Cell Surface Display of Cycloinulooligosaccharide Fructanotransferase Gene in Saccharomyces cerevisiae (Saccharomyces cerevisiae에서 Cycloinulooligosaccharide Fructanotransferase 유전자의 표층 발현)

  • The cycloinulooligosaccharide fructanotransferase (CFTase) gene (cft) from Paenibacillus macerans was subcloned into the surface display vector, pCTcon (GAL1 promoter). The constructed plasmid, pCTECFTN (9.0 kb) was introduced to S. cerevisiae EBY100 cell and then east transformants were selected on the synthetic defined medium lacking uracil and on the inulin containing medium. The surface display of CFTase was confirmed by immunofluorescence microscopy and its enzymatic ability to form cycloinulooligosaccharides(cyclofructans, CFs) from inulin. The total activity of the CFTase was reached about 5.52 unit/1 by cultivation of yeast transformant on YPDG medium. The optimized conditions determined were as follows; pH, 8.0; temperature, $50^{\circ}C$ ; substrate concentration, 5%; inulin source, Jerusalem artichoke. By the reaction with inulin, CFs consisting of cycloinulohexaose (CF6), cycloinuloheptaose (CF7), and cycloinulooctaose (CF8) were produced and CF6 was the major product.

  • Leuconostoc mesenteroides CJNU 0705 was isolated from a breast milk sample and identified by 16S rRNA gene sequencing and confirmed by its ability to produce dextran from tryptic soy agar plates supplemented with 2% sucrose. This strain can absorb various heavy metals including lead (Pb) and cadmium (Cd) which are both found in fine dust and have been shown to be harmful to human skin. In addition, Leu. mesenteroides CJNU 0705 has demonstrated antimicrobial activity against Propionibacterium acnes, the primary causative agent of acne. Given these traits it was natural to evaluate the use of this strain in the fermentation of several natural extracts from green tea, carrot, annual wormwood, parsley, broccoli, and corn silk, which are known to improve skin health, to see if it could increase their dextran content when supplemented with no sucrose, 2% sucrose, or 2% sucrose and 3% yeast extract. The extracts supplemented with both yeast and sucrose were found to produce the most dextran, which was confirmed by the scanning electron microscope (SEM) images. These results suggest that Leu. mesenteroides CJNU 0705 and its fermented natural extracts could be used as functional materials for improving human skin health.

Aroma Compounds Produced by the Yeast Hansenula saturnus var. saturnus Isolated from Soil (토양에서 분리한 Hansenula saturnus var. saturnus에 의한 휘발성 방향성분의 생성)

  • A yeast strain producing fruity-floral aroma was isolated from soil and identified as Hansenula saturnus var. saturnus. Glucose was found to be the best carbon source and sodium nitrate or phenylalanine as nitrogen source in terms of the nature and the intensity of the aroma produced by the isolated yeast. Seventeen compounds, mainly esters and alcohols, were identified in the ether-pentane extract of the culture broth by gas chromatography and/or coupled gas chromatography-mass spectrometry. Ethyl alcohol, isobutyl alcohol, isoamyl alcohol, phenethyl alcohol and their acetate esters together with ethyl caprylate were the major compounds in the aroma concentrate. Three unusual compounds, dibutyl disulfide, 3-methyl pentanoic acid and methyl pentanoate were also tentatively identified in the culture broth of the isolated yeast.

Effect of Feeding Live Yeast Culture on Performance of Laying Hens (생효모배양물의 급여가 산란계의 생산성에 미치는 영향)

  • In order to investigate the effect of feeding live yeast culture on the performance of laying hens, a feeding trial was conducted with 96 20-wk-old Hy4ine brown layers during their laying period of 60 wk. The live yeast culture used was a product from Saccharomyces cerevisiae that was cultured on the corn-based substrate followed by careful drying of whole material not to lose the viability of yeast. Three levels of yeast culture as 0.5, 1.0, and 2.0% for three treatments and 0% for the control were included in the experimental diets. The feeding trial was carried out for 60 wk from August 26, 1992 to October 26, 1993. To evaluate the performance of layers during cold or hot periods as affected by the yeast culture feeding, data from the 12-wk winter period and 12-wk summer period were separated and analyzed accordingly. During 60 wk of laying period hen-day egg production was slightly but significantly(P<.05) improved by feeding the yeast culture. The average egg weight and daily egg weight(g /day) were also increased by the yeast culture. Feeding the yeast culture did not increase feed intake but feed efficiency was improved significantly (P<.05). No significant difference was detected in egg or eggshell qualities between control and yeast culture-treated groups. Feed intake and egg weight were not affected by the yeast culture feeding under both cold and hot period, but egg production and feed efficiency during hot summer improved significantly by its feeding. This result indicates that the effectiveness of the yeast culture feeding is greater during summer than winter for laying hens.

Effects of polysaccharide (polycan) derived from black yeast in dexamethasone-induced muscle atrophy cell model (Dexamethasone으로 유도한 근위축 세포모델에서 흑효모 배양물 유래 polycan의 근위축 개선에 대한 효과)

  • Objectives : This study was conducted to evaluate the anti-atrophic effect of polycan in dexamethasone-induced skeletal muscle atrophy in vitro model. Methods : C2C12 myoblast were differentiated into myotube by 2% horese serum medium for 6 days, and then treated polycan extract at different concentrations for 24h. The effect of dexamethasone on the induction of muscle atrophy and expression of atrophy-related genes in differentiated C2C12 myotubes using a GSH, ROS, real-time PCR, western blots analysis. Results : The results showed that Treatment with polycan (100 and 200 ㎍/㎖) noncytotoxic levels on both myoblast and myotube. Polycan decreased the ROS level overproduced with dexamethasone and improved the depletion of GSH level. Dexamethasone showed a decrease in myotube diameter, which was associated with up-regulation muscle-specific ubiquitin ligases markers, such as atrogin-1, FoxO3, myostatin and muscle RING finger-1 (MuRF1), and down-regulation of myogenin, MEF2, Myogenic regulatory factor 5, 6 and MyoD. The results showed that polycan treatment significantly dose-dependently inhibited it. Furthermore, decreased expressions of PI3K/Akt signal pathway by dexamethasone were reversed by treatment with polycan. Conclusions : Thus, polycan suppresses dexamethasone induced muscle atrophy in C2C12 myotube in vitro model through activation of PI3K/Akt pathway and protective effect of improve skeletal muscle function.

Impact of Feeding Multiple Probiotics on Performance and Intestinal Microflora in Broiler Chicks (혼합 미생물제의 수준별 급여가 육계의 생산성 및 장내 미생물에 미치는 영향)

  • Feeding probiotics in broiler chicks still critical in several aspects. Thus, this study was conducted to investigate the impact of feeding multiple probiotics on performance, intestinal microflora, blood cholesterol and ND antibody vaccine titer in broiler chicks. Three hundred twenty one day old male broiler chicks(cobb ${\times}$ cobb) were divided into four levels of multiple probiotics(0, 0.1, 0.2, 0.3%) with five replicates for 35 days. Basal diets contained 21.5, 19.0% CP and 3,100 kcal/kg ME for starting and finishing period, respectively. Weight gain, feed intake and feed conversion were measured weekly. The number of Salmonella, E. coli, Lactobacillus, and yeast were examined from ileum and cecum at the end of experiment. ND vaccine titer, cholesterol were detected from sera. Weight gain of birds fed probiotics were 669.33, 679.75 at the level of 0.1 and 0.2% supplemental groups for starting period. It was also improved in those treatments for finishing period and higher than control for total period. Feed conversion tended to be improved compared to that of control by the supplementation of probiotics for the first three weeks and seemed to show the similar tendency for the rest of two weeks. It was 1.611, 1.621 for the entire feeding period and improved compared with control. Total salmonella, was not decreased in ileal digesta of birds fed the probiotics compared with control, whereas the number of yeast increased in 0.1% treatment. However, the number of Lactobacillus and yeast in cecum was higher than control. Even though the blood cholesterol seem to high in 0.1% probiotics treatment, the ratio of HDL to total cholesterol showed higher than control. ND vaccine titer of birds fed probiotics were significantly higher than control (P<0.05). These results 0.1% multiple probiotics would be possible to improve the performance of broiler chicks and ND vaccine titer.

Effects of Supplementation of Synbiotic Co-cultures Manufactured with Anaerobic Microbes on In Vitro Fermentation Characteristics and In Situ Degradability of Fermented TMR (혐기성 미생물로 제조한 synbiotics 혼합배양물의 첨가가 발효 TMR의 발효특성과 소실률에 미치는 영향)

  • This study was conducted to estimate the in vitro fermentation characteristics and in situ degradabilities of total mixed rations fermented by the synbiotic co-cultures composed of various anaerobic microorganisms in the rumen of cow. Seventy two TMR bags (4 treatments $\times$ 6 fermentation days $\times$ 3 replications) were manufactured for in vitro and in situ experiments. The experiment was composed of four treatments including the control, the mould and bacteria synbiotics (T1), the mould and yeast synbiotics (T2) and the bacteria and yeast synbiotics (T3). Each treatment had six fermentation days (1, 3, 5, 7, 14, 21 day) with three replications. Two rumen cannulated Holstein cows (550 ㎏ of mean body wt) were used for in situ trial, and a total of 96 nylon bags were retrieved from the rumen according to eight fermentation times (1, 3, 6, 9, 18, 24, 48 and 72 hr). The mean fermentation temperatures of TMRs by supplementation of anaerobic micoorganism co-cultures ranged from $22.97^{\circ}C$ to $26.07^{\circ}C$, and tended to increase steadily during the entire period. pH values of the F-TMRs ranged from 4.39 to 4.98 and tended to decrease with the extension of the fermentation period, and decreased by supplementation of synbiotics (p<0.05). The ammonia concentrations of F-TMRs were not affected by addition of synbiotic co-cultures during the early fermentation period (within 7 days), but was lowest (p<0.05) in T3 during the late fermentation periods (after 14 days). Lactic acid concentration of F-TMR was lowest in T3 at 1 day of fermentation, but was not different from treatments in the other fermentation days. Microbial growth rates of F-TMR reached a peak at 7 days of fermentation, and afterward tended to decrease. In in situ experiment, the DM disappearance rates were higher in T1 than the control during early fermentation times (within 3 hours), but was vice versa at 48 hours of fermentation (p<0.05). There was no significant difference in effective DM degradability among treatments. NDF and ADF disappearance rates in situ were similar to those of DM. From the above results, the supplementation of synbiotics, particularly the mould and bacteria synbiotics, resulted in improving the pH and concentration of lactic acid of F-TMR as parameters of fermentation compare to the control, and also had higher in situ disappearance rates of DM, NDF and ADF than the control at early fermentation time. However, effective DM degradability was not affected by supplementation of synbiotics.

Degradation of Tetrachloroethylene (PCE) by a Dechlorinating Enrichment Culture Fixed in an Anaerobic Reactor (탈염소화 미생물 부착 혐기성 고정막 반응기에 의한 테트라클로로에틸렌(PCE)의 분해)

  • A soil enrichment LYF-1 culture from a contaminated site, which could reductively dechlorinate 900 $\mu$M (ca. 150 mg/L) of tetrachloroethylene (PCE) stoichimetrically into cis-1,2-dichloroethylene (cis-DCE), was established and characterized. The enrichment culture can use yeast extract, peptone, formate, acetate, lactate, pyruvate, citrate, succinate, glucose, sucrose, and ethanol as electron donors for dechlorination of PCE. Addition of NO$_2$$^{[-10]}$ and NO$_3$$^{[-10]}$ as alternative electron acceptors showed complete inhibition of PCE dechlorination, but S$_2$O$_3$$^{-2}$ , SO$_3$$^{-2}$ and SO$_4$$^{-2}$ had no significant effect on PCE dechlorination. The enrichment culture was attached to ceramic media in an anaerobic fixed-bed reactor. The fixed-bed reactor showed more than 99% of PCE degradation in the range of PCE loading rate of 0.13-0.78 $\mu$moles/L/hr. The major end product of PCE dechlorination was cis-DCE.

Optimum Geometry of Glass Lined HOMEBASE Impeller for Gas-Liquid System of Low Viscosity Liquid (저점도 액 통기 교반용 글라스라이닝 홈베이스 임펠러의 최적 형상)

  • Glass lined impellers are corrosion resistant to most chemicals, including strong acids, and also have a smooth, non-stick surface, easy to clean and free from impurities in the process. Glass lined home base impeller is a multi-purpose impeller designed to stir a wide viscosity range of liquids from low viscosity fluids to high viscosity fluids, among others, cell culture, yeast culture, and beer fermentation pots, especially used for air-water system breathable stirring. The glass lining for HB impellers, which are simple in structure and competitive in performance, is essential to have upper and lower division in order to make the joint area between the impeller and shaft as small as possible. The upper and lower division of the impeller hardly affects the mixing performance, but the aeration performance. In this study, in order to optimize the shape of the Glass Lining HB impeller, a study was conducted on the effect of the angle between the upper and lower impellers, the clearance between the impellers, and the number of baffles on the aeration power. The optimal shape and baffle plate conditions for the Glass lined HB impeller were derived through the study results that the angle and the clearance between the upper and lower impellers decreased the ration of the power consumption with aeration Pg and that without aeration P0, Pg/P0.

Antimicrobial Activities of Natural Medicinal Herbs on the Food Spoilage or Foodborne Disease Microorganisms (식품부패 및 병원성미생물에 대한 천연약용식물 추출물의 항균효과)

  • Several medicinal herbs, which are nontoxic and have been used widely in traditional folk medicine, were extracted and antimicrobial activity of the extracts was investigated against various foodborne pathogens or food poisioning microorganisms. The ethanol extract of Sansa, Hwangryun, Cheukbaek, and Seokchangpo showed strong antimicrobial activities against Gram postive and Gram negative bacteria, whereas those of Sakunja, Sukjihwang and Baekji had little antimicrobial activities on microorganisms tested. Among medicinal herb extracts, ethanol extract of Coptis chinensis Franch (hwangryun) showed the strongest antimicrobial activity. Antimicrobial activity of ethanol extract of Coptis chinensis Franch was not destroyed by heating at $100^{\circ}C$ for 60 min and at $121^{\circ}C$ for 30 min, which is very stable over heat. The pH effect on minimal inhibitory concentrations (MIC) of the extract of Coptis chinensis Franch indicated that MIC was reduced with increasing the pH value of the medium. The inhibitory effect of partially purified substance from the ethanol extract of Coptis chinensis Franch on the growth of Listeria monocytogenes and Staphylococcus aureus was investigated. Growth of those strains occurred at the concentration of $100{\;}{\mu}g/mL$ and were inhibited at $500{\;}{\mu}g/mL$, whereas those strains was completely inactivated in the presence of $1000{\;}{\mu}g/mL$.
