  • 우리는 처음으로 바닷물에서 히알우론산 분해효소를 생산하는 균주인 Vibrio splendidus KCTC 11899BP를 분리하고 동정했다. KCTC 11899BP는 히알우론산(HA)이 기초 배지에 첨가 될 때에만 Hyaluronate lyase를 생산하며, 이 효소는 HA의 ${\beta}$-(1, 4) 결합을 분해하여 이당(disaccharide)을 생성시키는 효소로서 미생물에 의해 생산된다. 게놈 염기서열분석을 통해, KCTC 11899BP의 게놈은 2개의 염색체를 보유하는 다른 Vibrio sp.와 유사하게 각각 3,522 kb (contig 1)와 1,986 kb (contig 2)인 두 개의 원형 contig로 구성되어 있다는 것을 확인하였다. 또한 4,700개의 예측 오픈 리딩 프레임, G + C 함량 44.12%, 137개의 tRNA 유전자 및 46개의 rRNA 유전자를 포함하고 있다는 것을 확인했다.

골관절염 치료제 히알우론산 임상연구결과의 Systemic Review (Systemic Review of Hyaluronate for the Treatment of Osteoarthritis)

  • Background: The multitude of the therapeutic usefulness of intra-articular injection of hyaluronate on oateoarthritis of the knee is still in question. The objective of this systemic review was to elucidate both the therapeutic efficacy and the safety of intra-articular administration of hyaluronic acid for degenerative osteoarthritis of the knee joints. Methods: I searched MEDLINE and Korea Medical Database (KMbase) from January 1990 to April 2007 using a combination search terms for knee osteoarthritis and hyaluronic acid and a filter for randomized controlled trials. I extracted data on pain at rest, and during or just after movement, on joint function, and on adverse events. Results: Ten trials that reported usable quantitative information on any of the predefined end points were identified and included in the systemic review. Intra-articular injection of hyaluronic acid can decrease symptoms of osteoarthritis of the knee. The study revealed significant improvements in pain and functional outcomes with few adverse effects. However, there was significant between-study heterogeneity in the estimates of the efficacy of hyaluronic acid. Sub-group analysis showed that lower methodological quality such as a single-blind or single-center design resulted in higher estimates hyaluronic acid efficacy, and that patients older than sixty years of age and those with the most advanced radiographic stage of osteoarthritis were less likely to benefit from intra-articular injection of hyaluronic acid. Conclusion: According to the currently available evidence, intra-articular hyaluronic acid has been proven clinically effective for the patients bearing the knee osteoarthritis with NSAID-induced GI troubles or inapplicable to any surgery, and may be associated with lower risk of adverse events.

Klebsiella sp. L-10 의 NTG 50 변 이주로부터 생산된 고점성 히알우론산 복합체의 정제 및 특성 (Purification and Characterization of High Viscous Hyaluronic Acid Complex from Klebsiella sp. L-10 NTG 50)

  • High viscous hyaluronic acid complex from Klebsiella sp. L-10 NTG 50 mutant was purified by two-phase extraction system using PEG-K2HP04 and its physicochemical properties were Investigated. Viscosity of the purified hyaluronic acid complex was decreased as temperature and salts concentration were Increased and also showed low viscosity at below pH 5.0 and above pH 11.0. Hardness, cohesiveness and adhesiveness of the purified hyaluronic acid complex were 1, 20kg, 1.91 and 0.62, respectively. Water holding capacity was 6.9ml per gram of the purified hyaluronic acid complex powder.

마우스에서 히알우론산 나트륨이 항염효과에 미치는 영향에 관한 연구 (The effect of hyaluronic acid on anti-inflammatory action in mouse)

  • Purpose: The purpose of this study was not only to evaluate the relative mRNA expression of interleukin-$1{\beta}$(IL-$1{\beta}$), cyclooxygenase2 (COX-2) and prostaglandin E2 (PGE2) by RT-PCR analysis but to observe pattern of edema by light microscopic and electron microscope after topical apply of hyaluronic acid in inflammation-guided mouse. Material and methods: Mice of this study were devided into 4 groups: Control group (no inflammation guided), Positive control (inflammation guided + vaselin apply), Protopic group (inflammation guided + protopic apply), Hyaluronic group (inflammation guided + hyaluronic acid apply). Results: Hyaluronic group showed less expressions of IL-$1{\beta}$, COX-2, PGE2 than those of positive control & protopic group. Hyaluronic group revealed a decreased inflammation than positive control & protopic group in Light Microscope. Hyaluronic group appeared decreased edema of ear compare to positive control & protopic group in Elecron Microscope. Conclusion: It was considered that hyaluronic acid has an antiinflammatory effect for intercepting the gene expression of cytokines related to inflammation.
