A halophilic bacterium, H. subglaciescola DH-1, grew at 2.0 M salinity, but did not grow at 0.8 M salinity when cultivated at higher temperature ($40^{\circ}C$) than optimum ($30^{\circ}C$). When the cell extract of strain DH-1 was heated at $50^{\circ}C$ for 60 min in the absence of NaCl, isocitrate dehydrogenase and malate dehydrogenase lost their activities, but when it was heated in the presence of 2.0 M NaCl, the activity was maintained. Meanwhile, the cell extract of E. coli did not catalyze the reduction of $NAD^+$ to NADH coupled with the oxidation of isocitrate and malate at higher salinities than 1.0 M. The pH range for DH-1 was 7 to 10, and that for E. coli was 5 to 9. DH-1 was not grown in conditions with sodium salts other than NaCl.
Late blight, one of the most important disease in many agricultural crops, is caused by Phytophthora infestans. Fusarium wilt is a vascular disease of many plants caused by Fusarium oxysporum. Some bacteria isolated from rhizosphere were screened for their ability to inhibit the growth of F. oxysporum and P. infestans. Productions of siderophore, $\beta-1$,3-glucanase, hydrogen cyanide and chitinase by 4 isolated strains were examined. Among them, Bacillus sp. RFO41 most effectively inhibited the growth of F. oxysporum. The highest productions of siderophore and $\beta-l$,3-glucanase were shown in the culture of Bacillus sp. RFO41. Bacillus strain PS2 was most effective against P. infestans. PS2 showed the highest production of chitinase and hydrogen cyanide. A significant relationship was shown between the antagonistic effects of isolates against F. oxysporum and P. infestans and their production level of siderophore, $\beta-1$,3-glucanase, hydrogen cyanide, and chitinase.
A new alginate lyase gene of marine bacterium Streptomyces sp. M3 had been previously cloned in pColdI vector and transformed into E. coli BL21 (DE3). In this study, M3 lyase protein without signal peptide was overexpressed by induction with IPTG and purified with Ni-Sepharose affinity chromatography. The absorbance at 235 nm of the reaction mixture and TLC analysis showed that M3 alginate lyase was a polyG-specific lyase. When M3 lyase was assayed with substrate for 10 min, optimum pH and optimum temperature were pH 9 and $60^{\circ}C$. For the effect of 1mM metal ion on M3 lyase activity, $Ca^{++}$ and $Mn^{++}$ ions increased the alginate degrading activity by two-fold, whereas $Hg^{++}$ and $Zn^{++}$ ions inhibited the lyase activity completely. $Mg^{++}$, $Co^{++}$, $Na^+$, $K^+$, and $Ba^{++}$ did not show any strong effects on alginate lyase activity.

In the course of search for multifunctional microbial inoculants, three Bacillus strains (BS11-1,BS11-2,BS11-3) with biological control and biofertilizing effects were selected. In this study, their ability for solubilization of insoluble phosphate, production of indole-3-acetic acid (IAA), siderophore, and hydrolytic enzymes, and antagonism against phytopathogenic fungi were estimated. All strains produced IAA and siderophore depending on culture time and produced a visible clear zone on agar plate containing 0.5% carboxylmethyl cellulose as a carbon source. Also, these strains exhibited antifungal activities against phytopathogenic fungi, Botrytis cinerea, Cylindrocarpon destructans, Fusarium oxysporum, Rhizoctonia solani, and Phytophthora capsici.
A novel agar-degrading bacterium SL-5 was isolated from seashore of Homigot at Kyung-Buk province, and cultured in marine broth 2216 media. The bacterium SL-5 was identified as Thalassomonas genus by 16S rDNA sequencing with 96% identity. Growth rate was faster at $27^{\circ}C$ than at $37^{\circ}C$ and agarase was produced as growth-related. The optimum pH of the enzyme activity was 7.0 and the optimum temperature for the reaction was $40^{\circ}C$. Although the enzyme had no thermostability, the enzyme activity was remained over 80% at $60^{\circ}C$. The enzyme hydrolyzed neoagarohexaose to yield neoagarobiose as the main product, indicating that the enzyme is $\beta-agarase$. Thus, the enzyme would be useful for the industrial production of neoagarobiose.
The long-term effects of soil management history on microbial communities are still poorly understood. Our objectives were to determine the impact of long-term application of soil amendments on microbial communities in rice paddy fields. The treatments selected were control where crops were grown without any nutrient application (CON); nitrogen-phosphorus-potassium (NPK); NPK plus compost (CNPK); NPK plus lime (LNPK); and NPK plus silicate (WNPK). The long-term addition of organic and inorganic amendments significantly changed soil chemical properties. The amount of organic carbon increased in the treatments with fertilizer and amendments over that in the soil without inputs. However, we could not observe the differences of bacterial population among the treatments, but the number of aerobic bacteria increased by the addition of amendments. Isolates from the rice paddy soils before irrigation were Dactylosporangium, Ewingella, Geobacillus, Kocuria, Kurthia, Kytococcus, Lechevalieria, Micrococcus, Micromonospora, Paenibacillus, Pedobacter, Pseudomonas, Pseudoxanthomonas, Rhodococcus, Rothia, Sphingopyxis, Stenotrophomonas, and Variovorax. Dominant genera were Arthrobacter, Kocuria, Kurthia, and Bacillus in the long-term field. Microbial biomass was the highest in the compost treatment (CNPK), and was the lowest in the CON. Dehydrogenase activity in soils treated with rice compost straw was the highest and the activity showed an increasing trend according to treatment as follows: CON < WNPK < NPK = LNPK < CNPK. These results demonstrate that soil management practice, such as optimal application of fertilizer and amendment, that result in accumulations of organic carbon may increase microbial biomass and dehydrogenase activity in long-term rice paddy soils.
The purpose of this study was to investigate the antibacterial activity of green tea in mayonnaise against pathogenic bacteria (Escherichia coli O157:H7 and Salmonella typhimurium). Mayonnaise was prepared with salad oil, egg yolk, sugar, salt and vinegar, and green tea powder was added to the mayonnaise at 0.1, 0.3 and 0.5% for the experiment. Then, the mayonnaise samples with various levels of green tea were inoculated with about 10$\^$6/ cells/g of E. coli and S. typhimurium per 1 gram of mayonnaise and stored at 5$^{\circ}C$, 15$^{\circ}C$ and 25$^{\circ}C$ for 3∼9 days. Antibacterial activity of green tea was tested by the colony counting method for E. coli on violet red bile agar(VRBA) and S. typhimurium on xylose lysine desoxycholate agar(XLDA). The D-values of E. coli controls were 2.89, 2.73 and 2.13 days while those of S. typhimurium controls were 0.58, 0.53 and 0.52 days at 5$^{\circ}C$, 15$^{\circ}C$ and 25$^{\circ}C$, respectively Inhibitory effect of green tea in mayonnaise on the survival of E. coli and S. typhimurium was increased with increasing concentration of green tea and/or increasing storage temperature. The most effective antibacterial activity of green tea was shown against E. coli in mayonnaise during storage at 25$^{\circ}C$.
The algicidal effects of marine bacteria were investigated and a strain, which had the strongest algicidal activity against the harmful dinoflagellate, Cochiodinim polykrikoides was selected. The bacterium was isolated in seawater during the period of blooming of C. polykrikoides in Masan Bay. This algicidal bacterium was identified as Micrococcus sp. LG-5 by means of morphological and biochemical tests. The optimal culture conditions of Micrococcus sp, LG-5 were $25^{\circ}C,\;pH 7.0\;and\;3.0{\%}$ NaCl concentration. The algicidal activity of Micrococcus sp. LG-5 was significantly increased to maximum value in the late of logarithmic phase of cell cuture. In addition, the culture filtrate ($pore size,\;0.1{\mu}m$) of Microcoocus sp. LG-5 showed strong algicidal effects. The cell numbers of C. polykikoides were decreased from $1.2{\times}10^4 cells/ml\;to\;less\;than\;2{\times}10^3\;cells/ml$ within 3, 6, 30 hours at the concentrations of culture filtrate $10{\%},\;5{\%}\;and\;1{\%}$, respectively. These results indicated that the algicidal effect was mediated by certain substances released from Microooccus sp. LG-5.

Porphyromonas gingivalis는 치주질환을 야기하는 독성세균으로서, 구강상피세포에 p. gingivalis가 감염되었을 때, 세포형태에 변화를 초래함으로 인해 방어기작이 작동하게 된다. 치주질환과 관련되어 생성된 활성 산소종의 소거에 관여하는 항산화성분은 p. gingivalis 이 감염된 구강상피세포에서 그 분포와 발현수준이 달라지리라 예상된다. 따라서 이번 연구에서는 구강상피세포(KB 세포)에 p. gingivalis가 감염되었을 때 야기되는 활성산소종과 이를 소거하는 역할을 하는 항산화단백들의 역할들을 규명하고자 하였다. 활성산소종 형성을 조절하는 NADPH oxidase 중 NOX4와 Rac1 전사체는 구강상피세포에서 p. gingivalis세균에 의해 증가하였으며 $gp91^{phox}$, Rac2, $p47^{phox}$와 $p67^{phox}$는 세균에 의한 변화가 관찰되지 않았다. 반면에 $p40^{phox}$ 전사체는 감소하는 경향을 보였다. NOX1 전사체는 p. gingivalis 처리 30분 후 감소하였다가 60분 후에는 다시 증가하는 양상을 보였다. 같은 시간에 NOX 활성화 단백인 NOXA1은 감소하고, NOX 구성단백질인 NOXO1은 증가하는 경향을 보였다. p. gingivalis가 감염된 구강상피세포를 방어하는 항산화단백 발현수준을 평가한 결과, SOD1, 2, 3 모두 p. gingivalis 처리시간에 따라 증가하는 양상을 보였다. GPx 발현 양상도 SOD와 유사하게 나타났다. $H_2O_2$를 소거하는 Prx는 감염된 KB 세포에서 Prx4와 Prx5가 4-6배 증가하는 것을 알 수 있었다. 반면 endocytosis 과정 중 $H_2O_2$ 생산은 변화되지 않았다. 이번 연구의 결과, p. gingivalis의 감염은 KB 세포의 NOX4와 Rac1의 NADPH oxidase 발현을 증가시켰으며, NOX1은 NOXA1과 NOXO1의 조절에 의해 영향을 받음을 알 수 있었다. 또한 항산화기작으로는 SOD, GPx, Prx가 증가하였는데, 이것은 Prx4와 Prx5가 중요한 역할을 할 것을 시사하였다.

상온에 준하는 저온의 플라즈마를 발생시키는 장치들이 개발되면서, 저온 플라즈마와 생체조직간의 상호작용에 대한 연구가 큰 관심을 끌고 있다. 플라즈마에서 발생되는 다량의 이온과 활성종, 그리고 UV 등이 박테리아나 세포들과 작용함으로 해서 암세포 사멸, 치아 미백, 박테리아 살균/멸균, 지혈등의 효과들이 나타나고 있으며, 이러한 효과들을 극대화할 수 있는 장치 개발과 플라즈마와 생체조직간의 상호작용 메카니즘을 규명하는 것이 중요한 이슈가 되고 있다. 나노 금입자를 암세포의 막단백질인 FAK의 항체와 결합시킨 중합체를 만들어서, 암세포 표면에 나노 금입자붙이고, 플라즈마를 조사했을 때, 나노 금입자가 부착되지 않았을 경우에 비해서, 5배이상 사멸률이 증가하였다.[1] 변색된 치아에 미백제의 주성분인 과산화수소를 도포하고, 10분간 플라즈마를 조사하게 되면, 과산화수소만 도포했을 때에 비해, 치아 표면의 색이 3배이상 밝아지는 것을 관찰할 수 있었다. 과산화수소를 플라즈마에 노출시켰을 때, 활성종인 OH의 생성이 2배이상 증가하였고, 플라즈마에 의한 OH 생성의 촉진이 치아 미백효과가 증대되는 주된 요인인 것으로 추측된다.[2] 플라즈마에서 발생되는 O, $O_3$와 같은 활성종들은 살균력이 뛰어나기 때문에, 저온 플라즈마를 의료기구의 소독/멸균에 응용할 가능성이 아주 크다. 대장균이나 구강 세균이 플라즈마 처리로 5분이내에 멸균되는 것을 확인하였고, 핸드피스와 같은 의료기구를 오염시켜서 멸균 테스트를 수행하고 있다.
