Feasibility of Granular Activated Charcoal as a Detector in Fluorescent Tracer Tests (입상 활성탄을 이용한 형광물질 추적자시험 적용성 연구)

  • In recent tests using tracer have been frequently conducted by fluorescent tracers. In this study, granular activated charcoal (GAC) as a detector for the fluorescent tracers (rhodamine WT and uranine) was investigated through laboratory and field tests. In the laboratory tests, tracer concentrations of rhodamine WT and uranine determined by the GAC were slightly different from those of standard solutions but they were excellent in linearity. Results show that GAC is excellent as tracer detector when concentration of the fluorescent tracers is greater than 10 & micro; g/L whileas no obvious differences in mixed solutions of the two tracers due to interferences. Compared to conventional methods of water sampling, field results shows a high potential of GAC as a tracer in the field. Our results also show that wet analysis is better for the lower concentrations of tracers whileas dry analysis is good for high concentrations of tracers. This study demonstrates that fluorescent tracer detection using the GAC is very useful and economical for a hydraulic connection between target areas and very longer period of the tracer test.

  • Brassinosteroid-like substances in two japonica type Korean rices were investigated. The extracts from the shoots at the maximum tillering stage were purified by solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 chromatography, charcoal adsorption chromatography, preparative TLC, Bondesil chromatography and HPLC of normal phase and reverse phase, successively. Biological activity of each purification step were monitored by the rice lamina inclination test. Two cultivars tested in this experiment produced brassinosteroids and endogenous brassinosteroids showing similiarity between two cultivars.

  • An oxytetracycline as being a tetracycline-antibiotic substance displayed a general synergism in the antimicrobial activity by the interaction of ginseng saponin and antibiotics, but did not to Sarcina maginata. Penicillin G.Na as being a $\beta$-lactam-antibiotic substance displayed a synergism in the antimicrobial activities by the addition of ginseng saponin to microorganisms used, but changed the effects in the antimicrobial activity of penicillin G.Na against the genus Serratia. An antimicrobial activity by the addition of ginseng saponin to antibiotics showed a non-specificity, and it varied synergism or antagonism to Gram-positive or Gram-negative bacterium. It was assumed that a drug-resistance could be eliminated by the dual administration of ginseng saponin and antibiotics.

  • As a result of screening search for biologically active substances to weed seeds among higher plants, MeOH extract from Rhathiolepis ovata Briat was found to inhibit germination of test weeds considerably. In the course of purifying the active substances, pinoresinol which showed very similar behavior with the active fraction on various chromatographies, was isolated from the same source, spectrally identified and bioassayed. Pinoresinol exhibited germination inhibitory activity against the common purslane (Portulaca oleracea L.) only ; the inhibitory effect was about 55% at concentration of 5 mg/ml.

  • The methanol extract from leaves of Catalpa ovata 6. DoN showed DPPH (1,1-diphenyl-2-picrylhydrazyl) radical-scavenging activity, and its antioxidative compounds were studied. The ethyl acetate-soluble neutral fraction from the methanol extract was successively purified with silica gel adsorption column chromatography, Sephadex LH-20 column chromatography and HPLC. Three antioxidative compounds were isolated and identified as piperitol, pinoresinol and lariciresinol by HR-MS and NMR spectroscopic analyses. The DPPH radical-scavenging activity of the identified compounds decreased in the order of lariciresinol > pinoresinol > piperitol.

Brassinosteroid substances in immature Cassia tora seeds (결명자의 brassinosteroid 활성물질)

  • In order to explore the brassinosteroid-active component in Cassia tora, methanol extract of immature seeds was purified by sequences of solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 chromatography, charcoal adsorption chromatography and Bondesil chromatography. The activity of brassinosteroid was monitored by the rice inclination test and its presence could be confirmed in each purification step. The purified active components were separated by silica gel adsorption chromatography. Brassinosteroid substances in separated active fractions were identified as teasterone, castasterone, brassinolide by TLC and HPLC. Our work is probably the first report of endogenous brassinosteroid in Cassia tora. The content of brassinosteroid in Cassia tora as converted into brassinolide was $3.5{\sim}5.5\;ng/g$ fresh weight. The order of brassinosteroid contents was toward to be teasterone, castasterone, brassinolide.

Characterization and Anti-Helicobactor pylori Activity of Xanthium strumarium L. Extract on Lactic acid Fermentation (도꼬마리 추출물의 유산발효 특성 및 Helicobactor pylori에 대한 항균활성)

  • This study characterized the anti-Helicobactor pylori activity of Xanthium strumarium L. extract obtained by lactic acid fermentation. The growth of the Lactobacillus strains was typically robust upon lactic acid production in monocultures containing Xanthium strumarium L. extract. Lactic acid fermentation in mixed cultures of Lactobacillus brevis KCTC 3498 and Lactobacillus casei KCTC 3109 produced higher of anti- H. pylori activity than monocultures. Concerning antioxidant activity of fermented extracts, total polyphenol contents were the highest in co-cultures of Lactobacillus brevis KCTC 3498 and Lactobacillus helveticus KCTC 3545. Electron donating ability using diphenyl picryl hydrazyl (DPPH) showed 70% scavenging in Lactobacillus casei KCTC 3109.

칼럼실험을 통한 계면활성제 용액에서 phenanthrene의 선택적 제거에 관한 연구

  • 유해성 유기물질로 오염된 토양의 복원을 위한 토양세척공정에서 계면활성제를 선택적으로 재이용하기 위해 활성탄을 이용한 흡착 칼럼의 성능을 평가하였다. 계면활성제로는 Triton X-100을 소수성 유해 유기물질로는 다환방향족 탄화수소의 하나인 phenanthrene를 사용하여 그 성능을 평가하였다. 계면활성제의 흡착은 phenartthrene에 비해 빠른 흡착 특성을 보였으며 phenanthrene은 계면활성제의 포화흡착상태에서도 지속적으로 흡착이 이루어졌다. 이는 계면활성제의 흡착이 종료된 이후에도 유해성 유기물질의 지속적인 흡착을 기대할 수 있어 계면활성제의 지속적인 재이용과 활성탄 사용시간의 증대를 가져올 수 있음을 의미한다. 이와 같은 활성탄 칼럼에서의 유해성 유기물질의 우수한 선택적 흡착 결과는 기본적으로 소수성 정도의 차이와 size exclusion에 의한 기작뿐만 아니라 활성탄에 흡착된 계면활성제에 의한 다환방향족 탄화수소의 추가적인 흡착이 일어나 전체적인 성능이 향상되는 것에 기인한다.

Antifungal Effects on Plant Pathogenic Fungi and Characteristics of Antifungal Substances Produced by Bacillus subtillis SJ-2 Isolated from Sclerotia of Rhizoctonia solani (벼 잎집무늬마름병균(Rhizoctoniz solani)의 균핵에서 분리한 Bacillus subtilis SJ-2의 식물 병원 곰팡이에 대한 항균 활성 및 항균 물질의 특성)

  • 벼 잎집무늬마름병균(Rhizoctonia solani)의 균핵에서 분리한 길항세균 SJ-2를 대치 배양 방법에 의해 벼도열병균(Pyricularia oryzae)외 6종의 식물 병원균에 대한 억제 효과를 조사하였다. 형태 및 생리적 특성을 조사한 결과, 이 균은 Bacillus subtilis로 동정되었다. 이 균은 AM 1(antibiotic medium 1)액체 배지에서 항균 물질을 분비하였으며, 배양 2일째 가장 항균 활성이 높았다. Butyl alcohol로 배양액에서 항균 물질을 조추출하여 100$\mu\textrm{g}$/ml의 농도로 조제한 감자 한천 배지에서 P. oryzae의 15개균에 대한 생육을 조사한 결과 P. oryzae, R. solani, Cochliobolus sativus에 대해서는 100% 생장을 억제하였으며, C. miyabeanus, Alternaria alternata, Botrytis cinerea, Cladosporium fulvum, Colletotrichum gloeosporioides, Fusarium moniliforme, F. oxysporum에는 80% 이상의 억제 효과를 보인 반면 난균강에 속하는 병원균에 대한 효과는 낮게 나타났다. 활성물질을 분리 정제하여 동정한 결과 B. subtilis가 분비하는 항균 물질로 알려진 polypeptides계의 iturins로 밝혀졌다. 이러한 항균 물질은 벼 도열병균(P. oryzae)의 포자 발아 및 발아관의 팽대(swelling)를 야기했으며, 벼 잎집무늬마름병균(R. solani)에는 균사의 용균(lysis)현상을 일으켰다.

Brassinosteroid substances in immature Perilla frutescense seeds (들깨의 brassinosteroid 활성물질)

  • In order to explore the brassinosteroid-active component in Perilla frutescense, methanol extract of immature seeds was purified by sequences of solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 chromatography, charcoal adsorption chromatography and Bondesil chromatography. The activity of brassinosteroid was monitored by the rice inclination test and its presence could be confirmed in each purification step. The purified active components were seperated by silica gel adsorption chromatography. The seperated main and minor active brassinosteroid fractions were identified as castasterone and homobrassinolide, respectively, by HPLC. We acknowledge that our work is probably the first report of endogenous brassinosteroid in Perilla frutescense. The content of brassinosteroid in Perilla frutescense as converted into brassinolide was $0.5{\sim}0.8\;ng/g$ fresh weight.
