Nutritional Composition, Ginsenoside Content and Fundermental Safety Evaluation with Leaf and Stem Extract of Panax ginseng (인삼잎과 줄기 혼합 추출물의 영양성분, Ginsenoside 함량 및 기본적 안전성 평가)

  • This study was conducted to investigate the application possibility of leaf and stem extract (LSE) from the mixture of leaf and stem of Panax ginseng. This study measured the general nutritional composition, aminoacid minerals contents and fatty acid composition of LSE. We conducted analysis of the ginsenoside content by HPLC and the cell cytotoxicity tests in normal liver and kidney cells. The approximate composition of LSE was 2.51% of carbohydrate 0.53% of crude ash,0.20% of crude fat and 0.15% of crude protein, respectively. LSE contained 102.56 mg/100 g of K ion and high contents of acidic amino acids such as glutamic acid and aspartic acid. In addition to this, it contained all essential amino acids. The major compositions of fatty acids were 39.99% of palmitic acid 14.96% of linoleic acid, 13.31% of docosatetranoic acid and 12.91% of linolenic acid, The total ginsenoside was 0.82 mg/mL, and ratio of PD/PT was 0.68. Negative effects were not found from the results of the cell toxicity respection. These results imply that leaf and stem of Panax gineng could be used as possible food resources and functional food material and feed stuff.

Effects of mixed supplementation on Hoveni dulcis Thunb extracts and Ginseng-Berry extracts on hangover curves (헛개과병추출물과 인삼열매추출물의 혼합 음료 섭취가 숙취해소에 미치는 효과)

  • The purpose of this study was to investigate the effects of ingestion of rabies and ginseng fruit extracts on alcohol hangover, liver damage protection, fatigue recovery, and physical strength improvement. A total of 64 volunteers aged over 20 were participated in this study and the randomized and repeated measures design method was used to divide a group of participants with a random assignment. All participants were divided into 4 groups (n=16) treated with hoveni dulcis thunb extract + ginseng berry extract (ARI 1000), hoveni dulcis thunb extract, ginseng berry extract, and placebo. As a result of respiratory alcohol concentration change, the group treated with ARI 1000 was significantly lower than the group treated with hoveni dulcis thunb extract, ginseng berry extract, and placebo in 1 hour of drinking, and significantly lower than the placebo group in 2 hours and 3 hours of drinking (p<0.05). After 2 and 3 hours of alcohol consumption, blood alcohol concentration of the group treated with rabies ARI 1000 was significantly lower than those of the other 3 groups (p <0.05). In conclusion, ingestion of ARI 1000 before drinking may significantly reduce the respiratory and blood alcohol concentrations, which may induce an effect on the hangover effect.

Antimicrobial Plant Extracts as an Alternative of Chemical Preservative: Preservative Efficacy of Terminalia chebula, Rhus japonica (gallut) and Cinnmomum cassia Extract in the Cosmetic Formular (가자, 오배자, 계피 추출물을 이용한 화장품 제형에서의 방부효과)

  • This study was carried to investigate the efficiency of antimicrobial plant extracts as natural preservative in the cosmetic formulations. Ethanol extracts of different plants were tested using the disc diffusion (paper disc) method and the minimum inhibitory concentration (MIC) method for their antimicrobial activity against the common poultry pathogens. Terminalia chebula and Rhus japonica (gallut) extracts exhibited antibacterial activity against Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli. Cinnmomum cassia extract exhibited antifungal activity against Candida albicans and Aspergillus niger while the remaining plant extracts showed no activity. A study of the preservative efficacy of the cosmetic formular containing the T. chebula, R. japonica and C. cassia extracts demonstrated sufficient preservative efficacy against bacteria and eukaryotic test microbes. Also, the cosmetic formulations containing antimicrobial plant extracts more effectively inhibited the microoranisms than the mixture of traditional chemical preservatives. These results suggest that the mixture of antimicrobial plant extracts, T. chebula, R. japonica and C. cassia is incorporated as preservative in the cosmetic formulation and the mixture have considerable effect on its efficacy.

Antioxidant Effect of Aqueous Extract Obtained from Green Tea (녹차 물추출물의 항산화효과)

  • The antioxidant effects of aqueous green tea extract(AGTE) obtained from green tea and its combinations with several synergists on lard were investigated to equalize with that of crude catechin in green tea. The antioxidant effect of crude catechin was about 3-fold higher than that of AGTE. The suitable synergists for AGTE were ${\delta}-tocopherol$ and lecithin, while the tendency of their synergistic effects was different from each other. Addition of ${\delta}-tocopherol$ within 0.05% dosage to lard showed significant synergistic effect, and its synergistic effect decreased with the concentration of AGTE over 0.1% dosage. In combination of AGTE and lecithin, they gradually enhanced synergistic effect on lard with increasing the concentration. The combinations of AGTE, ${\delta}-tocopherol$ and lecithin showed intense synergistic effects and the optimum level of the each component was 0.1% AGTE, 0.05% ${\delta}-tocopherol$ and 1% lecithin. By adding the optimum level of three components to lard, the induction period was lenghtened approximately 8-fold than that of the control owing to their synergism. Comparing the antioxidant effect between the AGTE with synergists and crude catechin, the former was 60% and 20% higher than the latter at 0.1% and 1% dosage respectively.

마이크로웨이브 공정에 의한 마늘 올레오레진의 추출조건 설정 및 특성

  • 마늘을 MAE(microwave-assisted extractiona)방법에 의하여 물, 30% 에탄올 및 50% 에탄올로 추출하고 그 특성을 비교하였다. 마늘과 각 용매의 비율은 1:2.5로 하여 마이크로웨이브 60W로 각각 5분 및 20분 동안 추출하였다. 마늘 올레오레진 수율은 50% 에탄올로 20분동안 추출한 추출물이 14.1%로 가장 높았으며 polyphenol함량도 물로 20분동안 추출한 경우가 579.0mg%로 가장 많은 함량을 나타내었다. 전자공여작용 효과 및 피르브산 함량도 물로 20분 동안 추출한 추출물이 각각 32.7% 및 마늘 1g당 13.4$\mu$molus로 가장 높게 나타났다. 각 용매에 대해 20분동안 추출한 추출물들을 37$^{\circ}C$에서 8일동안 저장하면서 피르브산 함량과 전자공여작용의 변화를 조사한 결과, 피르브산 함량은 물 추출물이 에탄올 추출물보다 높은 함량을 나타내었고 저장 시간이 증가할수록 그 함량은 감소하는 경향을 나타내었다. 전자공여작용도 물 추출물이 가장 높은 효과를 나타내었으며 추출시간에 비례하여 증가하다가 저장 6일째부터 감소하는 경향을 나타내었다. 각 용매에 대한 추출물의 색깔을 조사한 결과 물 추출물이 가장 밝고 옅은 갈색을 나타내었으며 30% 에탄올 추출물이 가장 어둡고 푸르스름한 노란색을 나타내었다. 또한 각 추출물들을 37$^{\circ}C$에서 10일간 저장하면서 갈색화 정도를 측정해 본 결과 물 추출물이 에탄올 추출물이 에탄올 추출물보다 갈색화가 많이 진행되었으며 0.1% cysteine을 첨가한 추출물과 에탄올 추출물의 갈색화는 비슷한 경향을 나타내었다. 마늘 추출물을 추출한 후에 0.1% cysteine을 첨가하여 저장한 것 보다 추출하기 전에 첨가하는 경우가 갈색화 억제 효과가 뚜렷하였다.문에 밀가루에 일부를 대용한 wheat flour dough를 사용하고 가정용 제빵기로 구워 최종 단계에까지의 제빵성 결과를 산출했다. amarans folur 5%의 대체에는 빵의 비용적이 비교적 증대했지만 그 이상 amarans flour을 대처하면 확연히 비용적은 감소했다. amarans flour 10% 대체에 hemicellulase 1250U 이상을 첨가하면 비용적은 눈에 띄게 증대했다. farinograph에 있어서 반죽의 안정성은 amarans flour 10% 대용에 현저히 감소했다. 반죽의 점탄성(아축응력, 탄성률, 점성계수)는 amarans flour 10%를 대용한 것이 무첨가한 것보다 많이 단단해졌음을 알 수 있었다. 혼합중의 반죽의 조사형 전자현미경 관찰로 amarans flour로 대체한 gluten이 단단해졌음을 알수 있었다. 유화제 stearly 칼슘, 혹은 hemicellulase를 amarans 10% 대체한 밀가루에 첨가하면 확연히 비용적을 증대시킬 수 있다는 사실을 알 수 있었다. quinoa는 명아주과 Chenopodium에 속하고 페루, 볼리비아 등의 고산지에서 재배 되어지는 것을 시료로 사용하였다. quinoa 분말은 중량의 5-20%을 quinoa를 대체하고 더욱이 분말중량에 대하여 0-200ppm의 lipase를 lipid(밀가루의 2-3배)에 대하여 품질개량제로서 이용했다. 그 결과 quinoa 대량 7.5%에서 비용적, gas cell이 가장 긍정적 결과를 산출했고 반죽의 조직구조가 강화되었다. 또 quinoa 대체에 의해 전분-지질 복합제의 흡열량이 증대된 것으로부터 전분-지질복합제의 형성 촉진이 시사되었다.이것으로 인하여 호화억제에 의한 노화 방지효과가 기대되었지만 실제로 빵의 노화는 현저히 진행되었다. 이것은 quinua 대체량 증가에 따른 반죽의 안정성이 저하되어 버린 것으로 생

Cytotoxicity of Ethanol Extract of Cephaxus koreana on Human Breast Cancer Cell Lines (유방암 세포주를 애용한 개비자 나무의 에탄올 추출물의 항암 활성 분석)

  • This study was designed to investigate the cytotoxic effect of Cephaxus koreana on human breast cancer cell lines (SCC-1395 and SK-BR-3) us ins MTT(3- [4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay. RPMII -1640 medium with $10\%$ FBS and $1\%$ of penicillin-streptomycin was used. The ethanol extrart of Cephaxus koreana after adsoption resin treatment showed higher anticancer activity against SCC- 1395 than the ethanol extract of Cephaxus koreana without adsoption resin treatment. SK-BR-3 showed higher sensitivity activity than SCC-1395 at the same concentration of 20000 ppm.

Antimicrobial Effect of Lithospermum erythrorhizon Extracts on the Food-borne Pathogens (지치추출물의 식중독성 미생물에 대한 항균효과)

  • Antimicrobial effect of Lithospermum erythrorhizon extracts against food-borne pathogens was investigated. L. erythrorhizon was extracted with methanol at room temperature, and the extraction was sequentially fractionated using petroleum ether, chloroform, ethyl acetate, and methanol. Antimicrobial activity of L. erythrorhizon extracts was determined using paper disc method against food-borne pathogens and food spoilage bacteria. Ethyl acetate extracts of L. erythrorhizon showed the highest activity against Staphylococcus aureus and Shigella dysenteriae. Synergistic effect was found in combined extracts of L. erythrorhizon and Sophora subprostrata as compared with each extract alone. Growth inhibition curve was determined using ethyl acetate extracts of L. erythrorhizon, against S. aureus and S. dysenteriae. Ethyl acetate extract of L. erythrorhizon, showed strong antimicrobial activity against S. aureus at 4,000 ppm, retarding growth of S. aureus more than 48 hr and S. dysenteriae up to 12 hr.

Antioxidative Properties and Whitening Effects of the Eucommiae cortex, Salviae miltiorrhizae radix, Aurantii nobilis pericarpium and Cnidii rhizoma (두충, 단삼, 진피 및 천궁의 항산화 활성 및 미백 효과)

  • In an attempt to find natural sources of antioxidants and whitening agents, comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Eucommiae Cortex, Salviae miltiorrhizae radix, Aurantii nobilis pericarpium and Cnidii rhizoma were carried out. Comparison of the four ethanol extracts revealed that, Aurantii nobilis pericarpium had the highest electron-donating ability(79.0%),; however, Salviae miltiorrhizae radix had the highest SOD-like ability(21.9%). The xanthine oxidase experiment exhibited a hindrance effect of 79.3% in Salviae miltiorrhizae radix, 57.5% in Eucommiae cortex and 71.9% in Aurantii nobilis pericarpium. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 12.4% in the Eucommiae cortex, 22.8% in the Salviae miltiorrhizae radix, 27.5% in the Aurantii nobilis pericarpium and 59.5% in the Cnidii rhizoma. Based on these results, we suggest that the ethanol extracts of Eucommiae cortex, Salviae miltiorrhizae radix, Aurantii nobilis pericarpium and Cnidii rhizoma. can be used as food and cosmetic ingredients.

Comparison of Antioxidant, Wrinkle Improvement, and Whitening Efficacies of Extracts from Pinus koraiensis Cone Scale Using Extraction Methods (추출방법에 따른 잣나무(Pinus koraiensis) 구과피 추출물의 항산화, 주름개선 및 미백 효능 비교)

  • The present study evaluated the antioxidant activity, skin wrinkle reduction, and whitening activity of Pinus koraiensiscone scale extracts made with three different solvents (ethanol, supercritical fluid, and a mixture of both). Total polyphenol content was 11.03 mg/g GAE in the supercritical fluid extractand 33.79 mg/g GAE in the 70% ethanol extract. Electron donating ability of 1,000 ㎍/mL extract was 13.60% in the supercritical fluid extract, 91.37% in 70% ethanol extract, and 71.62% in mixed solvent extract. SOD-like activities in 100 ㎍/mL extract using supercritical fluid, 70% ethanol, and mixed solvents were 16.49%, 21.84%, and 10.7%, respectively. The ABTS+ radical scavenging activities of 1,000 ㎍/mL extract were 38.19%, 80.23%, and 78.72%for supercritical fluid extract, 70% ethanol extract, and mixed solvent extract, respectively. Tyrosinase inhibitory activities in 1,000 ㎍/mL extract were 24.54%, 36.55%, and 15.23% for supercritical fluid extract, 70% ethanol extract, and mixed solvent extract, respectively. Elastase inhibitory activities in 1,000 ㎍/mL extract were 15.62%, 22.56%, and 26.64% for supercritical fluid extract, 70% ethanol extract, and mixed solvent extract, respectively. Skin astringent activity (81.23% with 5000 ㎍/mL extract) was only detected in the 70% ethanol extract. Supercritical fluid and mixed solvent extracts showed no such activity. Our analysis of Pinus koraiensiscone scale extracts show that the highest aggregate activity was found in the 70% ethanol extract, followed by mixed solvent and supercritical fluid extracts. Therefore, our results oppose the hypothesis stating supercritical extract has the highest total polyphenol content and antioxidant activity.

Dough Characteristics and Biological Effects of Mixed Flour of Buckwheat and Wheat (메밀 혼합분의 반죽특성과 생리활성 검색)

  • This study was to investigate the mixed buckwheat flour quality by observing antimutagenic and cytotoxic effects of mixed buckwheat flour extracts using Ames test and SRB (sulforhodamine B) assay. Samples were prepared to the ratio of 100% (B1), 90% (BF1), 80% (BF2), 70% (BF3) and 60% (BF4) (w/w) flour buckwheat based on wheat flour weight. The initial pasting temperature in an amylograph was increased according to the increase of the buckwheat flour. The water absorption in farinograph decreased with the addition of buckwheat flour. The inhibition rates of B1, BF3 and BF4 extract (160 g/plate) were 45%, 37.3% and 42% against the mutagenesis of Salmonella Typhimurium TA100 induced by MNNG $(0.4{\mu}g/plate)$, respectively. In addition, the B1 at the same concentration showed 64% and 44.3% inhibition on the mutagenesis of Salmonella Typhimurium TA98 and TA100 induced by 4NQO $(0.15{\mu}g/plate)$, respectively. In SRB assay, human breast adenocarcinoma (MCF 7), human hepatocellular carcinoma (Hep3B), human stomach adonocarcinoma (AGS), human lung carcinoma (A549) and human cervical adenocarcinoma (HeLa) proliferations were inhibited by the increase in the sample concentration.
