Screening of Domestic Plants with Antibacterial Activity (국내 자생식물의 항균활성)

  • To select new useful plants with antibacterial activity, ninety five sample of eighty different species of wild plants were collected, and extracted with methanol. Antibacterial activity of the methanol extracts was tested against Bacillus subtilis, Staphylococcus aureus, Escherichia coli and Vibrio parahaemolyticus. The methanol extracts from Artemisia capillaris, Hemistepta lyrata, Youngia japonica, Prunella vulgaris, Lamium amplexicaule and Juniperus chinensis was effective against all bacterial strains tested, and eight methanol extracts including Ixeris dentata, Gnaphalium affine, Chelidonium majus and Spiraea prunifolia exhibited the antibacterial activity against at least 3 bacterial strains. Methanol extracts from leaf of Syringa vulgaris, Drava nemorosa and clove of Erythronium japonicum showed a selective antibacterial activity against two gram negative bacteria, V. parahaemolyticus, and B. subtilis, respectively. With investigations on antibacterial activity against a certain bacterial strains tested, metahnol extracts from clove of Erythronium japonicum, Spiraea prunifolia, leaf and twig of Camelia japonica, and Drava nemorosa showed strongest activities against B. subtilis, S. aureus, E. coli, and V. parahaemolyticus, respectively. Nine methanol extracts based on the results were successively fractionated with n-hexane, chloroform, ethyl acetate and water portions, which were examined antibacterial activity against B. subtilis and V. parahaemolyticus. Among the all fractions tested, chloroform fractions of Hemistepta lyrata showed strongest antibacterial activity against both B. subtilis (17mm) and V. parahaemolyticus (29 mm). Chloroform fractions of Youngia japonica, n-hexane fractions of Artemisia capillaris, Iexeris dentata and Prunella vulgaris, and ethyl acetate fraction of leaf and twig of Camelia japonica showed relatively a strong antibacterial activity. On the other hand, Juniperus chinensis and Equisetum arvense was distributed to all fractions except for water fraction.

  • Specimens of spider mites parasiting on conifer trees were collected from Korea during the period from May, 1986 to June, 1988. As a result, seven species belonging to the tribe Tetranychini, subfamily Tetranychinae, family Tetranychidae were identified as follows: 1. Panonychus citri(McGregor, 1916), *2. Oligonychus clavatus(Ehara,1959), *. O.hondoensis(Ehara,1954), *4. O.pustulosus Ehara, 1962, 5. O.karamatus(Ehara, 1956), 6. O.perditus Pritchard and Baker, 1955, 7. O. ununguis(Jacobi, 1905). Of which, three species with asterisk are newly recorded from Korea.

  • Antifungal activities of domestic plants were examined to develop natural anti-microbial agents, able to protect onion from spoiling microorganisms during their storage. A fungus was isolated from spoiling onion and identified to Penicillium sp. ON-211, based on the morphological characteristics of conidiophore and conidia. Chloroform extracts of Juniperus chinesis Linnaeus, Juniperus. chi. var. horizontalis, water extract of Thujs orientalis Linnaeus' seed and the n-hexane extract of Juniperus. chi. var. horizontalis N. showed remarkable antifungal activties against this fungus.

In vitro Anti-malarial Activity of Juniperus Chinensis Extract (향나무 추출물의 항말라리아 효과)

  • This study was carried out to investigate the anti-malarial activity of Juniperus chinensis by in vitro and in vivo system using Plasmodium falciparum chloroquine-sensitive(3D7) and P. falciparum chloroquine-resistant(S20) strains. According to cytotoxicty test on NIH 3T3 cell, the ethanol extract(EtOH), ethylacetate(EtOAc) fraction and aqueous fraction possessed significant anti-malarial activity against both 3D7 and S20 strains at non-toxic concentrations(<100 /). In vitro assay, EtOAc fraction showed notable activity against 3D7 and S20 strains of P. falciparum with $IC_{50}$ values of $37{\pm}2{\mu}g/ml$ and $36{\pm}6{\mu}g/ml$. In animal test using P. falciparum infected human erythrocytes, the treatment of EtOAc fraction significantly inhibited parasitaemia in mice in a dose-dependent manner that is parasitaemia of 42%, 34% and 31% in doses of 10 mg/kg, 20 mg/kg and 40 mg/kg, respectively. The study provides data to support the medicinal importance of the J. chinensis.

Dyeing Properties and Functionality of Methanol Extract from Juniperus chinensis Heartwood (향나무 심재 메탄올추출물의 염색성과 기능성)

  • In this study, we report dyeing properties and functionality of cotton and silk fabrics dyed with methanol extract from Juniperus chinensis heartwood. Non-aqueous dye of Juniperus chinensis heartwood extracted with methanol was expressed reddish color. It showed relatively good affinity to silk than cotton. Dye uptake of cotton and silk continued to increase according to the increase of the dye concentration. Cotton was not affected by the dyeing temperature and time, however, dye uptake of silk was increased as the dyeing time and temperature were increased. Dye uptake of cotton and silk were the highest at pH 8 and pH 6 respectively. Dyed fabrics showed various colors depending on mordant types and mordant concentration. Washing, light and perspiration fastness were not good. However, rubbing and dry cleaning fastness showed relatively good grade. Dyed fabrics of ultraviolet-cut ability and deodorant ability were improved. Also, dyed fabrics showed very good antimicrobial abilities of 99.9%.

  • The purpose of this study is to investigate dyeing properties and functionality of cotton and silk fabrics dyed with hot-water extract from Juniperus chinensis heartwood. Water-soluble dye of Juniperus chinensis heartwood extracted with distilled water was expressed YR color series. Dye uptake of cotton continued to increase according to the increase of the dye concentration. Its variation trends were shown to be similar to the isothermal absorption curve of the Freundlich. Dye uptake of silk was better than cotton, increased depending on increasing concentration of the dye, dyeing time and temperature. Its variation trends were shown to be similar to the isothermal absorption curve of the Langmuir. As the pH increased, the dye uptake of fabrics reduced and showed increased on red tinge. Colors of the dyed fabrics were various, depending on dye concentration and the mordant type and mordant concentration. Washing fastness, light fastness and perspiration fastness were not good. However, rubbing and dry cleaning fastness showed relatively good grade. Dyed fabrics of ultraviolet-cut ability and deodorant ability were improved. And dyed fabrics showed antimicrobial abilities of 99.9% against Staphylococcus aureus.

Influences of Sulfur Dioxide on the Growth of Ornamental Trees (아황산가스가 조경수목의 생육에 미치는 영향)

  • This study was to investigate ornamental trees injuries at the Tobong ornamental tree farm, which is located in Uichongpu City, Kyonggido. The injury was caused by $SO_2$ gas from the smoke of the vicinal factories. The results obtained are as follows: 1 The total concentration of sulfur in leaves was increased with incerased degree of injury The most severe injuries were found at the distance between 70-120m from the western boundary of the Tobong ornamental 4ree farm with a highest concentration of total sulfur at $0.5-0.6\%$. This means the injuries caused by the acute injury and the temperature changes. 2. Significant differences were found between normal and injured portion of the same trees in Juniperus chinensis. 3. Anatomical study of the leaf tissues, showed the plasmolysis occured both in spongy and epidermal cells, leading to shrinkage and destruction. An intercellular space and stomatal periphery became wider.

Termiticidal Activities of Chamaecyparis obtusa Endl. Heartwood (편백재의 흰개미 살충활성에 관한 연구)

  • Termiticidal activities of Chamaecyparis obtusa were quantitatively evaluated, and the activity differences between supporting materials such as woodmeal and filter paper or between species were defined based on the termiticidal activity value(TAV). It was found that TAV was high in the following order ; C. obtusa>Litsea coreana>C. obtusa var. formosensis>Ternstroemia gymnanthera. In particular, termiticidal activity of C. obtusa heartwood was stronger by 5 to 9 times than that of other three species. Median survival dosage(MSD) of C. obtusa was 108.8 mg. In case of woodmeal or filter paper tests with C. obtusa, termiticidal activities were inversely proportional to logarithms values of an added sample weight and median survival time(MST). The difference of termiticidal activities between woodmeal and filter paper in the methanol extracts was small, but that in the neutral fraction was enormous as 3.21 times. However, termiticidal activity of neutral fraction was corresponded to 17 to 53% of original woodmeal, and 47 to 83% of termiticidal activity was considered as a loss in test process.

Studies on the components in pycnial drops of Gymnosporangium haraeanum Sydow (배나무 적성병균의 병자적 성분에 관하여)

  • By way of paper chromatography, free sugars in pycnial drops of Gymnosporangium haraeanum Sydow were investigated in regard of their biochemical interrelation with free sugars of Chinese juniper and pear leaf. The free sugar in pycnial drops of Gynnosporangium haraeanum Sydow were identified to only Fructose spot. Free sugars in Chinese juniper leaf were identified to Glucose. Galactose and two unknown spots. Free sugars of another sample in pear leaf were identified to spots of Glucose, Furctose and Galactose. The Arbutin from pear leaf was crystalized and its structure was identified to Glucose and Hydroquinone. The acetone powder of Emulsin was incubated for 1 hour at $40^{\circ}C$ with 0.05 M Arbutin substrate in test tube and purified by general method with the purpose of analysis of its. metabolic products. And the paper chromatographic analysis showed it to be Glucose spot. From the above results, this Fructose in pycinal drops of Gymnosporangium haraeanum Sydow is presumed to be the exchangeable from free sugars in pear leaf or to be the hydrolyzed of $\beta-glycoside$ (Arbutin)-the metabolic isomerization of Glucose into Fructose by pycnia isomerase.

Hypolipidemic Effects of Korean Softwood Components (국내 침엽수재 추출성분의 혈액지질 저하 효과 연구)

  • As a basic study to find bioactive principles for the hypolipidemic effect from natural sources, various biological influences of MeOH extracts of Thuja accidentalis L. and Juniperus chinensis var. horizontalis Nakai on rats were examined. The concentrates obtained from the organic layer and aqueous layer fractions of each species were orally administered to rats and then their effect on the body weight, organ weight, serum lipids profile, and blood glucose concentration were examined. Among the samples tested, the organic layer of Juniperus chinensis var. horizontalis exhibited the possibility of the hypolipidemic effects. The corresponding layer was chromatographed to give three fractions and reexamined their effects on rats. The results demonstrated that the fraction corresponding for the R$_{f}$ 0.4~0.7 which was the mixture of many minor components showed the effect of lowering the blood-lipid bevel effectively without any side effects. Besides, communic acid, identified from and isolated as a major component of the species exhibited the possibe hypolipidemic effects.s.
