홍경천(紅景天)추출물(KH101)이 강제유영 흰쥐의 피로회복에 미치는 영향 (Effects of Rhodiola rosea (KH101) on Anti-fatigue in Forced Swimming Rats)

  • Objectives : Rhodiola rosea has been used in herbal medicine to treat various conditions, such as antimelancholia, antifatigue, improvement of work competence and prevention of altitude sickness. In this study, we investigated effects of Rhodiola rosea extract (KH101) on fatigue in forced swimming rats. Methods : Sprague-Dawley rats were induced with fatigue by forced swimming, then rats in each group were treated with KH101. We observed changes of glucose, LDH and cortisol in serum and LDH, glycogen, hexokinase, citrate synthase MDH, SDH and CK in muscle. Results : Obtained results were as follows: 1. Continuance times of exercise significantly increased in all groups at day 1, in the 50 mg/kg concentration group at day 2, in all groups at day 3 and in the 50 mg/kg conc. group at day 4. 2. In serum, glucose significantly decreased in all concentration groups. 3. In the soleus muscle, LDH significantly decreased in the 50 mg/kg concentration group. HK significantly decreased in the 100 mg/kg conc. group. SDH significantly increased in the 100 mg/kg conc. MDH were significantly decreased in all conc. groups. 4. In the gastrocnemius muscle, HK significantly decreased in all concentration groups, while MDH significantly increased all conc. groups. Conclusions : It is concluded that the KH101 has and anti-fatigue effect in rats. Additional studies are needed to find the mechanism of the association between each single herb.

홍경천(紅景天)이 뇌조직내출혈(腦組織內出血) 흰쥐의 뇌부종(腦浮腫)과 Matrix Metalloproteinase 발현에 미치는 영향 (Effects of Rhodiola Rosea on Brain Edema and Matrix Metalloproteinase Expressions Following Intracerebral Hemorrhage in the Rat)

  • Objectives : This study aimed at evaluation of the effects of Rhodiola rosea on brain edema and expressions of matrix metalloproteinases (MMPs) related to blood-brain barrier (BBB) disruption. Methods : Brain edema following intracerebral hemorrhage (ICH) was induced by the stereotaxic intrastriatal injection of bacterial collagenase type VII in rats (Sprague-Dawley). Then ethanol extract of Rhodiola rosea was treated once a day for 3 days. Brain edema % and water contents, and BBB leakage were examined. Immunohistochemistry was processed for MMP-9, MMP-12, and iNOS expressions in the brain sections and each immuno-labeled cells were analyzed with image analysis software. Results : 1. Ethanol extract of Rhodiola rosea reduced brain edema following ICH in rats significantly. 2. Ethanol extract of Rhodiola rosea reduced excessive brain tissue water contents following ICH in rats significantly. 3. Ethanol extract of Rhodiola rosea reduced BBB leakage in the cerebral cortex following ICH in rats. 4. Ethanol extract of Rhodiola rosea reduced cellular edema of neurons in peri-hematoma and the cerebral cortex following ICH in rats significantly. 5. Ethanol extract of Rhodiola rosea reduced MMP-9 positive cells in the cerebral cortex following ICH in rats significantly. 6. Ethanol extract of Rhodiola rosea reduced MMP-12 positive vessels in the cerebral cortex following ICH in rats significantly. 7. Ethanol extract of Rhodiola rosea reduced iNOS positive cells in the cerebral cortex and external capsule following ICH in rats significantly. Conclusions : These results suggest that Rhodiola rosea reveals protective effect against brain edema and cytotoxic edema of neurons by means of down-regulation of MMPs and iNOS expressions, and inhibition of BBB leakage.

홍경천 섭취와 운동수행이 비만 쥐의 인슐린 민감도와 골격근내 당수송 관련 단백질 발현에 미치는 영향 (Effect of Rhodiola Sachalinensis Administration and Endurance Exercise on Insulin Sensitivity and Expression of Proteins Related with Glucose Transport in Skeletal Muscle of Obese Bucker Rat)

  • Peripheral insulin resistance in obese/type II diabetes animals results from an impairment of insulin-stimulated glucose uptake into skeletal muscle. Insulin stimulate the translocation of GLUT4 from intracellular location to the plasma membrane. Soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs) is implicated in mediation of fusion of GLUT4-containing vesicle with the plasma membrane. Present study investigated regulatory effects of Rhodiola sachalinensis administration and exercise training on the expression of GLUT4 protein and SNAREs protein in skeletal muscles of obese Zucker rats. Experimental animals were randomly assigned into one of five groups ; lean control(LN), obese control(OB), exercise-treated(EXE), Rhodiola sachalinensis-treated(Rho), combine of Rho & EXE (Rho-EXE). All animals of exercise training (EXE, Rho-EXE) performed treadmill running for 8 weeks, and animals of Rho groups (Rho, Rho-EXE) were dosed daily by gastric gavage during the same period. After experiment, blood were taken for analyses of glucose, insulin, and lipids levels. Mitochondrial oxidative enzyme (citrate synthase, CS ; $\beta$-hydroxyacyl-CoA dehydrogenase, $\beta$-HAD) activity were analysed. Skeletal muscles were dissected out for analyses of proteins (GLUT4, VAMP2, syntaxin4, SNAP23). Results are as follows. Exercise and/or Rhodiola sachalinensis administration significantly reduced body weight and improved blood lipids (TG, FFA), and increased insulin sensitivity. Endurance exercise significantly increased the activity of mitochondrial enzymes and the expression of GLUT4 protein, however, administration of Rhodiola sachalinensis did not affect them. The effect of exercise and/or Rhodiola sachalinensis administration on the expression of SNARE proteins was unclear. Our study suggested that improvement insulin sensitivity by exercise and/or Rhodiola sachalinensis administration in obese Zucker rats is independent of expression of SNARE proteins.

홍경천 추출물이 Streptozotocin으로 유도된 당뇨쥐에 미치는 영향 (Effects of Rhodiola sachalinensis Extract in Streptozotocin-Induced Diabetic Rats)

  • The present study was undertaken to evaluate the effect of Rhodiola sachalinensis root ethanol extract (RS), on the body weight, organ weight, plasma glucose and plasma lipid in diabetic rats caused by streptozotocin (STZ). The body weight decreased more slowly in the RS group than in the diabetic, and the food intake increased significantly in all diabetic groups. The food efficiency was very low in all diabetic groups, but increased significantly in the RS groups than diabetic control (p<0.05). In comparing the weight of organ, the weight of liver and kidney were increased in all diabetic groups than in the control, and decreased slightly in RS groups. The weight of heart and spleen were not different among all test groups. The glucose in serum was decreased significantly in the RS groups fed the RS for 4 weeks, compared to the diabetic control (p<0.05). Total cholesterol, triglyceride and atherogenic index (AI) in serum were significantly higher in diabetic control, compared to the normal (p<0.05), and decreased $16.7\%,\;18.3\%\;and\;45.0\%$, respectively, in the RS fed $300\;\cal{mg/kg}$ of RS. HDL-cholesterol was increased slightly more in the $RS-300\;\cal{mg/kg}$, compared to diabetic control. These findings suggest that RS treatment has protective effect in diabetes.

흡착 공정을 활용한 홍경천(Rhodiola sachalinensis) 유산균 발효물의 이화학적 특성 및 항산화 활성 (Physicochemical Properties and Antioxidative Activity of Lactic Acid Bacteria Fermented Rhodiola sachalinensis using Adsorption Process)

  • Rhodiola sachalinensis fermentates by lactic acid bacteria were prepared using the adsorption process, and were investigated for changes of the main compounds and anti-oxidative activities during the adsorption and fermentation process. While the R. sachalinensis extract (RSE), which did not go through the adsorption process, showed little change in pH during fermentation and a significant reduction in the number of lactic acid bacteria, the pre-preparatory adsorption process was found to be helpful for promoting fermentation and for maintenance of bacterial numbers. The contents of total phenolic compounds mostly decreased during the adsorption process, but showed an increasing tendency to rebound during the fermentation process. The contents of salidroside and p-tyrosol in the RSE were 1153.3 mg% and 185.0 mg% respectively, and they did not significantly change after treatment with acid clay or bentonite as adsorbents, which were 1093.0 and 190.5 mg% by acid clay, and 882.2 and 157.3 mg% by bentonite. When the extract was fermented after treatment with acid clay or bentonite, the salidroside contents were decreased by 282.7 and 505.0 mg% respectively, but the p-tyrosol contents were increased by 714.0 and 522.4 mg% respectively. Compared to the DPPH radical scavenging activity of the RSE (66.8%) at the conc. of 0.1%, that of the fermented RSE, which went through adsorption process with acid clay or bentonite, was significantly increased to 79.4 and 72.7% respectively at the same concentration (p<0.05). Though fermentation by lactic acid bacteria was suppressed in the RSE, the results suggested that the adsorption process may promote fermentation without any change in the content of major active compounds. It is expected that fermentation by lactic acid bacteria could improve the antioxidant activity and various associated functionalities of R. sachalinensis.

COMPAREX 의 항염 및 진정 화장품 소재로서의 연구 (A Study on Anti-inflammatory and Skin Soothing Cosmetics Material of COMPAREX)

  • 최근 다양한 외부 요인에 의해 민감해진 피부를 진정시키기 위해 더욱 안전하고 효과적인 진정 소재 개발에 대한 관심이 증가하고 있다. 따라서 본 연구에서는 항염 효능으로 잘 알려진 마치현 추출물을 기반으로 애엽, 홍경천 추출물을 혼합함으로써 COMPAREX를 제조하고, 피부 진정 효능에 미치는 영향을 마치현 단일 추출물과 비교하였다. 실험 결과, COMPAREX는 마치현 추출물 보다 LPS에 의해 유도된 NO 생성을 더욱 억제시키고, 염증성 인자인 iNOS, COX-2, TNF-α, IL-6의 발현을 현저히 억제시킴으로써 염증완화 효능이 더 효과적인 것을 확인하였다. 또한, COMPAREX는 마치현 추출물 보다 SDS 자극에 대해 IL-1α의 유전자 및 단백질 발현을 더욱 억제시킴으로써 진정 효능이 더 효과적인 것을 확인하였다. 추가적으로, 모발세포에서 COMPAREX는 PM2.5에 의해 증가된 COX-2, IL-8의 발현을 억제시켜 두피 염증 완화 효능이 있으며, carbonylated protein을 저해시킴으로써 외부 자극으로부터 손상된 두피를 보호할 수 있는 효능을 확인하였다. 따라서, COMPAREX는 마치현 단일 추출물 보다 개선된 천연 진정 소재로의 활용 가능성을 보였고, 향후 더마 코스메틱 소재로 활용될 수 있을 것으로 기대된다.
