Screening of Natural Antimicrobial Edible Plant Extract for Dooboo, Fish paste, Makkoli Spoilage Microorganism (식용 식물로부터 얻은 추출물의 두부, 어묵, 막걸리 변질균에 대한 항균성 검색)

  • To extend the self-life of Dooboo (tofu), fish paste and Makkoli (Korean traditional rice wine) for which severe heat treatment are not expectable, main putrefactive microorganisms were isolated from each product and tested their growth inhibition by ethanol and water extracts of several edible plants. The ethanol extract of Phellodandron amurense Ruprs had the strong effect of growth inhibiting to all three isolates from Dooboo and Makkoli, and Eugenia caryophyllus, Pinus rigia Mill, Bletilla striata (Thunb) Reichb. Fill and Paeonia albiflora Pall were also same effect to isolates from fish paste. The ethanol extract was more effective inhibiting than water extract in all test microorganisms. The evident inhibition level of each extract was 2000 ppm of Pa ethanol for fish paste and 1000 ppm for Makkoli.

Characterization of an antifungal compound isolated from an antagonistic fungus Aspergillus terreus against phytopathogenic fungi (식물병원균 생육을 저해하는 Aspergillus terreus로부터 분리한 향균물질의 특성)

  • An antagonistic fungus inhibiting growth of various phytopathogenic fungi was isolated from greenhouse soils and identified. Mophological features of fruiting structures on potato dextrose agar and colorless globose to ovate heavy walled hyaline cells from the vegetative mycelium grown on MY20 agar indicate that this antagonist is Aspergillus terreus. The antagonistic activity is due to the production of antifungal compounds. An antifungal compound was purified from its culture filtrate using chloroform extraction, column chromatography, and thin layer chromatography. The purified antibiotic was effective to various phytopathogenic fungi and identified as butyrolactone I. $ED_{50}$ values measured by petri-plate assay through effective dosage(ED)-probit analysis were 9.7, 13.7, 23.3, 42.6 and 102.7 ppm on Botrytis cinerea, Rhizoctonia solani, Pythium ultimum, Phytophthora capsici, and Fusarium oxysporum, respectively.

  • Antifungal activities of domestic plants were examined to develope natural antimicrobial agents, able to protect citrus fruits from spoiling microorganism during their storage. A fungus was isolated from citrus fruits' storage and identified to Penicillium sp. CF-301, based on the morphological characteristics of conidiophore and conidia; flask shape of phialide, simple branching type of conidiophore, and columnar shape of conidial head, in malt extract agar and potato dextrose agar media. Chloroform extracts of Juniperus chinesis Linnaeus, J. chinesis var. horizontalis showed remarkable antifungal activities against Penicillium sp. CF-301.

  • The aim of this study was to increase production of leucocin A, a kind of bacteriocin, in a transformed variety of S. cerevisiae. We investigated optical density, total secreted protein, protease activity, and antibacterial activity for the transformed S. cerevisiae in different carbon sources. The production of leucocin A growth-associated, and antibacterial activity, according to carbon source, was in the order of sucrose, glucose, glycerol, and fructose. Antibacterial activity was $10.6\%$ higher in the presence of sucrose than glucose. This is the first report regarding the effect of carbon sources on the production of leucocin A in transformed S. cerevisiae, as far as we ascertain. Our results could prove useful in the industrial production of natural preservatives.

  • The synthetic procedures to synthesize polymethacryloylsulfadiazine were searched by DCC method and Acid Chloride mothod. Polymeric drug was synthesized by Acid Chloride method in high yield (72%) but DCC method in low yield (23%). The antimicrobial activities of polymeric drug were investigated in terms of minimum inhibitory concentrations by the common two-fold dilution technique. Polymeric drug revealed an excellent antimicrobial Bacillus subtillis ATCC 6633, Mycrobacterium phlei IFO 3158, Micrococus luteus ATCC 9341, Salmonella typhimurium KCTC 1925, Klebsiella pneumoniae KCTC 1560 and similar activity against Staphylococcus aureus IFO 12732, Escherichia coli BE 1186, Escherichia coli AB 0111, Pseudomonas aeruginosa IFO 13130. Polymeric drug have no antimicrobial activity against Candida albicans IFO 1594.

  • The antimicrobial and antioxidative activities were investigated to confirm the utility of wood vinegar. Antimicrobial activity was performed by paper disc method and liquid culture. The growth inhibition was observed in all microbial species at a dose of as low as 25 $\mu$m of wood vinegar by paper disc method. Also, in liquid culture, S. cerevisiae and P. aeruginosa were more inhibited the growth than others in the concentration of $2\%$ (v/v). For measuring of antioxidative activity, wood vinegar was fractionated with acidic, phenolic, basic and neutral fraction; and their antioxidant activities were measured by the radical scavenging effect on DPPH radical. In four fractions, phenolic fraction showed high antioxidative activity.

Effects of Gamma Irradiation on Antimicrobial Activity of Korean Medicinal Herb Extracts (한약재 추출물의 향균활성에 대한 감마선 조사의 영향)

  • Irradiated and non-irradiated Korean medicinal herbs were extracted by water and 70% ethanol. Antimicrobial activity of these extracts were invesigated against selected food hygiene microoganisms. The ethanol extracts of the non-irradiated Agrimonia pilosa ledebour japonica Nakai, Curcuma longa Linne and Angelica gigas Nakai were completely inhibited on four species of bacteria, such as Vibrio parahaemolyticus, Clostridium perfringenes, Bacillus subtilis and Staphylococcus aureus. Also, above four strains did not have antibacterial activity in the water-ethanol mixtures. Futhermore, the ehtanol extracts of the non-irradiated Agimonia pilosa ledebour Japonica Nakai, Curcuma Iedoaria Roscoe, Curcuma longa Linne and Scutellaria baikalensis George were shown inhibitory effects against Aspergillus flavus and Pen icillium islandicum. And the water extract of Scutellaria baikalensis George was the same effect to these molds. Essentially the same results were observed when samples irradiated at a dose of 10 kGy.

  • Water extract, and methanol extract, its chloroform and hexane fractions, and estragole from Agastache rugosa O. Kuntze were tested to find the inhibition effect on the growth of several microorganisms. The organisms used were Escherichia coli ATCC 1129, Staphylococcus aureus IAM 1011, Vibrio parahaemolyticus WP, Bacillus subtilis ATCC6633, Aspergillus oryzae KFCC 890, Aspergillus niger KCCM 11240. Water and methanol extracts at the concentration of 0.5%, and chloroform and hexane fraction at the concentration of 0.05% inhibited the growth of microorganisms from 1/5 to 2/3 of the control group. Estragole identified from the hexane faction as a major component, its authentic compound completely inhibited the growth of Vibrio parahaemolyticus completely at the concentration of 0.03%, and the other bacteria were at 0.05%.

  • 겨자의 가수분해 시간에 따른 향균력을 비교하고, 각종 미생물에 대한 겨자 물 추출물의 최소 저해 농도 및 최소 저해 투여량을 측정한 결과 다음과 같았다. 겨자의 자가분해 시간에 따른 물 추출물의 향균성은 처음에 아주 약하게 나타났던 것이 12시간 방치 후 크게 증가하였다. 최대 항균활성은 24시간 방치 후 나타났으며 이후 거의 변하지 않았다. 겨자를 첨가한 물의 산도는 시간이 지남에 따라 증가하였고, 겨자의 농도가 클수록 그 정도가 크게 나타났다. $37^{\circ}C에서$ 24시간 자가분해한 겨자 물 추출물의 MID는 Bacillus subtilils가 $70\mu1/disc로$ 다른 실험 균주에 비하여 낮았으며, 젖산균이 $95\mu1/disc로서$ 높게 나타났다. MIC는 Bacillus subtilils, Staphylococcus aureus, Salmonella typhimurium 및 Vibrio parahaemolyticus 균이 $70\mu1/ml로$ 다른 실험 균주에 비하여 낮게 나타났으며, 젖산균 곰팡이에서 $90\mu1/ml로$ 높게 나타났다. 자가분해 시간에 따른 겨자물 추출물의 중류성분은 자가분해 시간이 지남에 다라 점차 증가하는 경향이었고, isothiocyanate 류의 함량은 24시간 때에 최대치를 나타내었다.
