Studies on the types of hemoglobin and albumin in German shepherds (군용견(軍用犬) German shepherd의 hemoglobin 및 albumin형(型)에 관(關)한 연구(硏究))

  • The study was conducted to investigate the phenotypes of blood proteins, hemoglobin and albumin, of 104 German shepherds in Dag-gu, Yae-chum Kim-hae and Kwang-ju area by the starch gel electrophoresis. The results obtained were summarized as follows: 1. The hemoglobin phenotypes were observed three(HbBB, HbBb and Hbbb). Fast migrating bands were same and slow migrating bands were divided by heavy(HbBB), light(HbBb) and non-stained(Hbbb). The frequencies of appearance in HbBB, HbBb and Hbbb were 17.31%, 51.92% and 30.77%, respectively. 2. The hemoglobin phenotypes were controlled by two allelic genes, $Hb^b$ and $Hb^B$. The gene frequencies were calculated at 0.567 in $Hb^B$ and 0.433 in $Hb^A$. 3. The albumin phenotypes were observed three(AlbFF, AlbSS and and AlbFS), which were divided by fast migrating band(AlbFF), slow migrating band(AlbSS) and mixed migrating band(AlbFS). The frequencies of appearance in AlbFF, AlbFS and AlbSS were 5.77%, 28.85% and 65.38%, respectively. 4. The albumin phenotypes were controlled by two alleic genes, $Alb^F$ and $Alb^S$. The gene frequencies were calculated at 0.202 in $Alb^F$ and 0.798 in $Alb^S$.

Seismic Stratigraphy of the post-Paleozoic Sedimentary Section in the Main Pass area, Northern Gulf of Mexico (멕시코만 Main Pass 해역의 중생대-신생대 퇴적층의 탄성파층서)

  • Multichannel deep seismic reflection data in the Main Pass area of the northern Gulf of Mexico are interpreted in this study for the stratigraphy and the depositional history. Structural analysis of deep seismic reflection data provides new information on the locations of paleo-shelf margins and the basement. The basement occurs at about $7.5{\cal}km$ depth at the northern end of seismic line LSU-1 in the Mississippi shelf. The Jurassic and early Cretaceous shelf margins occupy approximately the same position, whereas the Oligocene shelf margin occurs about 28 km farther landward. Ten major seismic stratigraphic sequences are identified for the Mesozoic and Cenozoic sed-imentary section. Correlation of sequence boundaries defined in this study with those in other areas of the circum-Gulf region indicates that majo. regional unconformities formed at the mid-Miocene (10.5 Ma), mid-Oligocene (30 Ma), mid-Cretaceous (97 Ma), and top-Jurassic (131 Ma). Three distinct periods a.e recognized in the depositional history of the Main Pass area of the northern Gulf of Mexico: (1) shallow ma.me deposition du.ins the period from the opening of the Gulf to the mid-Cretaceous, (2) deep marine deposition in the Cretaceous to the mid-Oligocene, and (3) shallow marine deposition prevailed since the mid-Oligocene to present. A comparison of depositional rates between the Main Pass area and the Destin Dome area indicates that the northern Gulf of Mexico continental margin was initiated as a terrigenous sediment wedge province in the late Cretaceous.

  • Diacylglycerol kinase (DGK) phosphorylates the second messenger diacylglycerol (DAG) to phosphatidic acid and it may play a role in signal transduction in Escherichia coli as well as in eukaryotic cells. In addition, DGK is important for microorganisms to adapt to several physiological stimuli. In Bacillus subtilis, the effect of stress on dgk transcription was examined by northern hybridization. The high level of dgk transcription was induced against high osmolarity, low pH value and low temperature. Transcriptional analysis revealed that the dgk gene and dgk upstream locus (ORF2, ORF3 and ORF4) were transcribed as a polycistronic mRNA to form an approximately 2.5 kb transcript.

The Effects of Single Component of Ginsenosides on the Mechanism of Mediator Release in the Allergic Hypersensitivity (인삼 사포닌 단일물질이 알러지 과민반응의 매개체 유리기전에 미치는 영향)

  • Inflammatory diseases, allergic and asthmatic disorders are caused by the mediator release from the activation of the phospholipase C (PLC), phospholipase D (PLD), methyltransferase or adenylate cyclase etc. during IgG or IgE cross-linking of high affinity receptors on mast cells or basophil surface. One important enzyme activated after IgG or IgE receptor cross-linking is PLD, the enzyme which converts phosphatidylcholine (PC) to phosphatidic acid (PA). Under the hypothesis that these may be some differences in mediator release according to the difference in PLD activity, we attempted to confirm the ginseng saponin effects on the PLD activity. We examined the PLD activity during the passively sensitized mast cell activation in the presence of single component of ginsenosides $(Rc,\;Rg_1,\;Rg_2,\;Rg_3)$. We also measured the amount of mediators (histamine and leukotrienes) released by stimulating with ovalbumin (OA) or calcium ionophore (CaI), Guinea Pig lung mast cells were purified using enzyme digestion, count current elutriation, and discontinuous Percoll density gradient. In purified mast cells prelabeled with $[^3H]$ arachidonic acid or $[^3H]$ palmitic acid, PLD activity was assessed more directly by the production of labeled PEt by PLD-mediated transphosphatidylation in the presence of ethanol. Histanine release was determined by Spectrophotofluorometry, and leukotrienes by radioimmunoassay. The PLD activity during the passively sensitized mast cell activation is increased up to $3{\sim}5times$. The PLD activity during the passively sensitized mast cell activation in the presence of all ginsenosides is decreased up to $4{\sim}11$ times. $Rg_l\;and\;Rg_2$ ginsenoside pretreatment decreased histamine and leukotrienes by 50% in the OA-induced or by 40% in the Cal-induced mast cell after passively sensitization. Rc pretreatment poorly decreased histamine but leukotrienes decreased by 70% in the OA-induced or by 35% in the Cal-induced mast cell. $Rg_3$ ginsenoside pretreatment increased histamine release without challenging OA or Cal but leukotrienes decreased. These observations indicate that single unit of ginsenosldes may be an important contributor to inhibit the release of histamine and leukotrienes in the guinea pig lung mast cells, that inhibits the PLD-mediated formation of DAG evoked by mast cell activation.
