Simultaneous Determination of Various Pesticides : Analysis Utilizing GC/MSD (SIM mode) (잔류농약 다성분 동시분석법 : GC-MSD (SIM mode)를 사용한 분석)

  • An analytical method for simultaneous and rapid determination of organophosphorus, organochlorine, carbamate, and pyrethroid pesticides in polished rice was developed. The analysis is performed by gas chromatograph with mass selective detector (GC/MSD) in selected ion monitoring (SIM) mode. Pesticides were extracted from samples with acetone by automated Soxhlet apparatus and this extract was evaporated to dryness for the analysis. The residue was dissolved in hexane, followed by a treatment with a Sep-Pak florisil catridge. Pesticides were positively confirmed by GC/MS, retention times, and ion ratios. This analytical method allows a rapid, reliable, and a good recovery of hydrophilic pesticides. Except far captafol, captan, dichlofluanid and dichlorvos, recoveries of 42 pesticides were over 70%.

Synergistic Inhibition of IgY, Auricularia auricula, and Lactic Acid Bacteria from Kimchi and Tarak on Helicobacter pylori (Helicobacter pylori 의 생육억제에 대한 유산균, 난황항체 및 목이버섯의 상승효과)

  • The substances of lactic acid bacteria (LAB) isolated feom Kimchi and Tarak, L. mesenteriodes LAB kw5, and S. thermophilus LAB KW15 were investigated for growth effect of Helicobacter pylori with IgY and Auricularia auricula. Inhibition of H. pylori was confirmed at LAB KW5 and KW15 supernatants. Interestingly, anti-H. pylori substance in LAB KW5 and KW15 supernatants were sensitive to lipase, but insensitive to protein hydrolase and carbohydrate hydrolase. The inhibition zone toward H. pylori was not shown with the lipase-treated supernatants. Therefore, there seemed to be lipid-like substances in the cultures. By the analyses with gas chromatography, undecanoic acid ($C_{11:0}$), palmitic acid ($C_{16:0}$), stearic acid ($C_{18:0}$), and oleic acid ($C_{18:1}$) were detected at the culture substances from L. mesenteroides LAB KW5 and S. thermophilus LAB KW15, and more eicosadienoic acid ($C_{20:2}$) from L. mesenteroides LAB KW5. Anti-H. pylori substances of LAB with IgY and A. auricula extract were analyzed for inhibition effect of H. pylori. The inhibition increased more by the range from 57% to 86% by the mixture. The substances with IgY and A. auricula extract showed more effective inhibition of H. pylori than single or double trials.

  • This study was carried out to evaluate characteristics of Chrysanthemum sp. and to bioassay Tetranychus urticae, Nilaparvata lugens, and Aphis gossypii by different extracts part and method. Flower type of Chrysanthemum sp. line was anemone except 0706 line which was pompon. The flowering date was from April to June but 0721 line kept up November. They could overwinter except 0706 line. The Ethanol extracts of 0718 line of flower was high mortality as 63.8% on Tetranychus urticae. Ethanol mixed emulsifier extracts were the highest mortality as 69.4% at 5 days after. Mortality in water extract was lower than in ethanol and ethanol mixed emulsifier extracts. Ethanol mixed emulsifier extracts were higher mortality as 69.4% at 5 days after in 0718 line on Nilaparvata lugens. Mortality in water extract was lower than in ethanol and ethanol mixed emulsifier extracts. On Aphis gossypii, Chrysanthemum flower extracts could decrease the density of aphids in early days but the density of that was increase as time goes by because aphids produce offspring.

Identification of Pigment Separated from Cockle Shell (새조개 연체부위의 색소 동정)

  • Physicochemical characteristics of pigment separated from cockle shell were studied. Pigment on the cockle shell was peeled with distilled water, and soaked in 4% TCA solution for 30 minutes and filtered. Filtrate was washed 3 times with distilled water and Iyophilized. Dried pigment was dissolved with 2N NaOH solution and preparative TLC with 2N NaOH : acetone(1 : 1). UV/vis spectrum of pigment separated from cockle shell and melanin as standard was same one peak at UV area and λ$_{max}$was 226nm. Dissolved property of pigment was same as that of melanin. Pigment was not ommochrome but like melanin, because it was insoluble in formic acid, whitening in $H_2O_2, $ and pattern of UV-spectrum was same as melanin. Pigment separated from cockle shell was stable at high temperatures. In thermal treatment at $70^{\circ}C, $ $80^{\circ}C, $ $90^{\circ}C$ and $100^{\circ}C$ for 8 hours, retention ratio of pigment separated from cockle shell were 95.0%, 93.3%, 90.8% and 87.6%, respectively.

Antioxidant activities of blueberry hot water extracts with different extraction condition (추출조건에 따른 블루베리 열수추출물의 항산화 활성 비교)

  • Five extraction conditions (AE, autoclave extraction; OE, oven extraction; HWSE, hot water and sonication extraction; HWASE, hot water acidified with 0.5% (v/v) acetic acid and sonication extraction; and BE, boiling extraction) were examined to compare the effects of different hot water extraction methods on the antioxidant properties of blueberries. The extraction yields of the AE, OE, HWSE, HWASE, and BE were 7.94%, 8.35%, 8.55% 9.15%, and 8.50%, respectively. The polyphenol and flavonoid contents of AE were 3.47 mg GAE/g and 1.59 mg RE/g, respectively, which were highest centents among others. Those of OE were ranked second to the highest. The total anthocyanin content of HWSE (5.29 mg/g) was significantly higher than that of others whereas that of AE showed the lowest content (0.96 mg/g). The order of ABTS radical and alkyl radical scavenging activities was as follows: AE > BE > OE > HWSE > HWASE. The antioxidant properties were considerably correspondent with the total polyphenol and flavonoid content. DPPH radical scavenging activity was quite high in HWSE, AE, and BE extraction, however, there were no significant differences among the five extraction methods in the aspect of $Fe^{2+}$ ion chelating activities. Moreover, AE showed the highest SOD activity, and protected the dermal fibroblast the best against $H_2O_2$-induced cytotoxicity. In conclusion, it was suggested that the autoclave extraction (AE) would be the most effective method for preparing blueberry hot water extracts with relatively high antioxidant activities.

Isolation and Characterization of Microorganisms with Broad Antifungal Activity against Phytopathogenic Fungi (식물병원균에 광범위 항균활성을 가진 미생물의 분리 및 특성)

  • For the production of an antifungal compound, one strain (I-8) was selected from approximately 400 strains isolated from various soil samples. The optimum carbon source, nitrogen source and pH culture conditions for the production of the antifungal compound were investigated. ISP No. 2 medium (yeast extract 0.4%, malt extract 1% and dextrose 0.4%, at pH 8) was determined to be the optimum medium. Strain I-8 showed broad antifungal activity against the plant pathogenic fungi tested, including Sclerotinia sclerotiorum KACC 41065, as well as cellulase and chitinase activities in an agar plate assay. The extraction of antifungal compounds was performed using ethyl ether and ethyl acetate. In a culture broth of strain I-8, the ethyl acetate extract exhibited effective growth inhibition against 14 of the 20 phytopathogenic fungi tested. By mixing the ethyl acetate extract from I-8 with the ethyl ether extract from the fungus 13-16, which shows specific antifungal activity against Colletotrichum orbiculare KACC 40808, the antifungal activity of I-8 against phytopathogenic fungi was confirmed to be slightly increased. Strain I-8 showed strong growth inhibition against 16 phytopathogenic strains in agar plate tests.

Anti-aging Cosmetic Application of Novel Multi-herbal Extract Composed of Nelumbo nucifera Leaves, Saururus chinensis and Orostachys japonica (하엽, 삼백초 및 와송으로 구성된 식물복합추출물의 항노화 화장품 소재로서의 응용성 연구)

  • In this study, various plant extracts were screened for the effective and safe skin care ingredient according to the antioxidant activity assay guidelines. We selected an optimized combination herbal extract, Charmzone extract (CZE), which composed of Nelumbo nucifera leaves, Saururus chinensis and Orostachys japonica. CZE exerted free radical scavenging activity. It reduced reactive oxygen species formation and increased total antioxidant capacities in human keratinocyte (HaCaT) and normal human dermal fibroblasts (NHDF). CZE also leaded procollagen type 1 secretion in NHDF and decreased cellular melanin contents in B16F10. The production of nitric oxide was decreased by CZE in lipopolysaccharide-stimulated RAW264.7 in a dose dependent manner. Therefore, it is concluded that CZE can be applied for naturally derived anti-aging functional skin care ingredient for anti-oxidation, wrinkle enhancement, whitening, anti-inflammation and wound healing of skin.

The effect of Coptidis rhizoma, Chinese Galls and Phytoncide in Malassezia furfur (황련(Coptidis rhizoma), 오배자(Chinese galls), 피톤치드 혼합물(Phytoncide)이 비듬균(Malassezia furfur)에 미치는 영향)

  • The report explores the possibility that the single extract of natural substances and the mixture of single extracts of natural substances create a synergy effect to increase the antimicrobial activity. It also compares the previous researches to find out if the natural medicinal herbs’ extract has antimicrobial activity on dandruff causative organism, Malassezia furfur. For the experiment on the mixture of single extracts of natural substances, the results are like following: 1. Staphylococcus aures’s antibacterial activity is resistant to mixture of three natural substances. 2. Escherichia coil’s antibacterial activity is resistant to mixture with Coptidis rhizoma. 3. Candida albicans’ antifungal activity is resistant to mixture with Chinese galls, which means the different results may be expected when tested with each germ. 4. On the other hand, Malassezia furfur has no antifungal activity on the single extract of natural substances and shows weak antifungal activity, whose diameter is 3.20 mm when tested with the mixture of 50% of Coptidis rhizoma and 50% of Phytoncide. The result is totally different from the one on the same eumycetes, C. albicans. That is because M. furfur has more lipophilic chemicaled cell walls than C. albicans does and it also consists of lamella layer, inner plasma membrane and intermediate multiple layers.

Effects of Opuntia ficus-indica var. saboten Ripe Fruits on Protection of Neuronal PC-12 Cells and Cholinesterase Inhibition (백년초의 PC-12 신경세포 보호 및 콜린가수분해효소(cholinesterase) 저해 효과)

  • Oxidative stress caused by reactive oxygen species is ascribed to many neurodegenerative diseases like Alzheimer's disease. Phenolic antioxidants can reduce the oxidative stress. In this study, ripe fruits of Opuntia ficus-indica var. saboten (OFS) were extracted using 80% (v/v) aqueous ethanol. Total phenolic and flavonoid contents of the OFS fruits (100 g) were 409.9 mg gallic acid equivalents and 72.2 mg catechin equivalents, respectively. The OFS fruits had antioxidant capacity at 381.2, 298.2, and 3,219.9 mg vitamin C equivalents/100 g in ABTS, DPPH, and ORAC assays, respectively. The OFS fruits showed protective effects on PC-12 cells against oxidative stress in a dose-dependent manner, partly due to decrease of intracellular oxidative stress. Furthermore, the OFS fruits inhibited both acetylcholinesterase and butyrylcholinesterase. Consequently, these results suggest that the OFS fruits might be served as a source of functional materials to reduce oxidative stress in neuronal cells and to inhibit cholinesterases.

Bark Extractives of Several Populus Trees (몇가지 사시나무속 수종 수피의 추출성분)

  • The bark of P. alba × glandulosa, P. euramericana and P. nigra × maximounczii F1, several Populus trees, were collected, extracted with acetone-H2O(7:3, v/v), fractionated with hexane, chloroform and ethylacetate, and freeze dried to give some dark brown powder. Each fraction of the powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents and then identified by thin layer chromatography using TBA and 6% acetic acid as developing solvents. The structures of the isolated compounds were characterized by 1H, 13C and 2D-NMR tools including mass spectrometry. Most of the compounds were flavonoids and salicin derivatives as follows: (+)-catechin, taxifolin, aromadendrin, eriodictyol, naringenin, sakuranetin, sakuranetin-5-O-𝛽-D-glucopyranoside, neosaturanin, salireposide, p-coumaric acid, and aesculin from P. alba × glandulosa, (+)-catechin, salireposide, populoside and salicortin from P. euramericana and (+)-catechin, quercetin, padmatin, salireposide, populoside and salicortin from P. nigra × maximounczii F1.
