Antimicrobial Activity of Garlic Extracts according to Different Cooking Methods (조리방법을 달리한 마늘 추출물의 항균활성)

  • This study was conducted to find the antimicrobial activity of garlic extracts by various processing methods(boiled, pan fried, microwave heated, pickled). Ethanol and water extracts from garlic sample were prepared and antimicrobial activities were determined against 10 microoganisms ; food borne pathogens, food poisoning microoganisms, food-related bacteria and yeasts. The ethanol extracts from the fresh and pickled garlic showed antimicrobial activities for test microoganisms, except lactic acid bacteria and yeast. However, the antimicrobial activities were decreased by heat treatment. The minimum inhibitory concentration(MIC) of the fresh garlic extracts was determined to 0.1 mg/mL against an gram positive bacterium and 0.5 mg/mL against an gram negative bacterium. The antimicrobial activities of the ethanol extracts were affected by heating methods and not by pHs.

Antimicrobial Activity of Solvent Fraction from Cornus officianalis (산수유의 용매분획별 항균활성)

  • To develop natural food preservatives, ethanol and water extracts were prepared from the cornus (Cornus of officianalis) and antimicrobial activities were examined against 10 microoganisms which were food borne pathogens and/or food poisoning microoganisms, food-related bacteria and yeasts. Ethanol extract exhibited antimicrobial activity for the microoganisms tested, except lactic acid bacteria and yeast. Especially, minimum inhibitory concentrations (MIC) of the ethanol extracts were determined as 0.25 mg/mL against bacteria and 2 mg/mL against target lactic bacteria and yeasts. Antimicrobial activity of the ethanol extracts were not destroyed by the heating at 121$^{\circ}C$ for 15 min and not affected by pH. The ethanol extract of cornus exhibiting high antimicrobial activities were fractionated in the other of diethylether and butanol fractions to test antimicrobial activity The antimicrobial activity adjust bacteria test was highest in the ethanol fraction.

  • The cyclodextrin glucanotransferase (CGTase) gene from Bacillus stearothermophilus NO2 was expressed in Saccharomyces cerevisiae 2805 under the adhl promoter. The CGTase was purified from S. cerevisiae 2805/pVT-CGTS. The purified enzyme exhibited a optima of activity around pH 7.0 and $65^{\circ}C$. Thermal stability of the enzyme was increased fairly as compared with the CGTase of B. stearothermophilus NO2. The conversion yield of cyclodextrin (CD) and the production ratio of $\alpha$-, $\beta$,-, ${\gamma}$-CD from starch were showed similarly aspect to the CGTase of B. stearothermophilus NO2.

  • Fibrinolytic activities of culture concentrates of various yeasts were investigated. The concentrates of the culture broth of Saccharomyces cerevisiae Y99-7 showed the strongest fibrinolytic activity of 25 mm (clear zone). The fibrinolytic activity of Saccharomyces cerevisiae Y99-7 was more high in the culture concentrates from PD broth rather than that of yeast extract-peptone dextrose cultures (clear zone : 22.7 mm).

  • The goal of this study and potent whitening tyrosinase inhibitor-producing wild yeasts and further optimiz production of tyrosinase. Among non-pathogenic wild yeasts obtained from soils in Daejeon city and spice field of Geumsan in Chungcheongnam-do, Korea, we selected Saccharomyces cerevisiae(S. cerevisiae) WJSL0191 and Papiliotrema laurentii (P. laurentii) ON30 show 33.2% and 27.3% respectively. These selected strains formed and not pseudomycelium. S. cerevisiae WJSL0191 was sugar-tolerant as well as halophilic in 20% glucose-containing yeast (YPD) medium and 15% NaCl-containing YPD medium. S. cerevisiae WJSL0191 and P. laurentii ON30 showed 26.2% and 18.6% anti-wrinkle elastase inhibitory activities, respectively. aximal production of tyrosinase inhibitors obtained when S. cerevisiae WJSL0191 was cultured at 30 for 72h in YPD medium and P. laurentii ON30 was incubated at 20℃ for 24hr.

  • To screen for potent anti-inflammatory compound-producing yeasts, we evaluated nitric oxide production inhibitory activities in RAW 264.7 macrophage cells using cell-free extracts from 182 non-pathogenic yeasts. Rhodotorula graminis YJ36-1 and Meyerozyma guilliermondii YJ34-2 showed high inhibitory activities of 57.4% and 47.0%, respectively. The microbiological characteristics of these yeasts were investigated. Rhodotorula graminis YJ36-1 formed ascospores and pseudomycelium. This species grew well at $25^{\circ}C$ in yeast extract-peptone-dextrose (YPD) medium, vitamin-free medium, and 5% NaCl-containing YPD medium. Meyerozyma guilliermondii YJ34-2 was an asporogenous yeast and did not form pseudomycelium. This strain also grew well at $30^{\circ}C$ in YPD medium, vitamin-free medium, and 5% NaCl-containing YPD medium.

Establishment of a Leucocin A Producing Sccharomyces cerevisiae Cell (박테리오신 leucocin A를 생산하는 Saccharomyces serevisiae 세포의 제작)

  • In order to establish yeast cells that produce leucocin A, a bacteriocin, the 117 bp leucocin A gene with start and stop codons was synthesized and cloned in pAUR123, a yeast vector. Transformed yeast cells showed antibacterial activity against Bacillus subtilis. The leucocin A gene was confirmed by means of PCR methods with plasmid prepared from transformed yeast cells as template and two leucocin A-specific primers. In this results, yeast cells that produce mass amounts of bacteriocin to use as food preservative or antibiotics were established.

Antimicrobial Activity of Extract (삼백초(Saururus chinensis Baill) 추출물의 항균활성)

  • In order to develop natural food preservatives, the ethanol and water extracts of the Saururus chinensis Baill were prepared. Antimicrobial activity was examined against 10 kinds of harmful microorganisms. The ethanol and water extracts showed the most active antimicrobial activity against B. subtilis and E. coli. The ethanol extract showed stronger antimicrobial activity than that of the water extract. However, the extracts did not show any antimicrobial activity against lactic acid bacteria and yeast. The minimum inhibitory concentrations (MIC) of ethanol extracts against B. subtilis and E. coli were 5 to 10 mg/mL, respectively. Antimicrobial activity of the ethanol extract was not destroyed at 40-12$0^{\circ}C$ and pH 3∼11. The ethanol extract was fractionated in the order of hexane, diethyl ether, ethyl acetate, and water fractions. The highest antimicrobial activity was found in the diethyl ether fraction.

Antioxidant Activity and Inhibitory Activities of Xanthine Oxidase and Tyrosinase of Yeasts from Wild Flowers in Korea (우리나라 야생화에서 분리한 효모의 항산화 활성과 Xanthine Oxidase 및 Tyrosinase 저해활성)

  • We prepared supernatants and cell-free extracts of yeasts isolated from wild flowers in Korea, and their antioxidant activity and inhibitory activities of xanthine oxidase and tyrosinase were investigated. Among them, cell-free extracts of Kuraishia capsulata UL40-2 and Sporobolomyces ruberrimus 121-Z-3 showed significantly high xanthine oxidase inhibitory activity of 46.4% and 48.3%, respectively. Starmerella bombicola 80-J-1 also showed tyrosinase inhibitory activity of 36.2% in the cell-free extract. Other antioxidant activities and tyrosinase inhibitory activities were not detected or were below 20%. Maximal production of the xanthine oxidase inhibitors were observed when Kuraishia capsulate UL40-2 and Sporobolomyces ruberrimus 121-Z-3 were cultured in the yeast extract-peptone-dextrose media at $30^{\circ}C$ for 24 hour, respectively.

Breeding of Ethanol-producing and Ethanol-tolerant Saccharomyces cerevisiae using Genome Shuffling (Genome shuffling을 이용한 에탄올 생산 및 내성 효모 균주의 육종)

  • To improve yeast strains for bioethanol production, yeasts with ethanol tolerance, thermotolerance, and ${\beta}$-1,3-glucanase activity were bred using yeast genome shuffling. Saccharomyces cerevisiae $BY4742{\Delta}exg1$/pAInu-exgA, which has extracellular ${\beta}$-1,3-glucanase activity, and the Aspergillus oryzae and S. cerevisiae YKY020 strains, which exhibit ethanol tolerance and thermotolerance, were fused by yeast protoplast fusion. Following cell fusion, four candidate cells (No. 3, 9, 11, and 12 strains) showing thermotolerance at $40^{\circ}C$ were selected, and their ethanol tolerance (7% ethanol concentration) and ${\beta}$-1,3-glucanase activity were subsequently analyzed. All the phenotypes of the two parent cells were simultaneously expressed in one (No. 11) of the four candidate cells, and this strain was called BYK-F11. The BYK-F11 fused cell showed enhanced cell growth, ethanol tolerance, ${\beta}$-1,3-glucanase activity, and ethanol productivity compared with the $BY4742{\Delta}exg1$/pAInu-exgA and YKY020 strains. The results prove that a new yeast strain with different characters and the same mating type can be easily bred by protoplast fusion of yeasts.
