Effects of Dietary Fiber and Fat Sources on Lipid Contents of Serum and Tissues in Rats Fed Cholesterol Diet (식이섬유와 지방의 종류가 고콜레스테롤식을 섭취한 흰쥐의 혈청과 조직의 지질성분에 미치는 영향)

  • The purpose of this research was to investigate the effects of 10%(w/w) of various dietary fiber, pectin(P), lignin(L), cellulose(C) and fiber-free (F) on lipid content of serum, liver and intestinal tissues in rats fed 10% coix oil (C)-or lard(L)-1% cholesterol diets for four weeks. The lard pectin(LP) group was the lowest In body weight gain and feed intake. Liver weight was not affected by dietary fiber and fat sources, and intestinal weight was higher in pectin(CP, LP) groups than in the other groups. Serum cholesterol was significantly lower in pectin(CP, LP) groups than in fiber-free(CF, LF) groups, and was higher in lard fiber-free(LF) than In the other groups(p<0.05). Serum triglycerides was significantly lower in coix oil pectin(CP) and lignin(CL, LL) than in fiber-free(CF, LF) groups. Liver total lipid and cholesterol content was significantly lower In pectin than In lard fiber-free group, and liver triglycerides was lower in coix oil pectin(CP) than in fiber-free groups (p<0.05). Lipid content of intestinal tissue was not affected by dietary fiber source. But total lipid and triglyceride content of intestinal tissues was significantly lower, and cholesterol content was higher in coix oil than in lard group(p<0.05). These results indicate that pectin and lignin reduced serum cholesterol. Pectin also decreased total lipid, cholesterol and triglyceride contents of liver in rats fed either coix oil-or lard-cholesterol diets whereas lignin did not affect liver lipid content.

Studios on the Metagonimus fluke in the Daecheong Reservoir and the upper stream of Geum River, Borea (대청호 및 그 상류의 Metagonimus 흡충에 관한 연구)

  • The prevalences of the cuke belonging to genus Metagonimus hove been reported along the upper stream of inhabitants by several workers since 1980, however the taxonomical problems of the fluke was not yet settled. The larval flukes; cercaria and metacercaria as well as their intermediate hosts, and adult were studied in order to identify the Mepagonimus in the areas. The results obtained are summarized as follows: 1. The snails, Semisulcospira globus were collected (rom the three different localities along the upper stream of the River. The cercariae were found from 125(7.2%) out of 1,730 snails by natural emerging method, and were identified into 5 species including Metagenimus sp. (3.7%), Pseudexorchis major(1.4%), Cercaria nipponensis (0.9%), Cercaria incerpa(0.6%), and Cercaria yoshidae(0.6%). Cercariae of Metagonimus species had four to dye oral spines on its anterior of the first line. 2. The cercariae of Metagonimus were experimentally exposed to goldfish. nfection rate was 22.9% out of 105 goldfish, and the encysted metacercariae were found in fins(86.7%) and on scales (13.7%) of the fishes, but not in their muscle, head or visceral organs. 3. Seven species of ask were caught in the Daecheong Reservoir and the upper stream. Infestations with metacercaria of Metagonimus were found 100% in Opsariichtys widens and the parasitized numbers of the metacercariae were observed from 250 to 2,400 per fish. In the upper stream, Zacco termmincki, Z. platypus and Pseudogobio esocinus were infected 100% with the metacercaria, on the other hand, the fishes caught in the reservoir showed the lower infestation rates, and a few metacercariae found in the fishes Carassius carassius and Cyprinus carpio in the reservoir and the stream. The majority of metacercariae was detected only on the scales of fishes. 4. In order to know the infectivity and the distribution patterns in the intestine of hosts, rats and dogs were infected with the metacercariae obtained from O. bidens and Z. platypus. In addition the metacercariae obtained from Z. temmincki, P. esocinus and goldfish were given to the rats. The recovery rates of the worms in the small intestine of dogs were higher (63.3~65.8%) than those of the rats (3.5~31.6%). The flukes were found mostly in the middle and the lower part of small intestine of the rats and the dogs, but no worm was collected in the upper part of the intestine of rats. 5. The sixte of adult flukes varied by the hosts. In the adult cukes, oral sucker was smaller than ventral sucker, and the right and left testes were located diagonally, the uterine tubules circled around the upper left testis. The average egg sixte was $29.1{\times}1.7{\mu\textrm{m}}$. According to the above results, the cukes belonging to genus Metagonimus distributed along the Geum River was concluded to be identical with Miyata type of M. yokogawai as that Saito had proposed.

Radiation-Induced Apoptosis and Mitotic Death in the Small Intestinal Crypts of Rat (방사선에 의해 흰쥐 소장에서 발생되는 세포고사 및 유사분열사)

  • Purpose : We investigated the temporal alterations of apoptosis and mitotic death following irradiation in the rat's small intestinal crypts. Materials and methods : Male Sprague-Dawley rats were irradiated 2 Gy by 6 MV linear accelerator and sacrified at 2, 4, 8, 24, 48 hours after irradiation. The mean numbers of the apoptotic cells and mitotic cells per their small intestinal crypts were measured in the unirradiated control and irradiated groups. To compare with H & E staining, ISEL (In Situ End Labelling) were peformed in the group having the highest apoptotic count. Results : The mean number of the apoptosis per crypt in the control group was 0.14 and those at 2, 4, 8, 24, 48 hours after irradiation were 1.43, 3.19, 1.15, 0.26, 0.17, respectively. So the apoptosis development was increased upto 4 hours and then normalized around 24 hours following irradiation. The mean number of the mitotic cells per crypt in the control group was 1.29 and those at 2, 4, 8, 24, 48 hours after irradiation were 0.56, 0.47, 0.23, 0.65, 1.19, respectively. The mitotic cell counts following irradiation was decreased to 8 hours and recovered to the normal level about 48 hours. So the increment of apoptotic cell count was occurred earlier and more remarkable than the decrement of mitotic cell count after irradiation. According to the staining time, false positivity was found in the ISEL staining. Conclusions : The cell death in the small intestinal crypt developed by acute radiation damage was usually decreased to the normal level within $24\~48\;hours$ after irradiation and the apoptosis was thought to be more important process than the mitotic death.

  • An effect of carbon tetrachloride (CCl$_{4}$) was studied on the xanthine oxidase(XOD)activity of small intestine in male rats. Concomitantly a cause of increasing small intestine XOD was focused on an effect of actinomycin D and the kinetics of partial purified XOD frdm small intestine in CCl$_{4}$ intoxicated rats. An injection of CCl$_{4}$ to the rats showed an increase of small intestine XOD. In the pretreatment of actinomycin D before injection of CCl$_{4}$ to the rats, the XOD activities of small intestine were significantly decreased. In the partial purified enzyme preparation, the small intestine XOD in CCl$_{4}$ intoxicated rats showed the more increased Km and Vmax value with xanthine as substrate than that of control group.

  • The inhibitory activities of Amberlite active fraction, which was obtained from methanol soluble fraction of freeze dried slikworm urine, on the rat intestinal glycosidase-catalyzed enzymatic reaction were examined in in viro and in vivo experiments. Amberlite active fraction showed significant inhibitory effects on the hydrolysis of o-glycosidic bond, especially $\alpha$-1,4 bond. On the other hand, the inhibition on the hydrolysis of $\beta$-glycosidic bond was very weak. Oral administration of Amberlite active fraction resulted in a dose-dependent decrease in the blood glucose after an oral maltose load, and postprandial hyperglycemia in carbohydrate-loaded mice was suppressed by Amberlite active fraction at 60 mgHg in decreasing order of maltose, starch, sucrose and lactose. 60 mg/kg of Amberlite active fraction lowered the blood glucose level markedly after 18, 35, and 60 min after an oral maltose load and the antihyperglycemic activity was maintained upto 90 min. In alloxan-induced hyperglycemic mice, Amberlite active fraction at a dose of 100 mg/kg also significantly lowered blood glucose after an oral maltose load, and its efficacy was almost equivalent to that of acarbowe.

Effect of the Paclitaxel and Radiation in the Mucosa of the Small Bowel of Rat (흰쥐의 소장점막에 Paclitaxel(Taxol)과 방사선조사의 효과)

  • Purpose : Paclitaxel is a chemotherapeutic agent with potent microtubule stabilizing activity that arrests cell cycle in $G_2$-M Because $G_2$-M is the most radiosensitive Phase of the cell cycle, paclitaxel has potential as a cell cycle- specific radiosensitizer. This study was designed to investigate the ability of paclitaxel to increase the radiotoxicity in normal small bowel mucosa of the rat. materials and Methods : A sigle intraperitoneal infusion of paclitaxel (10mg/kg), and a single irradiation(8 Gy, x-ray) to the whole abdomen and combination of radiation(8 Gr, x-ray) 24 hours after paclitaxel infusion in the rats were done. The changes of jejunal mucosa, and kinetics of mitotic arrest and apoptosis in the jejunal crypt were defined at 6 hours - 5 days after each treatment histologically. Results : Paclitaxel blocked jejunal crypt cell in mitosis and induced minmal apoptosis. Mitotic arrest by paclitaxel was peaked at 6 hours after infusion and returned to normal by 24 hours. Radiation induced apoptosis and peaked at 6 hours and returned to normal by 24 hours. Combination of paclitaxel and radiation blocked crypt cell in mitosis at 3 days and induced apoptosis slightly at 6 hours and 24 hours and returned to normal by 3 days. The incidence of apoptosis in combined group at 6 hours was slightly higher than normal control but significantly lower than radiation alone group. The major changes of jejunal mucosa were nuclear vesicle and atypia which were appeared at 6 hours - 3 days and returned to normal by 5 days The degree of the mucosal changes are not different in 3 groups except for absence of inflmmatory reaction in radiation group. Conclusion : Mitotic arrest by paclitaxel was peaked at 6 hours and returned to normal by 24 hours and paclitaxel induced minimal apoptosis. Radiation induced apoptosis, peaked at 6 hours and returned to normal by 24 hours. Radiation-induced apoptosis was less in combined group which suggested that paclitaxel have a radioprotective effect when radiation was given 24 hours after paclitaxel infusion.

  • Evidence suggests that dietary indole, particularly rich in cruciferous vegetables, may reduce the risk of cardiovascular diseases. Endosulfan is a residual organochlorine pesticide, which is detected in fruits, vegetables, and crops. In this study, we investigated the effect of luminal indole on endosulfan transport in the small intestine in mesenteric lymph duct-cannulated rats. The duodenum was also cannulated and a fasting phosphate buffered saline-glucose solution was infused overnight at 3 mL/hr. After recovery, a lipid emulsion containing [2,3-$^{14}C$] endosulfan ($^{14}C$-endosulfan) was infused into the duodenum for 8 hours. The tested rats were infused with the same lipid emulsion, but with indole. Samples from the lymph-fistula were collected hourly, and the luminal contents and mucosa were collected at the end of the infusion. The lymph flow in the mesenteric lymph did not differ between the two groups. However, the intestinal absorption of both endosulfan and cholesterol were significantly decreased by indole. The amount of radioactive endosulfan, which remained in the mucosa, was greater in the indole-infused rats due to the decreased transport of endosulfan into the lymph. This study indicates that the indole decreases the intestinal transport of endosulfan into the mesenteric lymph.

Studies on Intestinal Trematodes in Korea XVIII. Effect of Praziquantel in the Treatment of Fibricola seoulensis Infection in Albino Rats (한국의 장흡충에 관한 연구 XVIII. 흰쥐의 Fibricolu seoulensis감염에 대한 프라지관텔의 치료효과)

  • The therapeutic effects of praziquantel on Fibricola seoulensis infection were studied by experimental infection of albino rats with the metacercariae obtained from the snakes. Total 51 albino rats were infected each with 500 or 1, 000 meta cercariae in number through intragastric tubes. One or four weeks la er the rats were treated with 1, 5, 10 or 20 mg/kg single dose of prazipuantel and sacrificed after 3 days or 10-120 minutes to search for the worms in their small intestine. The worm recovery rate at 3 days after the treatment was 10.7 % in 1 mg/kg dose group and o. 03~0.2% in 5, 10 and 20 mg/kg groups, while that of untreated control was 51. 3%. The minimum effective dose to treat F. seoulensis infection in the rats is considered to be 5 mg/kg in single dose. By observing the distribution pattern of worms in the small intestine after the treatment, dislodgement of the worms from the duodenum, their normal habitat, to the lower portions was recognizable within as early as 10 minutes. The majority of worms was found to have descended to the ileo-caecal portion after 60-120 minutes.

Effects of Dietary Calcium Levels on Lipid Metabolism in Rats Fed High Fat Diet with or without Supplemental Cholesterol (식이칼슘 수준이 콜레스테롤 첨가 또는 무첨가 고지방식 섭취 흰쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of dietary calcium on blood and tissue lipids of adult rats fed a high fat diet with or without supplemental cholesterol for 4 weeks. Male Sprague-Dawley rats were fed the experimental diets containing 18%(w/w) beef tallow and three levels of calcium 0.1%, 0.5% and 1.5%. The contents of total lipid, cholesterol, triglyceride and phospholipid in blood, liver, small intestine, aorta, small intestinal contents and feces were determined. Only in rats fed the diets containing 1% cholesterol the concentration of cholesterol in aortic serum and various tissues significantly increased, and then decreased with increasing dietary calcium intake. Another observation was that high Ca intake significantly facilitated the fecal lipid and cholesterol excretion and bowel movement. These results suggest that possible hypocholesterolemic effects of dietary calcium could be related to the hypercholesterolemia and to the increase in excretion of fecal lipid and cholesterol.

  • This experiment was carried out to investigate the effect of silkworm powder, mulberry leaves and mulberry root bark administered to S.D. rat on gastrointestinal function. Plot containing ten percent sericultural products(silkworm powder, mulberry leaves and mulberry root bark) was supplied with S.D. rat. The food efficiency ratio(FER) of S.D. rat fed with sericultural products were 0.1486~0.1573. The transit time also were 512.5~574.0 minutes, and transit speed were 25.05~29.11 mm/min. The pH of S.D. rat's feces fed with sericultural products were 5.95~6.72. The daily amount of S.D. rat's feces were 4.12~5.42g. As above results, sericultural products was evaluated to improve the function of S.D. rat's gastrointestine.
