This study was performed to determine whether the infusion of nucleosides and a nucleotide mixture directly ito intestinal lumen can induce a regenerative effect on impaired intestinal mucosa. The effects of massive small bowel resention and also total parenteral nutrition were induced by surgical creation of Thirty-Vella fistual in male Sprague-Dawley rats. The rats received saline solution (Control group) or nucleosides and a nucleotide mixture(lower concentration group(Nucl) or higher concentration group (Nuc2) every two days into the fistula. Mucosal protein, DNA , ornithine decarboxylase(ODC) activity, and morphometry were evaluated at 9 or 21 days postoperation in the fistual and also in the residual ileal segment. On the 9th day, mucosal protein, DNA content, and villous surface area in the fistula and also in the residual ileum increased in rats that received nucleosides and a nucleotide mixture of lower concentration (Nuc 1). On the 21 th day, there were no significant differences in intestinal mucosa between the control group and the lower level nucleoside nucleotide mixture-treated group. The fistula villous height of the higher nucleosides and a nucleotide mixture group was higher than in the control rats. Fistula mucosal ODC activities were not significantly different between groups although the mucosal ODC activity of the residual ileal segment was increased on the 9th day. Our data suggests that this animal model is suitable for studying the effect of dietary factors on intestinal mucosal growth and regeneration after villous stropy , differentiating direct effects of diet on the intestine from systemic effects. It is also suggested that external nucleosides and nucleotides have supportive effects on intestinal mucosal regeneration.
The effects of lactose on the formation of casein phosphopeptide (CPP), the increment of soluble calcium and the enhancement of calcium absorption by dietary casein in the intestinal tract were investigated. Rats were fed a lactose-free diet, 10% lactose diet and 30% lactose diet containing powdered milk. In rats receiving a lactose-free powdered milk diet, CPP formation was confirmed by gel filtration of the intestinal content on Sephadex G -25 and the amount of soluble calcium was increased in the small intestine and calcium absorption, measured by the ligated ideal loops in situ was enhanced. However, In rats receiving a powdered milk diet containing 10% lactose or 30% lactose, the similar effects were not seen. These observations indicate that CPP-stimulated effects on soluble calcium and calcium absorption in the small intestine are not dependent upon lactose.

This study was carried out to investigate the motor activity and glucose transport and metabolism of Gaewool-Whadam-Jian(GWJ) in rat gastro-intestinal tract. The motor activity of the rat gastro-intestinal tract has been investigated by means of measuring barium sulfate passage degrees. Atropine treatment significantly delayed barium sulfate transit, and GWJ pretreatment increased intestinal motor activity, but not significant. GWJ administration showed no toxicity to kidney and liver. Transport and metabolism of glucose were studied in everted sac of rat small intestine with incubation under several conditions. The transport and metabolism of glucose were greater at jejunum than ileum. So, everted jejunum of rat were used to study the effect of GWJ. When GWJ were treated, the concentration of glucose were higher than untreated group. This result was thought to be influenced by the glucose in GWJ. When 2, 4 dinitrophenol and phlorizin were treated, the transport and metabolism of glucose were decreased, but GWJ treated together, the concentration of glucose in serosal solution increased. Gastric juice secretion and total acidity significantly decreased by administration of GWJ through duodenum region. The mechanism of effect of GWJ was still unidentified, Dut through continuous investigation, the effect of GWJ should be investigated.

난소화성전분(resistant starch, RS)는 건강한 사람의 소장에서 흡수되지 않는 전분과 분해된 전분의 산물을 총칭하는데. 대장에서 발효와 미생물성장, 식후 혈당, 변부피, 장통과시간, 식품의 열량가 등에 영향을 미치므로 생리적인 이점을 갖는다. 본 연구에서는 고지방식이와 콜레스테롤을 섭취란 흰쥐에서 제조과정이 다른 RS의 섭취가 저 열량원으로서 체중감소나 혈액내 지방성분을 낮추는 효과가 있는지를 알아보는 것을 목적으로 하였다. 연구방법으로 실험동물은 생후 4주된 Sprague-Dawl ey계 수컷 흰쥐로, 지방을 식이 무게의 20%, 콜레스테롤이 식이 무게의 1%로 첨가 된 고지방식이로 6주를 사육시켜 비만과 고콜레스테롤증을 유발시켰다. 고콜레스테롤이 유발된 실험동물은 섭취하는 전분의 형태가 다른 실험식이로 4주간을 더 사육하였고 사육기간이 끝난 실험동물들은 희생하여 생화학적인 실험- 혈액과 간의 총지방양, 중성지방양, 콜레스테롤양을 측정하였고, 면역능력으로 비장세포 증식, 혈장내 IgG, $C_3$양을 측정하였고. 혈장내 leptin 양을 측정하였다. 실험결과는 체중과 체내 저장지방무게가 난소화성전분 혼합군(RSNE)에서 유의적으로 감소하였다. 혈장내 지방성분을 보면, 총 지방의 경우 고지방대조군(HC)이 저지방 대조군(LC)에 비래 유의적으로 높았으며, RS를 섭취하면 유의적으로 낮아지는 경향을 보여 저지방을 섭취한 LC군에 비해서 낮은 수치를 나타내고 있다. 혈장 중성지방은 저지방 즉 고 탄수화물식이를 섭취하는 LC군이 유의적으로 높았으며 고지방을 섭취하는 경우는 실험군에 차이가 없었다. 혈장 총 콜레스테롤은 LC군에 비해 고지방과 콜레스테롤을 섭취하는 HC군이 유의적으로 높았으며. RSNE군에서는 유의적인 감소를 보였다. HDL-콜레스테롤에서는 실험군간에 차이가 없었다. 간내 총지방과 콜레스테롤량, 중성지방량은 LC군이 다른 군에 비해 유의적으로 적었으며 고지방섭취군들은 섭취하는 전분의 종류에 따른 차이가 나타나지 않았다. 면역능력을담당하는 체내 기관인 비장세포의 중식능력은 실험군간에 유의적인 차이가 없으나 혈장의 Ig G 양은 RSNE군이 유의적으로 높고, $C_3$ 양은 난소화성 전분 추출군(RSEX)에서 유의적으로 높게 나타났다. 혈장에서의 leptin 농도는 고지방 대조군인 HC군이 유의적으로 높았으며, RSNE군이 LC군과 같이 유의적으로 낮았다. 결론으로 난소화성 전분(RS)를 식이무게의 10%로 첨가하는 수준에서 순수한 RS보다는 RS제조과정 중 순수한 RS를 분리하기 전 단계인 RS를 포함한 전분이 체중감소, 체내 저장지방감소, 혈액 내 콜레스테롤을 저하시키는 효과를 보였다. 그러므로 RS의 섭취는 섭취형태에 따라 저 열량원으로, 혈액 내 지방성분을 낮추는 효과를 가진 기능성 식품소재로 사용될 수 있을 것이다.
Probiotics have emerged as a potential treatment modality for numerous gastrointestinal disorders, including IBD. However, few probiotics have undergone appropriate preclinical screening in vivo. Kefir is considered a probiotic, benefiting the host through its effects in the intestinal tract. Despite numerous studies examining the action of probiotics on the host organism, few have analyzed the effects on intestinal environment. We assessed the protective effect of kefir for three weeks before inducing colitis with 2% dextran sodium sulfate for five days. The DSS loads were similar in all DSS treatment group. The results of the experiment are as follows. Food intake and FER of experimental groups were not significantly different each other, but water consumption tended to be higher in all DSS treatment groups as compared with the normal control. And visual inspection of feces revealed mild diarrhea in rat given 2% DSS. The anti-inflammatory activity of kefir was determined by myeloperoxidase activity during the DSS treatment, and there was no significant difference in any group. The levels of thiobarbituric acid reactive substances (TBARS) as a colonic lipid peroxidation were significantly lower in the kefir intake groups than in rats treated with 2% DSS alone. The DNA % in tail and tail moment values as a DNA damage level of the blood lymphocytes in kefir intake groups tended to be lower than 2% DSS treatment alone, especially tail lengths were significantly diminished. According to the colonic histopathological assay, there were a severe inflammation of lamina propria and submucosa and mild edema in mucosa and sub mucosa in DSS alone treated group. We found a slight regenerative change in kefir treatment groups. In our experiments, this means that ulcerative colitis related to oxidative injury might be prevented by kefir as a probiotic. Further studies of the potential benefits of kefir as a probiotic in inflammatory condition are encouraged.

This study was conducted to investigate the bioavailability of starfish calcium with substances enhancing calcium absorption. Three week-old young female rats (Sprague-Barley) were divided into 5 groups according to calcium sources and testing agents; calcium carbonate (C), starfish calcium (S), starfish calcium + casein phosphopeptide (S-CPP), starfish calcium+citrate-malate (S-CM), starfish calcium+isoflavone (S-ISO), and were fed experimental diets containing AIN-93G based Ca (0.35% w/w) diet with CPP, CM and ISO for 6 weeks. Blood, femur, urine and feces samples were collected. There was no significant difference among groups in terms of growth and food intake. Serum Ca concentrations were normal in all 5 groups. Serum P concentrations and ALP activities were not significantly different among groups. Ca absorption and retention were significantly increased both in S-CPP and S-CM groups compared to C group (p<0.05). p absorption was significantly higher in S-CPP group than in other groups. While the amount of soluble Ca of intestinal contents did not differ among groups, the amount of insoluble Ca was significantly lower in S-CPP, S-CM and S-ISO groups than in C and S groups. However, the weight, Ca and P concentrations of femur were not significantly different among groups. These results suggest that the addition of CPP and citrate-malate were more effective for enhancing the bioavailability, intestinal absorption and solubility of starfish calcium.

Excessive intake of trans fats is a risk factor for cardiovascular diseases. Previously, we showed that green tea extract lowers intestinal absorption of cholesterol and elaidic acid in rats. This study was conducted to investigate a possible role for sorghum extract on lymphatic absorption of trans fat and cholesterol in rats. Adult male rats with lymph cannulae were infused at a rate of 3.0 mL/h for 8 h via a duodenal catheter with a lipid emulsion containing $146.4{\mu}mol$ of trielaidin, $36.8{\mu}mol$ of trilinoelaidin, $452.0{\mu}mol$ of triolein, $1.0{\mu}Ci$ cholesterol labeled with $^{14}C$ ($^{14}C-cholesterol$), $20.7{\mu}mol$ of cholesterol, and $396.0{\mu}mol$ of Na-taurocholate without or with 100.0 mg of sorghum extract in phosphate-buffered saline buffer (pH 6.4). Lymph was collected hourly for 8 h. No significant difference was noted in lymph flow. However, the lymphatic absorption of elaidic acid and linoelaidic acid for 8 h was significantly lower in rats infused with sorghum extract than in those infused with no sorghum extract. Further, lymphatic absorption of $^{14}C-cholesterol$ was reduced by sorghum extract, which was observed previously. These data indicate that sorghum extract has an inhibitory effect on the intestinal absorption of trans fat and cholesterol. The mechanism(s) by which sorghum extract lowers intestinal absorption of trans fat warrants further study.
A histopathological study was carried out on the duodenum of mice and rats experimentally infected by F. seoulensis. Each mouse was infected with 500 metacercariae and killed after 1, 2, 3 days, 1 and 2 weeks from infection. Each rat was given 1, 000 metacercariae and was examined after 1, 2 and 4 weeks from infection. The duodenal tissue sections of mice and rats were stained with hematoxylin eosin, and PAS stained for the rats of 1 week group. The pathological findings are summarized as below. 1. There were no differences in mucosal findings between the mice and the rats, and between the location of duodenum, 1 and 5 em distal to the pylorus. 2. Each worm embraced a villus exclusively with its foliate fore body which was inserted into the intervillous spaces. The fluke pinched villous epithelia using its oral and ventral suckers. The tribocytic organ destroyed the villous epithelia deeply up to the stroma after 3 days from infection. 3. Apparent villous changes were observed in the mice after 3 days from infection. Villous changes were shortening, widening, blunting or fusion. The villous stroma showed edema, microscopic hemorrhage, capillary congestion, dilatation of lymphatics and inflammatory cell infiltration. The cells were lymphocytes, plasma cells, eosinophils and giant cells. Rarely submucosal and trans:nural inflammation was encountered.

Although the sesame lignans, sesamol, have been shown to possess antioxidative activity, less is known about the metabolism and antioxidative properties of sesamol, a major constituent of sesame oil. To determine the ability of sesamol to act as an antioxidant in vivo, we fed rats a diet containing 0.5% sesamol for 3 wk and studied its metabolism and its effects on oxidative stress. Body weight gain and weight of liver, kidneys were significantly higher in the rats fed sesamol than in rats fed the control diets. GST and GST-Px activities in rat liver microsomes were higher in rats fed sesamol and CAT activities were found to be significantly increased in rats fed sesamol. The formation of TBARS was decreased in the liver of rat fed the 0.5% sesamol diet than in controls. We detected sesamol metabolites in liver and kidneys of rats fed sesamol and its metabolites were present as conjugated glucuronides and sulfates. In contrast, not detected sesamol peak in other organs such as colon, small intestine and pancreas.

In case of young rats, DNA, RNA and protein content of liver, kidney and spleen was less in one- meal group than in ad-lib group : their compositions per DNA were, however, similar in both instances In case of adult rats, the weight of liver increased significantly and their composition per DNA except lipid increased in one - meal group, but the total DNA content in the liver did not show any significant difference. The size of stomach and intestine tend to increase in meal -feeding group, but the serum cholesterol showed no significant change.
