The 'Fantastic' in the René Laloux's movie (<죽은 시간들(Les Temps Morts), 르네 랄루(René Laloux) 작, 1964>의 환상성)

  • This research aims at showing specificity of the 'fantastic' in the movie < Les temps morts (on 1964) > directed by Ren$\acute{e}$ Laloux (1929-2004 ), all over the world recognized director. This movie has a particular style by composing four forms of expression: the real recording (movie), the recording embellishes with images by image (animation), the drawing, the photo. This film is the most strange among his all films. Even if we could catch the key meaning of the film, it offer for the audience the sentiment incertain and unclear. If we consider 'the fantastic' as a hesitation between the real and the unreal, in diverse levels, this movie offers to the spectators the fantastic feelings. In order to present the way this film shows us the fantastic, we divide the film into 15 sequences according to the criteria of the visual elements and the auditive elements. We analyze specificities of this fantastic in diverse levels. At first, the first style of the drawing of Roland Topor, does not let us escape easily from the feeling of fantasy. The four representation formats(drawing, photo, animation, movie) are integrated into one whole by auditive elements(music, narration). On the other hand, certain parts incomprehensible are not integrated into the entire. are fully integrated into the unity that does not understand that part, leaving them can. Laloux leads the audience into a reality toward the end of the film, but he leave incertain sequences at the last moment. Through which the audience is again hesitant between the real and the unreal, the fantastic is strengthened as a result of the work. Finally, the fantastic of the film could be found at three levels. First, the fantastic drawing style of Roland Topor. In the second place, the fantastic exposed through the entire composition and structure of work. Overall, these by leaving through the availability of the story incomprehensible to the audience is to provide a fantastic sentiment.

Monitoring of Feed-Nutritional Components, Toxic Heavy Metals and Pesticide Residues in Mushroom Substrates According to Bottle Type and Vinyl Bag Type Cultivation (버섯의 봉지재배 및 병재배 시 재배단계별 배지의 사료영양적 성분, 독성중금속 및 잔류농약 모니터링)

  • This study was carried out to monitor feed-nutritional components, toxic heavy metals (Cd, Pb and As) and pesticide residues through three cultivation stages (1st initial culture stage, 2nd mycelial growth stage, and 3rd fruit body-harvested stage) of king oyster mushroom (Pleurotus eryngii) produced by bottle type cultivation and oyster mushroom (Pleurotus osteratus) produced by vinyl bag type cultivation. For both cultivation types, compared with the initial culture, the weight reduction rate in spent mushroom substrates (SMS) after fruit body harvest was 29% for total wet mass, 21~25% for dry and organic matters and 19 ~22% for neutral detergent fiber. Two thirds to 3/4 of organic matter degraded and utilized by mycelia and fruit bodies was originated from fiber, of which the primary source (50~70%) was hemicellulose. The effect of mycelial growth stage on chemical compositional change in culture was little (P>0.05) for bottle type cultivation of king oyster mushroom but considerable (P<0.05) for vinyl type cultivation of oyster mushroom. Culture nutrients uptake by fruit bodies was very active for the bottle type cultivation. Compared with SMS, harvested fruit bodies (mushrooms) contained higher (P<0.05) crude protein, non-fibrous carbohydrate, and crude ash and lower (P<0.05) neutral detergent fiber. Regardless of stages, no culture samples were contaminated with toxic heavy metals and pesticide residues. In conclusion, the increase of fiber (neutral and acid detergent fibers) and indigestible protein contents and the decrease of true protein content in SMS indicated that the feed-nutritional value of SMS was significantly reduced compared with that of the initial culture and they were safe from toxic heavy metals and pesticide residues.

Establishment of an Optimal Washing Condition of a High Temperature Steaming System for the Production of High Quality Pork (고품질 돈육 생산을 위한 고온증기세척수의 최적 조건)

  • The objective of this study was to determine the efficacy of different water temperatures and time of spray-washing on the removal of bacteria contamination from surface of pork carcass and to obtain better meat quality with high temperature stream between 60 to 90℃ for the time ranged from 10 to 60 sec. Results showed that total plate counts were significantly decreased with increasing steam temperature(P<0.05). Similar results were found with the spray time(P<0.05). The lightness(CIE L*) value was significantly increased in both loin and ham cuts over 80℃(P<0.05). Ultimate pH of muscle and water-holding capacity were decreased with increasing steam temperature(P<0.05). There was a significant difference in solubility of sarcoplasmic protein between 70℃ and 80℃ of the steam treatments with higher value at 70℃(P<0.05). Again, longer than 30 sec spray at 70℃ steam, the lightness value of pork was significantly increased(P<0.05), while pH and water-holding capacity of muscle were significantly decreased(P<0.05). Therefore, a desirable pork quality would be achieved with spray-steam washing at 70℃ for 20 sec on pork carcass.

Study on characteristics of specific hazardous substances in the industrial wastewater effluent (사업장 방류수 중 특정수질유해물질 배출 특성 연구)

  • In this study, 165 wastewater discharge facilities in 10 business types were investigated with regard to 24 specific hazardous substances that included heavy metals, VOCs, CN, and phenol in the Gwangju city. Cu in the range from from 0.008 to 35.420 mg/L was detected in all business types and the detection rate was 46.8 %. Other heavy metals, such as Cd, As, Hg, Pb, and Cr+6 were detected as well. However, their detection rates ranged between 0.6 and 1.8 %. CN and phenol were detected in one and five facilities, respectively. 12 species of VOCs were detected: chloroform 80.6 % (0.42 to 81.60 μg/L), benzene 16.4 % (1.49 to 3.31 μg/L), trichloroethylene 11.5 % (1.78 to 6.02 μg/L), 1,1-dichloroethylene 10.3 % (1.23 to 5.89 μg/L), and dichloromethane 8.5 % (0.28 to 968.86 μg/L) in the detection rate order. The concentration of VOCs was detected in trace amounts, except for dichloromethane that exceeded the effluent quality standard in three business types, namely, metal manufacturing, food industry, and car washing facility. Chloroform was detected in all business types, where 24.88 μg/L were detected in the laundry business and 53.41 μg/L in the water supply business; the mean concentration of chloroform in these two business types was higher than elsewhere. Therefore, for the disposal of non-degradable specific hazardous substances in industrial wastewater, it is necessary to introduce physical and chemical processes, such as activated carbon adsorption, fenton oxidation, ozone treatment, as well as photocatalyst and the UV radiation.

Biodegradation of Aromatic Compounds by Strains of Pseudomonas (Pseudomonas속 세균에 의한 방향족화합물 생분해)

  • Thirty-six aromatic compound biodegraders; 10 strains for benzoate, 10 for salicylate, 6 for m-toluate, and 10 for DL-camphor were isolated and taxonomically characterized. A mutant Pseudomonas strain, Ben 6-2, derived from Ben 6 revealed remarkably improved ability to metabolize benzoate. Thus enhancement of the average substrate removal rate from 5.2 to 11.0mg/$\ell$/ hr was attained by the mutant. Both of strains Sal 7 and Tol 2, degraders of salicylate and m-toluate respectively, were classified as Pseudomonas sup. Both strains were found to be extremely effective in metabolizing each aromatic substrates. The average substrate degradation rates in minimal salt media containing 2,200mg/$\ell$ of the substrate were calculated to be 40.1 mg/$\ell$/ hr for strain Sal 7 and 33.0mg/$\ell$/ hr for Tol 2. Cam 10, a camphor degrading strain was demonstrated to be capable of mineralizing benzoate, phenol, toluene, octane, cyclohexane and xylene as well as camphor. Strain 1040 isolated from Cam 10 after repented adaptation to 1,000 mg/$\ell$ m-toluate gained the ability to utilize toluate as a sole carbon source. The mutant Brew actively at the expense of a mixture of car-bon sources; camphor, m-toluate, benzoate and phenol (each: 200 mg/$\ell$) and utilized the substances in the preferential order of camphor, phenol, benzoate, and m-toluate. Among the biodegraders examined Cam 1040 and Tol 2 were detected to harbor plasmid. The plasmid from Cam 1001 was determined to be about 98kb, and evidenced to encode the enzyme(s) for the degradation of camphor. For the further diversification of the metabolic potentials of Cam 1040, the NAH 2 plasmid of Pseudomonas putida NCIB 9816 was transferred to Cam 1040 by conjugation. The exconjugant obtained, Cam 1043, proved to gain an additional ability to metabolize salicylate and naphthalene.

Effects of High Molecular Hardwood Lignin on Anaerobic Digestion at Different Temperatures and Sludge Concentrations (혐기성 소화에 미치는 온도와 슬러지의 농도별 고분자 활엽수 리그닌의 영향)

  • Lignin is a major component of wastewater generated in the chemical processing of wood. Because it is recalcitrant, it inhibits biological treatment of wastewater of pulp manufacturing, especially high concentration of lignin may inhibit the anaerobic digestion. The objective of this study was to evaluate the toxicity of high molecular hardwood lignin (lignosulfonate, MW $\geq$ 20,000) on aceticlastic methanogens in the batch reactors at different temperatures with different sludge concentrations, using anaerobic serum bottles. The hardwood lignin was found to inhibit anaerobic conversion of acetate to methane and carbon dioxide, shown with a long lag-phase before methanogenesis started. The methanogens assumed not to be able to acclimate to the lignin were found to be acclimated slowly in the batch experiments, finally reaching non-toxic levels in which methane production could start. The hardwood lignin was found not to be bacteriocidal but bacteriostatic to aceticlastic methanogens. Hardwood lignin(lignosulfonate) at 1.3, 2.6, and 3.9%(w/w) inhibited the acetateutilizing methanogens of anaerobic digester sludge by 14.5, 17.8, 21.1 days(in noninhibitory condition it took 10 days) to produce the same amount of methane. The inhibitory effect of lignin was examined at temperature ranges of $30^{\circ}C$ to $50^{\circ}C$. When 2.6% of lignin was contained in wastewater, methane production was highest at $30^{\circ}C$ during initial 8 days. At $4^{\circ}C$, methane production rapidly increased after 12 days of digestion, the value became higher than that at $30^{\circ}C$ after 14 days. However, the methane production was completely inhibited during whole digestion period at $50^{\circ}C$. High ratio of lignin concentration to initial anaerobic sludge concentration gave tolerance to the inhibition. In this experiment, high molecular hardwood lignin was not degraded and decolorized.

Catalytic Wet Air Oxidation by TiO2 Supported Mn-Ce Based Catalysts (Mn-Ce계/TiO2 촉매에 의한 아세트산의 습식산화 반응특성)

  • Catalytic wet air oxidation of acetic acid over Mn-Ce based catalysts deposited on various supports ($SiO_2$, $TiO_2$, $ZrO_2$), $ZrSiO_4$, $ZrO_2(10wt%)/TiO_2$) have been carried out in high pressure microreactors. Also, promotional effects by small addition(O.5~1.0 wt%) of p-type semiconductors (CoO, $Ag_2O$, SnO) have been investigated. From the screening tests for initial activity ranking, both Mn(2.8)-Ce(7.2 wt%) and Ru(O.4)Mn(2.7)-Ce(6.9 wt%) supported on $TiO_2$ were selected as the promising reference candidates. In $Mn-Ce/TiO_2$ reference catalyst, addition of small amount of each p-type semiconductor (Co, Sn and Ag) resulted in activity promotional effect and the degree of the increase was in the following order: Co> Ag > Sn. Especially, $Mn-Ce/TiO_2$ promoted with 0.5 wt% Co gave the 2.6 folds activity increase compared to the reference case attributing to the surface area increase as well as synergy effect. In $Ru-Mn-Ce/TiO_2$ reference catalyst, only Co(1.0 wt%) promoted case showed a little reaction rate increase.

Water Quality Improvement of Stagnant Water using an Upflow Activated Carbon Biofilm Process and Microbial Community Analysis (상향류 활성탄 생물막 공정을 이용한 정체 수역 수질 개선 및 공정 내 미생물 군집 해석)

  • The capacity of natural purification was limited by the interruption of natural flow and the problems such as eutrophication were occurred by nutritive salts accumulation in stagnant stream. Moreover, the inflow of non-point sources causes non-degradable materials to increase in stagnant stream. In this study, an upflow biological activated carbon (BAC) biofilm process comprised of anoxic, aerobic 1, and aerobic 2 reactors were introduced for treatment of stagnant stream and SS, $BOD_5$, $COD_{Mn}$, $COD_{Cr}$, TN, and TP were monitored in the upflow BAC biofilm reactors with continuous cycling. In order to simulate stagnant stream, the lake water of amusement park and golf course were stored as influent in a tank of $2m^3$ and hydraulic retention time (HRT) was changed into 6, 4, and 2 hours. At HRT 4hr and the lake water of amusement park as influent, the removal efficiencies of SS, $BOD_5$, $COD_{Mn}$, $COD_{Cr}$, TN, and TP showed the best water quality improvement and were 69.8, 83.0, 91.3, 74.1, 74.7, and 88.9%, respectively. At HRT 4hr and the lake water of golf course as influent, the removal efficiencies of SS, $BOD_5$, $COD_{Mn}$, $COD_{Cr}$, TN and TP were 78.5, 78.0, 80.2, 74.9, 55.6 and 97.5%, respectively. As the results of polymerase chain reaction - denaturing gel gradient electrophoresis (PCR-DGGE), microbial community was different depending on influent type. Fluorescence in situ hybridization (FISH) showed that nitrifying bacteria was dominant at HRT 4 hr. The biomass amount and microbial activities by INT-DHA test were not decrease even at lower HRT condition. In this study, the upflow BAC biofilm process would be considered to the water quality improvement of stagnant stream.

Effect of the Application of Sucrose on Rapid Decrease of Soil Inorganic Nitrogen (Sucrose 처리가 토양 무기태질소의 신속 감소에 미치는 영향)

  • To solve the problems with excessive accumulation of soil inorganic N and resulting saline soils from overuse of nitrogen fertilizer, the effect of sucrose application on decrease of soil inorganic N content and electrical conductivity (EC) was studied. Sucrose treatment greatly reduced ${NH_4}^+$-N content in soil. The amount of reduction was greater as the amount of sucrose treatment was increased. When ${NH_4}^+$-N content was reached the lowest point (about 10 mg $kg^{-1}$or lower), the C/N ratio, which determines the amount of sucrose treatment, was around 10 regardless of initial ${NH_4}^+$-N content. For the rate of ${NH_4}^+$-N reduction 15~36 hours was required to reduce the initial ${NH_4}^+$-N content to half, and 36~69 hours to lower ${NH_4}^+$-N content to the lowest point (about 10 mg $kg^{-1}$or lower). In addition, sucrose treatment greatly lowered ${NO_3}^-$-N content. In case of C/N ratio above 10, initial ${NO_3}^-$-N content of 348 mg $kg^{-1}$ was reduced to the lowest of 14~21 mg $kg^{-1}$. As for the rate of ${NO_3}^-$-N reduction by sucrose treatment, it took 36~60 hours for ${NO_3}^-$-N content to reach the lowest point for C/N ratio of 10 or higher, and it took 3 weeks, comparably longer time, for C/N ratio of 5. Lowering soil EC from sucrose treatment showed the same trend as ${NO_3}^-$-N content. As an important energy and carbon source for humankind, sugar should not be wasted and must be carefully applied to soil. In principle, the best way of preventing salt accumulation in soil is to optimize the fertilizer input. However, when over-fertilization should be dealt with, the sucrose treatment would be a possible and effective counter-measure to reduce overdosed nitrogen sources in soil.

In Vitro Phosphorylation of Nuclear Proteins in Isolated Liver Nuclei from Rats Maintained in a Starvation State, Following Refeeding, and from Diabetic Rats with Insulin Injection (단식(斷食), 재급식(再給食) 및 인슈린 투여(投與) 후(後)에 쥐의 간(肝)으로부터 분리된 세포핵의 핵단백질 인산화)

  • Labelling of chromatin proteins with 32P was observed after incubating isolated liver nuclei with $[{\gamma}-32P]$ ATP for 5 minutes at $37^{\circ}C$. The pattern of labelling with 32P was examined on SDS polyacrylamide gel electrophoresis with nuclei from rats maintained in a starvation state for 48 hours, following refeeding for 12 hours; and from fed streptozotocin-diabetic rats with insulin injection 6 hours before sacrifice. With 48h starved rat liver nuclei the level of phosphorylation for 0.14M NaCl soluble proteins was decreased in the molecular weights between 41,000 and 200,000 daltons relative to normal controls. Refeeding the starved rats reversed the change of phosphorylation pattern over 12 hour The level of phosphorylation for five phenol soluble non-histone proteins with molecular weights above 59,000 daltons was somewhat decreased with 48h starved rat liver nuclei as compared with that of normal controls. Starvation also decreased the phosphorylation level of major histones in relation to normal controls. The experiment with insulin injection into fed streptozotocin-diabetic rats showed the tendency to increase phosphorylation of 0.14M NaCl soluble proteins (130,000 dalton protein) and phenol soluble non-histone proteins (155,000 dalton protein). The phosphorylation level of histones appeared to be invariant under the experimental conditoins employed here. These results suggest the possibility that the phosphorylation and dephosphorylation of 0.14M NaCl soluble proteins and $H_1$ histone precede those of other chromatin associated nuclear proteins, It is of interest to find that insulin signal was correlated to phosphorylation of nuclear proteins while glucagon signet dephosphorylated nuclear proteins.
