Effect of Dietary Supplementation of Extracts of Mushroom Mycelium on Survival and Growth of juvenile Flounder, Paralichthys olivacceus (버섯균사체 배양액 첨가사료가 넙치 치어의 생존 및 성장에 미치는 영향)

  • The effects of mycelium cultural extract supplemented diet, hematology and disease resistance against Vibrio anguillarum in juvenile flounder, Paralichthys olivaceus were evaluated. Fish were fed the Phellinus linteus with Coriolus militaris versicolor mixed mycelium cultural extract supplemented diet, Phellinus linteus mycelium cultural extract supplemented diet and Coriolus militaris tmycelium cultural extract supplemented diet a commercial diet for 12 week. The body weight and length gain from the fish fed on daily the phellinus with coriolus versicolor mixed mycelium cultural extract supplemented diet, phellinus linteus mycelium cultural extract supplemented diet and Coriolus versicolor mycelium cultural extract supplemented diet of each mycelium cultural extract were higher than the control and the Glutamic oxalacetic transaminase (GPT) were of lower than the control. The relative precent survival rate (RPS) after an artificial challenge with $7{\times}10^5\;CFU$ of Vibrio anguillarum per fish was higher than the control.

Induction of $\beta$-Lactamase on $\beta$-Lactam Antibiotics Resistant Bacterium ($\beta$-Lactam계 항생물질 내성균주의 $\beta$-Lactamase 생산의 유도)

  • Bacillus subtilis J105, high resistant bacteria against $\beta$-lactam antibiotics, become higher resistant through induction of $\beta$--lactamase in the presence of $\beta$--lactam antibiotics. When there is no antibiotics in medium, the production of resistance-inductive $\beta$--lactamase reached its plateau 15 hours later. But when there is ampicillin (500$\mu\textrm{g}$/$m\ell$) in medium, the production of enzyme reached its plateau 25 hours later since cultivating bacteria, whereas it is found that enzyme 2,900 units/$m\ell$ about 20 times as much as compared with not-presence of antibiotics was actived. In addition, as the result of MIC comparing applying ampicillin-treated and non-treated strain MIC of ampicillin-treated strain is about 2~27 times higher. It is considered that this strain induce $\beta$--lactamase production by ampicil-lin-treatment, then increasing it resistance. It is found that this resistant strain induce $\beta$--lactamase production against cephalosporin antibiotics as well as peni-cillin. As the result of examining the time of adding antibiotics for each phase of growth, it is concluded that logarith-mic phase is the most effective. As the aboves, it is suggested that this strain is a peculia strain that its resistance is induced high by various $\beta$--lactam antibiotics.

Identification of a Protein Kinase using a FITC-labelled Synthetic Peptide in Streptomyces griseus IFO 13350 (형광 Peptide를 이용한 Streptomyces griseus IFO 13350의 인산화 단백질 동정)

  • Streptomycetes is a group of Gram-positive soil bacteria that growas a branching vegetative mycelium leading to the formation of spores, and display a physiological differenti-ation related to the synthesis of many secondary metabolites including antibiotics. Their complex life cycle and multicellular differentiation require various levels of regulation and types of signal transduction systems including eukaryotic-type serine/threonine protein kinases and prokaryotic-type histidine/aspartic acid protein kinases. Akt kinase that was found in cells is a sorine/threonine kinase controlling signal pathway for multi-tude of important cellular events. The activation or inactivation of Akt kinase in the cell is one of the critical regulatory points to deliver cell proliferation, differentiation, survival or apoptosis signal. To find the regula-tory protein homologous to Akt in Streptomyces, the fluorescien-labeled synthetic peptide (FITC-TRRSR-TESIT) was designed from the consensus sequence of target proteins for Akt kinase. From the difference of the mobility between the nonphosphorylated and phosphorylated synthetic peptides on Agarose gel electro-phoresis, the Akt-phosphorylating activity was monitored. The cell-free extract prepared from Streptomyces griseus IFO 13350 and the Akt homologous protein was purified by ammonium sulfate fractionation and many steps of column chromatographies such as, DEAE-Sepharose, Mono Q, Resource Phenyl-Soporose and Gel permeation column chromatographies. As a result, the protein phosphorylating the fluorescien-labeled Akt substrate was identified and it's molecular weight was estimated as 39 kDa on SDS-PAGE.

Pathogenesis of Inflammation in H. pylori Infection

  • 위의 parietal cell 혹은 대식세포와 유사한 세포 내부에서 H. pylori가 발견된다는 보고가 있기는 하나 일반적으로 H. pylori는 Shigella와 같은 침습성 세균은 아닌 것으로 알려져 있다. 그럼에도 불구하고 H. pylori에 감염된 위점막에는 많은 수의 호중구를 위시한 염증세포의 침윤이 관찰되는데 H. pylori가 위상피세포에 부착 할 경우 위상피세포를 자극하여 interleukin-8을 위시한 cytokine 을 발현케하고 이에 의하여 호중구 등의 염증세포가 몰려들게 된다. 한편 고유층에 몰려든 호중구에서는 다시 interleukin-8을 위시한 일련의 호중구 활성화 chemokine을 분비하여 염증반응을 증폭해 나갈 것이다. 호중구에서 발현되는 myeloperoxidase나 활성 산소 등도 위점막의 조직 손상에 기여할 것이다. 위상피세포를 덮고 있는 점액층은 위상피세포를 보호한다고 알려져 있으나 H. pylori 감염의 경우 점액층에 의하여 H. pylori의 운동성이 증가하고 이것이 위상피세포로부터의 cytokine 발현을 자극하여 염증반응을 증폭하는데 관여할 가능성도 있다. H. pylori는 위상피세포에 대하여 apoptosis를 유도함과 동시에 고유층에 몰려든 호중구에 대하여는 apoptosis를 억제케하여 궁극적으로 염증반응을 증폭 및 지속시켜 나가는 쪽으로 작용한다. 한편 H. pylori는 위상피세포로부터 COX-2의 발현을 증가시키는데 이는 위상피세포의 apoptosis를 억제하는 방향으로 작용한다. 이외에 H. pylori의 urease에 의하여 발생한 암모니아나 H. pylori 자신이 분비하는 세포독소가 세포 손상을 유발할 가능성도 있다. 상술한 여러 독성 인자들 중 어느 하나가 단독으로 작용하기보다는 여러 인자가 같이 동시에 또는 시차를 두고 작용할 가능성이 많다고 생각된다.(\gamma-FeOOH)$, 침철광$(\alpha-FeOOH)$, 적금광$(\beta-FeOOH)$, 그리고 자철광$(Fe_3O_4)$이다. 인위적 부식에서는 전부 인철광의 부식물이 생성되었고 자연적 부식에서는 모두 침철광의 부식물이 생성되었다. 특히 철제 표면에 자연적으로 생성된 공식 녹을 XRD 분석한 결과 적금광으로 동정되었다. 이런 모든 시편들을 각 탈염방법에 따라 탈염처리한 후 XRD와 SEM-EDS으로 분석한 결과 인철광과 침철광은 어떠한 변화도 보이지 않았고, 다만 적금광으로 동정된 시편만이 잔존하지 않았다. 철기 제작별 $Cl^-$ 이온 추출량과 탈염효과에 대한 비교 실험은 이온 크로마토그래피 분석 결과와 마찬가지로 단조 철제유물이 주조 철제보다 $Cl^-$ 이온을 많이 가지고 있었으며, 탈염 처리 후에는 $Cl^-$ 이온은 전혀 발견되지 않았다. 이상의 결과 $K_2CO_3$와 Sodium 용액은 탈염처리에서 가장 적합한 탈염처리 용액으로 알수가 있었으며 특히 어떠한 탈염 용액으로 유물을 처리한다 해도 철제유물에 생성된 부식물은 제거되지 않는다는 것을 알게 되었다. 따라서 보존처리자는 유물 표면의 부식 상태만을 보고 처리하기 보다는 철기제작물로 고려하여 처리하는 것이 필요하다. 또한 금속에 부식을 야기시키는 $Cl^-$ 이온과 부식물을 완전하게 제거하여 탈염처리를 하는 것이 유물 부식을 최대한 지연시킬 수 있는 것이라 생각된다.TEX>$88\%$)였다.(P=0.063). 결론: 본 연구에서는 MTHFR C/T & T/T 유전자 다형성이 위암의 발생과 그 위치에 대해 관련이 있는 것으로 여겨지고, 흡연력, 음주력과는 관련이 없는 것으로 여겨진다.험이

항균제 임상시험 Guideline

  • 잠재적으로 항감염 약제로 사용될 가능성이 있는 약제들의 초기 연구과정에서 주된 관심사는 이들이 미생물(세균, 바이러스, 기타 기생충)에 대한 작용들이 있는가 하는 점이다. 이러한 작용들이 실험적으로 충분히 연구가 된 후에야 실험동물에서 그 효과를 연구한다. 항 바이러스 제제의 경우에는 세포배양을 통한 연구가 그 약제의 독성과 효용성을 나타내는데 필수적인 것이 된다. 여러 종류의 동물을 이용한 생체실험에서 약제의 일반적인 흡수와 배설, 분포 등에 관한 정보와 약제 자체와 동물 내에서의 대사적 변화에 대한 정보가 제공된다. 여러 가지 미생물로 감염을 시킨 적합한 동물과 여러 가지 용량으로 치료하는 실험을 통하여 약제의 항 감염 능력이 알려지게 된다. 동물에서의 생체실험과 실험관내에서 실험을 하고 나서야 사람에서의 연구가 이루어지게 된다. 소위 전임상시험에서 대표적 병원성 미생물에 대한 생물학적효과, 약리학적 효과와 독성 그리고 동물실험모델에서의 가능한 효과가 결정된 후에 임상시험에 들어가기 마련이다. 항균제의 임상시험에는 각각의 감염질환에 대한 진단 및 치료기준을 반영하는 것이 기본이다. 새로운 항균제의 임상시험에서는 안전성과 효과가 반드시 밝혀져야 한다. 1상에서는 인체에서의 약리효과, 안전성이 주목적이며, 2상과 3상은 겹쳐지는 점도 있으나 하나 또는 그 이상의 적응증에 대한 항균제의 효과와 단기간의 부작용은 2상에서 관찰하여야하며, 다수의 환자에서 제안된 적응질환의 무작위임상시험과 다수에서의 안전성도 3상에서 관찰하여야 한다. 4상에서는 이상에서의 자료로 시판된 후에도 계속해서 감시하는 것으로 지속적으로 안전성을 관찰하는 것이다. 이러한 기본사항외에도 소아, 임산부, 고령자등에서의 임상시험도 넓은 의미에서 포함되어야 할 것이며 또한 질적인 면에서 조절하는 Quality Assurance도 중요하다.양상은 세 용량군 간 차이가 없었으나, 시험기에서 발열의 발현율이 낮았으며, 발열일 수와 항생제 사용일 수가 짧았다. 결론: 골수억제 조절 효과는 용량에 따른 혈액소견에 미치는 영향, 부작용, 감염의 빈도, 감염발생에 따른 항생제 사용기간 등을 고려하여 그 임상 유효성 평가시, 제 3상 시험에 사용할 권장량 (recommended dose) 은 250 ug/$m^2$/d $\times$ 10d 으로 관찰되었다.5주에 부검한 랫드의 간에서 c-myc 종양단백의 발현은 모든 처리군들이 대조군에 비하여 높게 발현되는. 것이 관찰되었으나 시험개시후 26주에 부검한 랫드의 간에서 c-myc 종양단백의 발현은 대조군에 비하여 차이가 거의 없었다. 따라서 랫드에서 화학적으로 유도한 간암발생 과정에서 NK 세포활성이 현저하게 억제되는 것으로 생각되며, c-myc 종양단백의 발현은 시험개시후 15주에 그 발현이 확실한 것으로 사료되어 진다.에 영향을 주는 성분이 있음을 제시하였다.1과 항우울약들의 항혈소판작용은 PKC-기질인 41-43 kD와 20 kD의 인산화를 억제함에 기인되는 것으로 사료된다.다. 것으로 사료된다.다.바와 같이 MCl에서 작은 Dv 값을 갖는데, 이것은 CdCl$_{4}$$^{2-}$ 착이온을 형성하거나 ZnCl$_{4}$$^{2-}$ , ZnCl$_{3}$$^{-}$같은 이온과 MgCl$^{+}$, MgCl$_{2}$같은 이온종을 형성하기 때문인것 같다. 한편 어떠한 용리액에서던지 NH$_{4}$$^{+}$의 경우 Dv값이 제일 작았다. 바. 본 연구의 목적중의 하나인 인체유

Phenotypic Characteristics of Pseudomonas syringae pv. actinidiae Strains from Different Geographic Origins (지리적 기원이 다른 Pseudomonas syringae pv. actinidiae 균주들의 표현형적 특성)

  • Pseudomonas syringae pv. actinidiae is the causal agent of bacterial canker in kiwifruit (genus Actinidia). Multilocus sequence analysis of seven housekeeping and 11 type III effector genes differentiated the virulent P. syringae pv. actinidiae isolates worldwide into three groups designated as Psa1-Psa3. In this work, a total of 12 P. syringae pv. Actinidiae strains, including three Psa1, three Psa2, three Psa3 strains isolated from Korea and three Psa3 strains from Italy, were compared based on their phenotypic properties. Strains with different geographic origins had unique growth patterns as demonstrated by growth rate at several temperatures; all tested strains exhibited maximum growth at temperatures below $22^{\circ}C$, while the growth of Psa3 strains was completely inhibited above $30^{\circ}C$. Psa3 strains isolated from Korea had longer lag phases than the Psa3 strains from Italy. The Psa2 strains were different from Psa1 and Psa3 strains in the API 20NE test, in which the Psa2 strains could not utilize potassium gluconate, capric acid and trisodium citrate. Psa3 strains isolated from Korea could hydrolyze esculin. The API ZYM test showed that ${\beta}$-glucosidase activity was detected only from Psa3 strains. The strains belonging to the three Psa groups differed with regard to their susceptibility to ampicillin, novobiocin, and oleandomycin.

Shelf Life Extension of Steamed Bread by the Addition of Fermented Pine Needle Extract Syrup as an Ingredient (솔잎 발효액의 첨가에 의한 찐빵의 저장성 향상)

  • In order to improve the storage stability of steamed bread, fermented pine needle extract syrup potentially with several functional properties was added in manufacturing the products at levels of 8.3, 11 and 18% based on Brix degree of the dough. The manufactured bread was stored at ambient conditions and measured for quality attributes. Dough added with the fermented pine needle extract syrup maintained its pH and water activity at levels of $5.45{\sim}5.90\;and\;0.94{\sim}0.96$, which are normally suitable for yeast fermentation and for appropriate dough hardness. Addition of the extract syrup increased the bread volume by more than 20%. The bread with higher content of the pine needle extract syrup showed slower increase of bread hardness during storage, suggesting retardation of bread retrogradation. The addition of the pine needle extract syrup in bread dough also inhibited growth of the aerobic bacteria and molds on the bread surface (by $0.8{\sim}24$ in log (CFU/g) at 4 day storage). Use of higher than 11% concentration presented initially a strong fine needle flavor to the bread, which disappeared soon after 2 days. Generally addition of the pine needle extract syrup did not give negative effects on the bread quality including sensory quality. Therefore, the addition of the needle extract syrup could contribute to improving the storage stability and extending the shelf life of the bread.

Perchlorate Removal by River Microorganisms in Industrial Complexes (산업단지지역 하천 미생물에 의한 퍼클로레이트 제거)

  • Perchlorate ($ClO_4^-$) is an emerging contaminant of soil/groundwater and surface water. $ClO_4^-$ has been shown to inhibit iodide uptake into the thyroid gland and cause a reduction in thyroid hormone production. $ClO_4^-$ is highly soluble and very stable in water. Biodegradation by $ClO_4^-$-reducing bacteria (PRB) is considered the most important factor in natural attenuation of $ClO_4^-$. Rivers in an industrial complex have potential to be contaminated with $ClO_4^-$ discharged from point or non-point sources. In this study, water samples were taken from the rivers running through the Gumi industrial complexes and used for batch test to analyze $ClO_4^-$-degradation potential of river microorganisms. The results of 83-h batch culture showed that $ClO_4^-$-removal efficiency of all samples was 0.77% or less without addition of an external electron ($e^-$) donor. However $ClO_4^-$-removal efficiency was higher when an $e^-$ donor (acetate, thiosulfate, $S^0$, or $F^0$) was added into the batch culture, showing up to 100% removal efficiency. The removal efficiency was various depending on type of $e^-$ donor and site of sampling. When acetate was used as an $e^-$ donor, the highest $ClO_4^-$-removal efficiency was observed among the $e^-$ donors used in this study, suggesting that activity of heterotrophic PRB was dominant. The results of this study provide basic information on natural attenuation of $ClO_4^-$ by river microorganisms. The information can be useful to prepare a strategy to enhance efficiency of $ClO_4^-$ biodegradation for in situ bioremediation.

Fermentation Characteristics of Kochujang Prepared with Various Salts (소금의 종류를 달리한 고추장의 발효 특성)

  • Effects of sun-dried, refined, seaweed, and bamboo salts on the quality of kochujang were studied by measuring enzymatic, microbial, and physicochemical characteristics of kochujang during 12 weeks of fermentation. Yeast count was low in the bamboo-salt kochujang, whereas that of aerobic bacteria was low in the seaweed-salt kochujang. Acid protease activity was high in the bamboo-salt kochujang, whereas amylase activity did not show any remarkable difference. Color change was lowest in the bamboo-salt kochujang. Water activities of all kochujangs decreased during fermentation with the lowest shown in the refined-salt kochujang. Consistency of seaweed-salt kochujang was the highest. Total sugar content was higher, whereas ORP was lower in the seaweed- and bamboo-salt kochujangs. Titratable acidity changed slightly in the bamboo-salt kochujang. Reducing sugar content was the lowest, whereas ethanol content was the highest in the refined-salt kochujang. Amino and ammonia nitrogen contents of kochujangs increased up to the middle of fermentation period then decreased with lower changes observed in the sun-dried and refined-salt kochujangs. Results of sensory evaluation indicated that the bamboo-salt kochujang was more acceptable than the sun-dried salt kochujang in taste, color, and overall acceptabilities.

Antibacterial Activity of Oriental Medicinal Herb Extracts against Skin Pathogens (한약재 추출물의 피부 염증 유발 세균에 대한 항균활성)

  • The antibacterial activity of methanol extracts from 70 kinds of oriental medicinal herbs on four strains of Staphylococcus aureus and S. epidermidis was investigated. The results showed that C. japonica, C. sappan, R. javanica, R. tanguticum, and S. miltiorrhiza had an antibacterial activity on all the strains used. Among these, C. japonica and R. javanica, which showed excellent antibacterial activity, were extracted with water, ethanol, methanol, and ethyl acetate for further study of antibacterial activities. The results showed that the boiled water extract of C. japonica had the best antibacterial activity. Assuming that the antibacterial activity of C. japonica originated from berberine, which has been reported many times, the berberine content of the boiled water extract of C. japonica was analyzed and the result was 13.88%. To compare the antibacterial activity of berberine with that of other antibiotics, berberine chloride and three other kinds of antibiotics were investigated, which showed that berberine chloride had an antibacterial activity on KCCM 35494 S. epidermidis only when the concentration was higher than 600 mg/l, and it did not show antibacterial activity in the other strains. Based on these results, it was concluded that the antibacterial activity of the boiled water extract of C. japonica on the strains used in this study originated from high concentration of berberine or substances other than berberine. Therefore, identification of the substance will be necessary.
