Optimization of Anticoagulant Production from Phellinus linteus Mycelia (상황버섯(Phellinus linteus) 균사체로부터 항혈전 생산의 최적화)

  • To produce the functional food materials, 50 kinds of the mycelial extracts from edible mushroom were examined for anticoagulant activity and Phellinus linteus showed the highest activity through the activated partial thromboplastin test (aPTT). The maximum production of anticoagulant activity and the mycelial growth was observed in culture medium containing soluble starch 3.0%, peptone 0.1%, $MgSO_4{\cdot}7H_2O^{\circ}C$ 0.1%, $K_2HPO_4$ 0.1% and in the culture conditions controlled at initial pH 7.0, $30^{\circ}C$ and 150 rpm by the rotary shaker. In addition, the maximum production of mycelial dry weight was 7.5 mg/mL after 10 days under the optimal conditions, and anticoagulant activity was reached to 390 sec in 5 L-jar fermentor.

  • 이상의 모든 결과를 요약해 보면 어유의 경우 항혈전 효과에는 상당히 들기름과 함께 좋으나 막의 유연성과 다른 면에서 들기름은 항혈전과 세포막의 유연성 유지 등에서 상당히 유리한 기름으로 생각할 수 있다. 팜유는 우지나 참기름보다 항혈전 효과나 세포막 유연성 유지에서 좋으나 들기름보다는 떨어지는 것으로 생각할수 있다.

Evaluation of Anti-thrombosis Activities of Different Parts of Cannabis sativa L. (대마 부위별 추출물의 항혈전 활성)

  • The high fatality of COVID-19 (Corona Virus Disease 2019) is closely related to acute pneumonia and severe blood clot formation in brain, heart, kidney and lung. The use of low-molecular weight heparins has been shown to reduce the risk of thrombosis and reduce fatality rates among COVID-19 patients. In this study, we investigated the antithrombotic activity of different parts of C. sativa extracts to determine its potential in preventing blood clots in patients with thromboprophylaxis. The extracts of leaf, stem, root, immature flower and seed of C. sativa showed strong inhibitory activities on blood clot formation. In particular, the flower extract showed the strongest inhibitions against blood coagulation factors and thrombin. Strong hemolysis activities were observed in flower extract and seed extract, suggested that removal of the hemolysis active compounds in th extracts is necessary. This is a first report of anti-thrombosis activities of different parts of C. sativa extracts, and our results suggest that C. sativa extract has potential has a valuable bioresource for high-value products.

Effects of Methanolic Extract from Salvia miltiorrhiza Bunge on in vitro Antithrombotic and Antioxidative Activities (단삼 메탄올 추출물의 항혈전 및 항산화 효과)

  • Salvia miltiorrhiza Bunge is known to potentially prevent arteriosclerosis and hypertension, but its effects on platelet function are not clear. In this study, we evaluated the in vitro antithrombotic activities of the edible plant extract. Methanol extract of S. miltiorrhiza Bunge exhibited about 70% fibrinolytic activity compared to the plasmin control, and inhibited ADP- and collagen-induced platelet aggregation in a concentration-dependent manner with $IC_{50}$ values of 0.42 and 0.07 mg/mL, respectively. S. miltiorrhiza Bunge extract significantly prolonged the activated partial thromboplastin time (APTT) and prothrombin time (PT) compared with control. Moreover, 0.05 mg/mL S. miltiorrhiza Bunge extract contained 87.3% l,l-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity. In conclusion, S. miltiorrhiza Bunge seemed to enhance antithrombotic activity due to its radical scavenging activity. Based on these data, further examination is required to determine the mechanism of platelet-dependent antithrombosis and the effect of polyphenols on platelet function.

신물질 ASPALATPNE의 항혈전 작용

  • Acetylsalicylic acid (aspirin)와 인삼의 항산화 성분인 maltol의 축합반응으로 합성된 신물질 Aspalatene이 저용량: 장기 복용시 항산화활성과 더불어 지혈시간 연장효과가 탁월함을 보고 한 바 있다. Aspalatone은 aspirin과는 달리, 장기 복용시에도 위궤양 유발 작용이 거의 나타나지 않으므로 말초순환개선제로서의 개발이 기대된다. 본 연구에서는 Aspalatone의 항혈전작용을 in vivo, ex vivo 및 in vitro에서 연구하였다. Mouse thromboembolism test를 이용한 in vivo실험에서 Aspalatone은 aspirin과 유사하게 collagen에 의한 치사를 유의적으로 감소시켰으며 ADP에 의한 치사에는 영향을 미치지 않았다. 또한 10일간의 장기 투여 실험에서는 저용량에서 영향을 미치지 않았다. 또한 10일간의 장기 투여 실험에서는 저용량에서 항혈전 작용을 나타내며 투여 중단 4일 후에도 약효가 유지되는 것을 밝혔다. 이는 Aspalatone이 aspirin과 같이 cycloosygenase에 작용하여 항혈전 작용을 나타냄을 강력히 시사하고 있다.

Antithrombotic Effect and Antiplatelet Activity of Polygonum cuspidatum Extract (호장근 추출물의 항혈전 효능 및 혈소판 응집 억제작용)

  • We performed this study to develop antithrombotic agents from oriental medicine herb extracts. Polygonum cuspidatum has been traditionally used as an edible medical resources for the treatment of cancer, pyodermatitis, hepatitis, cystitis, and inflammation. However, the effects of Polygonum cuspidatum on thrombosis and platelet activation are not precisely understood. The antithrombotic and antiplatelet activities of Polygonum cuspidatum were investigated by assessing the effect of a 70% ethanol extract of Polygonum cuspidatum on blood coagulation, fibrinolysis, and platelet aggregation. Polygonum cuspidatum showed effective fibrinolytic activity at 10 mg/mL. Polygonum cuspidatum also inhibited adenosine diphosphate induced platelet aggregation. Furthermore, evaluation of anticoagulant activity showed that an extract of Polygonum cuspidatum prolonged coagulation time via activated partial thromboplastin time (APTT). Our results show that Polygonum cuspidatum can be a potential candidate for antiplatelet activity as well as a fibrinolytic agent.

Antithrombosis, Antidiabetes, and Antioxidant Activities of Houttuynia cordata (어성초의 항혈전, 항당뇨 및 항산화 활성)

  • Houttuynia cordata belongs to the Saururacease family and its leaves, stems, and roots have been used as oriental medicines to treat pneumonia, acute or chronic bronchitis, enteritis, and abscesses and to remove extravasated blood. Recently, the antioxidant, anti-inflammation, antibacterial, and anti-proliferation activities and protection abilities of H. cordata against liver and neuron cell damage have been reported. In this study, ethanol extract and its solvent fractions (fractions of hexane, ethyl acetate, butanol, and water residue) were prepared, and their antithrombosis, antidiabetes, antioxidant, and hemolysis activities were evaluated. The ethyl-acetate fraction of H. cordata (EF-HC) showed the highest polyphenol and flavonoids contents among the fractions and exhibited strong antithrombosis and antioxidant activities. The EF-HC at 5 mg/ml showed 2.09-folds of thrombin time, 2.19-folds of prothrombin time, and 1.69-folds of activated partial thromboplastin time compared to the their solvent control and 30.9, 19.9, and 49.6 ㎍/ml of RC50 against DPPH, ABTS, and nitrite radicals, respectively. Furthermore, the EF-HC did not show any hemolytic activity up to 1 mg/ml, whereas the hexane fraction of H. cordata showed 55% hemolysis at 1 mg/ml. This is the first report of the antithrombosis activity of H. cordata. Our results suggest that quercitirin, hyperoside, orientin, and isoquercitrin in EF-HC are related to its antithrombosis and antioxidant activities and that the EF-HC could be developed as a promising antithrombosis agent.

Antithrombotic and Antiplatelet Activity of Extract from Prunella vulgaris (하고초 추출물의 항혈전 효능 및 혈소판 응집 억제작용)

  • This study was performed to develop effective antithrombotic agents from traditional herb extracts. Prunella vulgaris L. has been used traditionally as a medical resource in cancer therapy, as well as treatment of hypertension and inflammation, and as a diuretic. However, the effects of Prunella vulgaris on thrombosis and platelet activation have not been clearly understood. Antithrombotic and antiplatelet activities of oriental medicinal herbs were investigated by evaluating the effect of the aqueous extract from Prunella vulgaris on the blood coagulation, platelet aggregation and fibrinolysis. Prunella vulgaris extracts showed effective anticoagulant activity in coagulation times such as activated partial thromboplastin time (APTT) and prothrombin time (PT). Prunella vulgaris also inhibited adenosine diphosphate (ADP)- and collagen-induced platelet aggregation. In addition, evaluation of fibrinolytic activity showed that the Prunella vulgaris extracts have high solubility. From these results, it is suggested that Prunella vulgaris can be a potential candidate for anticoagulants and antiplatelets, as well as fibrinolytic agents.

  • Antiphospholipid syndrome (APS) is defined as venous and/or arterial thromboses, recurrent fetal losses, thrombocytopenia in combination with repeatedly positive tests for the lupus anticoagulant (LAC), and anticardiolipin antibodies (aCL). The pulmonary manifestation is APS are relatively rare. We report a rare case of antiphopholipid syndrome with systemic lupus erythematosus in a patient who presented with pulmonary hypertension secondary to a chronic pulmonary thromboembolism. A bilateral thromboendarterectomy was performed satisfactorily and the incision was extended to the left intrapleural pulmonary artery.

Evaluation for the Antimicrobial, Antioxidant and Antithrombosis Activity of Natural Spices for Fresh-cut yam. (생마 신선편이 제조를 위한 천연 향신료의 항균, 항산화 및 항혈전 활성 평가)

  • To make functional fresh-cut yam using natural spices, the water or the methanol extract was prepared from different kinds of natural spices including ginger, wasabi, black pepper, mustard, cinnamon, chinese pepper, garlic, thyme, rosemary, chamomile, and peppermint. The extraction yields were varied as spice sources and extraction solvents used. Among the water extracts, peppermint extract showed strong antithrombosis activity with 26-folds extended thrombin time, and the $IC_{50}s$ of the extracts from cinnamon and rosemary were $61.3{\mu}g/ml\;and\;84.3{\mu}g/ml$ in DPPH scavenging assay, respectively. Whereas, the antimicrobial activity of all water extracts was ignorable. Among the methanol extracts, the extracts from thyme, ginger, and cinnamon showed antimicrobial activity against yam putrefactive bacteria. Strong antioxidative activity was found in the extracts from cinnamon and thyme. Based on these results, the functional fresh-cut yam was prepared using the water extract from peppermint or cinnamon. The antioxidative and antithrombin activities in the fresh-cut yam were maintained at $4^{\circ}C$ for 24 hr. Our results suggest that the natural spices could be applied to production of functional fresh-cut yam.
