Production of Citric Acid in a Flat-type Membrane Bioreactor (평판형 막 생물 반응기를 이용한 구연산의 생산)

  • A flat-type membrane bioreactor(FMBR ) for aerobic whole cell immobilization was developed and its performance for the citric acid production was investigated using Aspergillus niger (KCTC 1232). The reactor consisted of three layers. The top layer contained flowing air for oxygen supply, the middle layer had stationary cells, and the bottom layer had flowing aqueous nutrients. The initial pH of the culture medium played an important role in citric acid production and the lower initial pH of the culture medium resulted in a higher citric acid yield. Under air and pure oxygen aerations the volumetric productivity reached 0.20 and 0.40g/Lh. Furthermore, the productivity improved with the increase of the culture medium feed rate.

Degradation of BTX by Aerobic Microbial Consortium (호기성 미생물 컨소시엄에 의한 BTX의 분해)

  • In this study, a BTX degrading microbial consortium was obtained from the activated sludges of a BTX releasing sewage water and city sewage water treatment plant. The MY microbial consortium was developed for benzene and toluene degradation, whereas the MA microbial consortium was developed for xylene isomers. The major microorganism of the MA consortium was identified as Rhodococcus ruber DSM 43338T, whereas that of the MY consortium was Rhodococcus sp. In terms of the degradation of a single component, the removal rate of benzene was fastest and decreased in order; toluene, o-xylene, p-xylene and m-xylene. For degradation of mixed BTX, most BTX were degraded within 108 hours and the degradation rate showed either stimulatory or inhibitory effects depending on the composition. MA and MY microbial consortium obtained in this study may be used effectively to remove BTX biologically.

The Aerobic Nature of Arcobacter nitrofigilis (Arcobacter nitrofigilis의 호기적 특성)

  • The free-livein nitrogen fixing bacterium, Arcobacter nitrofigilis which has been known to be a microaerophile, exhibited aerobic growth in brucella broth. In a level of oxygen equivalent to an air atmosphere (21% $O_2$), the maximum cell growth was observed in brucella broth. Low level of cell growth occurred in a level of low oxygen equivalent to lower than 2%, unless any other terminal electron acceptors other than oxygen were wupplied in brucella broth. Membrane-bound cytochrome b and c, and soluble cytochrome c were found. The growth in an aerobic atmosphere, little growth at low oxygen level, and occurrence of cytochrome c mean that this species is an aerobe and obtains energy using energy-yielding respiration.

Effects of Oxygen and Salt on the Growth of Bifidus and Anaerobic Bacteria Isolated from Korean Traditional Fermented Foods (산소와 염농도가 한국전통 발효식품에서 생장하는 혐기성 세균과 Bifidus균의 생육에 미치는 영향)

  • Fourteen samples of kimchies and soy bean pastes were used to isolate strictly anaerobic bacteria on complex BL agar and on a selective BS agar for bifidus bacteria. About $10^7$ ~ $10^8$ colonies per g sample were developed on BL agar under strictly anaerobic conditions, while BS agar supported the growth of $10^3$ ~ $10^6$ colonies per gram sample at the same condition. All colonies developed on BS agar at anaerobic conditions grew in aerobic conditions and did not show fructose6-phosphate phosphoketolase activity. Type cultures of Bifidobacterium did not grow in PYG medium containig more than 3% NaCI. From these results it is conduded that salted fermented food cannot support the growth of strictly anaerobes induding Bifidobactenum.

Effect of Addition of Phosvitin and High Pressure Processing on Microbiological Quality and Lipid and Protein Oxidation of Minced Chicken Leg Meat (닭 다리 분쇄육에 초고압 처리시 Phosvitin의 첨가가 미생물학적 품질과 지방 및 단백질 산화에 미치는 효과)

  • The objective of this study was to investigate the effect of high pressure (HP) processing on shelf life, as well as the addition of phosvitin on lipid and protein oxidation stability of minced chicken leg meat. Minced chicken leg meat was mixed with yolk phosvitin at 500 or 1000 mg/kg meat levels, and divided into raw and cooked groups. Then, the samples were subjected to HP at 0.1, 300, and 600 MPa. The total aerobic bacteria, lipid and protein oxidation, along with instrumental meat color ($L^*$, $a^*$, and $b^*$value) of the samples were measured during storage for 7 d at $4^{\circ}C$. In raw meat, the number of total aerobic bacteria was decreased by HP at 300 MPa (4 Log reductions) and 600 MPa (5 Log reductions) after 7 d of storage (p<0.05). HP at 600 MPa increased lipid oxidation of samples at all storage days and protein oxidation of samples during storage at 3 and 7 d. HP induced the changes of meat color by increase of $L^*$ value and decrease of $a^*$ value (p<0.05). The total aerobic bacteria was not detected in the cooked samples, regardless of HP pressure, and the lipid or protein oxidation of the cooked sample treated by 600 MPa was higher than that of the control (0.1 MPa) on day 7 or control on day 3, respectively (p<0.05). The results suggested that HP can improve the shelf life of minced chicken leg meat. However, phosvitin might be a limited antioxidative agent for the improvement of oxidation stability induced by HP.

  • 종이 펄프 공장의 폐수는 다른 공업에 비해서 폐수량이 많고 오염도가 높다. 목재로부터 종이를 제조하는 경우에 수율은 종이의 종류와 제법에 의해 큰 차이가 있어서 한마디로 말하기는 어렵지만 평균 50~70%로 보는 경우, 목재 성분의 30~50%는 용해 또는 SS의 상태로 폐수중에 존재하기 때문에, 이들이 직접 하천이나 바다에 방류되면 오염의 원인이 된다. 그 때문에 각 공장에서는 목재자원의 종합적인 이용의 입장으로서 제품의 수율을 좋게 하고 동시에 폐수의 양을 감소시키고 폐수에서 유용한 자원을 회수하거나 증해약품의 회수 등을 위하여 연구 및 노력을 계속하고 있는 것이 현 상태이다. 우리 나라의 거의 모든 제지회사에서는 물리적, 화학적 처리법과 생물학적 처리법 중 호기성 처리방법이 주 처리방법으로 사용되고 있으며 탈묵 폐수나 리그닌을 다량 함유하는 폐수의 경우에는 혐기성 처리방법을 이용하기도 한다. 본 실험에서는 우유팩을 재생하여 화장지 제조의 원료로 이용하는 G제지공장의 폐수를 이용하였고, 공장의 원폐수를 부상법으로 1차 처리한 처리수를 실험실로 운반하여 활성슬러지법으로 실험하였다. 처리효율의 영향인자인 HRT를 G제지공장에 맞추어 12시간으로 조정하고 적절한 F/M비(0.23)를 위해 COD와 MLSS가 유지된 후부터 실험을 시작했다. $20^{\circ}C{\sim}560^{\circ}C$까지 $10^{\circ}C$간격으로 온도를 변화시키면서 미생물의 상태변화를 관찰하고, 또한 각 용도에서의 처리수질을 비교해 보았다. 실험조건은 pH는 중성상태(6.5~7.5), DO는 $2{\sim}4mg/{\ell}$, 영양원은 BOD : N : P = 100 : 5 : 1로 맞추고 BOD, COD, SS, SVI를 측정하였으며, 현미경을 통하여 미생물과 Floc를 관찰하였다. $20^{\circ}C$부터 종균제를 초기 운전시 $200mg/{\ell}$ (분말상)을 폭기조에 투여하고 매일 $200mg/{\ell}$를 정기적으로 투입하여 온도변화에 따른 미생물의 상태 및 처리수질을 비교해보았다. 특히 종균제로는 기존의 G제G지공장에서 사용하는 신일 미생물(Bio sound S)종균제와 청림C&C의 종균제(Cell bio P)를 비교 관찰하였다. 고온성 종균제를 두배 투입하여 투입량에 대한 처리효율을 비교하였으며 종균제를 사용하지 않고 운용함으로써 그 차이도 비교할 수 있었다.

Effect on Livestock Manure Composting by the Enriched Microbial Population (미생물에 의한 축산 폐기물 퇴비화에 미치는 영향)

  • Several kinds of thermophilic, aerobic microorganisms (Bacillus genus), metal leaching microorganisms (Thiobacillus, T. ferooxidans), and other nondegradable chemical-degrading microorganisms (Pseudomonas genus) were utilized to study the effect on composting livestock manure. Under the Carbon-Nitrogen ratio (C/N) of 35∼40 and water content of 50∼65% conditions, the composting in the cycling drum reactor showed slower composting and lower temperature increase than that of the manual reactor. Element analysis after composting indicated relatively high levels of mineral contents with the substitutional effect of chemical fertilizer. Metal analysis before and after composting showed lower As in all, Cr in pig, Pb in cow, Hg in chicken, and Cu in cow manure compost than the regulation values. Compost maturity was ascertained by the several maturity tests. Salmonella and E. cozi detection test by SS or EMB agar plate confirmed the safety from the pathogenic microorganisms. The results suggest that the inoculation of metal and some other chemical degrading microorganisms during composting might decrease metal contamination and increase composting rate.

Effect of Electron Beam Irradiation on the Microbial Safety and Qualities of Sliced Dried Squid (전자선 조사가 건조 오징어의 미생물학적 안정성 및 품질에 미치는 영향)

  • Electron beam irradiation was applied to examine the microbial safety and qualities of sliced dried squid. Sliced dried squid was irradiated at dose of 2, 4, 8, 12, and 16 kGy. Microorganisms contaminated in sliced dried squid were significantly decreased by irradiation. Decimal reduction dose (D/sub 10/ value) of total bacteria count, yeast and mold, coliforms in sliced dried squid were 8.57, 4.60, and 8.10 kGy, respectively. Electron beam irradiation caused negligible changes in Hunter color L, a, and b value. Sensory evaluations of irradiated sliced dried squid showed that there were no significant changes among the samples. These results indicate that electron beam irradiation improves the microbial safety and qualities of sliced dried squid.

  • Electron beam irradiation was applied to examine microbial safety and qualities of black pepper powder and commercial Sunsik. Whole black pepper powder and commercial Sunsik were irradiated at 2, 4, 8, 12, and 16 kGy. Microorganism contamination in black pepper powder and commercial Sunsik were significantly decreased by irradiation. Decimal reduction doses ($D_{10}\;value$) of total bacteria count in black pepper powder and commercial Sunsik were 5.32 and 1.56 kGy, respectively. $D_{10}\;value$ of yeast and mold were 2.54 and 2.14 kGy for black pepper powder and commercial Sunsik, respectively. Electron beam irradiation caused negligible changes in Hunter color L, a, and b values. Sensory evaluations of black pepper powder and commercial Sunsik showed no significant changes among samples. These results indicate electron beam irradiation improves microbial safety and qualities of black pepper powder and commercial Sunsik.

  • To evaluate the correlations of microbial populations with soil healthiness and crop production and establish the criteria for microbial population of soil types. We analyzed the microbial community structure of 13 soils which were different in physical and chemical properties and cultivation methods. According to the analysis of microbial population suing the dilution plate method, the large differences of the microbial population structures among soil types were shown: aerobic bacteria $2-27{\times}10^6$, fluorescent Pseudomonas $1-1,364{\times}10^5$, Gram negative bacteria $1-126{\times}10^4$, and mesophilic Bacillus $1-110{\times}10^5$. The density of Gram negative bacteria was highest on red pepper cultivating soils (sample no. 4 and 6) of Umsung and Gesan, Chungbuk, and the density of the fluorescent Pseudomonas was highest on greenhouse soil (sample no. 7) of Jinju, Kyungnam. The crop productivity of three soils was high as compared with those of other soils. It was supposed that the density of fluorescent Pseudomonas and mesophilic Bacillus were correlated with the incresed crop production. By MIDI analysis, 579 strains isolated from 13 soils composed of a variety of microbes including 102 isolates of Agrobacterium, 112 isolates of Bacillus, 32 isolates of Pseudomonas, 44 isolates of Kocuria, and 34 isolates of Pseudomonas. Among the 624 isolates of Gram negative bacteria, Pseudomonas including P. putida and p. fluorescens occupied the highest density (51%), and Stenotrophomonas maltophilia and Burkholderia cepacia also appeared at high density. From RAPD analysis, the fluorescent Pseudomonas strains isolated from 13 soil types showed a high level of strain diversities and were grouped into 2 - 14 patterns according to soil types. Many of unknown bacteria were recovered from the paddy soil, and needed to be further characterized on the molecular basis.
