without formworks is very effectively rapid constructions. This paper verified the lateral resisting capacity of CFT column-RC flat plate exterior connection in comparison with general RC column-flat plate connection and detected moment capacity and ductility capacity of connection according to moment-displacement ratio. We made and tested specimens which have different variables respectively and as a result derive a following conclusion. In CFT-E2 specimen a critical section was extended and maximum moment increased 20% respectively in comparison to general RC column specimen. In BME and CFT-E1 specimens generally shear governed behaviors and CFT-E2 specimen complemented with seismic band, flexure behavior region of slab was extended and also ductility ratio and energy absorptance increased.

Effects of the 3D Visual Feedback Exercise with Action Observation on the Posture Alignment and Cerebral Cortex Activation in Forward Head Posture (3D 시각적 피드백과 동작관찰을 이용한 운동이 전방머리자세의 자세 정렬과 대뇌겉질 활성도에 미치는 영향)

  • Purpose : The purpose of this study was to investigate the effects of exercise intervention combined with 3D visual feedback and motion observation on postural alignment and cerebral cortical activity in subjects with forward head posture (FHP). Methods : The study included 28 participants with FHP, randomly divided into a 3D visual feedback plus motion observation group (n=14) or control group (n=14). The experimental group received corrective exercise combined with 3D visual feedback and motion observation for FHP, three times a week for four weeks. We examined cervical spine radiographs in the lateral standing position with both arms crossed to measure the craniovertebral angle (CVA) and cervical lordosis (CL). Relative alpha (RA) and beta waves (RB) were measured by wireless dry EEG. Results : The CVA value was significantly different between the groups, and the CL value was significantly different only in the experimental group. RA and RB values were not significantly different before and after intervention in the control group. RB values were significantly decreased before and after intervention in the experimental group. Conclusion : Based on the results of this study, we suggest that interventions combined with motion observation and 3D visual feedback may be effective as exercise methods to improve postural alignment and cerebral activity in subjects with FHP. Further research is required to generalize our results on technical supplementation complemented with 3D visual feedback devices.

Development of the Design Prototype for Funtional Men's Outdoor-Wear Jacket (기능성 아웃도어 웨어의 등산용 남성 재킷 프로토타입 디자인)

  • The purpose of this study is to develop a design prototype of outdoor-wear which is suitable for trekking. For achieving this purpose, 6 factors(bodice, sleeve, hood, pockets, opening, hem-line) considered for clothing function were selected, classified, and complemented based on the survey of literature and the research of 185 designs in 12 outdoor-wear brands. The results are as follows; The removable hood and 4DM sleeve style with the elastic band and velcro for the cuffs was the most preferred in the detail design on the brand research. And also the ventilation function and 1 or 2 diagonal in-seam pocket, covered zipper was preferred. Based on this result, two design prototypes of men's outdoor-wear for trekking were suggested; jacket 1 has a slim waist line and a hip length. Front opening has a zip and placket for windproof effect. There is a detachable hood with stopper around the head line. 4DM sleeve and elastic band inserted at the end of sleeve, so that prevent the hem-line raising. Two inseam pocket is placed slightly diagonal. Jacket 2 is a hood jacket with a york. The Jacket has a little roomier in waist and a hip length. Front opening is fastened by a waterproof zip. It has a 4DM sleeve and a princess line to make it placed two pocket which have ventilation functionality.

Role of Two Sets of RND-Type Multidrug Efflux Pump Transporter Genes, mexAB-oprM and mexEF-oprN, in Virulence of Pseudomonas syringae pv. tabaci 6605

  • Pseudomonas syringae pv. tabaci 6605 has two multidrug resistance (MDR) efflux pump transporters, MexAB-OprM and MexEF-OprN. To understand the role of these MDR efflux pumps in virulence, we generated deletion mutants, ΔmexB, ΔmexF, and ΔmexBΔmexF, and investigated their sensitivity to plant-derived antimicrobial compounds, antibiotics, and virulence. Growth inhibition assays with KB soft agar plate showed that growth of the wild-type (WT) was inhibited by 5 μl of 1 M catechol and 1 M coumarin but not by other plant-derived potential antimicrobial compounds tested including phytoalexins. The sensitivity to these compounds tended to increase in ΔmexB and ΔmexBΔmexF mutants. The ΔmexBΔmexF mutant was also sensitive to 2 M acetovanillone. The mexAB-oprM was constitutively expressed, and activated in the ΔmexF and ΔmexBΔmexF mutant strains. The swarming and swimming motilities were impaired in ΔmexF and ΔmexBΔmexF mutants. The flood inoculation test indicated that bacterial populations in all mutant strains were significantly lower than that of WT, although all mutants and WT caused similar disease symptoms. These results indicate that MexAB-OprM extrudes plant-derived catechol, acetovanillone, or coumarin, and contributes to bacterial virulence. Furthermore, MexAB-OprM and MexEF-OprN complemented each other's functions to some extent.
