A Study on the Solubilisation of Excess Sludge using Microbubble Ozone (잉여슬러지 가용화를 위한 마이크로버블 오존 이용에 관한 연구)

  • This study was conducted with the experiment of solubilisation of excess sludge by microbubble ozone process. To improve ozone contact efficiency, microbubble ozones which its diameter were the avearge 30 ${\mu}m$, microbubble size less than 40 ${\mu}m$ occupied about over 90% of all. In treating sludge using microbubble ozones, in case microbubble ozones are injected at microbubble ozone dosage of 0.34 g $O_3/g$ SS or less regardless of sludge concentration, microbubble ozone consumption rate was found to be 100% with no emission of waste ozones. In treating sludges by each concentration, in case the initial SS concentration of sludge is set to 6,447 mg/L, 5,557 mg/L, 3,180 mg/L, 1,092 mg/L and 515 mg/L, the amount of removed SS tended to increase with increase in initial SS concentration for the same microbubble ozone dosage, and treatment of sludge with high initial SS concentration was effective in raising the oxidation efficiency of microbubble ozones. On the other hand, as a result of reviewing acid, alkali and microbubble ozone treatment as composite treatment of sludge, use of acid treatment for the pre-treatment of microbubble ozone was more effective than alkali treatment, and in case of treatment at microbubble ozone dosage 0.05g $O_3/g$ SS with the concentration of sulfuric acid infused in the sludge, the amount of removed SS, 153.9 g, was 1.9 times more than 81.2 g the amount of single treatment of microbubble ozone.

Characteristics of Temporal Variation on Water Quality (T-P, T-N, CODMn, SS, BOD5) in the Jungrang Stream during Rainfall Event (강우 시 중랑천 유역의 수질(T-P, T-N, CODMn, SS, BOD5)변화 특성)

  • Water quality variations were investigated at 4 locations of Jungrang river (upper, middle and lower basins) during a period of 3 rainfall events. During the rainfall, concentrations of $COD_{Mn}$, SS and $BOD_5$ significantly increased, while the concentration of T-N decreased and that of T-P remained relatively constant. This pattern became more apparent as the level of accumulative precipitation and rainfall intensity increased. Simple regression analysis showed that the accumulative precipitation was positively correlated with all water quality pollutants except for T-N. With increasing accumulative precipitation, the concentration of T-N decreased, while the others increased. $R^2$ of simple regressions of hourly average rainfall intensity and water quality pollutants, showed wider range of variation ranged from 0.483 to 0.992, which indicated a strong correlation. The stronger the hourly average rainfall intensity, the more T-N and T-P in the upper basin, more $COD_{Mn}$ in the middle and lower basins, more SS with gradual increase from upper to lower areas, and more $BOD_5$ with gradual decrease from upper to lower region. Simple regression showed that water quality pollution in the upper basin was more sensitive to an increase of rainfall discharge than that in the middle and lower areas.

Detection of Perfluorinated Compounds (PFCs) in Nakdong River Basin (낙동강 수계에서의 과불화 화합물(PFCs) 검출 특성)

  • The aims of this study were to investigate and confirm the occurrence and distribution patterns of perfluorinated compounds (PFCs) in Nakdong River basin (mainstream and its tributaries). 7 (PFOS, PFHpA, PFOA, PFNA, PFDA, PFUnDA and PFDoDA) out of 11 PFCs were detected in 29 sampling sites and PFOA and PFHpA were predominant compounds in upstream, but PFUnDA, PFDoDA and PFOS were predominant compounds in middle stream of Nakdong River basin. The total concentration levels of PFCs on February 2009 and on August 2009 in surface water samples ranged from 4.3. to 1168.2 ng/L and 16.4 to 627.8 ng/L, respectively. The highest concentration level of PFCs in the mainstream and tributaries in Nakdong River were Goryeong and Jincheon-cheon, respectively. The sewage treatment plants (STPs) along the river affect the PFCs levels in river and the PFCs levels decreased with downstream because of dilution effects.

A Comparative Study on the Measures Determining Optimal SAGD Locations Based on Geostatistical and Multiphysics Simulations (지구통계 및 다중 유체 거동 모사에 근거한 스팀주입중력법 적용 최적지 결정 척도 개발 연구)

  • In this study, two viable measures of mean length and cumulative thickness of sand layers as important spatial statistics responsible for optimal SAGD (Steam Assisted Gravity Drainage) location for oil sand development were compared. For the comparisons, various deposits composed of sand and clay media were realized using a geostatistical simulator and the extent of steam chamber is simulated using multiphysics numerical simulator (dualphase flow and heat transfer). Based on the spatial statistics of each realization and the corresponding size of simulated steam chamber, the representativeness of two candidate measures (cumulative thickness and mean length of permeable media) were compared. The results of the geostatistical and SAGD simulations suggest that the mean length of permeable media is better correlated to the size of steam chamber than the cumulative thickness. Given those two-dimensional results, it is concluded that the cumulative thickness of the permeable media alone may not be a sufficient criterion for determining an optimal SAGD location and the mean length needs to be complementarily considered for the sound selections.

Screening of Pesticidal Active Compounds from Various Domestic Wild Plants (국내 자생 식물자원 중 농약활성물질 탐색)

  • For the selection of plants contained pesticidal active conpounds, 31 families 59 species of native plants were collected and biological activites of their crude extracts against brown planthopper (Nilaparvata lugens), Xanthomonas oryzae pv. oryzae and allelopathy were examined. Among the screened plants, the crude extracts from the leaves of Ricinus communis and Sophora angustifolia showed 100% and 82% of mortality on brown planthopper at the concentration of 1% (w/v) respectively. Mixed crude extracts of Sophora angustifolia root and Melia azedarach seed exhibited 128${\sim}$155% of synergistic effects on the mortality of brown planthopper. In case of fungicidal activity, the crude extracts from the leaves of 8 plants including Chrysanthemum indicum were inhibitory of the growth of Xanthomonas oryzae pv. oryzae at 1%(w/v) soluction. Seed germination of radish (Raphanus sativas) and barnyardgrass (Echinochloa crus-galli) was inhibited by 0.5%(w/v) leaf extracts of Pinus densiflora and Quercus acutissima. MeOH extracts of Pinus denislora leaves gave 100% inhibition in seed germintion of radish at 2%(w/v) soluction and showed a complete inhibition in seedling growth of barnyardgrass as well as radish at 5% solution.

α-Glucosidase Inhibitory Effect of Vicine and α-Eleostearic Acid from the Seeds of Momordica charantia (여주씨에서 분리한 Vicine과 α-Eleostearic Acid의 α-glucosidase 저해효과)

  • BACKGROUND: Momordica charantia (MC) have been used in various parts of the world to treat diabetes. Despite the highest effects of phytochemicals than any other part of the plant, the main components of seed of MC grown in Korea and their properties have not been studied extensively. METHODS AND RESULTS: The seeds of MC were extracted into five different polarity solvents: 30%, 50%, 70%, 95% ethanol and distilled water. The 95% ethanol extract showed the most potent inhibition ($IC_{50}=88.7{\mu}g/mL$) against ${\alpha}$-glucosidase. To investigate the compounds responsible for this effect, activity guided fractionation of MC seeds by chromatography yielded two phytochemicals which were confirmed as vicine (1) and ${\alpha}$-eleostearic acid (2) based on their NMR and ESI-MS spectroscopic data. Among them, ${\alpha}$-eleostearic acid (2) possessed potent ${\alpha}$-glucosidase inhibitory activities with $IC_{50}$ values from $32.4{\mu}g/mL$. CONCLUSION: Collectively, the results from this present study strongly suggest that both extract and ${\alpha}$-eleostearic acid (2) have potent ${\alpha}$-glucosidase inhibitory activity. Furthermore, ${\alpha}$-eleostearic acid (2) as the most active ${\alpha}$-glucosidase inhibitor was proven to be present in high quantities in the MC seeds by a HPLC chromatogram.

Application of Multiresidue Analysis Method of Unregistered Pesticides in Korea for Imported Food (수입식품 중 국내 미등록 농약의 다성분 잔류분석법 적용)

  • BACKGROUND: Recently in Korea, the import of agricultural products is rising due to the increasing amount of trade. Unregistered pesticides, allidochlor, propachlor, propham, cycloate, diallate and pebulate are widely used as pesticides for rice cultivation in foreign countries, while they are not registered in Korea. Therefore, the residue amount of imported agri-foods should be verified using the proper official analytical method for each of them that has not registered in Korea. METHODS AND RESULTS: This work was conducted to apply the official method of Korea Food & Drug Administration (KFDA) for determining multi class pesticide multiresidues in agricultural commodities. Brown rice and orange which have different characteristics as a matrix were selected as representative samples for residue analysis. The recoveries of cycloate, diallate and pebulate by GC/MS in fortified brown rice and orange with levels of 0.04~0.4 mg/kg were ranged from 82.8% to 110.3%. The quantification limits of three pesticides in brown rice and orange were 0.04 mg/kg. CONCLUSION: As a result, this method can surely be used as an official method for routine analysis of unregistered pesticides in Korea for imported agri-food.

Effects of Transgenic Soybean Cultivation on Soil Microbial Community in the Rhizosphere (형질전환 콩 재배가 근권 토양 미생물상에 미치는 영향)

  • BACKGROUND: Soybean [Glycine max (L.) Merrill] is a legume and an important oil crop worldwide. This study was conducted to evaluate the possible impact of transgenic soybean cultivation on the soil microbial community. METHODS AND RESULTS: Microorganisms were isolated from the rhizosphere soils. Microbial community was identified based on the culture-dependent and molecular biology methods. The total numbers of bacteria, fungi, and actinomycete in the rhizosphere soils cultivated with transgenic and non-transgenic soybeans were similar to each other, and there was no significant difference between transgenic and non-transgenic soybeans. Dominant bacterial phyla in the rhizosphere soils cultivated with transgenic or non-transgenic soybeans were Actinobacteria, Firmicutes, and Proteobacteria. The microbial communities in transgenic and non-transgenic soybean soils were characterized using the denaturing gradient gel electrophoresis (DGGE). The DGGE profiles showed the different patterns, but didn't show significant difference to each other at 0.05 significance level. DNAs were isolated from soils cultivating transgenic or non-transgenic soybeans and analyzed for persistence of transgenes in the soil by using PCR. PCR analysis revealed that there were no amplified ${\gamma}$-tmt and bar gene in soil DNA. CONCLUSION(S): The results of this study suggested that microbial community of soybean field were not significantly affected by cultivation of the transgenic soybeans.

Forest Vegetation and Floristic Studies of Mt. Gaya National Park - Especially on the Danji - bong Area - (가야산국립공원일대의 식생 및 식물상 연구 -단지봉 지역을 중심으로-)

  • This study aims to investigate the flora of Mt. Gaya National Park, especially focused on the forested wetland vegetation of Danji-bong area. The flora of Mt. Gaya was investigated from April in 2003 to October in 2004. Vascular plants of this area consist of total 465 taxa, 4 hybrids, 12 forma, 75 varieties, 1 subspecies, 373 species, 268 genera, 91 families. It corresponds to 11.4% of totally 4,071 taxa appeared in Korea. Korean endemic plants were composed 11 families, 15 genera, 14 species, 1 variety and totaling 15 taxa. And also actual vegetation structures were analyzed the newly founded forested wetland and natural population Ajuga spectabilis on the Danji-bong area using the vegetation and the species composition by tree layer.

Investigation of Nonylphenols Contamination in Solvents and Solid-phase Extraction Cartridge, and its Removal Protocols (정밀분석용 용매 및 SPE의 Nonylphenols 오염평가 및 제거)

  • Nonylphenols are toxic compounds classified as endocrine disruptors. We investigated the nonylphenols clean-up procedures for the contamination control in the quantitative analysis. In this research we analyzed the residual nonylphenols in the solvent and the SPE cartridges. First, at the analysis of HPLC grade solvents (n-hexane, diethyl ether, ethyl acetate and its mixture), diethyl ether was confirmed the residue as 0.963 ${\mu}g/mL$, and we eliminated the contaminant through the distillation with $CaH_2$, Second, at the analysis of SPE cartridges (silica gel and Florisil), all products were showed the residue at 0.046~13.0 ${\mu}g/mL$, but unfortunately the residue in the cartridge were not easily removed with referenced methods in all tested SPE cartridges except in silica gel SPE cartridge with glass ware.
