Systematic Study on the fishes of the Family Cobitidae (Pisces, Cypriniformes) 4. The Analyses of Karyotype and Mitochondrial DNA between the Two Species of the genus Misgurnus from Korea (기름종개과(Family Cobitidae) 어류의 계통분류에 관한 연구. 4 미꾸리속 어류 2종의 핵형 및 mtDNA 분석)

  • 미꾸리속 어류 2종의 유전적 차이를 알아보기 위하여 염색체 분석과 미토콘드리아 DNA(mtDNA) 분석을 실시하였다. 일반염색에 의한 핵형 분석 결과 미꾸리(2N=50)와 미꾸라지(2N=48)는 염색체수에 차이가 있었으며. N-banding 분석 결과 두 종간에는 인형성 부위의 위치와 크기에 차이가 있었다. C-handing 겪과 미꾸라지는 1번 염색체쌍 동원 체부위에 넓게 C-band플 갖고 있었다. 미꾸리속 어류 2종의 mtONA를 7개의 6-base cutting 제한효소로 처리하여 절편 양상을 비교. 분석한 결과 2종 공히 mtDNA의 genome 크기는 약 16.OKb였으며 fragment homology(F)에서 미꾸리의 종내 집단간의 F값은 0.674. 미꾸라지는 0.862로 유사하게 나타났으나, 종간 F값은 0.207(0.074-0.417)로 낮았다 염기치환율은 미꾸리가 p=0.021, 미꾸라지는 p=0.002로 미꾸라지가 미꾸리에 비해 매우 낮은 염기치환율을 보였고. 종간 평균 염기치환율은 p=0.104로 차이 를 나타냈다. MtDNA 분석과 핵형 분석 결과 미꾸라지는 Robertsonlan translocation의 결과 미꾸리로 부터 분화된 것으로 추정 되었다.

Cytogenetic Analysis of Seven Angelica Species (Angelica속 식물 7종의 세포유전학적 분석)

  • Karyotypes were established in seven Angelica species cultivated in Korea. The somatic chromosome numbers were 2n = 2x = 22 with the basic number of x = 11 in all Angelica plants examined. Their metaphase chromosomes ranged from 3.56 ${\mu}M$. to 8.91 x. in length. Distinctive Karyotypes were found in two species, A. tenuissima with all metacentries, K(2n) = 2x = 22m, and A. genuflexa with all subtelocentrics, K(2n) = 2x = 22st. Karyotype formulas of A. gigas, A. acutiloha, A. sinensis, A. decursiva and A. dahurica were K(2n) = 2x = 20m + 2sm, K(2n) = 2x = 12m + 10sm, K(2n) = 2x = 16m + 6sm, K(2n) = 2x = 18m + 4sm and K(2n) = 2x = 10m + 10sm + 2st, respectively. Cytological data showed that chromosomal polymorphisms within species were observed in Angelica plants compare to other regions.

  • The comparative karyological analysis of the Korean treefrog, Hyla japonica and Hyla suweonensis performed by conventional giemsa-staining and NOR-staining method. The karyotypes of both species were the same (2n=24). However, according to the Karyological analysis, the 6th chromosomes of the two species distinctly different. Eventhough the 6th chromosomes of the two species shown the same % length, construction of the 6th chromosome of H.japonica was subtelocentric chromosome while H. suweonensis was submetacentric chromosome. This phenomena could explain that the morphological differences in the 6th chromosomes might be caused by pericentric inversion. The two species have 1 pair of NOR site in the 6th chromosome.

Chromosome Compositions of Four Cultivated Cucurbitaceae Species. (박과 작물 4종의 핵형분석)

  • The chromosome numbers and compositions were investigated in four cultivated species of Cucurbitaceae; Cucumis sativus L., Citrullus lanatus (Thunb.) Matsum. et Nakai, Cucumis melo L., Luffa cylindrica (L.) Roemer. through general aceto-orcein staining method. The chromosome compositions of four species were diploids of 2n=22, 2n=24 and 2n=26 respectively. The chromosomes were relatively small and showed gradual length degradation from $2.50\;{\mu}m$ to $2.16\;{\mu}m$ in Cucumis sativus, $3.71\;{\mu}m$ to $2.11\;{\mu}m$ in Cucumis melo, $3.20\;{\mu}m$ to $2.40\;{\mu}m$ in Citrullus lanatus and $3.17\;{\mu}m$ to $1.97\;{\mu}m$ in Luffa cylindrica. The chromosome types consisted of all metacentrics in Cucumis sativus, seven pairs submetacentrics and five pairs metacertrics in C. melo, four pairs of submetacentrics and seven pairs metacertrics in Citrullus lanatus, and two pairs submetacentrics and eleven pairs metacentrics in Luffa cylindrica (L.) Roemer.. The satellites were found in a pair of chromosomes in C. melo and two pairs in Luffa cylindrica. The chromosome compositions in these four species showed species-specific patterns and seemed to provide useful informations for breeding and molecular cytogenetic works on Cucurbitaceae.

  • Molecular cytogenetics allows the identification of unknown chromosome rearrangements, which is clinically useful in patients with mental retardation and/or development delay. We report on a 31-year-old woman with severe mental retardation, behavior development delay, and verbal performance delay. Conventional cytogenetic analysis showed a 46,XX,add(8)(p23.3) karyotype. To determine the origin of this unbalanced translocation, we performed array CGH and subtelomeric FISH. The results showed that the distal region of chromosome 8p was added to the terminal of chromosome 13q. This was confirmed the final result of 46,XX,der(8)t(8:13)(p23.3;q32.1)dn.

  • Backcross hybridization between Korean bullhead Pseudobagrus fulvidraco female and Korean bullhead P. fulvidraco $\times$ Ussurian bullhead Leiocassis ussuriensis hybrid male was performed, and early viability and karyotype of the backcross hybrids were examined along with their parental species. Mean fertilization rate (86.8%), hatching success (70.7%) and early survival rate (76.4%) of backcross hybrids were similar with those found in the maternal species, the Korean bullhead. From the chromosome analysis, modal chromosome numbers of Korean bullhead, Ussurian bullhead, their hybrid and backcross hybrid were the same as 2n = 52. However, their karyotypes were different among genotypes. The karyotype of backcross hybrid was 22 metacentric + 18 submetacentacentric + 12 acrocentric chromosomes.

  • The aim of this study was to test the validity of the hypothesis that the karyotypes of four species of Lepilemuridae were formed spontaneously from their ancestral hybrid karyotype. Hypothetical ancestral haploid Karyotype of Lepilemuridae is composed of 18 autosomes and X chromosomes. Lepilemur mustelinus karyotype has four tandem fused chromosomes and one Robertsonian translocated chromosome pairs. Lepilemur septentrionalis septentrionalis karyotype has only two pairs of translocated chromosomes. We reconstruct and suggest ancestral karyotype of LMU(ancLMU) and LSS(ancLSS), from which all four studied species were derived. Hybrids of ancLMU and ancLSS were formed and produce differently fused equilibrated gametes via circular form arrangement during gametogenesis. Five unit of trivalent homologous chromosome pairs were engaged in a circular form to give new gamete corresponding to the karyotype of L. dorsalis, orientation of one unit of trivalent was inversed in the circle to gave new gamete corresponding to the karyotype of L. leucopus. Seven homologous chromosome pairs were engaged in circular form to give haploid karyotype of Lepilemur ruficaudatus. Only one homologous chromosome pair is dissociated and the other chromosome pairs rearranged in the circle to form haploid karyotype of Lepilemur edwardsi. The new gametes could be produced from these circular forms. When the new gamete fertilized with the same type of gamete, The new homozygote is produced as existing L. dorsalis, L. leucopus, L. edwardsi and L. ruficaudatus. These results support the theory that new species could be formed in hybrid population through activated chromosome fusion, chromosome rearrangement in circular from at zygotene stage and production of equilibrated gametes to form homozygote new species.

Karyotype Analysis of Five Iris Species Native to Korea (한국 자생 붓꽃과 식물 5종의 핵형분석)

  • A detailed karyotype analysis was carried out in five Korean native Iridaceae species; Iris pseudoacorus, I. pallasii. var. chinensis, I. tectrum, I. dichotoma and Belamcanda chinensis. Chromosome compositions of the five species showed that they have different karyotypic formulae; I. pseudoacorus 2n=34=10m+16smn+8st including two pairs of satellite chromosomes, I. pallasii var. chinensis 2n=40=26m+12sm+2st including two pairs of satellite chromosomes, I. tectrum 2n=30=14m+16sm including five pairs of satellite chromosomes, I. dichotoma 2n=32=22m+10sm including two pairs of satellite chromosomes, and B. chinensis 2n=32=20m+10sm+2st including one pair of satellite chromosomes. These results will supplement the previous cytogenetic reports in Iridaceae species and enhance our understanding on the genetic structure, which will be useful in clarifying the unique characteristics of each species for practical breeding programs for horticultural and pharmaceutical purposes.

Karyotypes of Five species in Odontobutidae and Cottidae of Korea (한국산 동사리과 (Odontobutidae)와 둑중개과 (Cottidae) 5종의 핵형)

  • The chromosome numbers of five species in two families of Korea are investigated: Odontobutis platycephala, O. interrupta, and O. obscura in Odontobutidae, and Cottus koreanus and C. hangiongensis in Cottidae. In Odontobutis species, the three species showed the diploid chromosome number, 2n=44 (NF=44) telocentric chromosomes. In Cottus species, the mitotic chromosomes from 24 groups with two chromosomes each indicated that it is a diploid. The karyotype of C. koreanus and C. hangiongensis is 2n=48 and NF=52. These species is the first report on the chromosomes and the karyotype analysis except O. platycephala. Our findings provide cytotaxonomic evidence for the species distinctness of these five species whose descriptions were based primarily on external morphology.
