수종 생약에 의한 Lens Aldose Reductase의 억제 (Inhibition of Lens Aldose Reductase by Medicinal Plants)

  • To search for inhibitory activities for lens aldose reductase, twenty two medicinal plants were tested by the method of bovine lens aldose reductase inhibition test. In this test Rubia akane(68%), Artemisia selengensis(61%), Lespedeza cuneata (58%), Ligustrum lucidum(58%), Viola patrinii(58%) extracts were shown to have aldose reductase inhibitory activity. From the radix of Rubia akane, $2-methyl-1,\;3,\;6-trihydroxy-9,\;10-anthraquinone-3-O-(6'-O-acetyl)-{\alpha}-rhamnosyl-(1{\rightarrow}2)-{\beta}-D-glucoside$ was isolated and the $IC_{50}$ of this compound in the aldose reductase inhibition test was about $1.2{\times}10^{-6}M$.

Aldose Reductase Inhibitory Activity of Methanol Extracts from the Korean Plants

  • We examined methanol extracts prepared from the Korean plants for their inhibitory effects on rat lens aldose reductase (RLAR) activity in vitro. Among 41 plant extracts, the extracts of Lagerstroemia indica, Punica granatum, Eurya japonica, Liquidambar styraciflua, and Vitis coignetiae exhibited highest inhibitory potency, even more potent than tetramethylene glutaric acid (TMG), known as one of typical aldose reductase inhibitors $(IC_{50}\;value,\;0.51\;{\mu}g/ml)$. Especially, the extract of Lagerstroemia indica (Lythraceae), exhibited the most inhibitory potency $(IC_{50}\;value,\;0.069\;{\mu}/ml)$ on RLAR.

Purification of Aldose Reductase and Decolorization of Dye by the Enzyme

  • Aldose reductase was purified to electrophoretic homogeneity from porcine liver. The purified enzyme was a monomer of 36 kDa. The enzyme was strongly inhibited by $Cu^{2+}\;and\;Mg^{2+}$ ions. Incubation of the enzyme with pyridoxal 5'-phosphate led to complete inhibition of enzymatic activity, suggesting that lysine residue is involved at or near the active site of the enzyme. The enzyme exhibited a broad substrate specificity. Furthermore, the enzyme was capable of decolorizing Alizarin, an anthraquinone dye.

  • The present study was undertaken to elucidate the hypoglycemic effect and inhibitory effect of Cudrania tricuspidata root bark on aldose reductase activity. C. tricuspidata MeOH ext. 1,000 mg/kg showed a significant blood glucose lowering effect on alloxan-induced hyperglycemic rats and increasing body weight. C. tricuspidata MeOH ext. showed a potent inhibitory effect on bovine lens aldose reductase.

Aldose Reductase Inhibition Effect of PhenolicvCompounds Isolated from Paulownia coreana Bark

  • Nine compounds, caffeic acid, naringenin, apigenin, luteolin, kaempferol, verbascoside, isoverbascoside, isocampneoside II, and cistanoside F, were isolated from the EtOAc and n-BuOH fractions of P. coreana bark. The structures of these compounds (1-9) were elucidated by spectroscopic methods and literature data. All the isolates were subjected to in vitro bioassay to evaluate their inhibitory activity against rat lens aldose reductase. Among these, compounds 6 and 8 indicated the significant inhibitory activity on rat lens aldose reductase with $IC_{50}$ values of 2.67 and 5.59 ${\mu}M$, respectively. Especially, The inhibition activity of acteoside was 3.9 times better than that of quercetin as a positive control (10.6 ${\mu}M$). These results suggested that phenylethanoid glycosides are likely to be the potential compounds for the prevention and/or treatment of diabetic complications.

Inhibitory Effects of Naturally Occurring Flavonoids on Rat Lens Aldose Reductase

  • Aldose reductase (AR), the key enzyme of the polyol pathway, is known to play important roles in the diabetic complications. The inhibitors of AR, therefore, would be potential agents for the prevention of diabetic complications. In order to clarify the structural requirements of flavonoids for aldose reductase inhibitory activity, thirty flavonoids were examined. Among the thirty flavonoids, flavonols such as quercetin (5), reyneutrin (7), quercitrin (9), isoquercitrin (11), and avicularin (14) were found to exhibit much stronger AR inhibition. Lonicerin (10), amentoflavone (27) and sophoraflavanone B (30) were also showed strong inhibitory activity. Especially, quercitrin and reyneutrin exhibited the most inhibitory potency on rat lens (RL) AR. The results suggested that flavonol having the 7-hydroxyl and/or catechol moiety at the B ring exhibit strong activity. In addition, flavonols having 3-O-monosaccharide also showed stronger inhibition than free flavonols at the 3-position. These results suggested that quercitrin and reyneutrin are attributed to be the promising compounds for the prevention and/or treatment of diabetic complications.

Effects of Some Coumarin Derivatives on the Bovine Lens Aldose Reductase Activity

  • Nine coumarin dervatives were examined for thier inhibitory effects on bovine lens aldose reductase activity (bovine-LAR). More than 50% inhibition of BLAR activities was observed in the cases of treatments with decursin, decursinol, esculin, isoimperatorin, oxypeucedanine and isobergapten at the concentration of $10^{-4}$M. Especially, BLAR activity was completely inhibited by the treatment of decursin and decursinol at this concentration. At $10^{-5}$ M, only three coumarins-decursin, decursinol and isoimperatorin, were found to have still relatively higher inhibitory effect.

  • Aldose reductase (AR), the key enzyme in the polyol pathway, plays an important role in the development of the diabetic complications. None of Aldose reductase Inhibitor (ARI) has achieved worldwide use because of limited efficacy or undesirable side effects. Therefore, evaluating natural sources for ARI potential may lead to the development of safer and more effective agents against diabetic complications. Fifty two Chinese herbal medicines have been investigated for inhibitory activities on AR. Among them, twelve herbal medicines, Artemisia anomala (aerial part), Centella asiatica (aerial part), Scutellaria baicalensis (root), Senecio chrysanthermoides (whole plant), Gleditsia japonica (twig), Zizyphus jujube (twig, leaf), Citrus aurantium (fruit), Hydnocarpus anthelmintica (fruit), Potaninia mongolica (aerial part), Tribulus terrestris (fruit), Artemisia apiacea (aerial part) and Eclipta prostrate (aerial part) exhibited a significant inhibitory activity against AR. Particularly, Artemisia anomala, Centella asiatica and Scutellaria baicalensis showed four times more potent inhibitory activity than the positive control, 3,3-tetramethyleneglutaric acid (TMG).

Inhibitory activity of methanol extracts from different colored flowers on aldose reductase and HPLC-UV analysis of quercetin

  • The methanol extracts of flowers obtained from 67 plant species were screened for their inhibitory activity on aldose reductase (AR). Alnus japonica, Aster spathulifolius, Chionanthus retusus, Morus bombycis, Crysanthemum boreale, Persicaria tinctoria, Platycarya strobilacea, and Serratula coronata var. insularis exhibited potent aldose reductase inhibitory (ARI) activity. HPLC-UV analysis of quercetin, an AR inhibitory flavonoid, was performed on extracts showing strong ARI activity. Quercetin was detected in C. retusus, C. boreale, P. tinctoria, and S. coronata var. insularis at concentrations of 1.33, 1.56, 0.82, and $3.37mg\;g^{-1}$ extract, respectively, indicating that quercetin contributed to the ARI activity of these extracts. In the samples in which quercetin was absent, other compounds may be responsible for their potent ARI activity. These results serve as a basis for further studies regarding the bioactive components responsible for the inhibitory effects of various flower extracts on AR activity.
