To study the relationship between oxygen tolerance and enzyme activity in the oxygen metabolism of bifidobacteria, the activities of catalase, superoxide dismutase (SOD), NADH oxidase and NADH peroxidase from six typical bifidobacteria and other bacteria were assayed by spectrophotometry. Catalase activity was hardly detected in any of the bifidobacteria tested. SOD activity was detected in every species including the Clostridium species. In particular SOD activity was notably high in the aerosensitive Bifidobacterium adolescentis. This fact indicates that SOD activity is not a critical factor to ensure aerotolerance. Aerosensitive B. adolescentis showed very low NADH oxidative enzyme activity whereas other aerotolerant bifidobacteria exhibited considerable activity for the enzymes. It seems that detoxification of $H_2O_2$ by NADH oxidative enzymes might be an important factor in improving for aerotolerant bifidobacteria survival rates in an oxygen environment.

The effect of levanheptaose produced by levanase from Streptomyces sp. 366L on principle intestinal microflora was investigated. The reaction product, levanheptaose, was used as a carbon source for various intestinal microflora. As a results, Bifidobacterium adolescentis, Lactobacillus acidophilus, and Eubacterium limosum grew effectively in the in vitro experiment, whereas Clostridium perfringens, E. coli, and Staphylococcus aureus did not. Therefore levanheptaose seems to promote selectively the growth of B. adolescentis and L. acidophilus. In the in vivo experiment, the effect of levanheptaose on the growth of intestinal microflora, $\beta$-fructosidase activity, pH, and butyrate concentration were examined in rats. Apparently, the number of fecal Bifidobacteria, the amount of butyrate, and $\beta$-fructosidase activity were increased, whereas total aerobes and pH were reduced in rats fed by levanheptaose diets, compared with those of control diets. We concluded that those effects may be beneficial in improving gastrointestinal health.
The purpose of this research was to investigate the effects of nondigestible oligosaccharides (NDO) from red ginseng marc on the growth of Bifidobacterium spp. Red ginseng marc, a fibrous byproduct of ginseng extract from processing, was destarched by ${\alpha}$-amylase and amyloglucosidase treatment, and then treated with a commercial pectinase to produce NDO. The bifidogenic effects of NDO on B. adolescentis, B. animalis, B. breve, and B. longum were investigated in vitro. NDO significantly promoted the growth of Bifidobacterium spp. The growth, decrease of pH, and organic acid formation (acetate, lactate, formate) were markedly different among the species. B. adolescentis showed the best growth and produced the greatest amount of organic acids. When NDO was used as a carbon source in the cocultivation of Bifidobacterium spp. and Clostridium perfringens, the growth of Bifidobacterium spp. was not influenced by the existence of Cl. perfringens. The result strongly suggested that NDO from red ginseng marc could be used as a potential bifidogenic source.
항암 활성 유산균이 상용화되기 위해서는 장내 미생물균총의 개선에 의한 발암물질의 생성 억제능과 장내 면역계 활성화에 의한 암세포 증식 억제능 외에도 기초적인 생리활성능을 겸비하여야 한다. 본 연구는 항암 활성능이 우수한 유산균에 대한 기초 생리활성을 비교하므로서 살아있는 균주로서의 이용성을 확인하는데 그 목적이 있다. 암세포 억제율이 60% 이상인 5균주를 이용하여 내산성, 내담즙성, 항생제 내성, 우유 적용능, 안정성, 정착능을 확인하였으며, 그 결과는 다음과 같다. 내산성은 Lb. plantarum KCTC3099를 제외하고는 pH 3.5 이하에서는 내성이 없었으며, 유산보다는 HCl에 대한 내성이 강한 것으로 나타났다. 항생제 MIC에서 Lc. lactis와 Lb. plantarum KCTC3099는 cotrimoxazol(128 mg/1), B. adolescentis Maeil-K8과 B. infantis Maeil-K9은 doxycylin과 gentamicin(4 mg/1)에서 내성이 높았다. Caco-2 세포에 대한 정착률은 B. infantis Maeil-K9 균주가 3.1%로서 가장 우수하였으며, 나머지 균주들은 0.5% 이하로 매우 낮았다. 우유 배양능은 모든균주가 1 log cycle 이상의 균수 증가를 보였으나, Lb. plantarum KCTC3099를 제외하고는 산 생성능이 낮게 나타났다. 또한 우유 base에서의 안정성은 Lb. plantarum KCTC3099와 B. adolescentis Maeil-K8 및 B. infantis Maeil-K9이 우수한 것으로 확인되었다. 따라서 Lb. plantarum KCTC3099와 B. infantis Maeil-K9 균주가 항암 활성을 가지는 살아있는 균주로서의 활용성이 우수한 것으로 확인되었다.
The synergistic antimicrobial effect of Achyranthes japonica Nakai (AJN) and Bifidobacterium extracellular factors against Clostridium difficile were measured using a turbidity method. Each broth supernatant of Bifidobacterium infantis ($68.8{\pm}0.02%$) and Bifidobacterium adolescentis ($33.2{\pm}0.2%$) obtained by adding ethyl acetate soluble fractionate from A. japonica Nakai ethanolic extracts (AJNEA, 100 ppm, no inhibition) showed high synergistic antimicrobial activity against C. difficile. In addition, the antimicrobial activity in a laboratory medium and yogurt products against C. difficile were evaluated. In yogurt prepared with a starter 5 (Lactobacillus acidophilus: Streptococcus thermophilus: B. adolescentis =1 : 1 : 1) and a starter 4 (L. acidophilus: S. thermophilus: B. infantis=1 : 1 : 1) and 0.5% AJNEA powder, high antimicrobial effects were recorded that measured 79.0 and 65.2%, respectively. The results indicated the potential of AJN extract for use as an antimicrobial agent. In addition, the efficiency of the antimicrobial activity of the extracts was further improved in combination with lactic acid bacteria, which suggests that they have the potential to be used as a highly effective antibiotic-tolerant microorganism prevention system. Such a strategy can be used for alternative drugs or functional food additives for treatment of antibiotic-associated diarrhea.
Pseudomonas sp. K-52 유래의 levanase 에 의해 생산되는 levanoctaose 가 growth factor로서 각종 장내미생물에 미치는 영향을 조사하였다. In vitro 실험에서 0.5% levanoctaose를 탄소원으로 하여 glucose 일 경우와 상호 비교하여 분석한 결과 levanoctaose 는 Escherichia coli, Clostridium perfringens, Eubacterium limosum, Staphylococcus aureus 보다 Bifidobacterium adolescentis, Lactobacillius acidophilus, Bacteroid fragilis 등에 의해 효율적으로 이용되었다. 쥐를 이용한 in vivo 실험에서 탄소원의일부로 levanotaose를 제공한 경우, 장내 Bifidobacteria 수는 약 10배, butyrate 양은 2.3배, $\beta$-fructosidase 활성은 약 1.5배 증가하였다. 따라서 분리균주 Pseudomonas sp. K-2의 levanase로부터 생성되는 levanoctaose는 Bifidobacteria, Lactobacillus 와 같은 장내 유익균주에 선별적으로 이용되는 것이 확인되었다.
Bacillus polyfermenticus SCD was transformed by the recombinant shuttle vector for Bacillus and Escherichia coli containing 3 antibiotic resistant genes and an ${\alpha}$-amylase gene from Bifidobacterium adolescentis Int57. The ${\alpha}$-amylase gene fused to a secretion sequences was expressed under the control of the promoter of amylase gene from B. subtilis var. natto. The recombinant plasmid was maintained stably in the transformants producing the ${\alpha}$-amylase. The enzyme was secreted to outside of the cell and showed the similar enzyme activity as that of Bacillus subtilis BD170 under the same conditions of pH and growth temperature. Because of the relatively easy transformation and the secretion of the enzyme, the transformants of B. polyfermenticus SCD may give a new strategy in the production of foreign genes.
쥐의 사육실험결과 장내 균총 개선에 유효한 것으로 나타난 사과에 대하여 in vitro 배양실험을 통하여 이들의 유효성을 검증하였다. 사과로부터 분리한 crude pulp (CP), total dietary fiber (TDF), soluble dietary fiber (SDF), insoluble dietary fiber (IDF)와 시약용 사과 펙틴을 구입하여 PYF 액체배지에 첨가한 후 주요 장내 미생물의 표준균주들을 단독배양하여 O.D.와 pH를 측정함으로써 그 이용성을 조사하였다. 또한 이들 배지에 흰쥐의 분변 혼탁액을 종균으로 접종하여 혼합배양한 후 주요 장내 미생물의 균총변화를 조사하였다. 대부분의 장내 세균들이 사과의 IDF 첨가구에서보다 SDF 첨가구에서 더 잘 생육하였으며 특히 Bifidobacterium adolescentis, B. animalis, B. infantis, B. longum, B. thermophilum 등 Bifidus균들은 SDF 첨가구에서 비교적 높은 이용성을 나타냈으나 시약용 펙틴을 첨가한 시험구에서는 실험에 사용된 대부분의 균들이 거의 생육하지 않았다. 그러나 흰쥐의 분변 미생물들을 혼합배양하였을 때 사과의 섬유질 특히 수용성 식이섬유질(SDF)과 펙틴질 첨가구에서 포도당 첨가구에 비하여 Bifidobacterium의 수가 다량 검출되었다.
Adherence of probiotic bacteria to intestinal epithelium is found to be the most principal characteristics among the various physiological functionality. This study was conducted to investigate the effect of bifidobacterial growth properties and condition on the Caco-2 cell adherence and to construct a basic data on adherence-related research. Among 20 strains of bifidobacteris tested, when measured by cell surface hydrophobicity(CSH) and cell agglutination(CA), Bifidobacterium bifidum ATCC29521, Bif. adolescentis K8, and Bif. infantis K9 were selected. Using these strains, variations of Caso-2 cell adherence depending upon experimental condition were analyzed. The results obtained are as follows : Even though Bif. bifidum ATCC29521, Bif. adolescentis K8, and Bif. infantis K9 reached more 85% cell surface hydrophobicity there was no significant difference in cell agglutination, when reached 31.54$\pm$0.54mg/ml. By direct count method for adherence, viable cell count of M3, K1, K2, K8, K9 and K10 reached more 100 counts per 100 Caco-2 cells. When Bif. bifidum ATCC29521, Bif. adolescentistis K8, and Bif. infantis K9 were used to compare the adherence depending upon viable cell counts, reaction time, and growth phase, the more viable cell count, and the more adhered cell counts, the less adherence percentage. In addition, there was no difference in adherence percentage of bifidobacteria when bifidobacteria was incubated from 1 to 8 hrs after Caco-2 cells already formed monolayer. Considering of the effect of growth phase of bifidobacteria on adherence variation, all strains showed the highest adherence during the early stage of stationary phase. In conclusion, adherence of bifidobacteria was affected by strain specificity, viable cell count, and growth activity.
