Objective: Few data were available on glycemic control at the early prediabetic stage, though it is the important stage to prevent diabetes mellitus. The purpose of this research was to examine the glycemic control effects of herb medicine treatment (DA-9802) in prediabetic stage patients Methods: In this randomized, single blind, placebo-controlled study, we compared Sanyak extract (DA-9802) with placebo for the treatment of glycemic control effects in prediabetic stage patients. 45 volunteers who satisfied the requirements were enrolled in the study. They took Sanyak extract (DA-9802) or placebo treatment three times a day for 8 weeks. Blood sugar, Total Symptom Score (TSS) and blood serum were measured at weeks 0, 4, and 8. Results: In the DA-9802 group, fasting blood sugar changes between before 8 weeks significantly decreased. Fasting blood sugar, fasting blood sugar gaps between before and after treatment for 8 weeks, insulin changes and C-peptide significantly decreased at 8 weeks compared with those of the placebo group. Conclusion: Herb medicine treatment (DA-9802) is effective in glycemic control on prediabetic stage.

The present study was conducted to investigate the effect of dietary supplementation of sea tangle extracts on the excretion of neutral steroids and bile acid in diabetic rats. Sprague-Dawley rats were fed on AIN-76 based experimental diets containing methanol extracts (2%, w/w) or water extracts (4%, w/w) of sea tangle for 4 weeks. And then they were induced to be diabetic by receiving streptozotocin (45 mg/kg BW) intramuscularly. The dried fecal weight was increased significantly in water extracts group compared with control group. The fecal content of cholesterol was higher in extracts groups of sea tangle than in control group. But there was no significant difference in fecal excretion of cholesterol between methanol and water extracts groups. The fecal excretion of coprostanol was increased significantly in water extracts group compared with the control group. The fecal excretion of bile acid was increased significantly in sea tangle extracts groups compared to the control group. These data suggest that dietary supplementation of sea tangle extracts might reduce the incidence of atherosclerosis through increasing the excretion of neutral steroids.

The present study was carried out to investigate the effects of zinc and vitamin E on the antioxidative defense mechanism in the liver of streptozotocin(STZ)-induced diabetic rats. Levels of blood glucose of STZ-diabetic rats were higher than that of control, but ZDM($ZnSO_{4}$ 10mg/kg injection+STZ) group was lower than those of DM(STZ injection) and EDM(vitamin E 400mg/kg diet+STZ) group. Levels of plasma insulin were lower in all three STZ-diabetic groups than those of control. Thiobarbituric acid reactive substances(TBARS) peroxide values(LPO) in liver were increased 2.3-fold in DM group compared with those of control, while LPG in ZDM group was lower than that of DM group, and EDM group had similar tendency compared with that of control. Reduced glutathione(GSH) contents of liver were decreased in DM group compared with those of control, but increased 2.3, 1.7-fold in ZDM and EDM groups, respectively, compared with those of DM group. Oxidized glutathione(GSSG) was increased in DM group compared with control and GSSG in ZDM and EDM group were lower than that of DM group. GSH/GSSG ratio had similar tendency compared with results of GSH. The activities of free radical scavenging enzymes such as superoxide dismutase, glutathione peroxidase and glutathione S-transferase were significantly decreased in DM group compared to those of control, but higher in ZDM and EDM groups than those of DM group. The metallothionein contents in liver and kidney were increased in DM and EDM groups were remarkably increased 20, 5.3-fold in ZDM group, compared with those of control.

This study was designed to evaluate the anti-metabolic syndrome effects of capsule-filled cheonggukjang (CGJ) added with arrowroot (Pueraria thunbergiana) extracts on body weight, adiposity and lipid metabolism in ob/ob mice. Experimental groups were normal control group (NC: basal diet), positive control group (PC: 2% CGJ), CGJ added with arrowroot extracts group (AR: 2% arrowroot in CGJ), and capsule-filled CGJ added with arrowroot extracts group (ARC: 2% arrowroot CGJ capsule). Each group was fed experimental diet for 10 weeks. Final body weight gain and atherogenic index were significantly lower in the ARC than NC group. Serum levels of total cholesterol, LDL-cholesterol, and triglycerides, blood glucose and atherogenic index were significantly lower in the ARC than NC group. Furthermore, fatty liver and regional lipid accumultion in ob/ob mice were inhibited in the ARC group. The hepatic activities of superoxide dismutase, catalase and glutathione S-transferase were significantly higher in the ARC than NC group. Therefore, the anti-matabolic syndrome effects of the ARC group were higher than the AR group. In conclusion, these results indicated that CGJ added with arrowroot mediates its anti-obesity effects in ob/ob mice by improving lipid metabolism and antioxidant enzyme.
The aim of this study is to investigate the effect of Lespedeza maximowiczii var. tricolor Nakai (LMTN) on glucose metabolism. LMTN extract significantly enhanced the glucose uptake and lipid accumulation in 3T3-L1 adipocytes compared with control. Also, LMTN extract in 3T3-L1 adipocytes significantly increased the protein expression of peroxisome proliferator-activated receptor (PPAR)γ, insulin receptor substrate-1, and glucose transporter (GLUT)4. The regulatory effect on glucose uptake or insulin signal transduction of LMTM extract was lower than troglitazone or pinitol such as the positive control, but increased PPARγ activation. Additionally, LMTM extract has an insulin-mimetic effect. In db/db mice, LMTN extract (250 mg/kg BW) significantly reduced water and food intake, blood glucose, and level of plasma triglyceride and total cholesterol. Furthermore, the expression of PPARã and GLUT4 mRNA in adipose or muscle tissue effectively was increased by oral treatment of LMTN extract. Thus, our results suggest that LMTN extract improves the glucose metabolism through PPARγ and insulin-mimetic effect in 3T3-L1 adipocytes and db/db mice.

Rapid and accurate identification of marine microalgae causing harmful algal blooms (HABs) is a crucial tool for predicting and managing HABs. We previously developed a nuclease protection assay sandwich hybridization (NPA-SH) method for the in situ detection of blooming microalgae Cochlodinium polykrikoides. In this study, we improved the applicability of the NPA-SH method for the detection of C. polykrikoides by simplifying the reaction step. For this purpose, invertase (INV) was conjugated to the signal probe instead of using fluorescence, and sucrose was used as a reactant to induce a color reaction. The INV-signal probe conjugation was confirmed by SDS-PAGE and epifluoromicroscopy. The treatment time and appropriate amounts of the probe and sucrose that optimized the reaction were determined. As a result, the developed INV-SH reduced the treatment time in the field compared with NPA-SH, and also enabled the use of a relatively small volume and low-priced personal glucose meter, as well as an absorbance meter. INV-SH is the first C. polykrikoides species identification technology to which INV has been applied and could be an improved field technique.

This study investigated the effects of scopoletin (6-methoxy-7-hydroxycoumarin) supplementation on insulin resistance and the antioxidant defense system in chronic alcohol-fed rats. Rats were fed a Lieber-Decarli liquid diet containing 5% ethanol with or without two doses of scopoletin (0.01 and 0.05 g/L) for 8 weeks. Pair-fed rats received an isocaloric carbohydrate liquid diet. Chronic alcohol did not affect fasting serum glucose levels, although it induced glucose intolerance and hyperinsulinemia compared with the pair-fed group and led to insulin resistance. Both doses of scopoletin similarly improved glucose intolerance, serum insulin level, and insulin resistance. Scopoletin supplementation significantly activated phosphatidyl inositol 3-kinase, which was inhibited by chronic alcohol. Two doses of scopoletin up-regulated hepatic mRNA expression and activity of glucokinase as well as down-regulated mRNA expression and activity of glucose-6-phosphatase compared with the alcohol control group. Both doses of scopoletin significantly reduced cytochrome P450 2E1 activity and elevated aldehyde dehydrogenase 2 activity, resulting in a lower serum acetaldehyde level compared with the alcohol control group. Chronic alcohol suppressed hepatic mRNA expression and activities of antioxidant enzymes such as superoxide dismutase, catalase, and glutathione peroxidase; however, they were reversed by scopoletin supplementation, which reduced hydrogen peroxide and lipid peroxide levels in the liver. These results indicate that dietary scopoletin attenuated chronic alcohol-induced insulin resistance and activated the antioxidant defense system through regulation of hepatic gene expression in glucose and antioxidant metabolism.
Gallstone is the most common disease of the biliary system. Korean has experienced an increase in the percentage of cholesterol gallstones. The major risk factors associated with cholesterol gallstones are age, gender as well as obesity. This study was designed to determine the prevalence of gallstones in the last three years and evaluate the associated risk factors in the population who underwent health screening. The study population consisted of 2,484 males and 2,212 females who visited the health promotion center in Dalseogu, Daegu in Korea from January 2011 to December 2013. Each participant in the study had their biliary system gallbladder examined using ultrasonography. Classified as underweight, normal weight or overweight using the population of obese according to the body mass index, and classified according to mood diagnosis of diabetes presented by the American Diabetes Association. Fasting blood glucose and number of liver function, the divided the control group by referring to the normal liver function values used herein. The geological map, I was classified as NCEP APT III. A showed of total 148 people were found to have gallstones. The prevalence of sex among 148 patients (3.15%) 84 men (1.79%) and 64 women 1.36%) which shows significantly there is little difference. 1.84% 40 years and below, 3.38% 40's showed age prevalence was 4.66% in 50's and above. In addition, Total-cholesterol was at the most in 52 people, LDL-cholesterol in 398 people, Triglyceride in 36 people, HDL-cholesterol in 19 people. The abnormal group, was created from the total-cholesterol categories from a physical examination of a subject that has been found to be gallstones in the gallbladder. A result of conducting the univariate analysis shows the prevalence of gallstones, a correlation that is meaningful. The logistic regression analysis of multiple ages was chosen to show risk factors age independent cholelithiasis. In spite of the conclusion, gallstones are not displayed in relation to the metabolic syndrome but in order to clarify this, not only the subject of a health examination is needed but, a further study of the general public when possible.

This study investigated the relationship between Vitamin D levels and indices related to cardiovascular disease in Korean adults aged ${\geq}19years$. The data for analysis were obtained from the sixth Korea National Health Nutrition Examination Survey in 2013 and 2014. The results showed that the incidence of Vitamin D deficiency was 73.1% among Korean adults and that indices related to cardiovascular disease showed an increasing trend (55.6% of Korean adults). The relationship between Vitamin D levels and indices related to cardiovascular disease with controlled physical activity status was also studied. The odds ratios (ORs) for age were 1.72 for the 4-62 years age group (95% CI, 1.53-1.93) and 2.05 for the ${\geq}65years$ age group (95% CI, 1.71-2.45). For blood pressure, the OR for pre-hypertension was 1.30 (95% CI, 1.15-1.47) and that for hypertension was 1.31 (95% CI, 1.11-1.54). For body mass index (BMI), the OR was 1.36 (95% CI, 1.11-1.66) and that for waist circumference (WC) was 1.36 (95% CI, 1.11-1.66). For fasting blood sugar (FBS), the OR for impaired fasting glucose (IFG) was 1.37 (95% CI, 1.21-1.55) and that for diabetes mellitus (DM) was 1.31 (95% CI, 1.05-1.65). The OR for total cholesterol (TC) was 1.30 (95% CI, 1.11-1.52) and that for triglycerides (TG) was 1.20 (95% CI, 1.04-1.37) in Korean adults. There was a significant relationship between Vitamin D and indices related to cardiovascular disease in Korean adults with respect to age, blood pressure, FBS, BMI, TC and TG. Confirmation of a causal relationship between Vitamin D and indices related to cardiovascular disease may require further research consisting of more systematic cohort studies.

Background: Chronic obstructive pulmonary disease (COPD) is characterized by airflow limitation that is not fully reversible. COPD has systemic effects, such as skeletal muscle dysfunction and abnormal weight loss. It also has been suggested that COPD is related to other chronic disease, such as cardiovascular disease, osteoporosis, and anemia. The aim of this study was to evaluate a symptom questionnaire and laboratory findings in subjects with air flow limitation. Methods: We evaluated a symptom questionnaire and laboratory findings in subjects with airflow limitation detected by spirometry in conjunction with the Second Korean National Health and Nutrition Examination Survey. A total of 9,243 adults over the age of 18 were recruited. Among the adults, we finally analyzed 2,217 subjects who met the acceptability and repeatability criteria of spirometry, showed normal findings on chest radiography, and were older than 40 years of age. Results: There were 288 subjects with airflow limitation as determined by spirometry. The frequency of respiratory symptoms such as cough, sputum and wheezing were significantly higher in subjects with airflow limitation (p <0.01). Hemoglobin and hematocrit levels were higher in subjects with airflow limitation (hemoglobin level 13.98 mg/dL vs. 13.62 mg/dL, hematocrit 42.10% vs. 40.89%; p<0.01). The HDL cholesterol level was lower in subjects with airflow limitation (44.95 mg/dL vs. 45.60 mg/dL; p<0.01). There was no significant difference in the total cholesterol, triglyceride, blood urea nitrogen, creatinine, and fasting glucose levels. Conclusion: In subjects with airflow limitation, prevalence of respiratory symptoms was higher than in normal spirometry subjects and the levels of hemoglobin and the hematocrit were higher. The HDL cholesterol level was lower in subjects with airflow limitation.
