Antimicrobial Activity of Allyl Derivate in Multi-purpose Solution as Natural Preservative (다목적용액에서 천연보존제로서의 Allyl 유도체의 항균활성)

  • Purpose: The purpose of this study is to investigate antimicrobial activity of ally derivative of garlic against eye diseases, and to get basic data for an application development of the derivative, as a natural preservative, to multi-purpose solution (MPS). Methods: Antimicrobial activity of allyl derivative of garlic, diallyl sulfide (DAS), against Pseudomonas aeruginosa (P. aeruginosa) and Staphyloccus aereus (S. aereus) was determined. After DAS treatment in MPS of no microbial activity, its growth inhibition effect to bacteria was also determined. Results: DAS concentrations of $IC_{50}$ against P. aeruginosa and S. aereus were found when both bacteria were treated with DAS concentration of 0.25% and 0.64%, respectively. At MPS test, a product from Company A did not show any effects against both bacteria, but one from Company B showed antimicrobial activity against both bacteria. Antimicrobial activity test of the product from Company B in MPS did not showed any significant difference on both gram-negative and gram-positive bacteria. For a trial application of antimicrobial activity of DAS as a natural preservative, DAS was added to MPS of Company A which did not show any antimicrobial activity at treated concentration. Addition of DAS into MPS of Company A resulted in antimicrobial activity of DAS, which suggested a possible application of DAS as a germicide for contact lenses. Conclusions: This study showed allyl derivative of garlic has antimicrobial activity against both P. aeruginosa and against S. aereus, but more effects on P. aeruginosa. The result suggests that allyl derivative derived from garlic can be developed to a preservative of MPS as a natural substance.

  • Effective and alternative strategies to control methicillin-resistant Staphylococcus aureus (MRSA) are consistently needed. Previous study presented that 7,10-epoxy-octadeca-7,9-dienoic acid (EODA) was produced from 7,10-dihydroxy-8(E)-octadecenoic acid through one-step heat treatment. Further studies confirmed that EODA was highly active against broad range of pathogenic bacteria including MRSA, promising development of a novel antibacterial agent to control MRSA. However, there are some practical huddles for industrialization of EODA, especially high cost for fine purification. To address this problem, this study was focused on determination of any changes in the antibacterial activities of EODA when used as a crude extract. As a result, any significant changes in the antibacterial activities of EODA was not detected and additional synergistic effect for commercial antibiotics on antibacterial activity was sustained as it was.

급식소 조리종사자의 손 위생교육 효과 평가

  • 단체급식소 조리종사자의 손 위생실태를 알아보기 위해 일반세균ㆍ대장균군ㆍ황색포도상구균에 대한 미생물 검사를 실시하고, 조리종사자의 개인위생 개선을 위해 개인위생의 중요성ㆍ각종 식중독의 발생현황 및 예방대책ㆍ손 위생의 중요성 등에 대해 단계별 위생교육 프로그램을 실시한 후 최종 미생물학적 검증을 통해 위생교육 효과를 평가하였다. 위생교육 전 손 위생실태는 작업 전의 경우 일반세균수는 4.04∼1.49 log CFU/hand, 대장균군수는 6.52 log CFU/hand 이하였고, 황색포도 상구균의 경우 전체의 10% 정도가 검출되었다.   • 1991년∼2000년의 우리 나라 원인 균별 식중독 발생 상황을 보면 살모넬라균, 황색포도상구균, 그리고 비브리오균에 의한 식중독이 우리 나라 식중독 사고발생의 85∼90%를 차지하는데, 특히 전체 식중독 발생건수에 대해 살모렐라균에 의한 식중독이 차지하는 비율이 1993년에서 97년도 사이에 46.5%, 1999년도와 2000년에는 36.3%로 보고되었다. 이처럼 살모렐라균은 대표적인 식중독균으로서 우리 나라 뿐만 아니라 전세계적으로 세균성 식중독의 주요 원인이 되고 있는데 식육, 달걀 둥 동물성 식품 및 가공품, 어패류 등이 주요 원인식품으로 알려져 있다. Antibiotic Resistance in Staphylococcus aureus Isolated in Pusan (황색포도상구균의 항생제 내성 양상)

  • Antibiotic resistance patterns were determined for 14 strains of Staphylococcus aureus isolated at a hospital in Pusan during summer in 1989. Resistance to chloramphenicol or clindamycin was recorded in 100% of strains. Resistance to the other compounds tested was as follows: tetracycline 86%, gentamicin 79%, tobramycin 71%, kanamycin 71%, erythromycin 57%, ampicillin 57%, methicillin 50%, streptomycin 29%, cephalothin 29%, and trimethoprim 21%. All strains were sensitive to vancomycin and rifampicin. All strains showed multiple resistance to more than 3 antibiotics.

  • Seasonal distribution of various microorganisms was investigated in the effluent water in various sewage treatment plants from January to November in 2007. The target microorganisms were composed of total heterotrophic bacteria, Escherichia coli, coliform and enteric bacteria, Staphylococcus aureus, Yeast and Mold. The number of total bacteria and the indicator microorganisms, such as E. coli, coliform, and Enterobacteriacea, were increased during the summer season. S. aureus, however, was not influenced by the weather, showing higher number even in November and January in most sewage treatment plants investigated. Meanwhile, disinfection methods employed in various plants had no direct relationship with the growth of microorganisms, and small-scaled plants were found to be more effective than large-scaled ones in the sterilization of microorganisms.

  • Jeotgal is a generic term given to the high-salt-fermented seafood of Korea. This study aimed at developing an overview of the bacterial community present in Ojingeo-jeotgal, a highly consumed type of jeotgal, which is made with squid. Bacteria were isolated and purified from two samples on six different kinds of media and identified by 16S rRNA gene sequence analysis. Among the 121 total isolates, the most dominant genus was Bacillus, followed by coagulase-negative staphylococci (CNS) and lactic acid bacteria (LAB). CNS were detected in both samples, but LAB were observed in only a single sample. Six strains of Bacillus species inhibiting the growth of food pathogens, Staphylococcus aureus and Vibrio parahaemolyticus, were selected from the 121 isolates. These were found to inhibit the growth of both pathogens in addition to displaying proteolytic activities on media containing 6% NaCl and 2% skim milk.

  • This study was carried out to examine microbiological contamination of ready-to-eat foods (kimbab, sushi, salad, sandwich, sashimi) and to prove hazard of ready-to-eat foods by microbiological analysis. Collection of 440 samples of ready- to-eat foods were obtained from department stores, discount stores, super-markets in Seoul, Gyeonggi, Chungcheong, Gyeongsang, Honam areas. Sushi showed the highest detection rate of micro organism with value of20.4%, and then detection rates of kimbab, sashimi and salad were 13.0%, 12.5% and 6.9%, respectively. S. aureus was the most detected microorganism with value of 6.6%, and then Coliform and L. monocytogenes were detected 2.7% and 2.3%.

Prevalence of Food Poisoning Bacteria on Hands in Various Age Groups (손 위생에 대한 식중독 원인균 실태조사)

  • Spread of pathogenic micro-organisms through contaminated hands is a well recognized way of transmitting disease such as food poisoning. We investigated the prevalence of aerobic plate counts, coliform bacteria, and food-poisoning bacteria on hands in various age groups. The average number of aerobic plate counts was 3.3 log CFU/hand in kindergarteners, 3.4 log CFU/hand in elementary students, 3.2 log CFU/hand in middle school students, 3.4 log CFU/hand in high school students, and 3.3 log CFU/hand in adults. Two kindergarteners, 6 elementary students, and 2 adults were positive for the coliform bacteria. Among the food poisoning bacteria we tested, S. aureus was isolated from 47 individuals. Eight isolates of B cereus were all from kindergarteners. C. perfringens was isolated from 7 individuals. Among 47 isolates of S. aureus, 25 isolates produced toxins. Seven of eight isolates of B. cereus produced toxins. None of seven C. peifringens isolates produced toxins. All 47 isolates of S. aureus were sensitive to ciprofloxacin, trimethoprim/sulfamethoxazole, clindamyccin, imipenem, rifampin and vancomycin. Four isolates (8.5%) were resistant to cefepime, chloramphenicol, cefotetan, and gentamycin. Five isolates (10.6%) were resistant to oxacillin and 6 isolates were resistant to tetracycline. This study shows that it is needs to be established policy of school lunch and personal sanitation management.

Phosphate Concentration Dependent Degradation of Biofilm in S. aureus Triggered by Physical Properties (인산염 농도에 따른 물성 변화로 발생하는 황색포도상구균 바이오필름 제거 현상)

  • The objective of this study was to establish technology for removing bacteria with human- and eco-friendly material. Staphylococcus aureus as an important component for balanced equilibrium among microbiomes, was cultured under various concentrations of phosphate. Experimental observation relating to physical properties was performed in an addition of phosphate buffer. Statistically minimum value of size and hardness using atomic force microscope was observed on the matured biofilm at 5 mM concentration of phosphate. As a result of absorbance for the biofilm tagged with dye, concentration of biofilm was reduced with phophate, too. To identify whether this reduction by phosphate at the 5 mM is caused by counter ion or not, sodium chloride was treated to the biofilm under the same condition. To elucidate components of the biofilm counting analysis of the biofilm using time-of-flight secondary ion mass spectrometry was employed. The secondary ions from the biofilm revealed that alteration of physical properties is consistent to the change of extracellular polymeric substrate (EPS) for the biofilm. Viscoelastic characterization of the biofilm using a controlled shear stress rheometer, where internal change of physical properties could be detected, exhibited a static viscosity and a reduction of elastic modulus at the 5 mM concentration of phosphate. Accordingly, bacteria at the 5 mM concentration of phosphate are attributed to removing the EPS through a reduction of elastic modulus for bacteria. We suggest that the reduction of concentration of biofilm induces dispersion which assists to easily spread its dormitory. In conclusion, it is elucidated that an addition of phosphate causes removal of EPS, and that causes a function of antibiotic.
