In an attempt to improve biomass and bioactive compound production, we cultured adventitious roots of $Eleutherococcus$$koreanum$ in 250 mL Erlenmeyer flasks using Murashige and Skoog (MS) medium with different concentrations of auxins (IBA, NAA, IAA) and cytokinins (BA, kinetin, TDZ). Root biomass (fresh and dry weight) was enhanced at $5mg{\cdot}L^{-1}$ indole-3-butyric acid (IBA) after 5 weeks of culture. The content of total phenolics and flavonoids was also increased with $5mg{\cdot}L^{-1}$ IBA compared to ${\alpha}$-naphtalene acetic acid (NAA) or indole-3-acetic acid (IAA) treatments. The combination of $5mg{\cdot}L^{-1}$ IBA with $0.1mg{\cdot}L^{-1}$ thidiazuron (TDZ; N-phenyl-N'-1,2,3,-thidiazol-5-ylurea) enhanced root fresh and dry weight (1.4- and 1.6-fold, respectively) as well as the content of total phenolics and flavonoids compared to the relative control (without cytokinin). On the contrary, $N_6$-benzyladenine (BA) and 6-furfurylaminopurine (kinetin) did not significantly affect root biomass and bioactive compound production in adventitious roots of $E.$$koreanum$. These results suggested that $5mg{\cdot}L^{-1}$ IBA combination with $0.1mg{\cdot}L^{-1}$ TDZ supplementation was most suitable for both biomass and bioactive compound production from adventitious roots of $E.$$koreanum$.
Surfactant flushing for enhancing the removal of BTEX from contaminated sand/clay mixtures was investigated. Eight soil columns packed with relatively undisturbed BTEX contaminated soils, were leached with water, methyl alcohol and then flushed with surfactant with or without several additives. Initial concentrations of BTEX mixture range from 278mg/kg to 1975mg/kg. Initial BTEX removal efficiency was 98% when the contaminated soil was flushed with water of 850 pore volumes. Because of tailing effect, water flushing could not remove below 8mg/kg concentrations during the experimental period. Eventually, the most effective surfactant for flushing was turned out to be 4% SOFTANOL(equation omitted)-90 with 3% ethyl alcohol and 3% SXS. In interrupted flow conditions, the removal efficiency was 99.5% with the flushed water of 95 pore volumes. The BTEX mixture removed from the soil columns during the surfactant flushing ranges from 84.5% to 99.5% of the initial amount for both water leaching(850 pore volumes) and surfactant flushing(95-165 pore volumes), respectively. Test results indicated that surfactant flushing could enhance the removal of BTEX mixture from contaminated soils and could reduce the aqueous phase BTEX mixture concentration in leachate.
Chemical resistance of the cement mortar containing the Thermally Activated Diaomite(TAD) and H.M.(Heavy Metals) has been studied. The H.M.. extracted from EAF(Electrica1 Arc Furnace) Dust. were saturated with diatomite. The diatomite was then thermally activated at $750{\circ}C$ for 30minutes and powdeled. The powder was mixed with a portland cement on a weight basis from 0%. 2.5%. 5.0%. 10%. 20%. The optimum mixture. after those mixtures were subjected to compressive strength(7 and 28days) and leaching bchaviour of H.M.. was tested for its experiment on Wet/Dry cycles and chemical resistance(e.q. imrncrsion in 5%(Conc.) of H2S04, CaC12 and hlgSO4. It was shown that the cement, mortar containing 10% of' P.D. gave a rise to the remarkable increase in compressive strength. The compressive strength was generally decrease beyond the addition of 10% of P.D. The maximum $496kgf/cm^2$ of 28days compressive strength was acheiveti when 10% of P.D. was added to the cement mortar.
In the periods of July 31 to August 10. 1988 and March 9 to 13. 1989. sediment samples were collected from the South Sea stations (010] to 092]) located in the area from $N 32^{\circ}$/30' to $34^{\circ}$/30', of latitude and from E $123^{\circ}$ 30' to $128^{\circ}$30' of longitude. These samples were analyzed for the number of total heterotrophic bacteria and extracellular digesting enzyme activities. In the 1989 spring period the number of heterotrophic bacteria in the sediment surface layer was increased more than 100 times at the maximum compared to that in the 1988 summer period. The proportion of fresh water bacteria to total heterotrophic bacteria was also higher in the spring period than the summer period. The extracellular digesting enzyme activities were higher in spring season than summer. Although the water content of sediment in the spring period was lower than that the summer period. the ash weight indicating organic material content was higher. These results means that the diameters of sediment particles were larger in spring than summer but the input of organic material into the sediment was greater. Based on these results bacterial distributions in the sediment layer of South Sea depend greatly on the season due to the effect of fresh water. During the spring season plankton could grow extensively owing to the inorganic nutrients input by the vertical mixing in the water column, then be precipitated into the sediment. Organic nutrients supplied from enzymatic degradation of polymeric particle from plankton can increase the bacterial number, too.

The effects of forest fires on the activity of microbial communities in topsoil and subsoil were investigated. Samples were collected from Korean forest soils comprising mainly igneous and sedimentary rocks. Analysis of beta-glucosidase, found higher microbial activity in sedimentary rocks than in igneous rocks. Enzyme activity was not observed immediately after fire, but was restored over time. The enzyme activity of subsoil was inhibited by 33~46% compared with that in the topsoil, regardless of soil damage. The effect of fire on the availability of microbial substrate was investigated using EcoPlate. The percentages of average well color development values of damaged and normal topsoil were 52.7~56.8% and 62.3~83.6%, respectively. Forest fires appear to affect the diversity and substrate availability of the subsoil microbial community by accelerating the decomposition of soil organic matter. The Shanon index, representing microbial biodiversity, was high in the topsoil of all samples; it was higher for soil microorganisms in sedimentary rocks than in igneous rocks, and higher in topsoil than in subsoil.
In this study, we compare the quality of the mango cv. Irwin cultivated in Jeju and Okinawa by evaluating the total soluble solids (TSS) and antioxidant activity (AA) from ethanol extracts of the fruit. TSS and AA were higher in fruit cultivated at the Research Institute of Climate Change and Agriculture in Jeju (RICA) and Okinawa farm, where the low tree height cultivation method is used, than in fruit cultivated on farm in Jeju. Also, the mangiferin content of the Okinawa farm was higher than those produced on the RICA and Jeju farm house. These results suggest that the lowering tree height cultivation method improves the quality of the mango cv. Irwin.
The concentration of amino acids, total nitrogen, trehalose, lipids and the activities of respiratory, acid$\cdot$alkaline phosphatase, glutamic oxalozcetic transaminase and glutamic pyruvic transaminase during larval stage in Pine leaf gall midge, Thecodiplosis janensis Uchi. et Inouye were measured using Paper chromatographic method, micro-Kjeldahl method, Thin layer chromatographic method, Warburg's manometric method, Bessey-Lowry method and Reitman-Frankel method, respectively. Healthy specimens )yore chosen as samples of each larval stages; alrva in gall and larva in soil. Amino acids present in the alcoholic extracts were alanine, glutamic acid, glycine, histidine, methionine, proline, threonine, tryptophan and valine. The total nitrogen concentration reached to 31.348mg/g during the larva in gall and the larval stage in soil of the value was decreased to 29.027mg/g. The hemolymph sugar, trehalose value for larva in soil was about two times of the value for larva in gall. Total lipid, phospholipid, monoacylglycerol, triacylglycerol, sterol, free fatty acid and ester cholesterol were identified at larval stages in gall and soil. Triacylglycerol concentration reached high level in contrast with other lipid contents during larvae in gall and larva in soil. Free fatty acid, sterol except decreased lipids during larval stage in soil. Endogenous respiration, succinate of respiratory activities decreased at larval stage in soil compare with larva in gall. The activities of acid phosphatase decreased larval stage in soil but the activities of alkaline phosphatase increased remarkably. The activities of glutamic oxaloacetic transaminase and glutamic pyruvic transaminase reached high level of the larva in gall.
In order to enhance the efficiency of removal a series of ElectroKinetic Remediation (EKR) tests on ferrous soil contaminated by lead are carried out using acids, chelates and surfactant as flushing agents. The test results indicate that pH in the electrolyte rapidly reached at steady state as the introduce of flushing solution of the lower pH, the type of flushing solution have no effect the distribution of electrical voltage within the sample but the increasing of solution concentration increases it at x/L=0.9. In the distribution of the residual lead in the sample SDS is the highest. Also, the removal efficiency for acetic acid concentration of 1mM Is the highest but the concentration of acetic acid significantly have no effect.
This study was conducted to investigate the effect of activated charcoal on growth and yield of taro in different degree of activated charcoal concentration. The results obtained are summarized as follows. Stem length was shown the longest in activated charcoal of 10%∼20% , but shorter in control. Also stem diameter was shown the thickest in activated charcoal of 10%∼20%, but thinner in control. Length and thickness of main root and thickness of main root were good in activated charcoal of 10∼20% Quality and yield of taro was the highest in activated charcoal of 10∼20% . Therefore, optimum consistency of activated charcoal was 10∼20%.
In this study, a blast furnace slag with the alkaliphilic microorganism (Bacillus halodurans) alkaline activator was used to cement natural soils in the field. A low-cost and massive microbial solution for cementation of field soils was produced and compared with existing microbial culture in terms of efficiency. A field soil was prepared for three different cementation areas: a cemented ground with microbial alkaline activator (Microbially-treated soil), a cemented ground with ordinary Portland cement (Cement-treated soil), and untreated ground (Non-treated soil). The testing ground was prepared at a size of 2.6 m in width, 4 m in length, and 0.2 m in depth. After 28 days, a series of unconfined compression tests on the cement-treated and microbially-treated soils were carried out. On the other hand, a torvane test was carried out for non-treated soil. The strength of field soils treated with microorganism was 1/5 times lower than those of cement-treated soil but is 6 times higher than non-treated soil. The pH measured from microbially-treated soil was about 10, which is lower than that of cement-treated soil (pH = 11). Therefore, it is more eco-friendly than Portland cemented soils. The C-S-H hydrates were found in both cement- and microbially-treated soils through SEM-EDS analyses and cement hydrates were also found around soil particles through SEM analysis.
