A survey of canine heartworm (nirofilaria inmitis) infections among German shepherds in five areas of South Korea was performed from October 1994 to August 1995 using a microfilarial test (modified Knott's test) and an antigen test (DiroCHEKⓑ Synbiotics, USA). The infection rate of 127 German shepherds (71 males and 56 females) was 10.2% (13/127) by the microfilarial test, but was 28.3% (36/127) by the antigen test, revealing that 24 of the 36 antigen-positive dogs were microfilaria-negative in the peripheral blood . All dogs that were microfilaria-positive were also antigen-positive. Three of the microfilaria-negative and antigen-positive dogs contained 4-15 adult heartworms in the heart and pulmonary arteries upon necropsy. The infection rate among German shepherds was the highest in Hoengsong-gun (Kangwon-do, 84.4%) , while those of Yechon-gun (Kyongsangbuk-do) and Chungwon-gun (Chungchongbuk-do) areas were 20.0% and 14.3%, respectively. None of the dogs in the Kimhae-shi (Kyongsangnam-do) and Kwangiu areas was heartworm positive. The infection rates of heartworm in dogs at the age of 1-3, 4-6. and 7-11 years were 6.3%, 21.4%, and 56.4%, respectively. Based on the fact that the antigen test is more accurate than the microfilarial test, this study strongly indicates that the prevalence rate of canine heartworm in Korea may be higher than previously reported (3.1-23.0%) which utilized microfilarial tests.
Antigenic domain of jai or surface protein (p30) of Toxoplosmc Sondii was analyzed after polymerase chain reaction (PCR) of its gene fragments. Hydrophilic or hydrophobic moiety of amino acid sequences were expressed as glutathione S-transferase (G57) fusion proteins. Fragments of p30 gene were as follows: 737, total p30 open reading frame (ORF) ; S28, total ORF excluding N-terminal signal sequence and C-terminal hydrophobic sequence; Al9, N-terminal 2/3 parts of A28; A19, N-terminal 2/3 of S28; P9, C-terminal 2/3 part of S28; Z9. middle 1/3 of S28; and 29, C-terminal 1/3 of S28. respectively. Primer of each fragment was synthesized to include clamp sequence of EcoR I restriction site. PCR amplified DNA was inserted info GST (26 kDa) expression vector, PGEX-47-1 to transform into Escheri,hia coei (.JM105 strain). G57 fusion proteins were expressed with IPTG induction as 63. 54, 45, 45, 35, 36. and 35 kDa proteins measured by SDS-PAGE. Each fusion protein was confirmed with G57 detection kit. Western blot analysis with the serum of a toxoplasmosis patient revealed antigenicity in proteins expressed by T37. S28, and Al9 but not those by Pl8. X9, Y10, and Z9. Antigenicity of p30 seems to be located either in N-terminal 115 part in the presence of middle 1/3 part or in the oligopeptides between margins of the first and second 1/3 parts.
The olive flounder, Paralichthys olivaceus is susceptible to viral hemorrhagic septicaemia virus (VHSV) at $15^{\circ}C$ but no mortality at $20^{\circ}C$ even though the virus can grow well in vitro at $20^{\circ}C$. Thus, we designed an experiment to know immune response of olive flounder against VHSV when the host reared at $15^{\circ}C$ or $20^{\circ}C$. cDNA microarray analysis was performed to compare the gene expression patterns of the kidney cells between the host reared at $15^{\circ}C$ or $20^{\circ}C$. The expression of MHC class I, IL-8, myeloperoxidae and endonuclease G-like having function for the antigen presentation and chemokine-factor were up-regulted both the $15^{\circ}C$ and $20^{\circ}C$ during VHSV infection. MHC class II gene existing on antigen-presenting cells and B cell lymphocytes, immunoglobulin (Ig) genes and phagocytosis related genes were down-regulated at $15^{\circ}C$ but highly expressed at $20^{\circ}C$. It can be thought that innate immune related antigen presentation by MHC class I and phagocytosis reaction against VHSV are efficiently occur both the temperature but macrophage or B cell related antigen presentation via MHC class II fails to induce downstream immune reactions (adaptive immunity) to make antibody, and it can be one of the reason that causes high mortality only at $15^{\circ}C$.
To observe the antigenic protein fractions in saline extract of Spirometra mansoni plerocercoid (sparganum), the crude extract was separated in reducing conditions of sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE). The proteins, transferred by celctrophoresis to introcillulose paper, were reacted with sera from 15 surgically confirmed sparganosis and 24 cysticercosis patients for immunoblotting. Out of 30 identified protein bands in the extract, bands of 29 and 36 kilodaltons (kDa) were the strongest and the most frequently reacting with specific antibody (IgG) in sparganosis sera. Bands of highter molecular weight also reacted with the sera but their frequency of reactions was lower. Sera of cysticercosis reacted with different protein bands in saline extract of sparganum, but the cross reactions were observed in strong antigenic bands of 29 and 36 kDa.
Carcinoembryonic Antigen is one of most frequently checked tumor markers in cancer management. We performed statistical analysis with serum CEA data of 2626 persons who received regular health examination and were thought to be free of active disease to determine the cutoff values of serum CEA level in normal Korean adults and to study the factors influencing serum CEA levels in normal subjects. 1) The cutoff values of serum CEA in normal Korean adults in general were 9.28ng/ml for men, 5.90 ng/ml for women. 2) Serum CEA level was influenced by age, present smoking history, sex, and abnormal findings in chest X ray. 3) Serum CEA level had no correlation with the history of amount of alcohol consumption or obesity. 4) Cutoff values of serum CEA in normal Korean adults were tabulated according to age, sex, and smoking history. Remicade is a therapeutic biosimilar natural antibody in which the mouse variable domain has been linked to the human constant domain. It is a chimeric monoclonal antibody specific to tumor necrosis factor-alpha (TNF-α) and has been developed for the treatment of rheumatoid arthritis. To investigate the biological activity of the Remicade antibody, we carried out a bioinformatics study using a protein data bank to characterize the TNF-α antigen binding mechanism of the Remicade natural antibody. Because the production of the Remicade antibody is often limited by genetic instability of the natural antibody-producing cell, we generated a Remicade single-chain variable domain fragment antibody (Remicade) in which a heavy chain variable domain (VH) is joined with a light chain variable domain (VL) by a polypeptide linker. Furthermore, Remicade was fused to a leucine zipper (RemicadeScZip) for higher production and higher antigen-binding activity than Remicade. The Remicade and Remicade ScZip were expressed in Escherichia coli and purified by a Ni+-NTA-agarose column. As expected, the purified proteins had migrated as 28.80 kDa and 33.96 kDa in sodium dodecyl sulfate-polyacrylamide electrophoresis. The TNF-α antigen binding activity of Remicade was not observed by ELISA and western blot. In contrast, RemicadeScZip showed antigen-binding activity. Additional bio-layer interferometry analysis confirmed the antigen-binding activity of RemicadeScZip, suggesting that the leucine zipper stabilized the folding of RemicadeScZip in a denatured condition and improved the TNF-α antigenbinding activity.
It had been evaluated the recombinant Circumsporozoite(CS) protein of Plasmodium viva in serologic diagnosis of vivax malaria. Western blot was done to analyse the sera of malaria patients according to the days after onset. The sera which have the terms within 15 days were shown 43.8%(14/32) of positive rates and the sera over the 16 days were shown 94.4%(17/18) of positive rates. So the total positive rate was 62%(31/50). It was 22.6%(7/31) which was shown negative response in Western blot, even though they were shown positive response in Immuuofluorescent antibody test(1FAT) using whole blood stage antigens. The positive rate of non-epidemic area(Yechon-gun, Kyongsangbuk-do) was 10.7%(3/28), and epidemic area(Kangwha-gun, Inchon-shi) was 27.6%(13/47) in Western blot analysis using recombinant CS protein. In order to applicate the recombinant CS protein in seroepidemiological survey, blood samples of 422 inhabitants were collected who lived in malaria epidemic areas, Chosm-ri, Majeong-ri, Hyangyang-ri and Noejo-n in Paju-shi, Kyonggi-do. All of them were negative in microscopic examination and two(0.5%) of them were positive in Polymerase Chain Reaction. 42(10.0%) of them were seropositive in FAT using whole blood antigens and 71(16.8%) of them were seropositive in Enzyme-linked immunosorbent assay using recombinant CS protein. It was figured out the positive rates were much higher according to the distances of villages which were closed to the demilitalized zone(DMZ) in all kind of diagnostic methods, respectively.

Objectives: Since 1983, vaccination for Hepatitis B virus has been recommended for all neonates in Korea. The objectives of this study was to assess the changing trend of the positive rate of HBsAg and anti-HBs in university students in one province. Methods: The study population consisted of one university students who taken physical exam in 2004, which included 8,527 students (3,391 males and 5,136 females). Serum HBsAg and anti-HBs were detected by immunochromatography method. Results: The positivities of HBsAg and anti-HBs were 2.7% and 61.1%. The positivity of HBsAg was significantly higher in males(p< .05), while that of anti-HBs was significantly higher in females(p< .05). There was some differences of HBsAg positivities among age groups. But there was no correlation between age and the positivities. There was some differences of anti-HBs positivities among age groups. But there was no correlation between age and the positivities. Conclusion: The positivity of HBsAg has significantly decreased and that of anti-HBs has increased with years. It was suggested that the changes of positivity were caused by nationwide hepatitis B vaccination program.
돼지 생식기 호흡기 증후군 바이러스의 nucleocapsid와 반응을 하는 SDOW17 단크론항체를 이용하여 중성 포르말린에 고정시킨 자연감염된 포유자돈의 폐장에서 면역조직화학법을 이용하여 돼지 생식기 호흡기 증후군 바이러스 항원을 확인하였다. 서울대학교 수의과대학 병리학교실에 의뢰된 포유자돈들 중에서 병리조직학적으로 폐장에서 간질성 폐렴이 관찰된 포유자돈 7두를 임의로 선택하여 본 실험을 실시하였다. 간질성 폐렴의 병변으로 많은 수의 대식세포 침윤을 동반한 폐포벽 두께의 증가와 제II형 폐포세포의 비후가 관찰되었다. 검사한 7두 포유자돈중에서 6두에서 돼지 생식기 호흡기 증후군 바이러스에 대한 항체를 enzyme-linked immunosorbent assay에 의해 확인하였다. SDOW17 단크론항체를 이용한 면역조직화학염색과 간질성 폐렴의 대식세포에서 돼지 생식기 호흡기 증후군 바이러스의 항원을 검출하였고, 항원은 (주로)대식세포의 세포질에서만 진한 갈색의 양성반응이 관찰되었다. 이상 검사결과 돼지 생식기 호흡기 증후군 바이러스는 폐장의 간질과 폐포강에 분포되어 있는 대식세포에서 주로 증식하는 것으로 판명되었다. 본 실험에서 사용한 면역조직화학법은 돼지 생식기 호흡기 증후군 바이러스 감염여부를 바이러스 분리 또는 혈청검사 없이 진단하는데 사용할 수 있는 유용한 진단방법으로 판명되었다.
Our study investigated the prevalence of allergic conjunctivitis and skin prick test reactivity and type of allergen from 1352 individuals living in Gwangyang city. The prevalence of allergic conjunctivitis symptom were 40,2% of study subjects and diagnosised subjects with allergic conjunctivitis of them were 15.8%. In skin prick test, 20.2% of total subjects had at least one positive skin prick reaction and positive skin prick reaction with allergic conjunctivitis of subjects was 21.3%(n=103). Their major allergens were indoor allergen like dermatophagoides pteronyssinus, Dermatophagoides farinae.
