Production of Cellulolytic Enzymes by Trichoderma harzianum FJ1 in Solid State Fermentation. (Trichoderma harzianum FJ1의 고체상태배양에 의한 섬유소분해효소의 생산)

  • The cellulases production in solid state fermentation (SSF) of Trichoderma harzianum FJ1 with high cellulases productivity using cellulosic wastes was investigated. Physical and chemical conditions of the fermentation, such as moisture content, initial pH, and composition of mixed substrate (wine waste, rice straw, and soybean flour) on FPase (Filter paper activity) production were examined. The enzyme production was optimized in the conditions of moisture content of 70%, pH 5.0, 3$0^{\circ}C$, and 1:1:1 composition of mixed substrate containing wine waste, rice straw, and soybean flour. The highest activities of FPA, CMCase, Xylanase, $\beta$-glucosidase, and Avicelase in the optimized culture conditions were 15.2, 69.1, 83.9, 29.2, and 4.2 unit/g-SDW in 5 day cultivation, respectively. Economical and efficient production of cellulolytic enzymes by T harzianum FJ1 using cellulosic wastes in solid state fermentation will contribute to the biological saccharification of cellulosic wastes with enormous potential resource value in future.

The Effects of Aqueous Fibrolytic Enzyme Products Supplementation on In vitro Fermentation and Growth Performance of Hanwoo Steers (섬유소분해효소 발효액의 첨가가 In vitro 발효 및 거세한우의 성장에 미치는 영향)

  • The objective of this study was to investigate the effects of an enriched diet supplemented with aqueous fibrolytic enzyme products (FEP) on In vitro fermentation and growth performance of Hanwoo Steers. Total mixed ration combined with fibrolytic enzyme products showed a slight increase in the pH level on In vitro fermentation. Acetate ratio in volatile fatty acids, and the concentration of total volatile fatty acids were significantly greater at 12hr and 24hr after cultivation (p<0.05). Dry matter digestibility was significantly increased in groups supplied with aqueous fibrolytic enzyme products at 24hr after cultivation (p<0.05). In a rearing trial, the average daily gain and feed efficiency improved in the treatment groups supplied with aqueous fibrolytic enzyme products. Nutrient digestibility such as DM, crude fiber, NDF, and ADF digestibility were also significantly increased with the addition of aqueous fibrolytic enzyme products (p<0.05). Therefore, the aqueous fibrolytic enzyme products supplementation on In vitro fermentation is effective in improving the condition of rumainal fermantation. Also two kinds of aqueous fibrolytic enzyme products were found to be effective in improving the average daily gain and feed efficiency in Hanwoo Steers.

  • The multifunctional cation exchangers, sulfonated polypropylene-g-(acrylic acid/styrene) [PP-g-(AAc/Sty)] were synthesized by the irradiational grafting of AAc and Sty onto PP staple fabric with electron beam accelerator and its subsequent sulfonation. The highest degree of grafting obtained was 190% at a monomer mixture of 30 vol% AAc: 70 vol% Sty and a solvent mixture of 30 vol% water : 70 vol% methanol and the degree of grafting decreased with an increase of the AAc content in the monomer mixture at constant solvent content. Maximum ion exchange capacity of the copolymer was 4.6 meq/g. The Li$^{+}$ adsorption ability of the copolymer synthesized in the study was the best among PP-g- AAc, sulfonated PP-g-Sty, and sulfonated PP-g-(AAc/Sty).).

Effect of matrix on fatigue strength of carbon fiber composite materials (탄소섬유강화 복합재료의 피로강도에 미치는 모재의 영향)

  • In this study, the variation of fatigue strength in CF/PEEK and CF/EPOXY, the matrix and interfacial strength of which differ from each other, has been studied from the viewpoint of microfracture behavior. The results obtained are as follows; According as the fatigue strength moves from the lower cycle range to the higher cycle range, that of CF/PEEK shows higher curve than that of CF/EPOXY does. In the early stage of fatigue life, the characteristic of fatigue crack in CF/PEEK is mainly the fracture of longitudinal fiber, while that in CF/EPOXY is the fracture of transverse fiber. The difference of fatigue strength in these materials can be explained by the fracture criteria of transverse fiber and longitudinal fiber.

  • 근육 단백질과 우유 단백질간의 상호작용의 촉매제로써 TGase의 최적화를 위한 온도와 배양시간을 결정하고자 본 실험을 실시하였다. 돼지 등심에서 근원섬유 단백질을 추출하였고 배양온도는 4, $37^{\circ}C$로, 배양시간은 0, 0.5, 2, 4시간으로 설정하였다. 단백질의 열량분석, 점도, 겔 강도, 단백질 밴드의 변화를 측정하였으며, 그 결과 단백질 열량 변화는 각 단백질 별로 열량 변화 패턴이 상이하게 나타났으며 근원섬유와 카제인 염의 혼합액은 각각의 단백질의 피크와 유사하게 나타났고 배양시간과 온도에 따라 차이를 보여 $4^{\circ}C$에 비하여 $37^{\circ}C$에서 열량 변화가 크게 나타났다. 점도의 경우 배양하지 않은 것과 비교했을 때 $37^{\circ}C$에서 2시간 배양했을 때부터 유의적인 차이를 보이며 증가했으나 4시간 배양한 것과는 차이를 나타내지 않았다. 전기영동의 경우에도 $37^{\circ}C$에서 30분 배양한 처리구는 큰 변화를 나타내지 않았으나 2시간부터 저분자의 밴드가 소멸되고 고분자의 biopolymer를 형성되었다.

  • Fiber mats were made at five density levels, using fibers from kraft pulp screening rejects, rice straw and a 50/50 mixture of the two. They were soaked in the sulfur compounds. Specimens cut from the composite panels were tested in flexure at time intervals for one year to study the effect of aging. Modulus of elasticity (MOE) and modulus of rupture (MOR) were determined. Under optimum conditions of fiber mat preparation and saturation with molten sulfur and modified sulfur, composites were produced which exhibited mechanical properties comparable to conventional fiberglass in some properties and superior to conventional wood-based composition boards, For example. the moduli of elasticity of the reinforced composites made from pulp screening rejects, with a density of 0.35 gm/$cm^3$, were greater than 1,000,000 psi as compared 800.000 psi for high-density hardboard (1.28 gm/$cm^3$). Modulus of rupture of the best reinforced composites was about 7,000 psi, comparable to 6,000 psi of high-density hardboard.

  • Viable cells of periodontal ligament would be an important factor for the successful replantation of an avulsed tooth. Therefore, it is critical to choose the storage medium for the preservation of traumatically avulsed teeth. Growth factors and hormones could be considered for the therapeutic application of the maintenance of viable periodontal ligament fibroblasts (PDLFs). Epidermal growth factor (EGF) has been suggested as an important player for the regeneration and wound healing process on other tissues. Therefore, EGF was evaluated for the therapeutic application on avulsed teeth. In addition, the synergic effect of EGF with tri-iodothyronine (T3) and heparin-binding epidermal growth factor-like growth factor (HB-EGF). The cell proliferation of PDLFs was determined by MTT assay and increased dose-dependently up to 10 ng/ml in the presence of EGF. Maximum cellular growth was shown at the concentration of 10 ng/ml EGF. Also, EGF promoted the wound healing of PDLFs examined by in vitro wound healing assay. Combined effects of EGF with T3 or HB-EGF on the proliferation of PDLFs were also studied. Interestingly, EGF showed the synergic effect on the proliferation of PDLFs with T3 and HB-EGF. To find out the mechanism of the synergic effect of EGF and T3, the effect of T3 on the expression of endogenous EGF receptor was determined by RT-PCR. The result was that T3 enhanced the expression of EGF receptor in PDLFs. It suggested that EGF might be a good choice for a therapeutic application, which can be used as combination with T3 and HB-EGF.

Preparation of High-Fiber Bread with Soybean Curd Residue and Makkolli(Rice Wine) Residue (비지와 막걸리박을 이용한 고식이섬유 빵의 제조)

  • Soybean and residue contained 59.0% dietary fiber and makkolli residue contained 26.0% dietary fiber. The breads made from flour consisting of 90% wheat flour and 10% fiber materials were compared with that made with 100% wheat flour in terms of baking performance and consumer acceptance. Replacement of 10% wheat flour by soybean curd residue or makkolli residue increased water absorption and loaf weigh, but reduced loaf volume. Sensory panels could not accept favorably the bread made with the soybean curd residue or makkolli residue in flour replacement at 10% level. The protein content was increased from 13.5% to 15.4% in bread containing 10% soybean curd residue and to 16.4% in bread containing makkolli residue. The dietray fiber contents of the bread with soybean curd residue and the bread with makkolli residue were three-fold and two-fold higher than the bread with flour.
