Antimicrobial and Antitumor Activities of Seed Extracts of Camellia sinensis L. (차나무 종자 추출물의 항균활성 및 항종양활성)

  • Antimicrobial and antitumor activities of Camellia sinensis L seed extracts were investigated. Seed extracts showed antifungal activities against Candida albicans IFO 1594 and Cryptococcus neoformans. Inhibition zone of 20 mm was shown by 70% ethanol extract against C. albicans IFO 1594 at 100 mg/mL. Antifungal activity of seed extract was not decreased by heating at 80 and $100^{\circ}C$ for 30 min or at $121^{\circ}C$ for 15 min, indicating heat-stability of seed component. Growth-inhibitory effects were observed in 70 and 10% of tumor cell line SK-OV-3 and normal ceil line NIH/3T3 at $50{\mu}g/mL$, respectively.

The effect of nonionic surfactants on the antimicrobial activity of preservatives in cosmetic products (비이온 계면 활성제가 화장품의 방부력에 미치는 영향)

  • In order to investigate the effect of nonionic surfactants on the antimicrobial activity of preservatives in the presence and absence of p.0.E(20) Sorbitan fatty acid ester commonly used in cosmetics and pharmaceutical systems, these experiments were carried out by determining Minimum Inhibitory Concentration(MIC) values and MIC values of adaptation against test organisms. And also the inactivation of the preservative against each microorganism in formula added with various concentrations of P.0.E(20) Sorbitan monostearate were measured by use of a preservative death time curve The results obtained were as fort low : 1) Nonionic surfactant inactivated Methylparaben to varying extents, but not Imidazolidinyl urea. 2) A combined preservative system was inactivated to a little extent (range of 0.16-0.20% Conc.), no adaptation was observed for the 5. aureus ATCC 6538. Imidazolidinyl urea complex combined with Methylparaben had a broad antibacterial spectrum against the Gram(.) and the Gram(-) bacteria It was found that preservatives had a synergistic effect by use of mixed form of preservatives, 3) In formula preserved with 0.2% Methylparaben containing 0.5, 1.0 and 2.0% P.0.E(20) Sorbitan monostearate, E. coli ATCC 10s36 and P. aeruginosa NCTC 10490 died quickly within in 2hr 4) However, from Fig.5, S. aereus ATCC 6538 died more slowly within increasing surfactant concentration and the D-values(Decimal reduction time) were 5.2, 8 and 14 hr. for samples containing 0.5, 1 0 and 2.0% P 0. E(20) Sorbitan monostearate, respectively. 5) In the case of Methylparaben, no adaptation for the E. coli ATCC 10536 6) All of the nonionic surfactant, p.0. E(20) Sorbitan fatty acid ester used in the experiments decreased the effectiveness of Methylparaben, but not of Imidazolidinyl urea.

  • A drug transport carrier could be used for safe send of drugs to the affected region in a human body. The chitosan is adequate for the drug delivery carrier because of adaptable to living body. The gold, a metallic nanoparticles, tends to form a nano complex at rapidly when it combined with chitosan because of its negative charge. having energy from the other, outer gold nano-complex make heat due to its property to release the contained drugs to the target area. Silver could be also formed an useful biocompatible nano-composites with chitosan which should be used as an useful drug transfer carrier because its special ability to protect microbial contamination. Being one of the oxidized nano metals, $Fe_3O_4$ is nontoxic and has been used for its magnetic characteristics. In this study, the control of catalyst, reducing agent, and solvent amount. The chitosan-$Fe_3O_4$-gold & silver nanoshell have been changed to form about 100 nm size by ionic bond between the amine group, an end group of chitosan, and the metal. It was observed the change in order to seek for its optimum reaction condition as a drug transfer carrier.

Evaluation of Antioxidantive and Antimicrobial Activities of Garlic Stem and Red Cabbage, and Their Application to Pork Patties during Refrigerated Storage (붉은 양배추와 마늘종 추출물의 항산화 및 항균활성 평가와 냉장저장 중 돈육패티에 이용)

  • This study was carried out to investigate antioxidative and antimicrobial activities of ethanol extracted garlic stem and red cabbage, and their application to pork patties during refrigerated storage. Total phenolic compounds of ethanol extracted garlic stem and red cabbage were 3.13 g/100 g and 3.99 g/100 g, respectively. 1,1'-diphenyl-2-picryhdrazyl (DPPH) radical scavenging activity of garlic stem extracts increased up to 0.05%. Iron chelating activities of both garlic stem and red cabbage increased up to 0.5%, and red cabbage up to the concentration of 1% had similar iron chelating activity to those of EDTA. Pork patties containing garlic stem had higher Hunter L and b values, (p<0.05), whereas those with red cabbage had highest Hunter a values, resulting in similar value to those of control (p>0.05). Thiobarbituric acid reactive substances (TBARS) of pork patties with garlic stem were lower than those of the control (p<0.05). In addition, antioxidantive activities of pork patties with red cabbage were higher than those with garlic stem. No differences in microbial contents were observed between the control and treatments. In conclusion, garlic stem and red cabbage extracts might be used as natural antioxidants to be used in the manufacture of functional meat products.

  • Water supplies are decreasing in comparison to increasing clean water demands. Using nanofiltration is one of the most effective and economical methods to meet the need for clean water. Common methods for desalination are reverse osmosis and nanofiltration. However, pristine membranes lack the essential features which are, stability, economic efficiency, antibacterial and antifouling performances. To enhance the properties of the pristine membranes, graphene oxide (GO) is a promising and widely researched material for thin film composites (TFC) membrane due to their characteristics that help improve the hydrophilicity and anti-fouling properties. Modification of the membrane can be done on different layers. The thin film composite membranes are composed of three different layers, the top filtering active thin polyamide (PA) layer, supporting porous layer, and supporting fabric. Forward osmosis (FO) process is yet another energy efficient desalination process, but its efficiency is affected due to biofouling. Incorporation of GO enhance antibacterial properties leading to reduction of biofilm formation on the membrane surface. Pressure retarded osmosis (PRO) is an excellent process to generate clean energy from sea water and the biofouling of membrane is reduced by introduction of GO into the active layer of the TFC membrane. Different modifications on the membranes are being researched, each modification with its own advantages and disadvantages. In this review, modifications of nanofiltration membranes and their composites, characterization, and performances are discussed.

The effects of Chamaecyparis obtusa oil on anti-wrinkle, skin-barrier and moisturizing (편백 오일의 항주름, 피부 장벽 및 보습능 평가)

  • Chamaecyparis obtusa (C. obtusa) belongs to Cupressaceae and grows wild in southern area of Korea. C. obtusa has been used for furnitures because of the superior quality of the lumber and remaining branches and leaves were used for extraction of essential oil (C. obtusa oil). C. obtusa oil has numerous health effects, including anti-inflammation, antibacterial, deodorization, sedative effect. This study was performed to evaluate the effects of C. obtusa oil on anti-wrinkles, skin-barrier and moisturizing. First, DPPH and ABTS assay were conducted to evaluate antioxidant effect and C. obtusa oil showed significance scavenging activity on both radicals. And elastase activity was examined for anti-wrinkle effect. At the result, elastase activity was significantly reduced by C. obtusa oil. Next, real-time PCR was performed to evaluate gene expression. At the results, MMP-1 mRNA expression was significantly reduced by C. obtusa oil in HDF cells. In addition, filaggrin and HAS-2 mRNA expression significantly increased by C. obtusa oil in HaCaT cells. Taken together, these results indicate that C. obtusa oil has potential as a cosmetic material for anti-aging by enhancing anti-wrinkle, skin-barrier and moisturizing effects.

Quality Characteristics and Retarding Retrogradation of Sponge Cakes containing Red Yeast Rice(Monascus nuruk) Flour (홍국(Monascus nuruk) 분말을 첨가한 스폰지 케이크의 품질 특성 및 노화 억제 분석)

  • This study investigated the quality characteristics and retarding retrogradation of sponge cakes made with red yeast rice (RYR) flour. RYR (Monascus nuruk) is known to help digestion, smooth blood flow, and have anti-cancer, anti-microbial, and inhibitory effects against biosynthesis of cholesterol and blood pressure. This studys aim' was to find the optimal proportion of RYR flour in sponge cake. RYR sponge cakes were prepared with various levels (0, 5, 10, 15 and 20%) replacement of wheat flour and were designated as the control (without RYR), RYR5, RYR10, RYR15 and RYR20 respectively. Specific gravity was the lowest in RYR15 at 0.57, and the baking loss rate was not significantly different among the samples (p<0.05). The dough yield was the highest in RYR15 at 96.61. The moisture contents was highest in order, control, RYR5, and RYR15 at 28.67%, 28.18%, and 26.82% respectively. The L-value of crust tended to increase according to the level of RYR, but the L-value of crumb decreased in accorddance with the the content of RYR. The a-value of crust also decreased according to the level of RYR, although the a-value of crumb increased in response to higher levels of RYR. The b-value tended to decrease with increases of RYR (p<0.05). RYR5 exhibited the highest pH at 8.63, compared with RYR15 (8.57). The hardness, which was measured after cooling for 1 hour, was the lowest in RYR15 at $163.33g/cm^2$ and the springiness was not different significantly (p<0.05). Cohesiveness was the highest in RYR10 at 133.06%. The chewiness was the highest in RYR10 at $391.63g{\cdot}cm$ and lowest in RYR15 ast $169.62g{\cdot}cm$. Avrami equation showed that RYR15 and RYR20 had the lowest Avrami exponent (n) at 0.0664 and 0.4983 respectively. Time constant (1/k) was the highest in RYR15 at 200.00. Sensory evaluation revealed that RYR15 was the highest in color (5.50), flavor (4.95), sweetness (4.90), chewiness (4.75), and overall acceptability (4.60).

Biological Activities and Bioactive Compounds in the Extract of Acer tegmentosum Maxim. Stem (산겨릅나무 줄기추출물의 생리활성 및 유효성분 분리)

  • Acer tegmentosum (Acereaceae) has been used a source of traditional medicines for the treatment of hepatic disorders in Korea. This research was conducted to determine biofunctional activities of A. tegmentosum stem extract and to identify its bioactive components. Methanolic extract from A. tegmentosum stem was partitioned by using organic solvents, including n-hexane, ethyl acetate, n-butanol, and water. Two compounds were isolated by using an ODS column chromatography from ethyl acetate soluble fraction shown to the strongest antioxidant activity ($RC_{50}=3.15\;{\mu}g/m{\ell}$) among the fractions. The isolated compounds were analyzed by $^1H$ and $^{13}C$ NMR, IR, UV/VIS, MS spectrum data and identified as catechin, ${\rho}-Hydroxyphenethyl$ alcohol $1-O-{\beta}-_D-(6'-O-galloyl)-glucopyranoside$. The compounds have shown strong antioxidant activity, with similar activity to BHA ($RC_{50}=2\;{\mu}g/m{\ell}$). Especially, ${\rho}-Hydroxyphenethyl$ alcohol 1-O-{\beta}-_D-(6'-O-galloyl)-glucopyranoside$ was shown strong anti-lipid peroxidative activity. However, the compounds were not shown antimicrobial activities. In antimicrobial activity assays, ethyl acetate soluble fraction was effective to bacterial inhibition, such as Escherichia coli and Klebsiella pneumonia, with minimum inhibitory concentrations in $125\;{\mu}g/m{\ell}$. Otherwise, antifungal activity against Candida albicans was shown in n-hexane soluble fraction exhibiting $63\;{\mu}g/m{\ell}$ of minimum inhibitory concentration. In anticomplementary activity assays, water soluble fraction was the most effective exhibiting 24% inhibitory activity.

THE ANTIMICROBIAL EFFECT OF HORSERADISH (ARMORACIA RUSTICANA) ROOT EXTRACTS AS AN ENDODONTIC IRRIGANT (근관세척제로써 서양산 고추냉이(Horseradish, Armoracia rusticana) 뿌리 추출물의 항균효과)

  • Microorganisms are the main causative factors of pulpal and periapical diseases, therefore successful endodontic treatment is depend on the effective elimination of intracanal bacterial populations. Many studies have been reported antimicrobial effect of Allyl isothiocyanate (AIT) which the principle ingredient of Horseradish (Armoracia rusticana) root extracts. The purposes of this study are to evaluate the antimicrobial effectiveness of Horseradish root extracts against Enterococcus faecalis in root canals of extracted human teeth and compare to sodium hypochlorite (NaOCl). Extracted human mandibular premolar root canals were infected with E. faecalis for 21 days, and then irrigated with Horseradish root extracts, NaOCl solution and saline. After canal irrigation, first samples (S1) were taken. After first sampling, the canals were additionally incubated 7 days, and then second samples (S2) were taken. The samples were inoculated on EHI agar plate to determine the colony forming units (CFU). 1. Mean values of CFU in S1 were $5.815{\times}10^3$ CFU/ml at Horseradish groups, and $3.465{\times}10^3$ CFU/ml at NaOCI groups. There was no statistically significant differences (p=0.086). 2. Mean values of CFU in S2 were $3.100{\times}10^3$ CFU/ml at Horseradish groups, and $5.252{\times}10^5$ CFU/ml at NaOCI groups. There was statistically significant difference (p<.05). 3. There was no statistically significant differences (p=0.076) between S1 and S2 at Horseradish groups in the mean values of CFU. However, there was statistically significant differences (p<.05) between S1 and S2 at NaOCI groups in the mean values of CFU.

Quality Characteristics of Fresh Noodles With Perilla Leaves (들깻잎을 첨가한 생면의 품질특성)

  • This study investigates the optimal application of perilla leaves to fresh noodles in terms of a variety of beneficial effects and functions such as the anti-microbial function of the leaves. First, we measured the water contents of the fresh noodles and found that the ones added with 7% lyophilized perilla leaf extract showed the maximum water contents whereas the control noodles without perilla leaf extract the minimum. Increasing amount of raw perilla leaf extract or lysophilzed perilla leaves in the fresh noodles elevated the L value and significantly reduced the A value. The mass and volume of the fresh noodles were gradually decreased with raw perilla leaves added. Addition of raw perilla leaf extract and lyophilized perilla leaves decreased the turbidity of the fresh noodles. Hardness of the fresh noodles was increased by the addition of lyophilized Perilla leaves. Next, we tested the effect of the addition of sesame leaves on microbial growth. No microbes were found in the fresh noodles in the absence or presence of sesame leaves at day 0. When the noodles were stored for 3 days, the greatest number of bacteria was detected in the noodles without perilla leaves while addition of perilla leaves lowered the amount of bacteria in the noodles. We then performed the sensory test. For the raw perilla leaf extract addition, the noodles with 9% of extract exhibited the highest in appearance, flavor, color (6.47), texture (6.60), and overall acceptability (7.67). Texture was the highest in the ones with 3% (6.87) and 5% (6.20) of extracts added. Overall acceptability (7.07) was the best when 3% perilla leaves were added. Overall, 9% addition of raw perilla leaf extract or 3% addition of lysophilized perilla leaves showed optimal tastes.
