Optimization of blue berry extraction for beverage production using enzyme treatment (효소처리에 의한 블루베리 음료 생산을 위한 최적추출조건)

  • In this study, we tried to establish the best method for fresh blueberry beverage production using enzyme treatment as well as low temperature extraction. During extraction of physiologically functional materials, we used low temperature to prevent nutritional loss by heat. In addition, we investigated optimal blueberry extraction conditions using various enzyme treatments (cellulase, pectinase, cellulase:pectinase (1:1) mixture) to increase extraction efficiency and reduce turbidity. A variety and ratio of enzymes, extraction temperature, extraction time, and shaking speed were considered for the best extraction efficiency rate. We observed high extraction efficiency rates of 85.72-86.55% and 87.06-87.93%, respectively, upon cellulase or pectinase treatment. In addition, a mixture of cellulase:pectinase (1:1) showed an extraction efficiency rate of 86.84-88.14%. The best extraction efficiency rate was observed when crude blueberry was treated at $45^{\circ}C$ (87.91%), for 3 h (87.88%), in a 90 rpm shaker (89.19%). Sugar content and acidity of blueberry extract were not affected by the various treatments. However, total phenolic compounds were detected upon pectinase treatment (18.62 mg/g). Only fructose and glucose as free sugars were found in all samples regardless of treatments and extraction conditions.

A Study on the properties of hot water extracts of Korean dried tangerine peel and development of beverage by using it (국내산 진피열수추출물의 특성과 진피 음료 개발에 관한 연구)

  • This study was carried out to develop a traditional functional beverage by using hot-water extraction of dried peels of Korean tangerine. The pH of the hot-water extracts of dried tangerine peels decreased as the extraction time increased. The acidity and viscosity of the extracts increased as the extraction time increased. The antioxidative activity of the extracts during the extraction was monitored by measuring the electron donating ability. The electron donating abilities of the extracts were in the range of 80.93-83.27%. Extraction time did not affect the antioxidative activity of the extracts. The fibrinolytic activity of the extracts increased as the extraction time increased. The pH of the beverage made with the peel extract was not affected by the extraction time and the kind of sweetener added. The viscosity of the beverage increased as the extraction time increased. In sensory evaluation the highest score was obtained in the beverage samples made with the ones extracted for 180 min and added with sugar. The aboveresults indicate that the dried peel of Korean tangerine can be used as a functional material in beverage industry.

Optimization on preparation conditions of beverage using Opuntia ficus-indica stem (손바닥 선인장을 이용한 음료 제조조건의 최적화)

  • This study was conducted in order to monitor the extraction conditions for a gel-state beverage development of the Opuntia ficus-indica stem. Moreover, the organoleptic properties of the beverage prepared by the extract were optimized using the response surface methodology (RSM). The determination coefficient ($R^2$) value for the extraction yield of the stem was 0.95 (p<0.01). The maximum extraction yield was obtained at an extraction temperature of $93.02^{\circ}C$, 123 min of extraction time and 22.57 mL/g of water to sample. The beverage was prepared with the addition of xanthan gum, sugar and persimmon vinegar to the extract with a central composite design. The maximum organoleptic color of the beverage was obtained at 0.38% xanthan gum, 7.91% sugar and 0.76% persimmon vinegar. The maximum organoleptic flavor was obtained at 0.30% xanthan gum, 7.06% sugar and 1.26% persimmon vinegar. The maximum organoleptic taste was obtained at 0.22% xanthan gum, 10.36% sugar and 0.90% persimmon vinegar. The maximum overall palatability (3.92 score) of the gel-state beverage was obtained at 0.35% xanthan gum, 10.83% sugar and 1.21% persimmon vinegar.

Studies on the Determinations of Dissolved Oxygen in Beverages (음료중 산소분석법에 관한 연구)

  • This study was performed to establish a simple and accurate method for the determination of oxygen that is a processing aid in various beverage. The quantitative determination of dissolved oxygen (DO) contents in 30 cases of samples were performed by traditional titration method and polarography. As a result of the study, the analysis of DO contents in fruit-extract beverages containing oxygen by titration method was time consuming and large sample volumes were needed. Besides, serious interferences with compounds such as hydroxylamine and nitric oxide were observed, leading to false response. Although the polarography is easily affected by $H_2S$, proteins, and various organic compounds, it is a simple and practical method that provides inexpensive and relatively rapid analysis. The polarography is best suited to the routine determination of DO in a large number of samples and it is expected that the polarography can directly be applied to the quality control of the beverages containing added oxygen. The analysis results of DO contents in various fruit-extract beverages with oxygen and without oxygen were as follows: 23.10 ppm to 32.60 ppm for various frutis extract beverages with oxygen, 0.70 pp to 2.54 ppm for mixed beverages without oxygen, 7.63 ppm to 8.28 ppm for drinking water.

Development of Functional Mixed Drink using Extract of Hericium erinacium Cultivated with Artemisia capillaris and Black Garlic (인진쑥을 기질로 한 노루궁뎅이버섯 균사체 배양물과 흑마늘을 이용한 기능성 혼합음료 개발)

  • In this study, we examined the hepatocyte toxicity and protective effects of an extract of Hericium erinacium cultivated with Artemisia capillaris (HEAC), and also examined the hepatocyte protective and antioxidative effects of a mixture of the HEAC and black garlic. At a concentrations of more than 0.05 mg/mL, the HEAC extract significantly reduced cell viability. The extract of HEAC treated with the same ratio of water and ethanol at $80^{\circ}C$ showed the highest hepatocyte protective effect. No significant difference in the hepatocyte protective effect was observed between the mixtures of HEAC with and without black garlic. In addition, higher antioxidative activity was shown with the addition of less black garlic. As a result of the sensory evaluation, a significant difference of sweetness was observed with varying liquid fructose concentration, but there were no significant differences in bitterness, flavor, thickness and overall acceptability according to the liquid fructose concentration.

  • Lactobacillus spp. PAP1 과 Lactobacillus spp. MG-G2를 혼합구조로 사용하여 밤 발효제품을 제조하기 위한 제반 조건을 조사하였다. 밤 발효액은 phytone peptone, yeast exract, glucose를 각각 0.4, 0.2. 0.5%이상으로 첨가할 때 산도와 생균수가 가장 컸다. 첨가물 중 phytone peptone은 대두박이나 순물로 대체해도 같은 효과가 관찰되었다. 버섯추출물의 첨가는 산도와 생육을 약간 촉진하였으나 안정제는 약간의 감소를 가져왔다. 기본 발효액은 관능성적이 나빴으나 산도를 0.54%로 낮추고, 당을 10%로 보강하고, so-dium alginate를 0.2% 첨가하였을 때, 산미, 감미, 음용감이 크게 개선되었다

Physiological Activities of Mycelial Flammulina velutipes Cultured in Liquid Grain Media (곡물 액체배지에서 배양시킨 팽이버섯 균사체의 생리활성)

  • 팽이버섯 균사체를 액체배양하여 전통 장류 발효식품및 음료개발에 이용할 목적으로 7종류의 곡물에 배양한 팽이버섯 배양액의 생리활성을 검토하였다 혈전 용해능은 골목 배양액과 침전물사이에는 차이가 없었으며, 대체로 조, 대두박 및 검정콩 배지에서 혈전용해능이 높게 나타났다. 항균력은 S.aureus에 대해서는 밀, 보리 및 검정콩이 높았고, L. plantarum에 대해서는 조, 밀 및 보리가 높았으며, E.coli에 대해서는 밀과 보리 배지에서 배양한 배양액이 높게 나타났다. Linoleic acid에 대한 항산화 효과는 곡물배지 자체에서는 검정콩, 대두박 및 합성배지에서 높게 나타났고, 팽이버섯을 배양했을 때는 검정콩배지의 항산화력과 유사한 활성을 나타내었다. 비장세포의 증식능은 보리, 밀, 조 및 합성배지에 팽이버섯을 배양하였을 때가 곡물자체에서보다 약 20% 정도 높게 나타났다. ConA를 버섯배양 추출물에 혼합했을 때는 옥수수, 대두박 및 검정콩에서 팽이버섯을 배양한 배양액이 $22\sim26%$ 정도의 증식효과가 있었으며, LPS를 혼합했을 경우는 옥수수, 대두박, 검정콩배지에 배양한 배양액이 각각 45%, 25%,18%의 증식효과가 나타내었다.

A Survey on the Application of Preservatives to Processed Food Types (보존료의 가공식품 유형별 사용 현황 연구)

  • The application of color retention agents (3 items), preservatives (17 items), and bleaching agents (6 items) as food additives in processed foods were investigated by food type. Among color retention agents, sodium nitrite was used the most with 257 cases, mainly in seasoned jeoktal (71.21%), ready-to-eat foods (7.78%), and breads (4.87%). Of the benzoates (1,236 cases) used as a preservative, sodium benzoate showed up most, in 1,215 cases, while 81.16% of these were in beverages such as beverage base (39.51%), mixed beverages (22.47%), and ginseng/red ginseng beverages (8.89%). Grapefruit seed extracts (3,291 cases) were applied to 44 types of processed foods such as sauces (54.65%), liquid tea (10.46%), and other products (5.15%). Ethyl p-hydroxybenzoate (2,957 cases) was applied to products (total 96.44%) such as sauces (92.15%), blended soy sauce (2.77%), and pickled foods (1.52%). Potassium sorbate was applied to a total of 789 cases, mainly pickled foods (40.43%) and processed fishery products (47.15%). All 27 cases of sorbic acid were applied to fish paste (100%). Of the bleaching agents, sodium bisulfite and sodium hydrosulfite were mainly used in confectioneries, breads or rice cakes, and potassium metabisulfite, sodium metabisulfite, and sulfur dioxide were mainly found in alcoholic beverages including fruit wine, while sodium sulfite was mostly used in pickled foods. These results are deemed useful in applying food additives to processed foods.

Acid Production by Lactic Acid Bacteria in Soy Milk Treated by Microbial Pretense or Papain and Preparation of Soy Yogurt (미생물 Protease 또는 Papain으로 처리된 두유에서 젖산균의 산생함과 대두요구르트의 제조)

  • The soy milk prepared from soy protein concentrate was treated with microbial protease or papain. Growth and acid production by Lactobacillus acidophilus in soy milk containing partially hydrolyzed proteins were investigated. Sensory evaluation of yogurt beverage prepared from protease treated soy milk was also performed. Protease treatment of soy milk enhanced acid production by lactic acid bacteria, particularly in case of microbial pretense and simultaneous treatment by two types of protease showed synergistic effect. pH and number of viable cells were not affected markedly by pretense treatment. Microbial pretense treatment up to 15 minutes or papain treatment up to 45 minutes enhanced acid production, but further treatment up to three hours did not affect the acidity markedly. rho sensory evaluation showed that overall acceptability and taste of soy yogurt beverage were slightly improved when soy milk was treated with microbial pretense of 0.2% or papain of 0.2%. The amount of non-protein nitrogen considerably increased by pretense treatment of 15 minutes and it increased gradually by further treatment up to three hours.
