Potential use of Bacillus amyloliquefaciens as a probiotic bacterium in abalone culture (북방전복, Haliotis discus hannai 에 대한 Bacillus amyloliquefaciens의 probiotic 효과)

  • In comparison to the numbers of such studies of fish, few studies have been carried out on the immunity, physiology and ecology of abalone, while studies on abalone disease are also extremely rare. Moreover, mass mortality of cultured abalone due to pathogenic bacteria has not been reported in the southern coast of Korea. However, Vibrio-like bacteria have been isolated from dead abalone, which indicates that a review is required in order to determine the cause of abalone mortality. Use of an antimicrobial agent to minimize the damage caused by disease in abalone farms is common, but the therapeutic effects are insignificant. Demand for probiotics has increased, but research on the development of probiotics for use in abalone culture is very rare. Therefore, the present study isolated KC16-2 from fermented kimchi soup and investigated the characteristics of the isolate as a candidate probiotic bacterium in abalone. KC16-2 was identified as Bacillus amyloliquefaciens (B. amyloliquefaciens KC16-2) based on its biochemical properties and 16S rRNA gene sequence. B. amyloliquefaciens KC16-2 showed inhibitory effects against the growth of various vibrios in vitro, and kept the numbers constant until four days after inoculation in marine water at a temperature of $15{\sim}25^{\circ}C$, indicating the possible use of KC16-2 as a probiotic, except in the winter. The growth of KC16-2 was inhibited by bile salt, but the numbers increased over time suggesting the bacteria were still alive in the abalone's digestive tract. Abalone fed with a diet including KC16-2 for 12 weeks showed good growth, but showed no significant differences from the control group. However, the mortality of the abalone supplied the probiotic diet was reduced to half that of the control group in a challenge test with Vibrio tubiashii. Therefore, we suggest that B. amyloliquefaciens KC16-2 could be used as a probiotic bacterium for control of the mortality of abalone caused by opportunistic pathogenic vibrios.

The Physico-chemical Characteristics in the Garorim Bay, Korea (가로림만의 이화학적 수질의 시.공간적 특성)

  • The physico-chemical characteristics including water temperature, salinity, dissolved oxygen(DO), chemical oxygen demand (COD), chlorophyll-a(Chl. a), suspended particulate matter(SPM) and dissolved inorganic nutrients were investigated in the Garolim Bay, Yellow Sea, Korea in 2010 carried out six times per year at 11 fixed stations by Korea Fisheries Research & Development Institute. The water temperature, salinity, COD, dissolved inorganic nutrients, Chl. a and SPM showed significant difference between surface and bottom water but the other parameters didn't. There were not significant difference between stations. The water temperature showed typical change patterns of the temperate seawater. The annual average of salinity showed more than 31 so that there could not have occurred low saline water. The average of DO from June to August showed over than 3mg/L which showed higher than the below standard value of the hypoxic (oxygen-deficient) water. The average of Chl. a varied $1.68{\mu}g/L$ at surface, $2.38{\mu}g/L$ at bottom layer in June and $1.68{\mu}g/L$ at surface, $1.57{\mu}g/L$ at bottom layer at August. The dissolved inorganic nutrients showed high concentration in February and low concentration in August due to the limitation of the freshwater input in summer and phytoplankton used to the dissolved inorganic nutrients. The ratio of DIN/DIP showed 30.52 at surface and 37.89 at bottom layer in June which was higher than other month. The SPM was 44.15mg/L at bottom layer in February which was the highest value in this study due to the northwest monsoon. Because of the actively water change in the open sea without inflow of freshwater from land in Garolom Bay, there were not occurred low saline water and hypoxic water. thus, this Bay showed good water quality and required to be conserved continuously as important costal area for fisheries.

Egg Development and Larvae Morphology and Spawning Behavior of Five Striped Damselfish, Abudefduf vaigiensis (Pisces: Pomacentridae) Reared in the Laboratory (실험실에서 사육한 해포리고기 (Abudefduf vaigiensis)의 산란행동, 난 발생 및 자어의 형태발달)

  • The spawning behavior, development of eggs and larvae of the Five striped damselfish, Abudefduf vaigiensis were studied. The Five striped damselfish were caught at Dolsan Island, Yeosusi, Jeollanamdo from May in 2011. As a result of observation, male fish attracted female after cleaning the rock. Female left after spawning and male protected their eggs until they had hatched out. The fertilized eggs were elliptical in shape (mean long diameter: 1.06 mm; mean short diameter: 0.55 mm) and transparent. Larvae hatched at 53 hrs after fertilization in $24.5{\sim}26.5^{\circ}C$(mean $25.0^{\circ}C$). The newly hatched larvae were 2.55~2.86 mm (mean 2.71 mm, n=10) in total length and their mouth and anus were already opened. They began to eat rotifer and transformed to postlarva stage. 3 days after hatching postlarva was measured 2.74~2.97mm(mean 2.84 mm, n=10) in total length. 10 days after hatching postlarva was measured 3.85~4.20mm(mean 4.00 mm, n=10) in total length with dosal fin rays IV-5; ventral fin rays I-3; caudal fin rays 1~2.

Morphological Adaptation of Zostera marina L. to Ocean Currents in Korea (한국산 거머리말(Zostera marina L.)의 해류에 대한 형태적 적응)

  • The main purpose of this research is to prepare and provide basic materials for the propagational strategy of eelgrass by investigating on the morphological adaptation of Korean Zostera marina to ocean currents. An eelgrass plant mainly consists of rhizome, leaf sheath, leaves and roots. The rhizome is the horizontal stem of the plant that serves as the backbone from which the leaves and roots emerge. The leaf sheath is the bundle at the base of the leaves that holds the leaves together, protecting the meristem, the primary growth point of the shoot. Leaves originate from a meristem which is protected by a sheath at the actively growing end of the rhizome. As the shoot grows, the rhizome elongates, moving across or within the sediment, forming roots as it progresses. The aggregated leaves from the leaf sheath are found to have two cell layers on one side and multiple layers of airy tissues called aerenchyma on the other. The aerenchyma tissues are developed in multi-layered cell structures surrounding the veins which are formed in the leaf sheath. Generative shoots are made of rhizomes, which are circular or ovoidal, stem, and spathe and spadix. The transverse section of rhizome and the stem and central floral axis is found to be circular, ovoid and in the shape of convex respectively, and the vascular bundle, which is a part of transport system, has one large tube in the center and two small tubes on both sides. The layers of collenchyma cells numbered from 12 to 15 in the stem, and from 7 to 12 in the rhizome. The seed coat is composed of sclereids, small bundles of sclerenchyma tissues, which prevent the influx of sea water from the outside and help endure the environmental stress. In conclusion, alternative multi-layer structure in circular, convex type aggregated leaf base are interpreted to morphological adaption as doing tolerable elastic structure through movement of seawater. The generative shoots develop long slim stem and branches in circular or ovoidal shapes to minimize the adverse impacts of sea current, which can be interpreted as the plant's morphological adaptation to its environment.

A Study on The Consumer Expectation - Performance according to the Types of Internet Shopping Malls (인터넷 쇼핑몰 유형에 따른 소비자 기대-성과에 관한 연구)

  • To create and maintain comparative supremacy as a strategic tool of business, many organizations have introduced informational technology and system. By using this system, Some companies got a beneficial value for achieving organizational goals but others could not obtain their effectiveness and efficiency. In particular, a lot of organizations that tried to make strategic supremacy with e-commercial trade are under hard condition because of poor profit. It implies that it is essential to identify and analyse the consumer who uses e-commercial trade. This paper, therefore, focusing on internet shopping malls between business and consumer as one of areas of e-commercial trades, shows the difference between consumer expectation and performance. The results of this study are as follows: First, as for the significant difference of influencing factors to consumer satisfactions according to the types of internet shopping malls, there is a meaningful difference in consumer anxiety and internet usefulness, but not in consumer service. Prior to verify the differences in detail on consumer's anxiety and internet usefulness, we examined that there is any difference between expectation and performance. T-test was used for the variants of consumer anxiety and internet usefulness, and its meaningful probability was 0.000, which means that both showed statistically significant difference. Based on the results, we also found that regardless of the types of internet shopping malls, consumer expectation was greater than performance. although the difference between expectation and performance was not equal according to the internet shopping malls. Second, a regression analysis was performed to understand the relation between consumer service, internet usefulness, consumer anxiety, and consumer satisfaction, it was found that consumer service, internet usefulness, consumer anxiety had significantly effected on consumer satisfaction. Third, To verify the relation between consumer satisfaction and repurchase-intentions, intentions to spread out, Pearson correlation analysis was used. it was found that consumer satisfaction had positive effect on both intentions. This study has some limitations because of the shorts of money and time. since the sample of this study was consumers who have ever bought one or more products via internet shopping mall, this sample was appropriate. but the major parts of sample were college students, and the sample size was so small. therefore this results should carefully be generalized. For further study, it is required to select more precise samples and to include more variables.

Distribution of water Masses and Chemical Properties in the East Sea of korea in Spring 2005 (2005년 춘계 동해 중남부 해역의 수괴 분포 및 화학적 특성 연구)

  • In order to understand the water mass properties in the southern location of the East Sea in the Korean coasts, the vertical distributions (down to 2,000 m deep) of water temperature, salinity, and dissolved inorganic nutrients were determined in April of 2005. The water mass of the surveyed location showed distinct vertical layers; highly saline surface, surface mixing layer, and thermocline of low temperature and salinity. The water layer below 300 m was characterized by water temperature lower than $1^{\circ}C$ and salinity 34.06, showing a representative water mass of the East Sea. The inorganic nutrients rapidly increased from 200m in the northern and southern parts around Ulleung Basin. A marked environmental difference was found between two layers separated by thermocline. The upper layer of the thermocline was oligotrophic and the vertical distribution of nutrient was very stable. In the water layer between 100 and 200m the nutrients slightly increased but remained still stable. From southern coasts to northeastern Ulleung, the water mass properties were site specific; the thickness of the surface mixed layer and nutricline showed a trend diminishing toward the northern locations probably due to diminished influence of Tsushima water. Redfield ratio (N:P=16:1) based on the ratio of chemical composition in organism revealed that nitrogen value continuously decreased to less than 16 with the water depth down to loom from the thermocline. The value in the water layer deeper than 100 to 200 m, thereafter, showed an increasing trend (over 16). This result was further supported by the finding of lower chlrophyll a content in the layer.

Wind-and Rain-induced Variations of Water Column Structures and Dispersal Pattern of Suspended Particulate Matter (SPM) in Marian Cove, the South Shetland Islands, West Antarctica during the Austral Summer 2000 (서남극 남 쉐틀랜드 군도 마리안 소만에서 바람 및 강수에 의한 여름철 수층 구조의 변화와 부유물질 분산)

  • Time-series CTDT (Conductivity/Temperature/Depth/Transmissivity) were obtained at one point near tidewater glacier of Marian Cove (King George Islands, Antarctica) to present water column properties and SPM (suspended particulate matter) dispersal pattern in relation with tide, current, meteorological data, and SPM concentration. Four layers were divided from the water column characteristics measured in the interval of an hour for about 2 days: 1) cold, fresh, and turbid surface mixed layer between 0-20 m in water depth, 2) warm, saline, and relatively clean Maxwell Bay inflow between 20-40 m in water depth, 3) turbid/cold tongue of subglacial discharges compared with the ambient waters between 40-70 m in water depth, and 4) cold, saline, and clean bottom water beneath 70 m in water depth. Surface plume, turbid freshwater at coastal/cliff area in late summer (early February), had the characteristic temperature and SPM concentration according to morphology, glacial condition, and composition of sediments. The restrict dispersion only over the input source of meltwater discharges was due to calm wether condition. Due to strong wind-induced surface turbulence, fresh and turbid surface plume, englacial upwelling cold water, glacier-contact meltwater, and Maxwell Bay inflow was mixing at ice-proximal zone and the consequent mixed layer deepened at the surface. Large amount of precipitation, the major controlling factor for increasing short-term glacial discharges, was accompanied by the apparent development of subglacial discharge that resulted in the rapid drop of salinity below the mid depth. Although amount of subglacial discharge and englacial upwelling may be large, however, their low SPM concentration would have small influence on bottom deposition of terrigenous sediments.

  • This study was conducted to evaluate the purification efficiency in rearing water of the land based fish farm by screen filter and ultra violet (UV) irradiation. Purification efficiency for rearing seawater has been examined with screen filter of 60 $\mu$m pore size and UV irradiation at dose of 0.5 $mWS/cm^2$ for 5 months. Purification efficiency by changing of temperature, salinity, pH, DO, total bacteria and Vibrio species in rearing seawater by filtering and UV irradiation were not significant during 5 months, However, the removing rate of suspended solid and turbidity of rearing seawater were $43.8\~45.6\%$ (average, $44,7\%$) and $29.2\~33.2\%$ (average, $31,3\%$) by filtering, respectively. Also, Purification efficiency for the $NO_3^{-}-N,\;NO_2^{-}-N,\;NH_4^{+}-N$ and $PO_4^{3-}-P$ were $21.3\~21.9\%$ (average, $21.6\%$), $24.1\~25.2\%$ (average, $24.7\%$), $17.6\~17.8\%$ (average, $17.7\%$) and $19.0\~20.4\%$ (average, $19.7\%$) respectively by the system used on this study.

Detoxification of PSP and relationship between PSP toxicity and Protogonyaulax sp. (마비성패류독의 제독방법 및 패류독성과 원인플랑크톤과의 관계에 관한 연구)

  • The purpose of this study was to investigate the detoxifying effect on PSP-infested sea mussel, Mytilus edulis, by heating treatment and correlation between the PSP toxicity and the environmental conditions of shellfish culture area such as temperature, pH, salinity, density of Protogonyaulax sp. and concentration of inorganic nutrients such as $NH_4-N,\;NO_3-N,\;NO_2-N\;and\;PO_4-P$. This experiment was carried out at $Suj\u{o}ng$ in Masan, Yangdo in Jindong, $Hach\u{o}ng\;in\;K\u{o}jedo\;and\;Gamch\u{o}n$ bay in Pusan from February to June in $1987\~1989$. It was observed that the detection ratio and toxicity of PSP in sea mussel were different by the year even same collected area. The PSP was often detected when the temperature of sea water about $8.0\~14.0^{\circ}C$. Sometimes the PSP fox of sea mussel was closely related to density of Protogonyaulax sp. at $Gamch\u{o}n$ bay in Pusan from March to April in 1989, but no relationship was observed except above duration during the study period. The concentration of inorganic nutrients effects on the growth of Protogonyaulax sp., then effects of $NO_3-N$ was the strongest among them. When the PSP-infested sea mussel homogenate was heated at various temperature, the PSP toxicity was not changed significantly at below $70^{\circ}C$ for 60 min. but it was proper-tionaly decreased as the heating temperature was increased. For example, when the sea mussel homogenate was heated at $100^{\circ}C,\;121^{\circ}C$ for 10 min., the toxicity was decreased about $67\%\;and\;90\%$, respectively. On the other hand, when shellstock sea mussel contained PSP of $150{\mu}g/100g$ was boiled at $100^{\circ}C$ for 30 min. with tap water, the toxicity was not detected by mouse assay, but that of PSP of $5400{\mu}g/100g$ was reduced to $57{\mu}g/100g$ even after boiling for 120 min.

A Study on the Characteristics of Stream Flow Path and Water System Distribution in Gugok Garden, Korea (한국 구곡원림(九曲園林)의 하천 유로 및 수계별 분포 특성)

  • In this study, the water flow system by measuring the flow-way type and distance of flow path that composes the Gugok through literature survey, field survey, and map work on Gugok gardens in Korea whose existence has been confirmed, while investigating and analyzing watersheds, river orders, and river grades. It was intended to reveal the watershed distribution and stream morphological characteristics of the Gugok gardens and to use them as basic data for future enjoyment and conservation of the Gugok gardens. The conclusion of the study is as follows. First, Of the 93 Gugok gardens that have been confirmed to exist, it was found that 11 places(11.8%) were found to have a descending(top-down) type of Gugok that develops while descending along a stream. Second, As a result of analysis of the length of the flow path for each valley, Okryudonggugok(玉流洞九曲, Namsan-gugok) in Gimcheon, Gyeongsangbuk-do was found to have the shortest length of 0.44km among the surveyed valleys, while the flow distance of Muheulgugok(武屹九曲) located in Seongju-gun and Gimcheon-si, Gyeongsangbuk-do was 31.1km, showing the longest flowing distance. The average flow path length of the Gugok Garden in Korea was 6.24km, and the standard deviation was 4.63km, indicating that the deviation between the 'curved type'e and the 'valley type' was severe. In addition, 14(15.1%) Gugok gardens were found to be partially submerged due to dam construction. Third, As a result of analyzing the waters area where Gugok garden is located, the number of Nakdong river basins was much higher at 52 sites(55.9%), followed by the Hangang river basin at 27 sites(28.7%), the Geum river basin at 9 sites(9.7%), and the Yeongsan river and Seomjin river basins at 5(5.4%). Fourth, All Gugok gardens located in the Han river region were classified as the Han river system, and the Gugok garden located on the Nakdong river was classified as the main Nakdong river system, except for 7 places including 5 places in the Nakdong Gangnam Sea water system and 2 places in the Nakdong Gangdong sea water system. As a result of synthesizing the river order of the flow path where Gugok garden is located, Gugok, which uses the main stream as the base of Gugok, is 3 places in the Hangang water system, 5 places in the Nakdong river system, 2 places in the Geumgang water system, and 1 place in the Yeongsangam/Seomjin river system. A total of 11 locations(11.5%) were found, including 36 locations(38.2%) in the first branch, 29 locations(31.2%) in the second branch, and 16 locations(17.0%) in the third branch. And Gugok garden, located on the 4th tributary, was found to be Taehwa Five-gok(太華五曲) set in Yonghwacheon Stream in Cheorwon in the Han river system, and Hoenggyegok(橫溪九曲) in Yeongcheon Hoenggye Stream in the Nakdong river system. Fifth, As a result of the river grade analysis of the rivers located in the Gugok garden Forest, the grades of the rivers located in the Gugok garden were 13 national rivers(14.0%), 7 local first-class rivers(7.5%), and 74 local second-class rivers(78.5%) was shown.
